No effective antiviral therapy is proven yet, but clinical use of ribavirin (RBV) has been tried. We investigated the antiviral effect of RBV against SFTSV in vitro. Methods: To test for cytotoxicity of RBV, Vero cells were treated with different concentrations of RBV (3.90 to 500 μg/mL, two-fold dilution) and analyzed by cell viability MTS assay 48h post-infection. To determine antiviral activity of RBV against SFTSV, Vero cells were infected with SFTSV strain Gangwon/Korea/2012 at 100 TCID50 (50% tissue culture infective dose) per well in a 96-well plate, and RBV was added at the concentrations showing no or minimal cytotoxicity. Viral RNAs were extracted from the culture supernatants and quantifi ed using one-step real-time reverse transcription- PCR to amplify the partial large segment of SFTSV. Statistical analysis was done by one-way ANOVA with Tukey`s post hoc test. Results: Cytotoxicity due to RBV was not observed at RBV concentration =31.3 μg/ mL. Viral RNAs at 24h post-RBV treatment were reduced with increasing RBV concentrations (1-32 μg/mL), compared with those of mock-treated cells (P <0.01, Figure). Half maximal inhibitory concentration (IC50) of RBV was 3.69 μg/mL at 24h post-RBV treatment. Conclusions: Our study shows that RBV has antiviral effect against SFTSV in a dose-dependent manner. Further studies are required to evaluate the effi cacy of RBV in SFTS.

        Clinicopathological role of kidney injury molecule-1 in immunoglobulin A nephropathy

        Background: Urinary kidney injury molecule-1 (KIM-1) is an early and sensitivebiomarker of acute kidney injury, but it is unclear if it is a biomarker of chronicglomerulonephritis. We evaluated whether urinary KIM-1 levels in patients withimmunoglobulin A (IgA) nephropathy can be a marker to reflect clinicopathologicalseverity and predict the prognosis. Methods: We measured urinary KIM-1 levels in 40 patients (15 males; mean age36.6712.9 years) with IgA nephropathy and 10 healthy people (5 males; mean age37.379.6 years) as controls. The correlation of urinary KIM-1 levels with patients’clinical parameters, histological grades, and follow-up data were analyzed using themodified H. S. Lee grading system and tubulointerstitial change scores. Results: Urinary KIM-1 levels were higher in patients with IgA nephropathy thanhealthy controls (P¼0.001). Univariate and multivariate regression analyses showedthat urinary KIM-1 levels had a direct correlation with H. S. Lee grade andtubulointerstitial inflammation (P¼0.004 and P¼0.011, respectively). Conclusion: In patients with IgA nephropathy, urinary KIM-1 has a significantcorrelation with histopathologic severity.

      • Light-extraction efficiency control in AlGaN-based deep-ultraviolet flip-chip light-emitting diodes: a comparison to InGaN-based visible flip-chip light-emitting diodes.

        <P>We study light-extraction efficiency (LEE) of AlGaN-based deep-ultraviolet light-emitting diodes (DUV-LEDs) using flip-chip (FC) devices with varied thickness in remaining sapphire substrate by experimental output power measurement and computational methods using 3-dimensional finite-difference time-domain (3D-FDTD) and Monte Carlo ray-tracing simulations. Light-output power of DUV-FCLEDs compared at a current of 20 mA increases with thicker sapphire, showing higher LEE for an LED with 250-관m-thick sapphire by ~39% than that with 100-관m-thick sapphire. In contrast, LEEs of visible FCLEDs show only marginal improvement with increasing sapphire thickness, that is, ~6% improvement for an LED with 250-관m-thick sapphire. 3D-FDTD simulation reveals a mechanism of enhanced light extraction with various sidewall roughness and thickness in sapphire substrates. Ray tracing simulation examines the light propagation behavior of DUV-FCLED structures. The enhanced output power and higher LEE strongly depends on the sidewall roughness of the sapphire substrate rather than thickness itself. The thickness starts playing a role only when the sapphire sidewalls become rough. The roughened surface of sapphire sidewall during chip-separation process is critical for TM-polarized photons from AlGaN quantum wells to escape in lateral directions before they are absorbed by p-GaN and Au-metal. Furthermore, the ray tracing results show a reasonably good agreement with the experimental result of the LEE.</P>

        Ultra-energy-efficient analog-to-digital converters based on single-electron transistor/CMOS hybrid technology for biomedical applications

        <P>Ultra-energy-efficient analog-to-digital converters (ADCs) based on single-electron transistor (SET)/complementary metal-oxide-semiconductor (CMOS) hybrid technology are proposed as a solution to sense and process biomedical signals. Our results show the energy efficiency of 0.82 pJ/state, which is lower than that in previously reported energy-efficient ADCs. The performance and dissipated power of proposed ADCs are estimated and compared with those of CMOS ADCs by using Lee's SPICE model including non-ideal effects of the experimental data. While the proposed ADC shows an operating power lower by two orders of magnitude than that of the CMOS flash-type ADC, the number of required transistors is about 10% of that in the CMOS flash-type ADC. The peak-to-valley current ratio in Coulomb oscillation of SETs used in the circuit implementation has the range of 1.15–1.5, which is consistent with the experimental result of top-down approached Si-based SETs at <I>T</I> = 77–100 K. From the perspective of the immunity to the gate capacitance <I>C<SUB>cg</SUB></I> mismatch and the background charge <I>Q</I><SUB>0</SUB> noise, it is shown that the criteria of SET/CMOS hybrid ADCs are Δ<I>C<SUB>cg</SUB></I> ⩽ 0.02 × <I>C<SUB>cg</SUB></I> (with <I>C<SUB>cg</SUB></I> = 0.24 aF) and Δ<I>Q</I><SUB>0</SUB> ⩽ 0.23<I>q</I>, respectively.</P>

        Spontaneous retroperitoneal hemorrhage due to hemophilia A with impaired coagulopathy is very rare. Spontaneous retroperitoneal hemorrhage has been recorded as having originated from many retroperitoneal organs and blood vessels, and it may be due to local and/or systemic factors. In the majority of the patients, kidney and adrenal gland were the major site of hemorrhage. The systemic causes of spontaneous retroperitoneal hemorrhage are anticoagulation therapy and chronic hemodialysis. During the course of these treatments, hemorrhagic complications may occur at many site, including the retroperitoneal space. Blood dyscrasias including leukemia, polycythemia, sickle cell trait and hemophilia have been reported associated with spontaneous retroperitoneal hemorrhage.       • SEAWEED EXTRACTS AS A POTENTIAL TOOL FOR THE ATTENUATION OF OXIDATIVE DAMAGE IN OBESITY‐RELATED PATHOLOGIES

        <P>Recent studies suggest that seaweed extracts are a significant source of bioactive compounds comparable to the dietary phytochemicals such as onion and tea extracts. The exploration of natural antioxidants that attenuate oxidative damage is important for developing strategies to treat obesity‐related pathologies. The objective of this study was to screen the effects of seaweed extracts of 49 species on adipocyte differentiation and reactive oxygen species (ROS) production during the adipogenesis in 3T3‐L1 adipocytes, and to investigate their total phenol contents and 2,2‐diphenyl‐1‐picrylhydrazyl (DPPH) radical scavenging activities. Our results show that high total phenol contents were observed in the extracts of <I>Ecklonia cava</I> (see Table?1 for taxonomic authors) (681.1 ± 16.0 μg gallic acid equivalents [GAE] · g<SUP>−1</SUP>), <I>Dictyopteris undulata</I> (641.3 ± 70.7 μg GAE · g<SUP>−1</SUP>), and <I>Laurencia intermedia</I> (560.9 ± 48.1 μg GAE · g<SUP>−1</SUP>). In addition, DPPH radical scavenging activities were markedly higher in <I>Sargassum macrocarpum</I> (60.2%), <I>Polysiphonia morrowii</I> (55.0%), and <I>Ishige okamurae</I> (52.9%) than those of other seaweed extracts (<I>P </I><<I> </I>0.05). Moreover, treatment with several seaweed extracts including <I>D. undulata</I>, <I>Sargassum micracanthum</I>, <I>Chondrus ocellatus</I>, <I>Gelidium amansii</I>, <I>Gracilaria verrucosa</I>, and <I>Grateloupia lanceolata</I> significantly inhibited adipocyte differentiation and ROS production during differentiation of 3T3‐L1 preadipocytes. Furthermore, the production of ROS was positively correlated with lipid accumulation (<I>R</I><SUP>2</SUP> = 0.8149). According to these preliminary results, some of the seaweed extracts can inhibit ROS generation, which may protect against oxidative stress that is linked to obesity.         중소규모 사업장 근로자의 직무스트레스로 인한 결근, 조퇴 현황과 한국형직무스트레스 하부요인과의 관련성

        목적: 중소규모 사업장 근로자들의 직무 스트레스로 인한 결근, 조퇴의 현황과 한국형 직무 스트레스 하부 요인과의 관련성을 알아보고자 하였다. 방법: 중소규모 사업장 83곳 근로자들에 대해 한국형 직무스트레스요인 평가도구-단축형(KOSS-SF)과 사회 일반적 특성, 직업적 특정 및 직무스트레스로 인한 결근이나 조퇴 경험여부 등을 묻는 구조화된 자가 기입식 설문을 실시하였고, 응답한 4,350명에 대해 최종 분석을 시행하였다. 직무스트레스로 인한 결근, 조퇴와 한국형 직무스트레스 하부 요인들과의 관련성을 확인하기 위하여 로지스틱 회귀분석을 시행 하였다. 결과: 직무 스트레스로 인한 결근, 조퇴를 한 근로자는 남자에서 7%(196명), 여자에서 8%(113명) 로 조사되었고 한국형 직무 스트레스 하부요인과의 관련성에서는 남, 여 모두 직무요구, 관계갈등, 조직체계, 보상부적절, 직장문화 영역의 스트레스가 높은 군에서 통계적으로 의미있게 직무스트레스로 인한 결근, 조퇴율이 증가하였다. 결론: 중소규모 사업장 근로자들의 직무 스트레스로 인한 결근, 조퇴와 한국형 직무 스트레스 하부요인과의 관련성은 남자에서는 조직체계 영역이 가장 높았고, 여자는 직장문화 영역에서 가장 높은 관련성을 보였다. Objective: The aim of this study was to examine the actual conditions of absence and early leave due to job stress of 4,350 workers in 83 small and medium scale industries, and its relationship to job stress factors using Korean Occupational Stress Scale-Short Form (KOSS-SF). Methods: A structured self-reported questionnaire was used to assess each respondent's Sociodemographics, job stress, experience of absence and early leave due to job stress. Seven KOSS-SF domains of job stress were used. We estimated the relation of absence and early leave due to job stress and job stress factors using logistic regression analyses. Results: Absence or early leave due to job stress occurred in 7% of male workers and 8% of female workers. Multiple regression analysis revealed a significantly positive association job stress-related absence/early leave and job demand, interpersonal conflict, organizational system, lack of rewards and occupational climate. Conclusions: Absence and early leave due to job stress is significantly attributed to organizational System and lack of reward for men, and occupational climate and organizational system for women.

        골다공증 환자 혈청이 정상인 조골세포의 성장과 분화에 미치는 영향

        Osteoporosis is a condition in which an imbalance appears between bone resorption and formation, with bone resorption exceeding formation. Recent studies have shown that bone formation abnormalities in various forms of osteopenia result mainly from defective recruitment of osteoblastic cells. These abnormalities in osteoblast function and bone formation are associated with alterations in the expression or production of several growth factors, such as TGF-β which modulate the proliferation and activity of bone-forming cells. Bone transplantation is an absoulte requirement in several patholigical conditions. The growth factors, such as TGF-β, PDGF, were the effective in promoting growth of the bone in vitro and in animal models. We have investigated the effects of PDGF and TGF-β on the proliferation and differentiation of the normal human osteoblast in vitro culture. The normal human osteoblast from iliac bone were primarily cultured. The serums obtained from the osteoporotic patients and the normal was used to quantify PDGF and TGF-β from the osteoporotic patients serum and the normal serum. To clarify the effects of the various different the culture conditions such as 1×10 exp(4) cells/㎖, 2.5×10 exp(4) cells/㎖, 5×10 exp(4) cells/㎖, 10×10 exp(4) cells/㎖ (0.2∼2×10 exp(4) cells/well) of the osteoblast at 24 hours, 48 hours, 72 hours under 10% FBS, 10% normal human serum, 10% osteoporotic human serum, 3% normal human PRP, 3% osteoporotic human PRP. The cell proliferation and differentiation was determined by [^3H]-thymidine and SRB assay, and the magnitude of differentiation to osteoblast was confirmed by von Kossa staining and alkaline phosphatase stain and measuring the alkaline phosphatase activity during 48 hours and 72 hours. Statistical differences were evaluated using the scheffe's test. The ANOVA procedure of the SAS system. The obtained results were as follows ; 1. The age distribution of normal human was 36.9±4.4 old, osteoporotic human was 72.5±0.2 old with statistical significant difference(p<0.05). 2. The quantification of TGF-β of the normal human serum was 39658.38±11630.43 pg/㎖, the osteoporotic human serum was 30459.40±1704.92 pg/㎖ with statistical significant difference. The quantification of PDGF of the normal human serum was 3064.13±709.51 pg/㎖, the osteoporotic human serum was 2514.13±140.21 pg/㎖ with no statistical significant difference(p<0.05). 3. The DNA synthesis and protein assay of human osteoblast at 24 hours, 48 hours, 72 hours was similar increased to 10% normal human seurm, 10% osteoporotic human serum. There was no statistical significant difference between the normal human and the osteoporotic patients in 3% normal human PRP and 3% osteoporotic human PRP. 4. The optimal cell concentration was 5×10 exp(4) cells/㎖ among 1×10 exp(4) cells/㎖, 2.5×10 exp(4) cells/㎖, 5×10 exp(4) cells/㎖, and 10×10 exp(4) cells/㎖. The DNA synthesis was decreased after 72 hours in the normal human serum and PRP, the osteoportic serum and PRP. 5. The alkaline phosphatase activity was as the same result 10% FBS, 10% osteoportic serum and 10% normal human serum at 48 hours with no statistical significant, but the alkaline phosphatase activity was increased in 10% osteoportic human serum and 10% normal human serum except 10% FBS at 72 hours. From above result, the amount of TGF-β of the normal human growth factor was higher than the osteoporotic patients, but the growth factors of the osteoportic patients were enough the proliferation and differentiation of normal human osteoblasts such like the same effects of normal human growth factors.
