        An immunohistochemical observation on the somatostatin cells in the gastrointestinal tract of the Y?nsan Ogol fowl

        연산오골계의 위장관에 존재하는 somatostatin cell의 분포상태와 출현빈도를 알아보기 위해 면역조직화학적 방법을 이용하여 광학현미경적으로 관찰하였던 바 다음과 같은 결과를 얻었다. Somatostatin cell은 유문부에 밀집하여 출현하였으며, 그 다음으로 선위의 복합선에서 다수가 관찰되었다. 그리고 십이지장에는 소수가, 공장과 근위에는 드물게 분포하였으며, 회장 맹장 및 직장에서는 관찰되지 않았다. 또한 somatostatin cell은 부위에 따라 그 분포양상의 차이가 인정되었다.

        한우 및 한국재래산양 유래 말초혈액 림프구의 미소핵을 이용한 방사선 피폭의 생물학적 선량측정

        The frequencies of gamma-ray-induced micronuclei (MN) in cytokinesis-blocked (CB) lymphocytes at several doses were measured in three donors of Korean native cattle and Korean native goat. Measurements performed after irradiation showed a dose-related increases in MN frequency in each of the donors studied. When analysed by linear-quadratic model the line of best fit was : cattle : y=0.1016D+0.0118D2+0.0147, goat:y=0.1353D+0.0043D2+0.0087 (y=number of MN/CB cells and D=irradiation dose in Gy). The relative sensitivity of goat lymphocytes compared with cattle lymphocytes was estimated by best fitting linear-quadratic model based on the radiation-induced MN data over the range from 0 Gy to 4Gy. In the case of MN frequency with 0.05, 0.1, 0.2, 0.4 and 0.8, the relative sensitivities of goat lymphocytes were 1.106, 1.166, 1.140, 1.069 and 0.976 respectively. Our in vitro radiobiological study confirmed that the cytogenetic response obtained in blood from cattle and goat can be utilized for application in environmental studies.

        Murine lymphokines production in lymphoid organs during the various stages of lactation

        비유중기, 건유초기 및 임신말기의 C3H/HeN 마우스의 비장, 장간막림프절, 말초림프절(액와, 상완 및 샅림프절)유래 림프구에서 생산되는 lymphokine의 양을 비교하였다. 비장에서의 IL-2 생산양은 임신말기와 비유중기에 낮았지만, IL-4, IL-6와 $IFN_{\gamma}$의 생산양은 임신말기와 건유초기에 높았다. 말초림프절에서의 IL-4, IL-6 및 $IFN_{\gamma}$ 생산양은 임신말기와 건유초기에 높았지만, IL-2의 생산양은 임신말기, 비유중기 및 건유초기에 각각 감소하였다. 장간막림프절에서의 IL-4 생산양은 임신말기와 비유중기에 각각 증가하였으나, IL-2의 생산양은 임신말기에 감소하였다. 이와같이 전반적으로 건유초기와 임신말기에 IL-2의 생산양은 낮지만, IL-4, IL-6와 $IFN_{\gamma}$의 생산양이 높은 결과는 이 시기에 유방염의 발생비율이 높은 것과 연관성이 있는 것으로 추정된다.

        Effects of serum on lymphokines producing capabilities of CD8<sup>+</sup> T cells

        serum factor가 $CD8^+$ T cell의 lymphokine 생산양상에 미치는 영향을 알아보기 위하여 13-20주령의 BAL B/C 마우스로 부터 $CD8^+$ T cell를 분리한 후 serum-containing medium과 serum-free medium을 사용하여 배양하였다. serum-free medium에서 배양한 $CD8^+$ T cell이 분비하는 lymphokine의 양은 serum-containg medium에서의 결과와는 달리 IL-2의 생산양은 낮았으나 IFNr의 생산양은 상당히 높았다. 이와같은 결과로 미루어 serum-derived factor가 $CD8^+$ T cell의 lymphokine 생산에 영향을 미친다고 생각된다. The responsiveness of $CD8^+$ T-cell subpopulation according to serum-containing and serum-free conditions were investigated. Cells are freshly isolated from spleen of mature adult BALB/C mice between 13-20 weeks of age. $CD8^+$ T cells in serum-free conditions produce small amounts of IL-2, while significant amounts of IFNr following activation when compared the results of serum-containing conditions. These data indicate that serum-derived factors may play an important role in the alternations of $CD8^+$ T cell responsiveness.

        Dexamethasone을 투여(投與)한 Rat 유선조직(乳腺組織)의 병리조직학적(病理組織學的) 관찰(觀察)

        The present study was carried out to observe the histopathological changes in the mammary gland of lactating rats and rabbits injected with dexamethasone. White rats were intramuscularly injected with 0.25mg, 0.5mg or 1.0mg of dexamethasone sodium phosphate (containing $9{\alpha}$-fluoro-$16{\alpha}$-methylprednisolone, 5.0mg/ml) daily for 3 to 10 days on the 3rd day after parturition and white rabbits were intramammary infused with 4mg or 20mg of dexamethasone daily for 4 days on 7th day after parturition. The histopathological changes of the mammary glands, ovaries and adrenal glands of rats and rabbits were observed with light microscope. In the mammary glands of rats, the microscopic findings encountered were decrease of the milk in the alveolar lumina, necrosis and desquamation of epithelial cells, atrophy of alveoli, proliferation of fibroblasts and thickness of alveolar walls, destruction of alveoli, presence of fat droplets within the glandular epithelial cells, infiltration of mononuclear cells and proliferation of adipose tissue, which were relative to the dose and duration of injection. Especially, in the cases of the administration of large doses or long duration, there were severe fibrosis and focal necrosis of glandular tissue. In the mammary glands of rabbits, the morphological changes were similar to those findings in the rats. The milk in the alveolar lumina was decreased gradually according to the dose and duration of injection, while milk fat concentration regarded to increase. In the histological findings of ovaries, necrosis of granulosa cellos, vacuolization and necrosis of luteal cells, atrophy and necrotic foci in the corpora lutes were observed. In the adrenal glands, hyperemia, hemorrhage, vacuolization of adrenal cortical cells, necrotic foci and atrophy of adrenal cortex were observed.

        유선조직내에 출현하는 dendritic cell의 형태학적 연구 II. 전자현미경적 관찰

        In order to investigate the morphological characteristics of dendritic cells in the mammary gland of the mouse (C57 BL/6), rat(W), rabbit and cat, the fine structures of the dendritic cells have been observed by the electron microscope. The results obtained were summarized as follows: The dendritic cells with the well-developed processes had an irregular shape, and lacked the desmosome. The pinocytotic vesicles and tubular invaginations of the cell membrane were frequently observed, and the mitochondria with the well-developed cristae were located in the restricted region in the cytoplasm of the dendritic cells. The nuclei of dendritic cells were indented, In the mice and rats, the dendritic cells had a few Langerhans cell granules(Birbeck granules). From the above results, it is confirmed that the ATPase-positive dendritic cells in the mammary gland are the Langerhans cells.

      • 오리 선위 점액분비세포의 점액질에 대한 조직화학적 연구

        Histochemical sequential staining techniques, PAS, alcian blue pH 2.5 and pH 0.4, were applied to sections from the proventriculus of ducks to observe the histochemical properties of epithelial mucosubstances in the proventriculus of mallard and white Pekin ducks. The whole proventriculus obtained were divided systemically cross strips and processed routinely for histologic and histochemical study. Base on the histologic characteristics, we designated as upper portion which is the distal region of the esophageal-proventriculus junction, as middle portion, and as lower portion which is the proximal region of the proventricular-isthmus junction. The results obtained are as follows 1. Generally in the surface and foveolar epithelia showed the presence of the mixture of neutral and acid mucosubstances. 2. In the tubular gland showed the acidic nonsulfated and sulfated mucosubstance in addition to neutral mucosubstances with occasional neutral and acidic nonsulfated mucosubstances. 3. In the compound gland neutral mucosubstance with scant amounts of acidic mucosubstances or with lack of it were demonstrated. 4. There were recognized differences between speies in the staining intensity of all three types of mucosubstances. 5. There were recognized topographic difference in the acidity of epithelial mucosubstances according to portion.

      • Dexamethasone Disodium Phosphate를 투여한 생쥐 대장 점액질의 조직화학적 관찰

        This-experiment was performed in order to observe the effect of gulcocorticoid on the mucosubstances in the large intestine of mice. Adult mice were divided into control and hormone-treated group was daily injected intramuscularly with a dose of 100ug or 500 ug of dexamethasone disodium phosphate for 5 to 14 days and then were sacrificed under ether anesthesia on the 5th, 7th, 9th and 14th day after the injection. Specimens were collected from the cecum, the proximal protion of colon and the distal portion of colon and fixed in neutral buffered formalin. Paraffin sections were cut serially at 5 or 61um. Sections from each series were stained with hematoxylin and eosin, periodic acid-Schiff(PAS) reaction with counterstain, alcian blue pH 2. 5 with counferstain, alcian blue pH 1.0 without counterstain and aldehyde fuchsin pH 1.7-alcian blue pH 2.5 so as to demonstrate mucosubstances, respectively. The histochemical changes of the mucosubstances in the large intestine were observed with light miroscope. In the control grup, generally acid mucosubstances were more predominent throughout the large intestine than neutral one, but sulfated acid mucosubstances were predominent in the cecum and distal colon and nonsulfated acid mucosubstances were prominent in the lower crypt of proximal colon. In the dexamethasone treated group, from 7th day after the injection of a dose of 100 ug the decrease of mucosubstances in the cecum was appeared and these findings were remarkable on proportion to the duration of treatment and dose. During these duration, the decrease of blue-purple or purple were more remarkable. The similar findings were also found in the proximal and distal colon and generally the decree of sulfated acid mucosubstances were the prominent type seen.

        유선조직내에 출현하는 dendritic cell의 형태학적 연구 I. ATPase-positive dendritic cell의 분포양상

        In order to investigate the morphological characteristics of dendritic cells in the mammary gland, the appearance on the clear cells(CLs) or ATPase-positive dendritic cells(APDCs) have been observed by the light microscope. The results obtained were summarized as follows: CLs were observed in the mammary tissues of the experimental animals, such as mice, rats, guinea pigs, rabbits, cats, dogs, pigs, cows and Korean native goats, and these CLs were confirmed as the ATPase-positive cells of typical dendritic appearance(APDCs), The APDCs were distributed in between the secretory epithelial cells, between the secretory epithelial cells and the myoepithelial cells, the basal area of the secretory epithelial cells, the interalveolar and interlobular connective tissues, and in between the epithelial cells of secretory duct. The APDCs were observed more frequently during the middle period of lactation than the other periods, and were irregularly or uniformly distributed according to the location. During the middle period of lactation, there were notable quantitative differences in the APDSs depending on the mammary glands of mice, rats, guinea pigs, rabbits and cats, The most prominent differences were recognized among the mice, guinea pigs and cats. The number of AP DCs per unit area was statistically fewer in the guinea pigs($209.07{\pm}51.75cells/mm^2$) than in the mice($221.00{\pm}50.94cells/mm^2$) and cats($223.56{\pm}49.68cells/mm^2$) (respectively, p<0.05, p<0.05). Among the A/J, DBA/2, C57BL/6 and NIH(GP) mice, the mean densities of APDCs was statistically significantly fewer in the DBA/2($196.65{\pm}43.47cells/mm^2$) than in the C57BL/6($248.40{\pm}41.40cells/mm^2$) and NIH(GP) ($235.98{\pm}55.89cells/mm^2$) (respectively, p<0.0000, p<0.0000), however no significant difference between the C57BL/6 and the NIH(GP) was recognized (p>0.1). Among the F344, SD and W rats, the statistical analysis were confirmed that there were significantly fewer APDCs in the F344($198.72{\pm}47.61cells/mm^2$) than in the SD($227.70{\pm}41.40cells/mm^2$) and W($223.56{\pm}49.68cells/mm^2$) (respectively, p<0.0000, p<0.0001), however no significant difference between the SD and the W was recognized(p>0.1). The mean difference between the inbred and the noninbred counts in the mice was statistically significant (p<0.0001), and the similar result was presented in the rats(p<0.0000).
