        H9c2 심근 세포주에서 외인성 nitric oxide가 허혈에 의한 세포 독성에 미치는 영향

        Background: Nitric oxide(NO) is known to have protective effects on an ischemic heart and to exert triggering effects on ischemic preconditioning. However, the effects of NO during the ischemic period have not been investigated. To investigate the role of exogenous nitric oxide in a model of ischemic heart cell death, we studied the effects of ischemic preconditioning and ischemia in a normal and an ischemic buffer. Methods: Rat cardiac myoblast cells(H9c2) were cultured in a normal and an ischemic buffered medium. For the ischemic culture of heart cells, the cells were cultured in a dessicator with GasPak for 5 hrs. In ischemic preconditioning, the cells were pretreated with ischemic buffer for 5 min and then perfused with normal medium for 30 min. For the measurement of the cytotoxicity, a MTT(3-4-Sdimethylthiazol-2-yl-2,5-diphenyl tetrazolium bromide) assay was performed. A DAPI(4',6-diamidino-2-phenylindole dihydrochloride) staining procedure and a flow cytometry analysis were performed to confirm apoptotic cell death by ischemia. Results: Cell viability, as determined by using a MTT assay, showed that the preconditioned group treated with NO showed more cell death than with the not-preconditioned groups in both normal and ischemic buffers. But, In normal medium and not-preconditioned groups, NO showed protective effect according to the concentrations(100,1000μM) . No treatment with NO produced the different results. In normal medium, the protective effect of ischemic preconditioning was demonstrated, but no protective effect of ischemic preconditioning could be seen in the case of the ischemic buffer. The DAPI staining and flow cytometry analysis of heart cells showed characteristic apoptotic features. Conclusion: NO added in the ischemic phase had deterious effects on heart cells. Ischemic preconditioning was more harmful than ischemia alone. The toxicity of the cells was characteristic apoptosis.

        In this paper, the composition of lead glass was studied to lower the melting temperature. The composition of lead glass was silica, lead monoxide, sodium hydroxide, and other metal oxides which were blended in alkaline aqueous solution. The blend was dried and melted to produce transparent glass material. Using the composition suggested in this paper, it was possible to manufacture transparent lead glass easily at 700~900 ℃. And the dried bled material had very low softening temperature around 200℃ which can be applied in inorganic adhesives.

        스킨스쿠버다이빙의 한국도입과 협회의 창설

        스킨스쿠버다이빙이 한국의 수권스포츠 중 수중분야로서 도입 된 것은 현재 약 반세기에 근접한 자취를 보이고 있고, 아직 수중스포츠 분야의 연혁에 관련된 선행연구가 없는 관계로 그 이력을 사료(史料)를 통해 상고(詳考)하여 한국체육사 분야의 기초자료로 제공할 필요성이 있다. 따라서 본 연구의 목적은 스킨스쿠버다이빙의 한국 도입 초기 상황에 대하여 고증하는 것이다. 이 목적의 실현을 위한 연구방법으로 신문, 잡지, 단행본 등을 이용한 문헌고찰을 하며, 이것에 따른 결과는 다음과 같다. 첫째, 수중스포츠 단체 창설 이전의 상황에서 수중활동의 시작은 군사적 목적으로 이루어졌다. 둘째, 수중스포츠 단체 창설 이후의 상황에서는 초대 단체장으로 약 18년간 재직한 김상겸의 노력과 총 네번의 단체의 명칭 변화 과정이 있었다. 셋째, 단체창설 후 활동에서는 그것의 확산을 위해 강습회 및 대회를 이용하였음을 알 수 있었다.       • 녹동균 세포외막 단백질 백신 CFC-1-101의 안정성 및 면역원성 검토 : 임상 제 Ⅰ/Ⅱa상 시험

        목적 : 제일제당에서는 녹농균의 세포외막 단밸질을 유효성분으로 하는 백신인 CFC-101을 개발하였으며, 동물시험에서 이 백신의 안전성과 유효성을 입증하였다. 본 연구에서는 이 녹농균 백신의 인체에 대한 안전성과 면역원성을 평가하는 동시에 인체 접종시의 최적 투여 용량을 결정하기 위하여 제 I/Ⅱa상 임상시험을 수행하였다. 방법 : 건강한 성인 남자를 피험자로 선별하여 각 용량군에 백신투여자 6명, 위약투여자 2명을 배정하였다. 백신 투여군은 0.25mg, 0.5mg 또는 1.0mg 용량의 녹농균 백신을 7일 간격으로 3회에 걸쳐 근육주사 하였으며, 위약 투여군에게는 세포외막 단백질을 제외한 동일한 성분을 투여하였다. 백신접종 후 국소적 또는 전신적인 반응의 발생여부를 관찰하고, 혈액시료를 체취하여 백신의 역가와 유효성을 검정하였다. 결과 : 녹농균 백신 CFC-101은 모든 접종자에서 양호한 내약성을 보였다. 또한 0.5mg 과 1.0mg 백신 투여군에서는 100%의 항체양전율을 나타내었다. 생성된 항체는 녹농균 세포외막단백질에 특이성을 보였고, 녹농균 감염에 대해 방어효능이 있었다. 결론 : 이와같은 결과로부터 이 녹농균 백신은 인체에 안전하게 투여할 수 있으며, 높은 항체 생성능으로 감염방어 효능을 보이고 0.5mg과 1.0mg이 최적용량인 것으로 판단되었다. Background : We developed a Pseudomonas aeruginosa outer membrane protein(OMP) vaccine CFC-101, and the prophylactic efficacy of which has been demonstrated in animal models. In order to evaluate the safety and immunogenicity of the P. aeruginosa vaccine, we carried out a phase I/Ⅱa clinical trial in healthy male volunteers. Methods : Groups of eight volunteers, including two placebo subjects, were vaccinated intramuscularly with three doses of 0.25, 0.5 or 1.0 mg of the vaccine at one week intervals. Sings of systemic and local reactions observed after vaccination were recorded for each vaccinee for 5 days. Physical examinations were performed on days 0, 1, 7, 8, 14, 15, 21, and 42, and clinical laboratory tests were done on days 0, 3, and 21. Blood samples for assay of serum antibody levels were obtained up to 42 days after the first vaccination. Results : The vaccine was generally well tolerated by all vaccinees, showing no significant side effects. In the three dosage groups, all vaccinees, except one receiving the 0.25 mg dose, showed significant elevation in serum IgG antibody titers against the vaccine proteins, indicating 100% seroconversion in 0.5 and 1.0 mg groups. The human antibodies induced by the vaccine were specific for P. aeruginosa OMPs, as confirmed by western blot analysis and immunoprecipitation assays. The capacity of the human antisera to enhance opsonophagocytic killing activity by polymorphonuclear leukocytes and to confer protection against P. aeruginosa infections indicates that the antibodies elicited by the vaccine have protective efficacy. Conclusion : We conclude that the P. aeruginosa OMP vaccine is safe and effective for human use and its optimal dose to be 0.5 or 1.0 mg.

        리처드 크래쇼(Richard Crashaw)는 죤 던(John Donne)을 기점으로 하는 형이상학파 시인들(Metaphysical Poets)중의 한 사람이면서도, 또한 앵글로 색슨 시대 이래로 영문학 사조 안에 끊이지 않고 내려오는 신비주의(mysticism)를 계승하고 있는 종교시인이다. 그런데 얼핏 서로 이율배반적인 것처럼 보이는 이 두 가지 요소-과학적이고 철학적인 논리를 바탕으로 하는 형이상학파 시의 특성과 지극히 주관적이고 비논리적인 개인의 체험을 위주로 하는 신비주의적 특성-는 크래쇼의 시세계가 갖는 독창성을 아주 잘 나타내주고 있다. 크래쇼는 형이상학파의 시가 갖는 기교적인 특징을 사용하여 자신의 신앙심을 통하여 보는 신의 모습을 누구보다도 선명하게 그려낸 시인이기 때문이다. 여기서 그가 신의 모습을 선명하게 그려냈다고 하는 말은 그가 신의 모습을 누구보다도 더 정확하게 그려냈다는 말이 아니라, 그가 다른 어떤 시인보다도 직관적이고 감각적으로 자신의 신을 이해했다는 뜻이다.

      • Diphenylhydantoin이 배양중인 치은 섬유아세포에 미치는 영향

        The purpose of this study was to evaluate the effect of diphenylhydantion on humna gingival fibroblasts in culture. Cell proliferation assay was performed to determine the effect of PHT on the DNA synthesis rate of human gingival fibroblasts by means of [^3H]-thymidine incorporation method. Protein synthesis assay was designed to detect the collagen production by [^3H]-proline incorporation method. The results were as follow : 1. In case of DNA synthesis assay, the contents of DNA were significantly increased more than the control group(1 ㎍/ml : p<0.05, 5 ㎍/ml, 10 ㎍/ml : p<0.01). 2. In case of collagenase-digestible protein synthesis assay, the contents of CDP were significantly increased on PHT 5 ㎍/ml group compare to the control group(p<0.05), and PHT 1 ㎍/ml, 5 ㎍/ml group were increased but there were statistically not significant. 3. In case of noncollageneous protein synthesis assay, the contents of NCP were significantly increased on PHT 1 ㎍/ml group(p<0.05), and PHT 5 ㎍/ml, 10 ㎍/ml group were increased but there were statistically not significant. 4. The ability of collageneous-digestible protein synthesis was great increased than the cell proliferation. 5. The effects of PHT increasing the ability of collagen synthesis, were cell proliferation and increased of protein synthesis.

      • 태권도 수련생의 윤리관 정립 방안

        The purpose of this study was to improve the ethic corruptness by focusing on the fact that Taekwondo is our own traditional property as a whole, and to establish the morals and ethics of Taekwondo trainee, athlete, coach, administrator, and veteran of Taekwondo. The problems of Taekwondo ethic values lie on the absence of right guidance and education, repeated violence in the stadium, the questioning of the referee's judgements, commercialization of Taekwondo that leads to excessive materialism and 'win at all costs' phenomena, and the lost manners. These are the methods to ameliorate the above mentioned problems. First, guide and educate the ethic values of the Taekwondo trainee. Second, research a program that establishes the ethic values of the trainee. Third, breed referees with rigid ethic values. Fourth, establish ethic values through Taekwondo culture creation . Fifth, establish the origins of ethic values through Taekwondo spirit. 1) Establishment of Koryo, Chosun Dynasty's traditional spirit and ethic value. 2) Establishment of ethic value through the unique Taekwondo spirit. Sixth, establish ethic value by education for the whole man. Seventh, establish ethic value through fair play. Therefore this study was carried out to contribute to innovating culture creation of Taekwondo through upright moral values as well as ethic values, and to succeed and develope the superb Taekwondo spirit and ethic values to Taekwondo families all over the world who have dreams and hopes.

      • 성견 2급 치근이개부 병변 치료 시 이종골 이식 및 혈소판 농축 혈장의 골재생에 관한 효과

        New techniques for regenerating the destructed periodontal tissue have been studied for many years. Current acceptable methods of promoting periodontal regeneration are basis of removal of diseased soft tissue, root treatment, guided tissue regeneration, graft materials, and biological mediators. Platelet Rich plasma has been reported as a biological mediator which regulates activities of wound healing progress including cell proliferation , migration, and metabolism. The purpose of this study is to evaluate the effects of using the Platelet Rich plasma as a regeneration promoting agent for furcation involvement defect. Five adult beagle were used in this experiment. The dogs were anesthetized with Ketamin HCl (0.1 ml/kg, IV) and Xylazine hydrochloride (Rompun^ⓡ, Bayer, 0.1 ml/kg, IM) and conventional periodontal prophylaxis were performed with ultrasonic scaler and hand instruments. With inrasulcular and crestal incision, mucoperiosteal flap was elevated. Following decortication with 1/2 high speed round bur, degree Ⅱ furcation defect was made on mandibular third(P3), forth(P4) and fifth(P5) premolar, and stopping was inserted, After 4 weeks, stopping was removed, and bone graft was performed. Ca-P was grafted in P3(experimental group I), combination of Ca-P and plasma rich platelet were grafted in P4(experimental group Ⅱ), and P5 was remained at control group. Systemic antibiotics(gentamicin sulfate) and anlgesics(phenyl butazone) were administrated intramuscular for 2 weeks after surgery. Irrigation with 0.1% Chlorhexidine Gluconate around operate sites was performed during the whole experiment period except one day immediate after surgery. Soft diets were fed through the whole experiment period. After 4, 8 weeks, the animals were sacrificed by perfusion technique. Tissue block was excised including the tooth and prepared for light microscope with Gomori's trichrome staining. At 4 weeks after surgery, there were rapid osteogenesis phenomenon on the defected area of the platelet produced by activated osteoblast. Bone formation was almost completed to the formix of furcation by 8 weeks after surgery. In conclusion, Platelet Rich Plasma can promote rapid osteogenesis during healing of periodontal regeneration.
