Gel electrophoresis has proven to be one of the most useful of DNA separation and purification. The new technique of pulsed field gel electrophoresis (PFGE) is high resolution separation of large size DNA moleculs. Conventional continuous gel electrophresis can not be separation of large DNA fragments(20~50 k base). Field inversion gel electrophoresis(FIGE) is very useful for large DNA molecules. We have found that a pulse ratio ; 2 : 1, time ; 24hrs., volts ; $10^{volts}/_{cm}$, start ; 0.45sec, end ; 1sec, is most effectively resolves DNA fragment in the 6~50k base.

Tuberculin(HCSM) 반응유우 60두에 대하여 가열살균처리한 우결핵균에서 얻은 PPD-BS tuberculin을 이용한 추벽피내재검사법과 석탄산살균처리한 우결핵균에서 얻은 SFA tuberculin을 이용한 경측피내재검사법간의 민감성과 특이성을 비교하였다. 또한 SFA tuberculin과 현행 조형 PPD(PPD-A) tuberculin을 이용한 비교검사법의 가치를 판독기준에 따라 분석하였다. 병소우군에서 PPD-BS와 SFA tuberculin간에 민감성은 차이가 없었으나 무병소우와 감염우동거군에서 SFA tuberculin은 PPD-BS tuberculin에 비하여 비특이반응이 현저히 낮았다(P<0.01). SFA와 PPD-A를 이용한 비교검사법은 판독기준에 따라 가양성반응과 가음성반응에 크게 영향을 주었으며, 피내반응차이 4mm를 판독기준으로 할 때 병소우와 무병소우를 감별할 수 없었다. 이 연구에서 SFA tuberculin은 PPD-BS에 비하여 병소우에서 민감성간에는 차이가 없었으나 무병소우에서 특이성이 현저히 높았다는 점으로 보아 앞으로 HCSM tuberculin 반응우에 대한 재검사는 현행 PPD에서 SFA tuberculin으로 대체함으로써 비특이반응우를 더욱 감소시킬 수 있다는 것을 의미한다.

Milk urea nitrogen (MUN) determination is being used an indicator of the protein-energy balance in dairy herds. A faulty balance can be corrected to optimize milk production and animal health. This parameter is regarded as a potential tool to evaluate suboptimal feeding practices and reproductive disorders. Therefore, the purpose of this study was to investigate the response of milk composition by regular feeding analysis and to compared the relationship between MUN and milk protein(MP) and fertility at the insemination period in Holstein dairy cows. Total of 355 artificial insemination (AI) for 150 Holstein cows in the herd were used to examine the relationship between MUN and MP content and conception rate. The AI occured for the cows 50 to 150 day in milk, and MUN and MP concentration were determined using automated infrared procedures. The mean${\pm}$standard deviation of MUN and MP concentration in the herd were $15.6{\pm}2.1mg/dl$ and $3.23{\pm}0.38%$, respectively. MUN contents of bulk milk were increase by elevated crude protein intake. The conception rate was lower in the cows in which the level of MUN was lower than > 8.0mg/dl (10.0%) or > higher than 25mg/dl (15.4%) relative to the cows in MUN content of 12.0~17.9 mg/dl (36.7%) at the time of insemination. Also, lower MP than 3.0% or higher MP than 3.25% were associated with a lower conception rates. Consequently, MUN and MP analyses may be used serve as a monitoring tool of protein and energy nutritional balance to improve reproduction efficiency in Holstein dairy cows.

The present study was undertaken to develop the kit for indirect immunofluorescence antibody test(IFAT) and to investigate the prevalence of N caninum in cattle and dogs in Korea. The neo-antigen kit of IFAT developed in our laboratory was proved diagnostic efficacy by compared with the kit of veterinary medical research and development(VMRD). A survey of N caninum infections among cow and dogs in elevn areas of southern part of country was performed using a IFAT. The infection rate of 190 nationwide cattle was 8.4%(16/190) but was 75%(45/60) at P area in Chunnam province where the cattle showed the abortion repeatedly. Any of the dogs was not N caninum -positive in the Kangwon and Kyonggi areas.
Leukocyte adherence inhibition test를 이용한 Mycobacterium 속균(屬菌) 감작(感作)기니픽의 세포면역반응(細胞免疫反應)의 특이성(特異性)

In order to measure in vitro cell mediated immunity in the guinea pigs sensitized with the killed bacilli of Mycobacterium bovis ($AN_5$), M avium (serotype 2), M tuberculosis and M intracellulare (serotype 8), leukocyte adherence inhibition (LAI) test was established using the antigens of purified protein derivatives (PPD) tuberculin. By using LAI test, specificity of cell-mediated immune responses of the guinea pigs inoculated with various Mycobacterium spp was investigated, and comparison between values of LAI and skin test was also made to evaluate the specificity of the newly designed test. The results obtained throughout the experiments were summarized as follows; 1. The optimal concentration of PPD antigens for LAI test was 1 to 2mg per ml of medium. 2. When the leukocytes of guinea pigs sensitized with both M bovis($AN_5$) and M avium (serotype 2) for 2 to 8 weeks were incubated with homologous or heterologous PPD antigens, mean values of LAI test were $61.2{\pm}11.2$ and $65.6{\pm}5.1%$ in homologous PPD antigens respectively, while $30.0{\pm}3.7$ and $32.8{\pm}5.7%$ in heteNlogous PPD antigens, showing the prominently high value of LAI in the homologous syst,em (p<0.01). 3. When the leukocytes of guinea pigs sensitized with both M tuberculosis and M intracellulare (serotype 8) for 2 to 8 weeks were incubated with homologous and heterologous PPD antigens, mean values of LAI test were $67.9{\pm}2.9$ and $66.9{\pm}5.0%$ in homologous PPD antigens, while $27.4{\pm}7.4$ and $24.4{\pm}7.1%$ in heterologous PPD antigens, showing the prominently high value of LAI in the homologous system (p<0.01). 4. Comparing with the specificity of LAI and skin tests on the basis of the value obtained from the homologous system, deviation of reaction was revealed to be 49.5 to 100.2 in LAI test, and -15.9 to 52.0 in skin test.

Beef plates were sanitized with 3% acetic acid and 3.5% sooium chloride before stored at 4c and 85±5% relative humidity. Organotrophic, Bacteriologic, pH and Volatile basic nitrogen analysis were followed for 9 days. The results obtained were summarized as follows; Slime was detected within 4-5 day in the control, 9 day in the sanitized samples with corresponding Psychrotrophic count of about 10⁸c.f.u/cm². Isolates from Psychrotrophic colony have the characteristics of Pseudomonas. Coliforms and Lactic acid bacteria count increased slowly but the relative population load to psychrophiles decreased with time. The pH in control showed the little changes until spoilage was detected. The pH in sanitized samples dropped to 4.7, rose to original level until about sixth day of storage. The equation of the regression between the Volutile basic nitrogen(V) and the Psychrophiles count(P) was V=-1.58+3.07 Log P with a correlation coefficient of 0.84(P<0.01). The treatment with 3% acetic acid and 3.5% sodium chloride prolonged the microbial spoilage by 4-5 days compared to the control samples, but the treated samples evolved acetic acid odors and showed the discolorization.

The present surveys were conducted in attempts to investigate the health situation of horses in Korea through mass-screening the samples serologically, bacteriologically and clinically. A total of 575 horses were sampled randomly, comprising 126 from the Korean Horse Affairs Association, 288 from the Korean Equestrian Federation and 161 from the Jeju ponies. Each of the samples taken was tested for diagnoses of 18 horse diseases including African horse sickness. Summarised below are the results obtained from this surveys. 1. From results of the serological survey it is evident that Korea is currently free from African horse sickness, dourine, glanders, vesicular stomatits, equine piroplasmosis, equine viral arteritis, Venezuelan encephalomyelitis and contagious equine metritis. Constant vigilence with strengthened quarantine measures is thus vital for maintaining freedom of any those diseases in Korea. 2. No clinical case was observed with any of signs or symptoms of infectious lymphangitis, anthrax and infestations with ringworm, mange or scab. However, continuous follow-up is required for establishing the evidence of no occurrence of the diseases in Korea. 3. One case of seropositive to equine infectious anemia may fully justify systematic and regular testings for the whole population of horse in Korea. 4. It is manifested that equine rhinopneumonitis, Japanese encephalitis and Getah virus infection are well established in Korea, together with the presence of equine infectious abortion(Salmonella abortus equi). This strongly entails preventive precautions before entry into Korea for the horses participating in the 1986 Asian Games and the 1988 Seoul Olympics.
