        Previous studies have shown that Magnoliae cortex and Ginkgo biloba extracts were showed on the antimicrobial and anti-inflammatory action, in vitro. The purpose of this study was to evaluate on the effect of antimicrobial and anti-inflammatory activity of Magnoliae cortex and Ginkgo biloba extracts containing dentifrice in gingivitis. 70 subjects with gingivitis were divided into an experimental group which performed normal oral hygiene procedure with Magnoliae cortex and Ginkgo biloba extracts containing dentifrice and a control group which also performed normal oral hygiene procedure with the same dentifrice without the natural extracts and completed a double-blind, cross-over study. At baseline and 3 weeks, subjects were assayed for clinical study by plaque index, gingival index, pocket depth, GCF rate, and microbiological study by subgingival dental plaque bacterial morphotypes by phase contrast microscopy, total anaerobes, total aerobes, Black pigmented bacteroides, A-actionomycetemcomitans, A.viscosus, C.rectus, S.sanguis, P.gingivalis, P.intermedia by bacterial culture and immunofluorescence microscopy. After 3 weeks using their respective dentifrices, reductions in the clinical indices of subjects were similar between the experimental dentifrice group and a control dentifrice group except for statistically significant much reductions in PI, GI, and GCF rate in the experimental dentifrice group as compared to control dentifrice group. Also statistically significant reductions in the motile rods and Spirochetes were found in both experimental group to compare with control group, however statistically much reduction in total anaerobes, Black pigmented bacteroides, and P.gingivalis, P.intermedia were found in the experimental dentifrice group as compared to control dentifrice group. This results indicates that Magnoliae cortex and Ginkgo biloba extracts containing dentifrice might be useful for elimination of gingival inflammation.

        Three-dimensional CFD analysis of hydrogen-air-steam explosions in APR1400 containment

        <P><B>Abstract</B></P> <P>The present study concerns the three-dimensional CFD analysis of hydrogen explosions in APR1400 containment. Initial conditions for combustion simulations are obtained by a cost-effective coupling with a lumped parameter code for preceding mixture distribution analysis. Three severe accident scenarios are examined: Small-Break Loss-Of-Coolant Accident (SB-LOCA) and Station Black-Out (SBO) with activated as well as deactivated three-way valve. Only in the last case, thermal ignition of the hydrogen-air-steam mixture could be triggered. Flame propagation in the containment and particularly in the In-containment Refueling Water Storage Tank (IRWST) is investigated in detail. Additional generic scenarios demonstrate the methods ability to reproduce strong Flame Acceleration (FA) and even the hazardous Deflagration-to-Detonation Transition (DDT).</P> <P><B>Highlights</B></P> <P> <UL> <LI> Full-scale 3D CFD analysis of hydrogen explosions in APR1400 containment is presented. </LI> <LI> A new cost-effective lumped parameter code to CFD coupling strategy is proposed. </LI> <LI> In the Station Black-Out accident, the three-way valve is a major safety improvement. </LI> <LI> Most critical conditions are found in the In-containment Refueling Water Storage Tank. </LI> </UL> </P>

        치은섬유아세포와 치은인대세포의 형태와 화학주성에 미치는 대조추출물의 효과에 관한 연구

        The most important object of periodontal treatment is the perfect regeneration of destructed periodontal tissue. The healing of periodontal lesion is affected by several cells & factors, which result in formation of long juntional epithelium, root resorption, bony ankylosis or connective tissue attachment. And ideal healing is enhanced by epithilial exclusion or periodontal ligament cell activation. In this investigation, I studied the effect of Zizyphus Fructus extract which enhances biologic activity& collagen synthesis, on the chemotaxis & cell nature. The cells were obtained from interdental area & middle third area of the freshly extracted teeth for the orthodontic purpose. And they were fully incubated in${\alpha}-MEM$ solution containing $100{\mu]g/ml$ penicillin & $100{\mu]g/ml$ streptomycin followed by 6 generation incubation. The test cells were collected by trypsin-EDTA & centrifuge in the fully incubated cells, counted by Hernacyotmeter, incbated $5{\times}10^5/ml$ cells for 24 hours, re-incubated 24 hours in media containing natural extract and photographed. The cells were incubated for 4 hours in 48 well microchemotaxis chamber bisecting upper & lower chamber by 8ug/m pore polycarbonate membrane coating 5mg/ml gelatin solution. The migrated cells in microscope were counted, which meaned cell chemotaxis activity. The study had shown that the morphology of cell was spindle-shaped as the control group, and the subextract test groups were not significantly different. In gingival fibroblasts, the chemotaxis effect of PDGF was statistically significant compared to control group. The Zizyphus Fructus extract was more or less enhanced chemotaxis effect and in $1{\mu}g/ml$ concentration the chemotaxis effect was slightly elevated compared with $10{\mu}g/ml$ concentration. But, among the subextracts, it was not significantly defferent. In PDL cells, the chemotaxis effect of PDGF in statistically significant, and the zizyphus Fructus extract had shown the enhanced effect. The effect was slightly higher in $1{\mu}g/ml$ concentration than 10g/ml concentration,and no significance among the subextracts.

      • 頭鍼이 中風患者의 단기간 NIHSS와 MBI에 미치는 영향

        Objective ; The aim of this study was to investigate the effect of Scalp acupuncture on recovery of motor disorders in stroke patients. Methods ; Twenty two patients with poststroke-hemiplegia were randomized into two groups. Ten patients(test group) treated by 2 methods-Scalp acupuncture and body acupuncture. The other twelve patients(control group) treated only by body acupuncture. The activity of daily living was measured with a National Institutes of Health stroke scale(NIHSS) and Modified Barthel Index(MBI). The therapy was performed one a day for 2 weeks. Results ; In terms of score of NIHSS, the test group showed statistically meaningful decrease after 2 week treatment, but the control group showed statistically meaningful decrease after 1 week(p<0.05). And in terms of score of MBI, the test group did not show statistically meaningful increase but the control group showed statistically meaningful increase after 1 week(p<O.05). There was no statistically meaningful difference after 1 and 2 week treatment between the groups. Conclusions ; These results support that test group has almost same effectness compared with control group in improvement of the activity of daily living of poststroke-hemiplegic patients.

        델파이 설문조사를 통한 토마토 재배시설 평가지표 개발

        이 연구의 목적은 토마토 재배시설을 종합적으로 평가할 수 있는 지표를 개발하는데 있다. 이를 달성하기 위해 관련 문헌 및 선행 연구들을 분석하여 예비 평가항목을 추출하고, 전문가 집단을 대상으로 델파이 설문조사를 실시하여 평가 항목, 등급 및 기준으로 구성된 평가지표를 개발하였다. 토마토 재배 및 시설 관련 교수, 연구원, 농업인(현장 사용자) 등으로 구분하여 100명의 전문가 패널을 선정하였으며, 1차부터 3차까지 폐쇄형 질문 방식의 설문조사를 진행하였다. 그 결과 최종적으로 확정된 평가 항목은 4개 대분류 영역과 39개 세부 평가항목으로 이루어졌다. 대분류 영역은 시설 구조적 요소, 시설 설비적 요소, 재배적 요소, 생산기반적 요소이며, 요소별로 각각 9개, 15개, 7개, 8개씩 세부 평가항목으로 구성되었다. 또한 전문가 의견을 수렴하여 39개 평가항목별로 가중치를 산정하고, 등급 및 기준을 설정하였다. 이 연구에서 개발된 평가지표는 새로운 시설 모델 개발, 사용 중인 시설에서 우선적으로 보완해야 될 사항을 판단하거나 정부 지원사업의 효율성을 높이는데 중요한 역할을 할 것이다. This study aimed to develop the comprehensive indicators which can be used for evaluating greenhouse for tomato cultivation. To achieve this aim, the study developed the evaluation indicators composed of evaluation items, grades and criteria by extracting preliminary evaluation items through analyzing the related papers and preceding studies, and conducting Delphi survey on an expert group. During the three surveys, the questions of closed-ended type were given to a panel of 100 experts - professors related to tomato cultivation and facilities, researchers and farmers (practical users). As a result, the finally established evaluation indicators consist of 4 categories and 39 specific evaluation items. The 4 categories are the structural factor of greenhouse, equipment factor of greenhouse, cultivation factor, and infrastructure factor. These factors consist of specific evaluation items of 9, 15, 7 and 8, respectively. In addition, on 39 specific evaluation items, weighted values were calculated and grades and criteria were established by collecting opinions of the experts. The newly developed evaluation indicators through this study will play an important role in developing new greenhouse models, considering things that should be complemented preferentially regarding in-use facilities, and improving the efficiency of projects supported by the government.

        상온 유동층에서 층내 수평전열관의 마모특성

        대부분의 유동층 연소장치에서 충물질과 비산된 입자에 의한 충내 전열관과 water wall의 전열관 표면의 마모는 계속적으로 골칫거리가 되고 있다. 본 연구는 단면적이 0.15m×0.30m인 상온유동층장치에서 마모하기 쉬운 아크릴 관을 사용하여 유동층에 파묻힌 전열관에 대하여 유동조건과 전열관의 배열효과에 대한 실험적인 연구를 나타냈다. 유동층의 단면적은 0.15m×0.30m이고 높이는 1.0m이다. 층물질은 강모래 평균입도 0.73, 1.24mm의 두 가지와, 평균입도 0.9mm의 무연탄 희재를 사용하였다. 유동층 높이는 0.45m이었다. 마모실험은 조건마다 100시간동안 유동화속도 1.2-1.8m/s 까지 변화시켜 수행하였다. 실험결과 마모율은 유동화속도가 증가할수록 증가하였고, 공기분배기로부터 떨어진 거리가 증가할수록 증가하였다. 그리고 공기분배기로부터 높이 0.6m의 비산영역에 위치한 전열관에 심한 마모를 나타냈다. 전열관의 원주방향에 대한 마모경향은 다른 부분과 비교하여 시계방향으로 4시 방향에서 8시 방향까지 심한 마모가 나타났고, 특히 5시 방향과 7시 방향은 심한 마모를 보였다. Erosion of in-bed tubes and water wall heat transfer surfaces by bed materials and elutriated particles have persistently plagued most fluidized bed combustion (FBC) systems. This papere presents a systematic experimental study of the effects of tube arrangement and flow condition on embedded tube erosion by using erosion-prone acrylic cylinders in a 0.15m×0.30m bench-scale cold fluidized bed. The bed material was two different sizes of river sand and anthracite ash with the average diameter of 0.73, 1.24, 0.90㎜, respectively. The static bed height was 0.45m. Erosion test was performed with the variation of fluidization velocity of 1.2 to 1.8m/s for 100 hours per set. The result shows that the erosion rate increased with fluidizing velocity, with distance from the distributor. The erosion was severe at the tube located in the splash zone of 60㎝ above the distributor. The trend with radial direction shows severe erosion at the 4-8 o'clock clockwise compare to other area, especially 5 and 7 o'clock clockwise.

        Background: PPL photography would be a quantitative assessment method for the examination of the properties of human skin. In our previous study, the quantitative difference of the light reflected from skin surface could have been analyzed by PPL photography. Objectives: So, we designed this study to improve the technique using various LEDs of different colors. In addition to PPL, in-house skin conductance meter was used to measure the skin hydration for comparison. Methods: A total of 40 healthy subjects were participated in this study. PPL imaging was taken on mid-cheek, volar aspect of forearm and anterior shin, using white and green LED illuminators. In order to assess skin conductance value, we designed our own probe. Results: First of all, as for glossiness on extremities, the L* value was positively correlated(r=0.33417), a* and b* value were correlated negatively(r= -0.32046, -0.33128) under the green illuminator. Skin conductance value was positively correlated with glossiness(r=0.65258). For dryness of extremities, the b* under the green illuminator was positively correlated(r=0.23339) and negatively with skin conductance (r= -0.80447). Skin conductance value itself showed correlations with CIELAB coordinate L* and a* from green light(r=0.24305, -0.22102). Conclusion: In conclusion, analytic PPL image technique combined with LED would be an efficient way to evaluate the properties of human skin and the results implicated the validity of our in-house conductance meter.

        국내 하천 퇴적물 건강성평가를 위한 Microtox 독성시험 조건확립 연구

        1. 하천 퇴적물 공극수, 증류수 추출용액 및 유기용매 추출액을 pH6.0~6.5 범위로 조절한 후 Microtox 독성을 측정하는 것이 퇴적물내의 암모니아 독성이 배제된, 유해화학물질 오염에 의한 퇴적물 독성을 평가하는 방안임을 확인하였다. 2. 퇴적물에 대해 공극수, 증류수 추출물, 유기용매 추출물 구분하여 Microtox 독성을 구하는 것이 퇴적물 내 유해화학물질의 독성발현 특성을 예측할 수 있는 기법이라고 사료된다. 3. 조사정점 내 시료 채취 지점간 독성에 편차로 인하여, 동일 정점의 반경 50~100m 내에서 최소 4지점 이상의 퇴적물을 확보하는 것이 해당 정점의 독성 대표값을 산출할 수 있는 방안이 된다고 판단된다. 4. 한강수계인 남한강 지천인 복하천과 양화천, 금강수계인 청주 미호천, 대전 갑천, 낙동강수계인 대구 금호강, 영산강수계인 황룡강 그리고 익산 만경강을 대상으로 한 예비 조사한 결과, 집중연구대상 하천으로의 조건은 금호강이 가장 우수하였다. 5. 하천 퇴적물의 증류수 추출액과 유기용매 추출액의 Microtox 독성값 간의 상호 상관성이 높았다. 그러나, 농축 과정이 용이하므로 정점간 비교를 위한 독성 측정시 유기용매 추출액을 사용하는 것이 바람직하다고 사료되었다. Six rivers were selected as preliminary screening sites to determine the test conditions of Microtox in assessing the toxicity of the sediment. In addition, a pH range of 6.0--6.5 was established in testing pore water, aqueous extracts and organic extracts. Each extractable fraction of sediment showed different toxicities. Therefore, in order to properly examine the toxicity in the sediment, all extractable fractions of sediment samples needed to be tested with Microtox. Thus, sediment samples were additionally collected from at least 4 secondary sites within 50~100 m area of the primary sampling site to reduce any variation or deviation in toxicity assessment. From all sediment toxicity data that was collected from this study, it was concluded that the Keumho river was the most polluted with the highest sediment toxicity of all the rivers analyzed and needed further detailed research on its pollution problem.

        셀레콕시브 및 그 합성유도체들의 항암활성 스크리닝

        Selective COX (cyclooxygenase)-2 inhibitors including celecoxib have been shown to induce apoptosis and cell cycle changes in various tumor cells. New inhibitors are recently being developed as chemomodulating agents. We evaluated celecoxib and screened 150 synthetic compounds for anti-proliferative activities in vitro. Effects of celecoxib on COX activity, cell growth, cell cycle distribution, and apoptosis induction were determined in A549 COX-2 overexpressing human non-small cell lung cancer (NSCLC) cells. The COX inhibition of celecoxib increased with concentration up to 82% at 1μM after 24 hr exposure. Forty μM and 50μM of celecoxib induce G_1 arrest, and TUNEL-positive apoptotic cells, respectively. Among 150 compounds, several compounds were selected for having greater COX-2 inhibitory activity and higher selectivity than celecoxib with growth inhibitory activity. Celecoxib showed concentration-dependent COX inhibitory activity, and ability to induce cell cycle arrest and apoptosis in human NSCLC cells in vitro. Among synthetic analogues screened, several compounds showed promising in vitro activity as COX-2 inhibitory anticancer agents, which warrant further evaluation in vitro and in vivo.

        Quantitative Proteomic Analysis Identifies AHNAK (Neuroblast Differentiation-associated Protein AHNAK) as a Novel Candidate Biomarker for Bladder Urothelial Carcinoma Diagnosis by Liquid-based Cytology

        <P>Cytological examination of urine is the most widely used noninvasive pathologic screen for bladder urothelial carcinoma (BLCA); however, inadequate diagnostic accuracy remains a major challenge. We performed mass spectrometry-based proteomic analysis of urine samples of ten patients with BLCA and ten paired patients with benign urothelial lesion (BUL) to identify ancillary proteomic markers for use in liquid-based cytology (LBC). A total of 4,839 proteins were identified and 112 proteins were confirmed as expressed at significantly different levels between the two groups. We also performed an independent proteomic profiling of tumor tissue samples where we identified 7,916 proteins of which 758 were differentially expressed. Cross-platform comparisons of these data with comparative mRNA expression profiles from The Cancer Genome Atlas identified four putative candidate proteins, AHNAK, EPPK1, MYH14 and OLFM4. To determine their immunocytochemical expression levels in LBC, we examined protein expression data from The Human Protein Atlas and in-house FFPE samples. We further investigated the expression of the four candidate proteins in urine cytology samples from two independent validation cohorts. These analyses revealed AHNAK as a unique intracellular protein differing in immunohistochemical expression and subcellular localization between tumor and non-tumor cells. In conclusion, this study identified a new biomarker, AHNAK, applicable to discrimination between BLCA and BUL by LBC. To our knowledge, the present study provides the first identification of a clinical biomarker for LBC based on in-depth proteomics.</P>
