      • 스페르민-DNA 복합체의 이례적 흡광도-온도 양상에 미치는 DMSO의 영향

        DNA의 응축을 유발시키는 spermine 농도 존재시 downward peak(trough)를 갖는 이례적 흡광도-온도 양상에 hydrophobic environment를 조성하는 DMSO의 영향을 조사하였다. DMSO 농도증가에 따라 downward peak의 깊이가 점차 얕아졌으며 그 phase transition enthalpy와 relative cooperative length(n)가 melting region보다는 downward peak(trough) phase transition에 더 예민하게 영향을 받았다. 따라서 본 data는 spermine에 의하여 유발되는 응축된 DNA 구조에 이르는 hydrophobic interaction에 의한 tertiary structure 형성이 온도상승에 따라 유리해짐으로써 downward peak가 형성된다는 추리를 확인하는 실험결과가 될 것이다. In an effort to improve our knowledge on the mechanism of the collapse of DNA into a compact structure by polyamines, we investigated the effect of dimethyl sulfoxide (DMSO), a hydrophobic environmental reagent on the anomalous absorbance-temperature profile of condensed DNA, induced by spermine and having a trough preceeded by the initiation of the melting phase. The trough peak size can be decreased by increased concentration of DMSO, and the cooperativity of the phase transition to the region in the anomalous absorbance-temperature profile is more sensitive to DMSO than that of the melting regions of the profile. The present data indicate that the anomalous absorbance-temperatuve profile is due to the thermal stabilization of the tertiary structure of the spermine-induced collapsed state of DNA and the importance of hydrophobic interaction in the stabilization of the condensed state of DNA induced by spermine.

      • 폐결핵 환자 혈청에서 수용성 Interleukin 2 수용체 및 Adenosine Deaminase 활성도에 관한 연구

        Interleukin-2 (IL-2) induces T cell proliferation in an autocrine manner and provides a means by which antigen triggered T cells can be clonally expanded in vitro. During the following activation, the activity of IL-2 is mediated by specific high affinity IL-2 binding membrane receptors which are expressed shortly after activation. In this process, a 42 KD-fragment (soluble IL-2R) is continuously cleaved off and circulates as a soluble marker of T lymphocyte activation. Elevated level of soluble IL-2R has been identified in the serum of patients with malignant autoimmune and allergic disorders, systemic parasitic infection, undergoing graft versus host disease, acute or chronic lymphocytic leukemia and HIV-infection. ADA (adenosine deaminase) completes the process of differentiation of T cell and is essential for progression of T cell maturation. Therefore level of ADA is to correlated with magnitude of T cell immune response. The fact that expression of sIL-2R and ADA activity increases in peripheral blood T-lymphocytes of patients with active pulmonary tuberculosis suggests that T cell activation might have a major role in the pathogenesis of pulmonary tuberculosis. In order to evaluate the T cell immune response in pulmonary tuberculosis, we measured the serum concentration of sIL-2R and ADA activity in 17 patients with current pulmonary tuberculosis, 10 chronic inactive pulmonary tuberculosis, 10 as normal controls. (1) Current pulmonary tuberculosis had significantly higher levels of sIL-2R (237.24±95.47)when compared with those of inactive tuberculosis (78.6±11.06). and the control (68.17±15.4) group. (2) ADA activity in current pulmonary tuberculosis was significantly increased (34.41±20.63) when compared with those of inactive tuberculosis (24.7±14.36) and control (17.65± 5.94) group. (3) There was good correlation between sIL-2R concentration and ADA activity in serum in current pulmonary tuberculosis group. (4) sIL-2R concentration and ADA activity was decreased significantly 6 months after anti-tuberculosis drug medication. In conclusion, sIL-2R concentration and ADA activity in serum in current pulmonary tuberculosis group was increased when compared with those of inactive pulmonary tuberculosis and the control group. By the way, T cell mediated immune response was enhanced in current pulmonary tuberculosis, but in inactive chronic pulmonary tuberculosis who had treated by antituberculosis drug medication, the concentration of sIL-2R and ADA activity was nearly normal.

      • QPSK 고속 변복조기의 설계 및 구현

        본 논문에서는 전 디지털 방식의 QPSK 고속 변복조기 설계방식을 제시하였고 이를 CPLD칩으로 구현한 결과를 제시하고 검토하였다. Multiplier, Pulse Shaping Filer로 구성된 변조기는 심볼당 4샘플을 취하는 Zero=IF 방식으로 설계되었으며, 비트 동기를 포착하는 STR(Symbol Timing Recovery) loop과 반송과 동기를 포착하는 CPR(Carrier Phase Recovery) loop으로 구성된 복조기는 DD-Gardner 방식과 Decision-Directed 동기화 방식을 이용하여 설계하였다. STR loop에서는 샘플 해상도를 높이기 위하여 보간기를 채용하였다. 설계한 모뎀은 Altera사의 Design Compiler를 이용하여 칩으로 합성 구현되었다. 합성결과 QPSK 변조기는 약 50Mbps, 복조기는 약 12Mbps의 전송속도까지 동작 가능하였다. 실제 ASIC으로 구현할 경우, CPLD 칩으로 구현한 속도의 6-7배 까지 고속화 할 수 있으므로 향후 고속 무선 모뎀에 적용될 수 있을 것이다. In this paper, a design method for high-speed QPSK MODEM based on all digital implementation scheme is presented, and implementation results of QPSK MODEM using a CPLD chip are examined. The modulator that consists of multiplier, Pulse Shaping Filter is designed on a basis of the Zero-IF sampling theory, i.e., 4 samples per symbol. DD(Decision Directed)-Gardner algorithm is chosen for timing error detector of STR loop and Decision-Directed algorithm is used for CPR loop design. To increase the time resolution, interpolation between received signal samples is also utilized in the desine of QPSK demodulator. The maximum speed of the QPSK modulator synthesized in FLEX10K chip by using the Design Compiler of the Altera corporation is guaranteed approximately 50Mbps and that of the QPSK demodulator is guaranteed approximately 12Mbps.

      • 의약품중 잔류유기용매 시험법 확립에 관한 연구

        Headspace-GC(딘5-GC)를 이용하여 ICH 잔류웅매 가이드라인 class 1, 2에 속하는 28종의 잔류유기웅매 분석방법을 연군한 결과 SPB-5, BB-WAX column을 동시 이용하여 각각을 확인, 정량할 수 있었다.또한 염의 참가효과, 평형시간,평형온도가 headspace분석에 미치는 영향을 검토한 결과 평형 시간 30분,평형온도 85℃,Na₂SO₄Ig을 첨가하여 시험할 때 최적 부넉조건을 나타내었다. 회수율은 1,1-achloroethene(68.3%)를 제외하고 90.9 ~ 114.5%로 패체적으로 양호하였다. 따로 HS-GCJECD, DB-524 column을 이용하여 9종의 잔류유기용매에 대하여 각각의 검랸선을 작성하고 검출한계를 구하였다. 실제 의약품 원료 10종및 제제 5종에 적용시킨 결과 모두 불검출되었으며 본 시험법은 의약품중의 잔류유기용매 분석에 적용할 수 있는 방법이라고 판단된다. The headspace-fC/FID(HS -GC/FID) method was performed for test method developnlent of residu;31 organic solvents in pfrarmaceuticals. Using SPB-S and BB-WAXcolumn, 28 kinds of solvents iri ICH residual sof)·ents guidetine class 1, 2 couBd be individuallt-identified and quantitated. The foIBowing residoal soIYents were not detected by the headspaceinjection condition : N,N-Cmethylacetamide, N,N-dimeth)rlformamide, ethyfeneglycol, formamide,2-methoryethanot, N-rnethylpyrrolidone, suBforane. The effects of the addition of salts,equilibration time, and equilibration temperature on headspace analt·sis were invesugated. Theoptimum conditions weiFe obtained with addtion of Ifa2s04 19 as a salt, simultaneously. thetime and temperature of equitibration were 30min and 8i'f, respectivety. The recovery havefound between 90.9 and 114.5% except 1,1-dichloroethene(fi8.3%). Using DB-624 column &HS-GC/:ECD method, 9 kinds of residual solvents could be individually identified andfuantitated. This HS-GC method can be applied to test the residua3 organic solvent in thepharmaceuticals.

        We have investigated the effect of hydrophobic environmental reagents (urea and DMSO) on the anomalous absorbance-temperature profile of condensed DNA, induced by spermine. The cooperativity of the phase transition to the trough region in the anomalous absorbance-temperature profile is more sensitive to the hydrophobic environmental reagents than that of the melting region, and the trough peak size can be decreased by increased concentration of the hydrophobic reagents. The present data indicate the importance of hydrophobic interaction in the stabilization of the condensed state of DNA induced by spermine, and that the anomalous absorbance-temperature profile is due to the thermal stabilization of the tertiary structure of the spermine-induced collapsed state of the DNA.

      • Pragmatic TCM의 구조분석 및 성능평가

        기존의 Ungerboeck TCM에서는 path의 병렬천이, branch metric계산방법, 그리고 trace back등의 구조가 기존의 Viterbi복호기에 적용되는 방식과 많이 다르기 때문에, 기존의 Viterbi복호기를 TCM에 그대로 적용하여 사용할 수 없다. Pragmatic TCM은 (2,1,m) Viterbi 복호기를 이용하여 수신되는 Trellis coded 8PSK신호를 복조할 수 있도록 고안된 방식이다. 본 논문에서는 Pragmatic TCM의 기본 구조를 살펴보았고, 위상 섹터의 분할에 따른 Pragmatic TCM의 성능을 분석하여 최적의 위상섹터 분할 수를 결정하였다. The customized Viterbi decoder chip cannot be used as it is in TCM applications because the decoding schemes in TCM are quite different from those in customized Viterbi decoder chip. In this paper, we examine the structure of pragmatic TCM which was devised to decode the transmitted information from trellis coded 8PSK signal with customized (2,l,m) Viterbi decoder. Also we analyze the performance of progmatic TCM as a function of the number of phase sector.

        정상 월경주기에 따른 혈액 응고 기능의 변화 : 혈전탄성묘사도를 이용한 평가

        Background: As the menstruation has been thought to be associated with abnormal hemostasis and increased fibrinolytic activities, some surgeons and even anesthesiologists tend to postpone elective surgery. Many investigators have studied individual coagulation profiles and fibrinolytic activity during the menstruation, but conventional coagulation screens are frequently inadequate for the purpose of monitoring coagulation dynamically. This study was designed to evaluate the whole dynamic hemostatic process globally during the normal menstrual cycle by the thromboelastography (TEG). Methods: Thirty six-healthy, normally menstruating female volunteers wrote a menstruation diary about their menstruation cycle. Blood samples were obtained for TEG analysis 3 days before menstruation, the 1st day of menstruation, the 2nd day of menstruation, the last day of menstruation and 5days after menstruation. TEG coagulation indices (R value, K value, α angle, WBCL160 during menstruation compared with pre- or post-menstruation period, but there was a significant decrease in the MA value on the 1st day of the menstruation. Conclusions: The result of our study showed the possibility of abnormal hemostatic function on the 1st day of menstruation. However, the more study is needed to confirm the possibility of bleeding tendency during surgery on the 1st day of menstruation. (Korean J Anesthesiol 2002; 43: 413~417)

      • RNase A에 對한 Spermine의 妨害效果로 부터 變性 DNA에 依한 救濟

        Recently we have reported that the susceptibility of denatured calf thymus DNA to DNase 1 is suppressed by spermine, while that of the native DNA is activated by spermine. We have also noted that the susceptibility of RNA to RNase A is suppressed by spermine, by being subjected to form inermolecular aggreates. In the present work, we find that the inhibitory effect of spermine on the susceptibility of RNA to RNase A can be relieved by the denatured DNA, but not by the native DNA. The possible explanation, based on our data, is that the relieving effec of the denatured DNA is brought about by RNA-DNA (denatured) interactions induced by spermine, thereby decreasing the aggregation of RNA molecules.

      • 충북 지역 초등학교 과학교육의 실태 조사

        A survey was carried out comprehensively to investigate the present status of science education in primary schools 496 teachers in 199 primary schools in Chungbuk area participate in the survey. They questionnaire used in this study included items regarding goals or objectives for science education, approaches for instruction, teaching-learning strategies, assessment for instruction, teaching-learning strategies, assessment for instruction, computer use in science classes students' activity and the status of experimental equipment or tools for science education. Items on science textbooks and the understanding of the 7th national curriculum were also included in it. The results based on the teachers' responses were summarized and provided in this paper and it showed practices of science education in Chungbuk area.
