        동과자 에탄올 추출물이 비알코올성 지방간 세포 모델에 미치는 효과

        Objectives : In this study, we investigated the effect and the underlying mechanism of ethanol extract of Benincasa seeds on a cellular model of non-alcoholic fatty liver disease (NAFLD) established by treating HepG2 cells with palmitate. Methods : We evaluated ethanol extract of Benincasa seeds (EEBS) for its hepatic lipid-lowering potential in fatty acid overloaded HepG2 cells. After incubation in palmitate containing media with or without EEBS, intracellular neutral lipid accumulations were quantified by Nile red staining. We also investigated the effect of EEBS on lipogenesis and ${\beta}$-oxidation. $LXR{\alpha}$-dependent SREBP-1c activation, expression of lipogenic genes, and expression of ${\beta}$-oxidation related genes were determined with or without pretreatment of EEBS. Results : EEBS significantly attenuated palmitate-induced intracellular neutral lipid accumulation in HepG2 cells. EEBS suppressed fatty acid synthesis by inhibiting $LXR{\alpha}$-dependent SREBP-1c activation. EEBS also repressed SREBP-1c mediated induction of lipogenic genes, including ACC, FAS, and SCD-1. However, EEBS had no effect on ${\beta}$-oxidation related CPT-1 and $PPAR{\alpha}$ gene expression. Conclusions : Our results suggest that EEBS has an efficacy to decrease hepatic lipid accumulation, and this effect was mediated by inhibiting the $LXR{\alpha}$-SREBP-1c pathway that leads to expression of lipogenic genes and hepatic steatosis. Therefore, the Benincasa seeds may have a potential clinical application for treatment of this chronic liver disease.

        휴대용 폐기능 검사기의 재현성과 기대치에 관한 연구

        Objectives : The object of this study was to evaluate repeatability of portable spirometry and to determine an appropriate equation for predicted value. Materials and Methods : The subjects were 76 men and 45 women recruited from among Kyunghee University oriental medical students. Portable spirometry was performed via the procedure recommended in ATS guidelines. Repeatability of spirometry was checked against ATS repeatability standards and NHLEP QC grade. Percentage of predicted value of non-smokers was calculated through a Caucasian population-based regression equation(by Morris et al.) and Korean population based regression equation(by Kim et al.). These were compared. Result : 91.7% of subjects matched repeatability standards of ATS and 95% of subjects got interpretable NHLEP QC grades of A, B, and C. Mean percentage of predicted value by Morris et al. were as follows: Mean of FVC% was 90.86, FEV1% was 96.66, FEV1/FVC% was 107.48. Mean percentage of predicted value by Kim et al. were as follows. Mean FVC% was 92.38, FEV1% was 89.77, FEV1/FVC% was 96.95. Conclusions : Results of this study show that more than 90% of subjects meet repeatability criteria. This supports a role for the portable spirometer as a primary oriental medical office tool. Further, results are a verify that they Korean population-based predicted value equation is a more appropriate standard than the Caucasian population-based one, but a still more accurate standard is needed for the Korean adult population.

        자하거(紫河車) 약침의 기관지 천식 치료 가능성에 대한 소고(小考)

        Objectives: The aim of this study was to find out some rationales for using Hominis placental extract(HPE) pharmaco-acupuncture on asthmatic patients, and to suggest an appropriate clinical research of HPE pharmaco-acupuncture on asthma. Methods: Articles regarding immunologic experiments about HPE and clinical reports about HPE-contained pharmaco-acupuncture on asthmatic patients were reviewed by on-line and off-line searching. Results and Conclusions: HPE is suggested to have several pharmacological effects on allergic diseases including asthma as well as autoimmune diseases. There are some observational studies of HPE-contained pharmaco-acupuncture on asthmatic patients. However, the designs of these studies are not sufficient to investigate the exact effect of HPE pharmaco-acupuncture on asthma. To investigate HPE pharmaco- acupuncture as a novel asthma therapy, it will be needed to perform further experiments about HPE`s pharmacological mechanism on asthma and to try further advanced clinical researches such as well-designed randomized controlled trial of HPE pharmaco-acupuncture.

        토양 소수성에 따른 미량원소의 흡착 및 이동

        소수성 토양이란 물방울이 떨어져도 자연스럽게 젖지 못하고 표면 위에 그대로 존재하는 토양을 말한다. 소수성 토양은 토양입자들 사이 혹은 토양입자와 그 중간물질의 사이에 소수성 유기물질이 침투하여 입자를 코팅하여 소수성을 띠게 되는 것이다. 토양이 소수성화가 되면 심한 침식과 침출, 농약의 급속한 침출, 식물이 이용할 수 있는 많은 양의 물과 영양분의 손실 그리고 농작물과 목초의 생산성 감소 등을 야기한다. 본 실험에서는 미량원소가 어떤 흡착과 이동 메커니즘을 갖는지를 배치 실험과 컬럼 실험을 통해 비교 평가하였다. 배치 실험에서는 Cu<SUP>2+</SUP>, Mn<SUP>2+</SUP>, Fe<SUP>2+</SUP>, Zn<SUP>2+</SUP> 및 Mo<SUP>5+</SUP>를 이용하여 일반 토양과 소수성 토양의 수착의 차이, 소수성 정도에 따른 차이, 각각의 미량원소 별 차이를 비교하였고, 컬럼 실험에서는 피흡착물질인 미량원소 Fe<SUP>2+</SUP> 및 Mo<SUP>5+</SUP>에 대한 탈착성능 비교와, 특정시간에 따른 공간적 농도 분포를 측정하였다. Fe<SUP>2+</SUP>와 Mo5+의 경우 비소수성 토양보다 소수성 토양에서 더 높은 흡착을 나타났다. Fe<SUP>2+</SUP>의 흡착성능은 소수성 토양별 40% 〉 15% 〉 0% 순으로 나타났다. 특히 Mo5+의 경우 0%, 15% 및 40%의 소수성 토양에서 초기에는 비슷한 경향을 보였으나 시간이 지나면서 소수성 토양에서 비소수성 토양보다 빠른 탈착속도를 보였다. Fe<SUP>2+</SUP>와 Mo5+의 파과곡선 분석결과 소수성 토양에서 더 빠른 이동속도를 나타내었다. Water repellency which affects infiltration, evaporation, erosion and other water transfer mechanisms through soil has been observed under several natural conditions. Water repellency is thought to be caused by hydrophobic organic compounds, which are present as coatings on soil particles or as an interstitial matter between soil particles. This study was conducted to evaluate the characteristics of the water repellent soil and transport characteristics of trace elements within this soil. Capillary height of the water repellent soil was measured. Batch and column studies were accompanied to identify sorption and transport mechanism of trace elements such as Cu<SUP>2+</SUP>, Mn<SUP>2+</SUP>, Fe<SUP>2+</SUP>, Zn<SUP>2+</SUP> and Mo<SUP>5+</SUP>. Difference of sorption capacity between common and repellent soils was observed depended on the degree of repellency. In the column study, the desorption of trace elements and the spatial concentration distribution as a function of time were evaluated. The capillary height was in the repellency order of 0% 〉 15% 〉 40% 〉 70% 〉 100%. No water was absorbed in soil indicating 〉70% repellency. Using trace elements, Fe<SUP>2+</SUP> and Mo<SUP>5+</SUP> showed higher sorption capacity in the repellent soil than in non-repellent soil. The sorption performance of Fe<SUP>2+</SUP> was found to be in the repellency order of 40% 〉 15% 〉 0%. Our results found that transfer of Mo5+ had similar sorption tendency in soils having 0%, 15% and 40% repellency at the beginning, however, the higher desorption capacity was observed as time passes in the repellent soil compared to in non-repellent soils.

        위장 비율동성 당뇨환자에서 족삼리(足三里)($ST_{36}$) 전침처치의 위전도 조절 영향 사례군 연구

        Objectives : This study was to investigate the regulatory effect of electro-acupuncture on Jogsamni ($ST_{36}$) in bradygastric condition or gastroparesis which is a neuropathic complication of type 2 diabetes mellitus(T2DM), assessed using 4-channel electrogastrography (EGG). Methods : It was a case series study. Each patient was given electro-acupuncture(EA) one point, Jogsamni($ST_{36}$) or Gyeonu($LI_{15}$) bilaterally for thirty minutes with 2Hz bipolar square wave frequency and moderate intensity of stimulation. With attaching electrodes around stomach region throughout pre-acupuncture, acupuncture, and post-acupuncture session, these parameters were produced; percent rate of bradygastria power compared to sum of power in all kind of gastric rhythm(% bradygastria), dominant frequency(DF) and dominant power(DP). Parameter was analysed per each channel's variable change from pre-acupuncture session by paired t-test. Results : There was statistically significant decrease in % bradygastria parameters on one of the $ST_{36}$ treated patients from pre-acupuncture session and to post-acupuncture session(P=0.015, P=0.008 respectively). A marginal significant decrease of DP was shown in the other $ST_{36}$ treated patient from pre-acupuncture session through post-acupuncture session(P=0.049). Combined data of two $ST_{36}$ Treated patients showed that there was significant decrease of %bradygastria from pre-acupuncture session to acupuncture session(P=0.020), and was decrease of DP from pre-acupuncture session to post- acupuncture session(P=0.020). Conclusions : EA at $ST_{36}$ resulted in statistically significant decrease in %bradygastria and DP for dysrhythmic condition in T2DM gastroparesis patients. Considering this limited results, further study will be needed to elucidate the effect of multiple-acupuncture on gastric myoelectrical activities.

        만성기침에 대한 보조제제로써 청인(淸咽)트로키 복합투여의 임상적 유효성 평가

        Objectives : We designed this study to evaluate the effectiveness of Chungin-troche(CIT) preparation as an adjunctive medicine to a herbal extract of Chungsangboha-tanggagambang(CSBH). Methods : A paralled, comparative study of CSBH+CIT and CSBH was conducted on 42 patients of chronic cough who attended Division of Allergy, Immune & Respiratory System, Department of Internal Medicine, College of Oriental Medicine, Kyung Hee University between December, 2007 and June, 2008. The treatment in each group lasted for two weeks. Patients were asked to fill in the Leicester Cough Questionnaire (LCQ) before and after administration of each treatment legimen. Results : 42 patients completed the trial and no adverse effects were reported. Both CSBH +CIT group and CSBH group showed statistical significant improvement in total LCQ score after 2 weeks from baseline. Social domain score and total score in patients taking CSBH+CIT showed a statistical significant increase compared to those scores in CSBH patients. Conclusion : CSBH+CIT compositive preparation and CSBH separate preparation appeared to have some benefits in the treatment of chronic cough. However, additional CIT preparation to CSBH can improve social domain in LCQ of chronic cough patients.

      • 하절기 한지형 잔디 재배 시 침수 및 고온으로 인한 잔디의 생육 불량 현상

        한지형 잔디는 하절기 온도가 $30^{\circ}C$ 이상까지 올라가는 우리나라에서 뿌리와 줄기의 성장이 급격히 저하되고 심하면 고사하게 된다. 기온의 상승으로 인한 지온 상승은 잔디뿌리에 스트레스가 되며 그 스트레스는 호르몬의 불균형적인 형성으로 지상부 생장 및 광합성, 증산작용 등의 여러 매카니즘에 많은 영향을 미치게 된다. 이러한 문제를 해결하기 위해 토양내 수분함량을 조절하기도 하고 유기물 함량을 조절하여 지온의 상승을 제한함으로서 잔디의 생육부진을 해소하는 연구도 수행되었다. 본 실험은 토양 수분의 변화에 따른 토양 온도의 변화 양상을 분석하고, 잔디 재배지에서의 토양 깊이와 수분 함량에 따른 열 전도 기작과 열 환경 변화에 따른 직접적인 잔디 생육(생산량, 녹도, 건조피해, 고온피해 등)에 미치는 영향을 연구하였다. 공시 초종으로 Kentucky bluegrass(Poa pratensis L.)를 선정하였고 각각의 실험구는 수분함량에 따라 25%, 33%, 40%의 3가지로 구분하여 처리하였다. 토양온도측정 센서를 각 실험구에 깊이 12cm 지점과 2cm 지점에 각각 2개씩 즉, 한 실험구당 4개의 토양온도측정 센서를 설치하여 매 5분마다 온도변화를 측정하였다. 수분함량에 따른 온도변화는 수분함량이 33% 일 때 가장 낮게 측정되었다. 잔디의 생육상태를 비교하여 볼 때 수분 함량이 33% 일 때 생육이 가장 양호하였으며 40% 일 때 가장 생육이 불량하였다. 하절기 고온 환경에서 한지형 잔디인 Kentucky bluegrass의 생육은 토양 수분함량에 따라 큰 영향을 받지만, 장마기에 잔디가 침수된 환경에서 토양 수분 함량을 최소한 33%까지 감소시켰을 경우에는 잔디 생육에 큰 영향을 받지 않을 것으로 사료된다. The growth of root and shoot normally decline dramatically in mid-summer of Korea, moreover the cool-season turfgrassgrass eventually wither to death over $30^{\circ}C$. The increase of air temperature also drives the heat of soil, that makes stress on root system. The heat stress affects physiological mechanisms of hormonal unbalance that stimulates shoot growth, photosynthesis, and transpiration. To solve those problems, many studies have been carried out to control soil moisture and OM content to decrease soil temperature for dissolving the growth retardant by heat stress. This study initiated to analyze the change of soil temperature with soil moisture, and the effect of soil depth and moisture content on heat transmit and thermal changes on turfgrass growth(productivity, green color, and damage by dryness and high temperature). Kentucky bluegrass plots prepared with 25%, 33%, 40% soil moisture treatments. Soil temperature was measured every five min. with four thermo-sensors at 12 and 2 cm soil depth. The most acceptable growth showed at 33% soil moisture, but the worst result showed at 40%. The soil moisture seriously affected on the growth of Kentucky bluegrass, however the quality of turfgrass may acceptable if we can control soil moisture down to 33% when the flooding season of monsoon.
