        Multiplex PCR Detection for 3 Events of Genetically Modified Maize, DAS-59122-7, TC6275, and MIR604

        A multiplex polymerase chain reaction (PCR) method was developed to simultaneously detect 3 events of genetically modified (GM) maize. The event-specific primers were used to discriminate the following 3 events of GM maize (DAS-59122-7, TC6275, and MIR604) using multiplex PCR method. The zein gene was used as an endogenous maize reference gene in the multiplex PCR detection. The primer pair Zein-F/R producing a 99 bp amplicon was used to amplify the zein gene. The primer JI-Das-F1/R1 for DAS-59122-7, JI-TC6275-F3/R3 for TC6275, and JI-MIR F1/R1 for MIR604 yielded an amplicon of 130, 162, and 197 bp, respectively. The detection limit of multiplex PCR was 1% for DAS-59122-7, TC6275, and MIR604 for one reaction.

        가감청영탕(加減淸營湯)에 효과를 보인 열성 아토피 피부염 환자 6례 증례 보고

        Objective : The purpose of this study is to investigate the effect of Gagamchengyoung-tang($Ji\bar{a}ji\check{a}nq\bar{i}ngy\acute{i}ng-t\bar{a}ng$) for atopic dermatitis patients diagnosed as the febrile tendency. Method : This case study was done on 6 atopic patients with febrile tendency who have visited Korean Medicine Clinic from 2013.08.01. to 2014.04.30.. Gagamchengyoung-tang($Ji\bar{a}ji\check{a}nq\bar{i}ngy\acute{i}ng-t\bar{a}ng$) was prescribed to all 6 patients and evaluated the symptom change through photographs and questionnaire. Objective SCORAD Index(OSI) was used to evaluate the objective symptom and VAS was used to evaluate the subjective change. Results & Conclusion : The Objective SCORAD index and VAS were decreased in all patients. The average OSI reduction score was 26.3. Most symptoms of 6 patients were improved. Gagamchengyoung-tang ($Ji\bar{a}ji\check{a}nq\bar{i}ngy\acute{i}ng-t\bar{a}ng$) can be effective in patients who were diagnosed as febrile tendency.

        커뮤니티공간으로서의 어린이공원 조성에 관한 연구

        This study deals with the participatory design process of renovating a children's park. The process should proceed with two major elements. Firstly, the children's park should be designed from the perspective of children. Secondly, it should be designed in consideration of the context of the community. This study is divided into two parts. First is the review of theory concerning the meaning and roles of the children's park as a community space, the importance of children's participation in the design and the participatory method of the children. The second is the case study. The site for the case study is Ssial Park in Nowon-Gu, Seoul. The entire process of the case study is with community participation, from identifying the park's problems to the actual construction. At every step, through the participatory programs, the community participates in the decision making. In addition, mutual understanding among participants is pursued through diverse means of communication. The following results of the case study were found. Firstly, diverse participatory programs and the active trial to enable the community to communicate contribute to the sense of ownership and responsibility concerning the park Secondly, the community can negotiate the differences in opinions without needing the help of experts. Of course, there are limits in the case study, such as the fact that the community organization, which is the core of communication and maintenance concerning the park, is not organized through the process of a case study. Another is that more diverse methods, which inform all community members of the participatory process, should have been used.

No effective antiviral therapy is proven yet, but clinical use of ribavirin (RBV) has been tried. We investigated the antiviral effect of RBV against SFTSV in vitro. Methods: To test for cytotoxicity of RBV, Vero cells were treated with different concentrations of RBV (3.90 to 500 μg/mL, two-fold dilution) and analyzed by cell viability MTS assay 48h post-infection. To determine antiviral activity of RBV against SFTSV, Vero cells were infected with SFTSV strain Gangwon/Korea/2012 at 100 TCID50 (50% tissue culture infective dose) per well in a 96-well plate, and RBV was added at the concentrations showing no or minimal cytotoxicity. Viral RNAs were extracted from the culture supernatants and quantifi ed using one-step real-time reverse transcription- PCR to amplify the partial large segment of SFTSV. Statistical analysis was done by one-way ANOVA with Tukey`s post hoc test. Results: Cytotoxicity due to RBV was not observed at RBV concentration =31.3 μg/ mL. Viral RNAs at 24h post-RBV treatment were reduced with increasing RBV concentrations (1-32 μg/mL), compared with those of mock-treated cells (P <0.01, Figure). Half maximal inhibitory concentration (IC50) of RBV was 3.69 μg/mL at 24h post-RBV treatment. Conclusions: Our study shows that RBV has antiviral effect against SFTSV in a dose-dependent manner. Further studies are required to evaluate the effi cacy of RBV in SFTS.

        A flow of fluid has been studied in various fields of fluid engineering. The unsteady flow such has pulsation and liquid hammer in pipes was raised to hydraulic engineers as a trouble. We intend to approach a formalized mathematical model by a very exact and reasonable polynomial for fluid transmission lines. We observed most precedent studies for pipe line were studied without consideration of viscosity and temperature though press loss due to the friction of fluid inside pipe line, but learned change of peak within the 300Hz~400Hz with frequency characteristics according to temperature change after we study that considering temperature.

      • 원형관로내 층류 맥동유동의 속도와 압력 분포에 관한 연구

        In this study basic equations for the pulsatile laminar flow in a circular rigid pipe containing a slightly compressible fluid are derived and analytical solutions for the distributions of wave propagation factor K, cross-sectional mean velocity u_(m) and pressure P are schematically developed and confirmed experimentally. The pulsatile flow with 0 ≤ f ≤ 48Hz and 0 ≤ Re ≤ 2500 generated by summing the oscillatory flow made by ball valve into steady one. The wave form of pulsatile flow is described as a longitudinal wave and particle velocities measured experimentally are group velocities of the wave packet which are always smaller than phase velocities which are approximate to the sonic velocity C when the radial dispersion and wall friction are negligible. Therefore wave number is approximated to k = ω/c for high angular velocity region. The axial pressures and centerline velocities were measured by strain-gage type pressure transducers and Pito-static tubes, respectively. The cross-sectional mean velocities calculated from centerline velocities with the parabolic distributions of the laminar flow and confirmed by using the ultrasonic flowmeter. We knew that axial distributions of oscillatory velocity and pressure coincide with thoeretical and depend on the Reynolds number and angular velocity.

      • 계지의 항염 효과에 관한 연구

        Objectives : Cinnamomi Ramulus (CR), the young twing of Cinnamomi loureirii nees, has been used for treating symptoms related to pain. rheumatic arthritis and inflammation in Korean herb medicine. This study was carried out to investigate the anti-inflammatory effect of CR in vivo and in vitro. Methods : Extracts of CR were prepared and the chemical components of the extracts were examined by gas chromatography-mass spectrometry (GC-MS). The extracts were administrated to the rat paw edema model induced by carrageenan to evaluate the anti-inflammatory effect of CR. The expressions of nitric oxide (NO). prostaglandin E2(PGE2), inducible nitric oxide synthase (iNOS) and cyclooxygenase (COX)-2 were also quantified in lipopolysaccharide (LPS)-induced RAW 264.7 macrophages to survey the effect of CR in vitro. The main components were cinnamaldechyde and coumarin. Results : We examined the anti-inflammatory activity of the 80% ethanol extract of Cinnamomi Ramulus in vivo by using carrageenan-induced rat paw edema model. Maximum inhibition of 54.9% was noted at the dose of 1000㎎/㎏ after 2 hours of drug administration in carrageenan-induced rat paw edema and this showed a potent anti-inflammatory effect. Conclusions : The results showed that Cinnamomi Ramulus suppressed dose-dependently LPS-induced NO production in RAW 264.7 macrophages and also decreased iNOS protein expression. Cinnamomi Ramulus also showed a significant inhibitory effect in LPS-induced PGE2 production and COX-2 expression.

      • 한국산 자리공 자리공 열매의 적색소를 이용한 식용색소 : Ⅱ. 활성 사포닌을 제거한 색소의 안정성과 가공적성 Ⅱ. Color Stability and Characteristics as Food Colorant of Saponin-Free Pokeberry Pigments

        우리나라산 자리공 열매의 색소 함량은 산지에 따라 건물 g당 48.56에서 62.16mg으로 천연색소 자원으로 높은 편이였으며 산지간의 격차가 크지 않아 자원성이 좋은 편이었ㄷ. 자리공 열매의 적색소를 absolute ethanol 침전법으로 분획하는 과정에서 활성 사포닌이 95% 이상 제거되며 이 색고 침전물을 silica gel column이나 Sephadex G-25 column으로 정제함으로서 색소의 순도를 높일 수 있었다. Absolute ethanol 처리와 column 정제법을 응용하여 활성 사포닌을 제거 한 실험용 자리공 열매색소 분획 모두 색소 안정성이나 가공적성에서 뚜렷한 차리를 보이지 않았다. 따라서 absolute ethanol만 처리한 색소제품이나 이를 column chromatography로 정제한 색소분획 모두 고순도의 천연색소 첨가물로의 응용 가능성은 분명하였다. 이들 활성 사포닌을 제거한 자리공 열매색소의 색소안정성과 가공적성은 다음과 같이 요약 할 수 있다. 즉, pH 안정범위는 4.8-6.0이었으며, 열에 대한 안정성을 조사한 결과는 4℃에서 45일 저장했을 때는 70%의 색소 잔존율을 나타낼 정도로 안정한 편이었지만 25℃에서는 10일째에서 70% 잔존율을, 50℃이상의 온도에서는 수 시간 정도에서 50%이하의 색소 잔존율을 보여 전반적으로 열 안정성은 불량한 편이었다. 한편 이들의 존재 하에서는 급격한 변색이 진행되었다. 자리공열매 색소에 대하여 ascorbic acid나 palmityl ascorbate의 첨가는 색소 잔존율에 큰 영향을 미치지 않았으며 chlorogenic acid, resocinol 그리고 catechin의 첨가도 마찬가지였다. 포도당, 과당, 맥아당, 설탕, 펙틴, 셀루로스 그리고 cyclodextrin 등의 탄수화물도 전혀 영양을 미치지 않았다. 1000ppm 이내의 EDTA나 citric acid, malic acid 또는 oxalic acid 등의 산도 미치지 않았다. 금속이온으로서, Ca^(2+)과 Al^(3+)의 첨가는 색소의 잔존율에 큰 영향을 미치지 않았으나, Cu^(2+)나 Fe^(2+)의 첨가는 심한 변색을 초래하였다. 자리공 열매즙의 무수 알코올 처리물(적색 색소 침전물)은 soft gel, firm gel 또는 캔디 등의 고형 식품에 잘 어울리고, 음료에서는 변색이 빠르게 진행되며, 특히 요구르트의 경우가 가장 민감하였다. 결론적으로 보면, 우리나라산 자리공 열매에서 활성 사포닌을 제거하고 확보한 색소제품은 사탕무의 betanin과 거의 같은 색소안정성과 가공적성을 보여 줌으로서 사탕무색소 첨가제인 beet red를 대신할 수 있으며 색소의 함량이나 순도라는 관점에서는 훨씬 우수할 것으로 사료된다. To evaluate the basic data on the color stabilities of saponin-free pokeberry pigment products, % remain of pigment were calculated from absorbances of solutions stored at various conditions. The pigment products were estimated to be very stable at pH 4.8-6.0. Thermostability of pH 5.0 solution of pigment products could be kept stable below 4℃. Addition of Cu2+ or/and Fe2+ decreased the % remain of pigment, whereas most food components had no offset the stability of pokeberry pigment. The color changes of some model foods having pokeberry pigment were checked by Hunter color differerence meter for 45 days. In solid foods like gels and candy the changes of color difference were very slower than that of model beverages. Application of pokeberry pigment to soft drink and yogurt were found to be effective at the refrigerated condition.

      • 고량강의 항염증 작용에 대한 연구

        Objective : The use of herbal therapy is becoming an increasingly attractive approach for the treatment of various inflammatory disorders. The Alpiniae officinari Rhizoma is popular in Aisa as a traditional herbal medicine. Alpiniae officinari Rhizoma is a species of the ginger family(Zingiberacease). Method : This study was performed to investigate the anti-inflammatory effect of Alpiniae officinari Rhizoma extract by the methods of "carrageenan induced paw edema" and "Lipopolysaccharide-induced inflammatory mediators in mouse macrophage RAW 264.7 cells". Result : We suggest that Alpiniae officinari Rhizoma extract decreased paw volume induced by plantar injection of carrageenan. Also Alpiniae officinari Rhizoma extract inhibited nitric oxide, prostaglandin E₂production and induced nitric oxide synthase, cyclooxygenase-2 protein expression in Mouse macrophage RAW 264.7 cells stimulated with lipopolysaccharide. Conclusion : This study shows that Alpiniae officinari Rhizoma extract seems to have anti-inflammatory effect by inhibition of nitric oxide, prostaglandin E_(2) production and nitric oxide synthase, cyclooxygenase-2 protein expression.
