      • Atropa belladonna 모상근의 생장량과 tropane alkaloid 생산에 관한 연구

        Agrobacterium rhizogenes strain R1000에 의해 유도된 Atropa belladonna 모상근을 배지별, NAA, IBA, IAA, 2,4-D를 농도별(0.01-100 mg/l)로 첨가한 NN액체배지에서 생장량과 tropane alkaloid 함량을 HPLC로 이용하여 정량하였다. 모상근 배양 30일째의 각 auxin의 농도별에 따른 생장율은 고농도보다 저농도일때 대체적으로 양호하였으나, 0.1mg/1 2,4-D농도에서는 callus화 하였고 10mg/l 2,4-D, 1mg/l IBA일때 2.96g, 2.36g으로 대조구보다 2.3배의 생장량을 보였다. 농도별에 따른 Tropane alkaloid 함량은 atropine이 대조구 0.141%, 원식물체가 0.059%로 나타났으며, 0.01mg/l NAA와 0.1mg/l IAA에서 배양한 모상근이 각각 1.378%, 1.814%의 생산을 보여 대조구와 원식물체보다 9-30배의 생산을 보였다. 또한 scopolamine 함량은 대조구와 원식물체는 각각 0.109%. 0.050%에 비해 0.01mg/l NAA에서 배양한 모상근에서 0.505%로 대조구 원식물체보다 4-10배의 생산량을 보였다. The study aimed to measure growth rate of Atropa belladonna hairy root cultured under medium, various auxin(NAA, IBA, IAA, 2.4-D) and auxin concentration(0.01-100mg/l) and investigated tropane alkaloid content using HPLC. The growth rate of hairy root cultured for 30 days in MS and NN liquid medium were higher in NN medium as 3.51g than MS medium, growth rate of belladonna hairy root cultured in NN medium added various auxin and auxin concentration was the most effective in low concentration than high concentration, especially, was best in the medium supplemented with 0.01mg/l 2,4-D. Atropine content of hairy root cultured in 0.01mg/l and 0.1mg/l IAA produced 9-30 times than control and normal plant. scopolamine measured each 0.109%, 0.050% in control and normal plant, hairy root cultured in 0.01mg/l NAA produced 4-10 times than control and normal plant.

      • 차나무 callus 의 catechin류 함량분석

        본 연구는 차나무의 신초를 이용하여 기내에서 여러가지 배양조건에 따른 캘러스의 catechin류 함량을 분석하여 생산성이 높은 배양조건을 찾고자 실시하였다. 신초배양시 캘러스는 0.5mg/L NAA와 1mg/L BA 조합 처리구에서 가장 잘 유도되었다. 이 조건에서 유도된 캘러스를 가지고 thiamin-HCI, PVP, taxifolin과 quercetin을 농도별로 처리하여 4주간 배양하여 생장 및 catechin류 함량을 조사하였다. 캘러스의 생장은 30mg/L thiamin-HCL, 200mg/L PVP, 1 mM taxifolin, 0.01 mM quercetin처리시 대조구에 비해 2~3배의 생장를 보였다. 차나무 잎의 주성분인 catechin류를 분석하고자 신초와 여러조건에서 배양된 캘러스를 가지고 분석한 결과를 보면, 캘러스에서 높은 함량을 보였다. 또한 신초에서의 catechin류는 EGC, EGCG, ECG등은 검출되었으나, EC는 검출되지 않았다. 그러나 배양된 캘러스에서는 4가지 성분이 모두 검출되었다. 각각의 성분들의 최적 배양조건을 보면, EGC는 500mg/L PVP, EC는 100mM quercetin, EGCG는 10 mg/L thiamin-HCI, ECG는 10mM taxifolin처리구에서 가장 높은 함량을 보였다. This study aimed to measure the growth of callus cultured under MS medium with thiamin-HCI as a vitamine. PVP as the antioxidant. taxifolin and quercetin as precursors of catechins and to select the clone producing catechins highly. Callus induction from Thea sinensis young leaf was great in the MS medium with 0.5mg/L NAA and 1 mg/L BA. When components of callus were compared with those of young leaf. Four kinds of catechins(EC, EGC, EGCG, ECG) were detected in cultured callus whereas EC not in young leaf. Catechins were detected most in the medium with 10mg/L thiamin-HCI. Expecially, EC was most as a 6.39mg/g. The content of catechins in PVP treatment was not different each other but was higher in 100 mg/L and 200 mg/L PVP than in control. When the callus was cultured in the medium with taxifolin and quercetin the caffeine content was lower than in the medium with not them. Also, the content of catechins in taxifolin and quercetin treatment was not different each other but it was highest in 10mM taxifolin and 100mM quercetin treatment.

        방사선 조사 건고사리의 수화 복원성, 색상 및 조직감 특성

        The gamma irradiation effect of dried fernbraken was investigated on the physical and sensory qualities during and after soaking process. Dried fernbraken packaged in vacuum was irradiated up to 7 kGy with γ-ray produced by 132 Cs at 0℃. Moisture content and rehydration rate of gamma-irradiated fernbraken after soaking increased significantly depending on the irradiation doses(p<0.05). Lightness(L), redness(a) and yellowness(b) values of Hunter color system increased significantly in the gamma-irradiated fernbraken after soaking, compared with control, while the hardness and chewiness by the texture analyser decreased significantly in irradiated ones. Sensory evaluation results showed that the mean scores of flavor, appearance, taste, texture and over-all acceptability in soaked fernbraken irradiated were not significantly different from those in control, except higher preference score of texture in irradiated ones. From these results, it might be concluded that the irradiation does not affect the physical and sensory qualities of dried fembraken during and after soaking, especially in texture.

      • 패류중 중금속함량에 관한 연구

        본 연구는 패류중 중금속함량 측정을 위한 분석효율이 높고 간편한 분해법을 찾아내어 그 방법에 의끓 유종중인 패류중 중금속함량을 파악하고자 수행하였다. 시료를 분해시키는 Microwave와 습식, 건식회법을 비교한 결과 변형습식회화르가 가장 적합하였다. 변형습식회차딘는 려린0,3+C104+H2SOf의 혼찰산을 쪼 · j 1 (v/v)의 비율로 혼합하여 사용하였 을 때 회수을 Pb린틴. Cd 97.8, Cu 94.2, Zn 92.8%를 나타내었근 분해시간은 2~2.5시간 소요되었다. 유통중인 패류중 중금속함량 」최소~최대(평균), mgAgl 은 Pb 0.028~o.528(0.153), Cd 0.OO7~1,832(0.587), Cu 0.360~37.375(4.6691, Zn 6.317~233.2iO(38.881), Hg 0.001 ~O.151(0.012)mg/ltg으로 나타났다. 또한 유통중인 5종의 패류중 중금속평균항량이 생산지 하재지역에서 구입한 패류에 비해 모든 중금속언써 약간 높은 함량을 보였으나 오염된 수준이 아닌 자연함유량수준이었다. This study was conducted to improve the digestion method for the determination of heavy metals in shellfishes and to estimate the contents of heavy metal in commercial shellfishes by their digestion method. This digestion method was compared microwave with wet and dry ash digestion for shellfish samples, obtained results showed that the modified wet ash Ⅱ digestion was convenient and easy to use. For the modified wet ash Ⅱ digestion, The mixed acid of HNO₃+HClO₄+H₂SO₄(25:5:1, v/v) was used. Under this condition, Recoveries were Pb 94.3, Cd 97.8, Cu 94.2 and Zn 92.8% and digestion time was ∼2.5 hours. The contents of heavy metal [minimum∼maximum (mean), mg/kg in commercial shellfishes were as follows ; Pb:0.028∼0.528(0.153), Cd:0.007∼1.832(0.587), Cu:0.360∼37.375(4.669), Zn:6.317∼233.250(38.881), Hg:0.001∼0.151(0.013). The contents of heavy metal in commercial shellfishes of five species were slightly greater than those of shellfishes purchased at Hajae. There was no great difference between the contents of heavy metal of Hajae and those of shellfishes purchased at Whole market.

      • 유통 방기의 화학적 분석

        방기는 대한약전 및 중국약전에 식물학적 및 화학적으로 다른 식물이 동일한 이름으로 수재되어 있어서 운통 및 사용에 혼란을 초래하고 있다. 현재 우리나라에서 유통되고 있는 방기는 패부분 중국 및 베트남에서 수입되고 있으며 방기 외에도 중국산 시장품인 분방기, 광방기가 다수 유통되고 았는 것으로 알려져 있다. 특히' 광방기는 함유성분인 ariStOBDChiC acid의 광범위한 발암성, 신독성이 알려지면서 각 국에서 사용을 금지하고 있으나 우리 나라예서는 유통 현황에 대한 실태조차 파악되지 않고 일는 실정이다. 따라서 방기에 대한 정확한 감별법과 품질기준을 마련하고 이에 따라 aristolochic acid를 함유하고 있는 광방기에 대하여는 수입 및 유통을 금지시킬 필요가 있다. 현재 방기는 외형, 현미경 검사 및 알카로이드 확인시험법으로 감별하게 되어 있으나 다양한 유사품의 정확한 식별을 왜해서는 알카로 이드 타입 및 함량 분포에 러한 연구에 기초한 화학적 감별의 필요성이 요구되고 있다. 이에 본 딴군에서는 방기류에 포함되꺼 있는 주요 알카로이드를 대상으로 확인 및 정량할 수 있는 동시분석법을 확립하였으며, 국내 유통방기 딘움목에 대하여 시험한 결과 유통품의 72.3%에서 독성물질인 aristolochic acid가 검출되어 rlris,!of ocho 속 광방기로 추정되었으며, a7.7%도 방기, 분방기, 목방기와는 다른 방기류 생약이 아닌 강츠로 추정되었다. 광방기 중의 aristolochic acid 함량은 평균 0.31 mg/g으로 측정되었다. 따라서, 유통방기의 대부분이. 약전품 방기가 아니어서 방기의 수입검사와 품질관레, 약효에 심각한 문제점이 있음이 딱혀졌다. 그러므로 광방기 및 유사품의 수입 및 사용을 금지시키고 방기류 생약에 대하여 화학적 성룬검사에 의한 엄격한 수입검사 기준을 설정하여 관리, 검사할 필요가 있다. ln Korean PhaγmaEopia(KP), Sinomeni stem is recorded to he originated from 51'norleriufl ucu#url(fvrenispennaceae 1. These results demonstrate that the present status of circulated Sinomef stem hasserious problem. When the amount of aristolechic acids were determined by HPLC, the contentwas quite high in the range of 0.i2-1.6smg/g of dry plants. Therefore, it suggests thatAristolochia fangi should be proHbited by chemical analysis method due to the toxic effectattributed to the toxic effect of aristolochic acids.

      • 농산물중의 잔류농약 실태에 관한 연구(Ⅰ) : 고체상 추출에 의한 농산물중 잔류농약 분석에 관한 연구 Analysis of Residual Pesticides in Agricultural Products by Solid-Phase Extraction

        신속 · 정밀하고 효율적인 짠류농약 검출을 위하여 극싱칼럼 (Extrelut-3')을 사옹한 SPE법을 시도하였다. 시료는 채소류체 사용하는 농약중 잔류성이 있고 식품공전상 시험 전처리가 각각 다른 Chlorothalonil등 6종의 농약을 선정하고 그들 표준액에 대한 PPIR수준의 농도로추출용라의 종류 및 용매량 등의 추출조건을 달리하여 회수율 등을 측정한 결과 최적의 분석조건을 얻었다. 1) 추출 용매량은 증가할수록 추출 수율이 높았고,용매 종류에 대한 추출수율은 각 농약별로 다소 차이는 있었으나 극성 정도가 비교적 높은 Ethyl acetate애서 평균 추출수율이 가장 높았다 2) 최대 평균회수율은 Ethyl acetate 60mL를 사웅할 경우로서 93.6%였다. 3)표준액을 시료에 첨가한 시험에서도 역시 Ethyl ace늘to가 푼출수율이 가장 높았으며 불순물 분리제거 효과도 LLE법 보다 월등히 우수하였다. 4) 각 농약별 검출한계는 치소 0.001ppin으로 농산물 중 미량 잔류하는 농약을 검출할 수 있을 것으로 사료된파. 그러므로 SPE법은 LLE법보다 경제적인 방법으로 판명되었으며, 앞으로 농산물 중 잔류농약 분석에서 LEE법을 대치할 쑤 있을 것으로 기대된다. Solid-phase extraction by polar column(Extrelut-3') was attempted to develope the fast and efficient method of detecting pesticide of farm product.5·ix kinds of pesticide used in farming fieldfrequently and different in pretreatment, were chosen from Korea Food Cord. Optfmal analysis condi-tions were determiBed from t31e recovery rate of standard pesticides according to extraction solvent andextraction volurae. Extraction yietd was increased as solvent polarity and extraction volume. Maximumrecovery rate was acquired at 93.6% when 60mL of ethyl acetate was used as eluent. Tllis method wasmore effective than liquid-liquid partition extract·ion method to eliminated the impurity and had 0.001ppm of detection limit. Therefore. solid-phase eEl=raction was expected to be economical substitute fortraditional liquid-liquid extT·action method.

        Formyl Peptide Receptor 2 Is Involved in Cardiac Repair After Myocardial Infarction Through Mobilization of Circulating Angiogenic Cells : WKYMVm Promotes Cardiac Repair through Fpr2

        <P>Increasing evidence suggests that circulating angiogenic cells (CACs) promote repair of ischemic tissues. Activation of formyl peptide receptor 2 (Fpr2) has been reported to stimulate repair of ischemic heart. This study was conducted to investigate the role of Fpr2 on CAC mobilization and cardiac protection in myocardial infarction (MI). WKYMVm, a strong agonist for Fpr2, was administered in a murine model of acute MI, and mobilization of CACs including endothelial progenitor cells (CD34(+) Flk1(+) or Sca1(+) Flk1(+) cells) in peripheral blood was monitored. CAC mobilization by daily injection of WKYMVm for the first 4 days after MI was as efficient as granulocyte colony-stimulating factor and provided myocardial protection from apoptosis with increased vascular density and preservation of cardiac function. Transplantation of bone marrow (BM) from green fluorescent protein mice showed that BM-derived cells homed to ischemic heart after WKYMVm treatment and contributed to tissue protection. Transplantation of BM from Fpr2 knockout mice showed that Fpr2 in BM cells is critical in mediation of WKYMVm-stimulated myocardial protection and neovascularization after MI. These results suggest that activation of Fpr2 in BM after WKYMVm treatment provides cardiac protection through mobilization of CACs after MI, which may lead to the development of a new clinical protocol for treating patients with ischemic heart conditions.</P>

        Korean dance is handed down the type of pedigree and background ideas and occurs depending on the region. The term has been referred to variously as much as Various types of dance. Korea dance moves of culture and ideology holds, the term itself, you can experience the Korean sentiment. Korean dance of efficient practical training and systematic theory in this study to cleans up the term of a term that can complement existing dance. Korean dance moves presented in this study, the structure was divided into the lower body and upper body, the whole body. The dancing in the breathing, breathing and dancing to match the dance 6. Lifting, opening, and extension is represented by inhalation to dance, dance by exhalation down, closed, is represented by the movement of the bend. The upper body by breathing is open, firmware, and down, closed, bending that are common in the lower body and upper body choreography. The dance term for a variety of South Korea on the type of dance that can be accepted for this study to propose and Korean dance choreography cultural values as a practical education than Korea Dance efficiently and systematically, it can forward theoretical research that can be sustained by expectations should.

        아쿠타가와 류노스케(芥川龍之介)의 『히나(雛)』고찰

        This work is of a story describing deep emotion and discord of former days before making over a traditional doll that had been kept preciously in a Japanese home to a foreigner by moving to the era of civilization of Meiji from Edo. The work is also describing well the description of ruin of a home that was living well owing to the changes of the times, for generation of an ancestor. And owing to the ruin some dependent cases occur. It is that between mother and son who are persisting in tradition not other, a big discord has been experienced due to the difference of opinion. In the discord with mother who has own former modern way of thinking, Eikichi"s expectation that was firm in his conviction has been frustrated. And the family that kept good position in the era of Edo, has been rather frustrated owing to changes of times so called modern ages, and also lowly people can enjoy occupation and freedom that they wanted to have after times are changed, taking an example of the lowly people as rickshaw. As a result, owing to arrival of the era of Meiji, it shows well that the society is becoming equalization slowly.
