Expression Profiles of Apoptosis Genes in Mammary Epithelial Cells

To investigate apoptosis in HC11 mammary epithelial cells, we compared the gene expression profiles of actively growing and serum-starved apoptotic cells using a mouse apoptosis gene array and 33P-labeled cDNA prepared from the RNA of the two cultures. Analysis of the arrays showed that expression of several genes such as clusterin, secreted frizzled related protein mRNA (sFRP-1), CREB-binding protein (CBP), and others was higher in the apoptotic cells whereas expression of certain genes including survivin, cell division cycle 2 homolog A (CDC2), and cyclin A was lower. These expression patterns were confirmed by RT-PCR and/or Northern analyses. We compared the expression of some of these genes in the mouse mammary gland under various physiological conditions. The expression levels of genes (clusterin, CBP, and M6P-R) up-regulated in apoptotic conditions were higher at involution than during lactation. On the other hand, genes (Pin, CDC2) downregulated in apoptotic conditions were relatively highly expressed in virgin and pregnant mice. We conclude that certain genes such as clusterin, sFRP-1, GAS1 and CBP are induced in apoptotic mammary epithelial cells, and others are repressed. Moreover, the apoptosis array is an efficient technique for comparing gene expression profiles in different states of the same cell type.
The Changes of Steroid Receptors by Irradiation in rat Mammary Tumors

To centralize the determining estrogen and progesterone receptor concentrations in metropolitan area which are essential to decide the therapeutic method in managing the breast cancer patients and to realize the changes of hormonal receptors after irradiation authors induced mammary tumors in female rats by single intragastric feeding of 20㎎ 7, 12-dimethylbenzanthracene(DMBA). The estrogen and progesterone receptor concentrations were estimated before and four weeks after irradiation using the dextran-coated charcol procedures and the tumor size was checked before and weekly after irradiation for 4 weeks. 1. Only one of the nine positives was changed into negative state in estrogen receptor concentration after irradiation without statistical significance. 2. The positive seven cases were changed into negative state in progesterone receptor concentration except one and statistically significant reduction was observed after irradiation. 3. The average tumor size of single irradiation group before irradiation was 11.45±4.49㎝^2 and changed to 5.55±2.88㎝^2 at irradiated 2 weeks, 0.89±0.36㎝^2 at irradiated 4 weeks. 4. The average size in divided dose group was 16.17±7.22㎝^2 and changed to 11.85±7.81㎝^2 firstly irradiated 2 weeks, 7.85±5.07㎝^2 at secondarily irradiated 2 weeks.

N/A A clinical study on 18 cases of sheehan's syndrome, which were available for combined ptiuitary stimulation test consisting of insulin induced hypoglycemia followed by TRH and LHRH stimulation tests, was made. The results are followings; 1) The age at the time of diagnosis of sheehan's disease was ranged from 28 to 55 and averaged 44 years. The mean duration of appearance of clinical sumptoms after episode of postpartum hemorrhage is 11 years. 2) Basal pituitary function was within normal rage in most patients. 3) The responses of anterior pitutary hormones to combined pituitary stmulation test were impaired more than three in most cases (89%). 4) Panhypopituitarism was observed in only 5 (28%) out of 18 patients. 5) Varying degrees of pituitary response to combined pituitary stimulation test were noted among the various anterior pituitary hormones.
