        The objective of this research was to characterize viscosity of sea tangle extract isolated from sea tangles in Korea to obtain basic data for production of dietary fiber materials with new functional properties. The viscosity of sea tangle extract was increased as the the extraction time increased. However, these values increased inginficantly up to 1 hour of extraction time and then slow increased in case of hot water extraction and enzymatic hydrolyzed sea tangle extract. As for addition of sugar and salts, the values of viscosity of sea tangle extract was very low in the additon of EDTA-2Na regardless of concentration. But in case of sucrose and NaCl, the viscosity of sea tangle extract were tended to decreased up to 2.5% concentration.

        제주연안에 서식하는 망둑어과 어류의 소화관 구조 및 조직학적 연구

        제주연안에 서식하는 망둑어과 어류인 바닥문절, S. geneionema과 별망둑, C. gulosus 소화관의 형태와 조직학적 특성을 조사하였다. 바닥문절은 소화관 길이가 짧고 (RLG 0.45), 단순한 형태를 가지고 있으며, 위와 유뮨수는 가지고 있지 않다. 별망둑은 소화관 길이가 짧고 (RLG 0.43). 단순한 형태의 위를 가지고 있으며, 유문수는 가지고 있지 않다. 별망둑은 위선이 잘 발달된 위를 가지고 있다. 바닥문절 소화관의 점막주름은 다양한 형태를 가지고 있다. 장에 있어서 근육층의 두께, 점막주름의 길이는 유사한 양상을 하였다. 장 점막주름의 길이는 두 종 모두 전장부에서 다른 부분보다 길었다. 장 근육층의 두께는 두 종 모두 전장부에서 후장부로 갈수록 두꺼워지는 양상을 나타내었다. 점액을 분비하는 배상세포는 바닥문절의 경우 후장부, 별망둑의 경우 중장부에 가장 많이 분포하였다(P〈0.05). The morphology and histological characteristics of the digestive tract of two gobiid species, hairychin goby (Sagamia geneionema) and gluttonous goby (Chasmichthys gulosus), were investigated in the coastal area of the Jeju Island, Korea. The hairychin goby has a short (relative length of gut 0.45), simple gut and no obvious stomach and pyloric caeca. The gluttonous goby has a short (relative length of gut 0.43), simple gut and stomach, but no obvious pyloric caeca. Gastric gland cells are well developed in the stomach of gluttonous goby. In the mucosal fold of the intestine, hairychin goby has a regular branched form and gluttonous goby has a rather complex branched form. The length of mucosal fold and thickness of the muscle layer were similar in the two species. The length of mucosal fold was longer in anterior intestine portion, and the thickness of muscle layer was thicker in posterior intestine portion. In the number of mucous - secreting goblet cells, the hairychin goby was higher in the posterior intestine portion, and the gluttonous goby was higher in the mid-intestine portion (P< 0.05).

      • 포도나무가지 추출물의 프리라디칼 소거 작용 및 염증 발현 매개인자 생성 억제 효과

        Free radical scavengers or quenching agents for reactive oxygen species(ROS) present in consumable fruits, vegetables, and beverages have received considerable attention as potential antioxidants, and thus uses for treatment of several human diseases. In this study, grapevine shoot extract(GSE) containing high concentration of resveratrol and viniferine was evaluated for antioxidant potential and inhibition of pro-inflam-matory medicator production. Utilizing 1, 1-diphenyl-2-picrylhydrazyl(DPPH) scavenging activity and xanthine oxidase(XOD) inhibition assay the GSE showed inhibitory effects of DPPH radical scavenging and XOD activity with the IC_50 values of 34.5 ad 155㎍/ml, respectively. In addition, GSE also exhibited the inhibition of prostaglandin E_2(PGE_2) and nitric oxide(NO) production in lipopolysaccharide(LPS)-induced mouse macrophage RAW264.7 cells with the IC_50 value of 6.4 and 14.5 ㎍/ml, respectively. This suggests that grapevine shoot extract has the potential activity as a natural antioxidant or antiinflammatory agent.

      • 자궁경부, 자궁내막 및 난소병변에서 Nucleolar Organizer Regions(NORs)에 관한 연구

        Nucleolar organizer regions (NORs) is a DNA loop which has a ribosomal RNA gene, and it can be readily demonstrated by argyrophilia of NORs-assocated proteins (Ag NORs). The number of Ag-NORs reflects the cellular activities and proliferation rates, and it is known that it correlates well with the DNA flow cytometrlc data. Recently, the Ag-NORs has been actively studied on many organs under the assumption that Ag-NORs may be useful in determining the malignant potential of the lesions. A silver staining technique to identify Ag-NORs was applied to eighty-one paraffin sections from the normal, premalignant and malignant lesions of uterine cervix, endometrium and ovary. The numbers of Ag-NORs in the nuclei of 100 cells of each case were counted on x1000 magnification, and the mean number of Ag-NORs for every 100 cells was calculated. The results were summarized as follows: 1. The mean numbers of Ag-NORs per nucleus in squamous metaplasia, mild, moderate and severe dysplasia of cervix were 1.41±0.29 (mean±S.E), 1.19±0.39, 1.31±0.49 and 1.40±0.40 respectively. In constrast, the mean numbers of Ag-NORs per nucleus in CIS (3 cases) was 2.46±0.30. In cervical invasive squamous cell carcinoma, the mean numbers of Ag-NORs in stage Ⅰa (5 cases), stage Ⅰb (5 cases), stage Ⅱa (7 cases) and more than stage Ⅱb(6 cases) were 2.69±0.41, 2.54±0.36, 2.61±0.22 and 2.63±0.24 respectively. The differences between the numbers of Ag-NORs in nuclei of squamous metaplasia and cervical squamous cell carcinemas were statistically significant (P <0.01-0.001). 2. In endometrium, the mean numbers of Ag-NORs per nucleus in proliferative phase (3 cases), secretory phase (5 cases), endometrial hyperplasia (5 cases) and endometrial carcinoma (9 cases) were 1.73±0.04, 1.44±0.09, 1.60±0.15 and 2.59±0.35 respectively. The difference between the numbers of Ag-NORs in nuclei of endometrial hyperplasia and endontetrial carcinoma was statistically significant (P<0.001). 3. Among mucinous and serous tumors of the ovary, the mean numbers of Ag-NORs per nucleus in benign (9 cases), borderline (9 cases) and invasive tumors (9 cases) were 0.91±0.11, 1.71±0.16 and 2.39 ± 0.31 respectively. The differences between these groups were statistically significant (P<0.001). In conclusion, firstly, the numbers of Ag-NORs is useful in differentiating between premalignant and malignant lesions in uterine cervix. But it has no correlation with the clinical stages of cervical cancer Secondly, the number of Ag-NORs is also useful in differentiating between endometrial hyperplasia and endometrial carcinoma. Lastly, the Ag-NORs are useful in classification of the ovarian common epithelial tumors into benign, borderline and malignant tumors.

      • 백혈병 치료 중 발생된 간비장 칸디다증에 대한 Caspofungin 치험 1예

        Hepatosplenic candidiasis (HSC) or chronic disseminated candidiasis (CDC) is an invasive fungal infection that affects neutropenic patients. Standard empirical therapy consists of the administration of amphotericin B or fluconazole. In many cases, candidiasis is not easily eradicated, despite the adminstration of these drugs. Caspofungin, a member of the new echinocandin class of compounds, may be an effective alternative that has fungicidal against most candida species and fungistatic against aspergillus species. We report a case of hepatosplenic candidiasis developed during chemotherapy for acute myelocytic leukemia and cured with 5 weeks of caspofungin treatment.

        각종 소화성질환에서의 Campylobacter Pylori의 검출에 관하여

        Peptic disorders are among the most common maladies in korea as the world. In most cases their etiologies not be established and their pathophysioolgy remain obscure. Although the persence of fastric bacteria has been logn established, recently campylobacter pylori seems to be etiologic agent of peptic disorders. Campylobacter pylori is visualized in most cases of peptic disorders and not visualized after treatment of bctericidal agents, bismuth compound or metronidazole. We had a following results for identification of campylocacter pylori in stomach and duodenal mcuosa via endoscipc biopsy. 1. We studied 54 patients who had dyspeptic symptoms, 29 males. 2. Most patients are in 5th decades, next in 3rd and 6th in order. 3. Ptients were composed of 27 peptic ulcer, 22 gastitis and 5 gastric carcinoma. 4. Campylobacter pylori was identified in 69% of total patients, peptic ulcer 63%, gastritis 77%, stomach carcinoma leastly.

      • 수출용 돼지고기 부산물의 냉장저장 중 미생물, pH, VBN 변화 및 유통기한 설정

        본 연구는 저장기간동안 수출용 돼지 부산물(간, 신장)의 미생물, pH 그리고 VBN을 측정하고 이를 토대로 유통가능 기간을 예측함으로서 수출 부산물에 품질과 취급에 대한 기초자료를 얻고자 실시하였다. 총균수는 간과 신장 모두 저장 4일까지 3 logCFU/㎠ 수준을 나타내었으며, 저장6일까지는 6 logCFU/㎠을 유지하였다. 내냉성 균수도 총균수와 비슷한 양상을 보여 주었고, 신장은 간보다 초기오염도가 높고 미생물 증식도 빨라 저장기간이 상대적으로 짧을것으로 사료된다. 저장 1일차의 pH는 간에서 6.31이었고, 신장은 6.78이었다. 저장기간 중 pH의 변화는 간이 6.31∼6.37, 신장은 6.70∼6.78의 범위로 신장이 간보다 pH가 다소 높게 나타나 미생물과 pH와의 관계에서 볼 때 신장의 저장성이 상대적으로 낮을 것으로 사료된다. 저장기간 중 VBN의 변화를 보면 신장은 초기 17.93 mg%에서 8일차에 이르러 40.12 mg%를 나타내었지만, 간은 초기 30.26 mg%에서 저장 6일차에 50.82 mg%에 도달하였다. 유통기한 설정에 있어서는 5.8일∼6.9일까지의 결과가 나왔으나 부패 판단시점은 모두 7일 이상으로 나타났다. 그러므로 초기 오염이 일반적인 식육의 초기 미생물 수준일 경우 일주일 정도가 돼지의 간, 신장에 있어서의 적절한 유통기한이라고 사료된다. This study was conducted to determine the changes in microbe, pH, Volatile Basic Nitrogen (VBN) of exportation by-products of pork and establish their shelf-life during storage at 4℃. Exportation pork by-products of pork were obtained from commercial slaughter house. The number of total plate count (TPC) and psychrotrobes increased (P<0.05) with storage periods. Both TPC and psychrotrobes were significantly (P<0.05) higher in kidney, pH of kidney was singnificantly (P<0.05) higher than liver, whereas VBN of liver was significantly (P<0.05) higher in all storage periods. The shelf-life in both kidney and liver were 7 days at 4℃.

      • 산삼 배양액 급여가 돈육의 이화학적 특성에 미치는 영향

        Berkshire를 이용하여 70일간 산삼 배양액 급여 시 육의 이화학적 특성에 미치는 영향은 다음과 같다. 산삼 배양액의 급여에 의해 육의 수분, 조단백, 조지방, 회분, pH, 가열감량, 콜레스테롤 함량, 육색, 조직감의 응집성과 검성, 포화지방산 및 불포화지방산의 비율은 차이를 보이지 않았다. 보수력, 지방색 L^(*)값, 지방산 중 linoleic acid(18:2), arachidonic acid(20:4), 필수지방산의 함량, 아미노산 중 threonine, glutamic acid, valine, isoleucine, leucine, histidine, lysine 및 총아미노산 함량은 산삼 배양액의 급여에 의해 감소하였다. 전단가, 조직감의 표면경도, 경도, 씹힘성, 지방산 중 stearic acid(18:0), oleic acid(18:1) 함량, 아미노산 중 serine, glycine, cystine, methionine, tyrosine 및 phenylalanine의 함량은 산삼 배양액의 급여에 의해 증가하였다. 본 연구결과를 종합하면 산삼 배양액의 급여는 돈육의 경도를 증가시키는 효과 외에 뚜렷한 이화학적인 차이를 나타내지 않았다. 이러한 이유는 급여된 산삼 배양액 내에 함유된 생리활성 물질의 함량이 낮고, 급여수준이 낮은 결과로 사료되어진다. A total of 60 pigs (Berkshire) were used to investigate the effect of dietary artificial culture medium of wild ginsengs (CMG) on physico-chemical characteristics of pork. About 60 kg pigs were randomly alloted into one of two experimental diet groups (C:commercial diet feed; T:commercial diet + 1 L CMG per day for 70days). Pigs were slaughtered at approximately 110 kg live weight, and chemical composition and physico-chemical characteristics were measured in pork loin. Moisture, crude protein, crude fat and ash were not significantly different by dietary artificial culture medium of wild ginsengs. Water-holding capacity was decreased by dietary artificial culture medium of wild ginsengs, while shear force was increased. However, pH, cooking loss and cholesterol contents were not significantly different between two different treatments. In color, L^(*) of fat was decreased by dietary artificial culture medium of wild ginsengs. Brittleness, hardness and chewiness were increased in dietary artificial culture medium of wild ginsengs group. In fatty acid, stearic acid (18:0) and oleic acid (18:1) were higher in dietary artificial culture medium of wild ginsengs group, whereas linoleic acid (18:2) and arachidonic acid (20:4) were higher in control group. In amino acid, serine, glycine, cystine, methionine, tyrosine and phenylalanine were higher in dietary artificial culture medium of wild ginsengs group, whereas threonine, glutamic acid, valine, isoleucine, leucine, histidine and lysine were higher in control group.

        가시오갈피의 생리·생태적 특성 (Ⅰ) : 산지별 순광합성, 기공증산, 기공전도도 Net Photosynthetic Rates, Stomatal Transpirations, Stomatal Conductances of Leaves

        이 연구는 가시오갈피 엽의 생리적 특성을 밝히고자 광합성, 기공증산, 기공전도도의 생리반응을 측정한 것으로 결과를 요약하면 다음과 같다. 1. 광보상점은 양엽이 약 30 μ mol m^(-2) s^(-1), 음엽이 약 15 μ mol m^(-2) s^(-1) 이었다. 2. 광포화점은 양엽이 약 1,000 μ mol m^(-2) s^(-1), 음엽이 약 300 μ mol m^(-2) s^(-1) 이었다. 3. 우리나라 및 중국산 가시오갈피 상엽의 순광합성속도는 약 8.0-8.8 μ mol m^(-2) s^(-1) 로 산지간에 유의차가 인정되지 않았다. 제주산 섬가시오갈피 상엽은 약 6.9 μ mol m^(-2) s^(-1) 로 다소 낮았다. 4. 모든 산지에서 중위엽의 순광합성속도는 상엽의 40-65%, 하위엽은 상엽의 30% 정도이었다. 그러나 섬가시오갈피 하엽은 상엽의 약 71%로 다소 높았다. 5. 상엽의 기공증산속도는 1.1-1.4 mmol H₂O m^(-2) s^(-1), 중위엽은 0.7-1.0 mmol H₂O m^(-2) s^(-1), 하위엽은 0.5-0.6 mmol H₂O m^(-2) s^(-1)로 산지별로 유의차가 없었다. 6. 기공전도도는 상엽이 약 70-90 mmol H₂O m^(-2) s^(-1)로 산지별 유의차가 인정되지 않았다. 그러나 제주 섬가시오갈피 상엽은 약 380 mmol H₂O m^(-2) s^(-1)로 타산지보다 현저히 높았다. This study was made on the physiological reactions of photosynthesis, stomatal transpiration, stomatal conductance of Acanthopanax senticosus of leaves. The resuits obtained are as follows : 1. The light compensation points were approximately 30μ㏖ m^(-2)s^(-1) in sun leaves and 15μ㏖ m^(-2)s^(-1) in shade leaves. 2. The light, saturation points were approximately 1,000μ㏖ m^(-2)s^(-1) in sun leaves and 300μ㏖ m^(-2)s^(-1) in shade leaves. 3. There was no significant between various mountains distributed, net photosynthetic rates were approximately 8.0 to 8.8μ㏖ m^(-2)s^(-1) in upper leaves. However, net photosynthetic rate in upper leaves of Acanthopanax koreanum in Jeju island was slightly low about 6.9μ㏖ m^(-2)s^(-1). 4. Net photosynthetic rate in middle leaves of all mountains was ranged from 40% to 65% of upper leaves, and that in lower leaves was approximately 30% of upper leaves. But, the net photosynthesis in lower leaves of Acanthopanax koreanum in Jeju island was slightly high about 71% of upper leaves. 5. There was no significant between various mountains distributed, stomatal transpirations were ranged from 1.1 to 1.4 mmol H₂Om^(-2)s^(-1) in upper leaves, 0.7 to 1.0 mmol H₂O m^(-2)s^(-1) in middle leaves, and 0.5 to 0.6 mmol H₂0m^(-2)s^(-1) in lower leaves, respectively. 6. There was no significant between various mountains distributed, the stomatal conductance in upper leaves was ranged from 70 to 90 mmol H₂O m^(-2)s^(-1) . However, stomatal conductance in upper leaves of Acanthopanax koreanum in Jeju island was approximately 380 mmol H₂O m^(-2)s^(-1), its remarkedly higher than any other mountain.

      • 신경교세포 및 RAW 264.7 세포에서 Protein kinase의 활성에 의한 유도성 Nitric oxide synthase의 발현

        NO(nitric oxide) plays an important role as neurotransmitter or cytokine, and pathologic factor for some diseases by the large amount production with iNOS(inducible NO synthase) expression in macrophages or glial cells. The expression of iNOS is regulated by various cytokines, protein kinases and transcription factors. In this experiment, to investigate the roles of progein kinase and NF-kB for iNOS expression, the effects of PMA(phorbol 12-myristate 13-acetate), cAMP, and various protein kinase inhibitors on LPS(lipopolysaccharide)-induced iNOS mRNAN expression and nuclear NF-kB binding complex were examined in C6 glial cells and RAW 264.7 cells. In C6 glial cells, iNOS mRNA expression by LPS was induced from 1 hour and peak at 3 hour after treatment. In RAW 264.7 cells, the mRNA was observed from 3 hour and peak at 6 hour. PMA enhanced markedly LPS-induced iNOS mRNA expression and NF-kB binding complex in C6 glial cells, but did not much influence on LPS-induced iNOS mRNA expression in RAW 264.7 cells, in spite of increased LPS-induced NF-kB binding complex at 30 min. cAMP(dibutyryl cAMP) did not much influence on LPS-induced iNOS mRNA expression, by increased LPS-induced NF-kB binding complex in C6 glial cells. However, in RAW 264.7 cells, cAMP increased slightly LPS-induced iNOS mRNA expression without change of NF-kB binding complex. Staurosporine did not influence on LPS-induced iNOS mRNA expression and NF-kB binding complex in C6 glial cells, but in RAW 264.7 cells, decreased LPS-induced iNOS mRNA expression and NF-kB binding complex. Ro-31-8220 did not much influence on LPS-induced iNOS mRNA expression and NF-kB binding complex in C6 glial cells, but in RAW 264.7 cells, decreased significantly LPS-induced iNOS mRNA expression in spite of increased LPS-induced NF-kB binding complex for 3hours. G 6976 did not much influence on LPS-induced iNOS mRNA expression with decreased NF-kB binding complex in C6 glial cells, but in RAW 264.7 cells, decreased iNOS mRNA expression without influence on LPS-induced NF-kB binding complex. Genistein did not influence on LPS-induced iNOS mRNA expression and NF-kB binding complex in C6 glial cells, but in RAW 264.7 cells, decreased LPS-induced iNOS mRNA expression inspite of increased NF-kB binding complex. These results suggest that LPS-induced regulation of iNOS expression or NF-kB activity in C6 glial cells, might be different from RAW 264.7 cells through various protein kinases or other factors.
