한국산 씀바귀(Lactuca dentata Makino)의 ethylacetate 추출물에서 6 종류의 flavonoid 성분이 TLC에 의해서 검출되었으며, 이들은 극성에 따라서 A에서 F가지 명명되었다. 그들 중 주성분으로 생각되는 물질 E를 column chromatography을 이용하여 분리정제 하였다. 물질 E의 녹는 점은 249.5℃-251℃로 나타났으며, 그 구조를 밝히기 위하여, 물질 E와 이를 가수분해한 genin에 대한 여러가지 shift reagent에서 UV와 IR분석을 하고 또한 물질 E와 그의 acetyl 유도체에 대한 NMR 분석을 하였으며 물질 E에 결합된 당은 TLC에 의하여 확인하였다. 이와같은 분석 결과에서 물질 E는 luteolin 모핵에 glucose가 결합된 luteolin-7-0-β-D-glucoside로서 확인되었다. The ethylacetate extract of Lactuca dentata Makino showed 6 flavonoidal components as detected by ferric chloride solution. The flavonoidal constituent of Lactuca dantata Makino was isolated and purified by the series of column chromatography. The chemical structure of one of the flavonoidal component named as compound E was identified by UV, IR and NMR spectrometry. The melting point range of compound E was 249.5℃-251℃. The UV and IR spectra of purified compound E, and its genin were measured with the various shifting agents. The results of UV analysis showed the free state of hydroxy group at 3rd and 4th carbon and binding of sugar at the 7th carbon of compound. The sugar bound to the compound E was identified as glucose by TLC. The IR spectrum showed the presense of hydroxy group, conjugated carbonyl group and aromatic group. The analysis of NMR spectrum was done to the purified compound and its derivatives. The chemical shifts against hydrogen atom, hydroxy group, and the moiety of luteolin were observed in the NMR spectrum along with their position and number as well as type of sugar bound. The isolated and purified compound was identified as luteolin-7-0-β-D-glucoside.

시판 농후 발효유의 저장 중에 유산균수, Bifidus균수의 경시적 변화를 조사하였다. Bifidus균의 검출에서 혐기적으로 BL배지에서 NNL용액에 5ml을 첨가하여 배양하는 방법이 간편 방법으로 적합하였으나, 시료별 유산균수에 비하여 Bifidus균의 검출이 감소되었으며 5℃에서 유효기간 동안 Bifidus균의 변화는 거의 없었다. 총 유산균수(혐기적 배양)는 ?? ∼?? 정도였으며 Bifidusr균은 시료별 많은 편차를 보였다. B,N,Y,S회사제품은 유효기간 동안 ??을 유지하여 큰 차이를 보이지 않았으나 M 회사 제품만이 유산균수, Bifidus균수도 적게 검출되었다. Bifidus균의 증식촉진 여부를 알아보기 위하여 SMP배지에서 oligo당을 첨가한 결과, 산도나 pH, Bifidus균수의 차이를 발견하지 못하였으며, 이는 SMP배지 자체가 생장 영양원으로 충분하였던 것으로 판단되었다. The counts of Bifidobacterium in commercial products of stirred yogurt were detected in order to investigate the consecutive changes of this bacteria. An anaerobic culture method was chosen to isolate the lactic acid bifidobacteria. This method using BL agar media containing NNl solution was found to be the most convenience and suitable for detect the total counts of Bifidobacterium. With this method, it were found that total counts of Bifidobaterium in samples were ranged over ?? ∼?? whereas that of total lactic acid bacteria cultured in anaerobic condition were ?? ∼?? /g. Among sample products made by B, N, S and M company, only a sample of M company was found to have lower level of total counts of lactic acid bacteria than other samples, B, N, Y and S. Commercial samples from B, N, Y and S company indicated consistently the average total counts of Bifidobacterium to ?? /g.
동의나물 Caltha Minor Nakai 뿌리의 saponin에 관한 연구

Three crude saponins having the same aglycone were isolated from the root of Caltha minor NAKAI which has been known to be of effect as emetics in the folk medicines of Korea. And they were confirmed to their saponoidal properties through the methods of hemolytic test and LIEBERMANN-BUCHARD reaction.

Two triterpenoid saponins were isolated from the methanol extract of Caltha minor leave(Ranunculaceae). The structure of these saponin were elucidated as hederagenin-3-0-alpha-rhamnopyranosyl(1->2)-alpha-L-arabinopyranoside and 3-0-alpha-L-rhamnopyranosyl(1->2)-alpha-L-arabinopyranosyl hederagenin 28-0-alpha-L-rhamnopyranosyl(l->4)-beta-D-glucopyranosyl(l->6)-beta-D-glucopyranosyl ester.

Objectives: The present study investigated Inflammatory effect of Coptidis Rhizoma in lipopolysaccharide-exposed rats and Raw 264.7 cells. Methods: The plasma concentration of IL-1β, IL-6 and TNF-α peaked at 5 h after LPS injection, and the values of the Coptidis Rhizoma extract groups were lower than those of the control group. In the increment of cytokines concentration at 2 h and 5 h after LPS injection, the Coptidis Rhizoma groups were lower than that of control group. The plasma concentration of IL-10 peaked at 5 h after LPS injection, and the values of the Coptidis Rhizoma extract groups were higher than those of the control group. In the increment of cytokines concentration at 2 h and 5 h after LPS injection, the Coptidis Rhizoma groups were higher than that of control group. Liver cytokines measurement was done at 5 h after LPS injection. The concentration of liver IL-1β and IL-6 in the Coptidis Rhizoma groups was lower than that of the control group. The concentrations of liver TNF-α, and IL-10 showed no significant differences among all the treatment groups. Results: In the studies of lipopolysaccharide-exposed Raw 264.7 cells, the concentration of IL-1β, IL-6 and TNF-α in the lipopolysaccharide-exposed cells groups was higher than that of control group (normal group), and in the lipopolysaccharide-exposed cells groups, these values showed a tendency to decrease in the Coptidis Rhizoma groups. The concentration of IL-10 in the lipopolysaccharide-exposed cells groups was higher than that of control group (normal group), and in the lipopolysaccharide-exposed cells groups, the values showed a tendency to increase in the Coptidis Rhizoma groups. Conclusions: These results indicate that the Coptidis Rhizoma extracts have an functional material for Inflammatory activities.
