      • Breeding of Tetraploid in Platycodon grandiflorum (Jacq.)A. DC. by Colchicine treatment

        Present studies were carried out to produce tetraploid plants by colchicine treatment using seeds, seedlings and shoot tips of Platycodon grandiflorum in Campanulaceae. The most successful colchicine treatment for tetraploid production in P. grandiflorum was soaking treatment using 0.01 and 0.5% colchicine solution for 1 hour and 12 hours, respectively. Morphological characteristics of both diploid and tetraploid were similar, but tetraploid plants had more leaves. Compared to diploid, tetraploid had the larger stomata, but less number of stomata. Fresh weight of tetraploids was 20∼40% heavier than that of diploid.

        Background: The diabetic patients are at significantly increased risk of developing vascular disease. It's etiology may involve oxidative damage by free radiacals and protection againse such damage can be offered by antioxidants. We investigated that oxidative stress as assessed by measurement of total antioxidant status may play a role in development of diabetes mellitus. Method: We measured total antioxiant status using merchandised kit, glycated hemeglobin(HbA1c) in 46 type 2 diabetes mellitus patients and 50 healthy matched control subjects. Result: The total antioxidant status(TAS) was 2.10(±0.04) mmol/L in uncontrolled type 2 DM patients, 2.60(±0.03) in controlled type 2 DM patients and 2.70(±0.16) in healthy control subjects. TAS was significantly lower(P<0.05) in uncontrolled type 2 DM patients, but no significant association between in controlled type 2 DM patients and healthy control subjects. The TAS was 2.00(±0.17) mmol/L in complicated type 2 DM patients and 2.10(±0.29) uncomplicated type 2 DM patients. There was no significant associations between complicated type 2 DM patients and uncomplicated type 2 DM patients. Conclusion: Poor glycemic control is associated with reduced TAS in type 2 DM patients. TAS was thought indirect index that predict glycemic control of type 2 DM patients.

      • Breeding of Tetraploid in Codonopsis lanceolata (Sieb. et Zucc.) Trautvetter by Colchicine Treatment

        Present studies were carried out to produce tetraploid plants by colchicine treatment using seeds, seedlings and shoot tips of Codonopsis lanceolata. Three tetraploid plants of C. lanceolata were produced from seeds which absorbed 0.1 % colchicine solution for 12 hours, and 0.5% colchicine solution for 1 and 6 hours from seedlings, respectively. But tetraploid was not produced from shoot tips treated by colchicine solution. Compared to diploid, tetraploid plants had larger stomata, but less number of stomata. Fresh weight of tetraploid plants was 1.4∼3.6 times heavier than diploid plants.

      • 유도 업어치기 패턴에 따른 하지의 운동학적 분석

        The purpose of this investigation was to analyze the changes of the lower limbs angle, when performing Morote-Seoinage(M.S.N.:two arm shoulder throw) in judo who was 5 females university representative judokas of light weight category in judo, and filmed on video cameras(60field/s). The data of this study digitizied by KWON3D 2.1 program computed the average and standard deviation calculated individual 5 trials with Programing Lab view 6i. From the data analysis & discussion, the following conclusions were drawn : 1) A pattern : Right vknee angle was flextion and extension Kuzushi phase and flexed in Tsukuri to Kake phase. Light knee angle flexed in the whole phase. Right·Light hip angle weren't difference changle of angle. 2) B pattern : R. knee angle had flextion and extension in Kuzushi phase and flexed in Tsukuri to kake phase. Light knee angle slowly flexed in the whole phase. Suddenly, flexed in Kake 1·2 stage. Right hip angle raised flextion and extension in Kuzushi phase and suddenly flexed in Tsukuri to Kake phase. Light hip angle slowly raised in the whole phase. 3) C pattern : C pattern was larger than A, B pattern's change of Right knee angle, this time angle was 30.4˚. Light knee angle was flextion in the whole phase. Right hip angle flexed in Kuzushi phase and largely extended, again flexed. Light hip angle firstly flexed and extended, flexed on the Tsukuri to Kake phase.

Was construed that weight lifters are difference that is attentive being higher than Taekwondo player weight lifting and Taekwondo player's body special quality comparison. 2. Showed laziness or difference by thing that weight lifters are tall in weight lifter and Taekwondo player's cycle and comparative analysis result maximum blood pressure of heart exhaust ability and minimum blood pressure in weight lifting and Taekwondo player's cycle and heart exhaust ability special quality comparison. There is no difference in heart beat, capacity of the lung, standard capacity of the lung, %Capacity of the lung. 3. According to weight lifter's age change state of physical strength as result that analyze comparison age high maximum oxygen intake amount, sit-up, getting down push-up, appeared by standing high, difference that die and appeared by thing that increase from one leg stand, and keep in mind from other physical strength measurement item is not. Appeared difference A that is not that keep in mind according to result that compare Taekwondo player's physical strength by rain parameter statistics. Therefore, can see that there is no special in difference physical strength in athletes. In this research is looked should be developed more various program for development that have at athlete type playing the first stones for 8 weeks station and Taekwondo training that influence in health physical strength and body development know can but present correct measurement data because forward research develops various program with little more many number of persons and athletes' health physical strength and bodily balance.

      • Magnetic bead capture 를 이용한 염색체 띠 7q11, 7q22 에 위치한 cDNA 선별

        Recently, surmountable amounts of genes are being cloned and it has become neccessary to develope new techniques for discovering genes with known chromosomal location and their possible functions. We have developed one such a method and applied it to search for genes that may play a functional role during the development of the central nervous system. To observe the distribution of 18 weeks old fetal brain cDNAs on the chromosome, fluorescent in situ hybridization (FISH) was performed with biotin labeled 18 weeks old fetal brain cDNAs. Among the chromosome bands showing strong hybridization with the cDNAs, 7q11 and 7q22 were microdissected and amplified by PCR. Each band-specific probe pool was confirmed by FISH and band specific cDNAs were selected by magnetic bead capture method. Selected cDNAs were subcloned to plasmid vectors and the nucleic acid sequences were analysed. As a result 41 different clones from 7q11 and 43 different clones from 7q22 were obtained. They were categorized as 25 clones of well characterized genes, 22 clones showing low homology with known genes, 13 clones of simply registered uncharacterized human cDNAs, and 24 clones of unknown genes. From these results, it may be suggested that our technique is very useful to clone the genes expressed in the developing human brain with confirmed chromosomal location. In addition, this cloning technique can be used to discover the new genes related with neural development in combination with functional screening methods.

      • 급성골수백혈병에 대한 관해유도화학요법 후의 Granulocyte Colony-stimulating Factor의 효과

        Granulocyte colony-stimulating factor(G-CSF) have been shown to hasten the recovery of neutropenia following anti-cancer chemotherapy. There are controversial opinions on the use of G-CSF in acute myelogenous leukemia(AML) because clonogenic studies have shown that G-CSF stimulates leukemic colonies as well as granulocyte colonies. In this study, we evaluated the effectiveness and safety of recombinant human G-CSF after induction chemotherapy with DAV regimen(Ara-C 100mg/㎡ day 1-8, Doxorubicin 45mg/㎡ day 3-5, VP-16 100mg/㎡ day 6-8) in 9 patients with AML. G-CSF therapy(200 ㎍/㎡/day) was begun 2 days after the end of chemotherapy and continued for 10 days. 17 AML patients who recieved the same chemotherapy before the onset of this study were used as historical control. G-CSF shortened the duration of granulocytopenia (less than 500/㎣) significantly (13 vs 23 days, p<0.001), but it had no effect on platelet recovery. Although the incidence of febrile episodes was almost the same, the duration of febrile episodes was shorter in the group treated with G-CSF( 5 vs 12 days, p=0.03). There was no evidence that G-CSF accelerated the regrowth of leukemic cells and the complete remission rates between the 2 groups were not different. These results show that G-CSF accelerates the recovery of granulocytopenia and shortens the febrile days after chemotherpy in patients with AML, without affecting the regrowth of leukemic cells.

      • 급성골수성백혈병 환자에서 DAV 병용화학요법 후의 장기생존율

        Background : Despite substantial progress, the treatment of acute myeloid leukemia(AML) has produced complete remission in 60-80% of patients receiving induction chemotherapy, and median remission duration is about 12 months and only 20% to 35% of patients undergoing consolidation chemotherapy achieve long-term disease-free survival(DFS). We evaluated the long-term outcome of AML patients treated with doxorubicin/Ara-C/VP-16(DAV) induction chemotherapy and consolidation/intensification therapy. Method : Induction therapy : From January 1986 to December 1991, twenty three patients with previously untreated acute myelogenous leukemia received a course of 45mg/㎡ doxorubicin daily intravenously for three consecutive days with Ara-C at 100mg/㎡ by continuous intravenous infusion for eight consecutive days and VP-16 at 100mg/㎡ daily intravenously for three consecutive days. A second course of treatment was started if leukemia persisted on 22 days after treatment. Post-remission therapy : Three to six cycles were given at three or four months interval with Ara-C/doxorubicin/VP-16 regimen or other therapy. Results : Twenty two pateints were evaluable and complete remission was achieved in 16 of 22(73%). Median duration of complete remission was 8 months. The relapse rate was 81% and 63% relapsed in first year. 4-year survival rate of patients entering complete remission(n=16) was 19% and median survival duration was 14.5 months. The postremission chemotherapy was the only significant prognostic factor influencing long term disease free survival. No significant correlation was observed between the probability of survival and age (40< or >40), sex, FAR subgroup, and leukocyte count at diagnosis. The median survival duration were 21 months and 12.5 months for patients who received, or not received postremission chemotherapy respectively(P=0.035). Conclusion : Our results show that DAV combination chemotherapy is a useful therapeutic regimen in remission induction and postremission chemotherapy offering survival advantage in patients with AML entering complete remission.

      • 항정자항체 평가에 있어서 정장액 interleukin-6 측정의 유용성

        Breakdown of the blood-testis barrier by infection, trauma, or operation allows sensitization of spermatozoal antigens. Sperm antibodies may be a relative cause of infertility in infertile males. Multiple assay techniques have been developed to detect antisperm antibody. Older tests have been supplanted by the newer immunoglobulin specific assays. Immunobead test was used widely. Interleukin(IL)-6 is necessary for formation of antibody. So, this study was performed to investigate the value of interleukin-6 in the detection of immunologic cause of infertility. The antisperm antibody titers and interleukin-6 was measured in 10 healthy males and 30 patients with chronic prostatitis using immunobead methods and enzyme immunoassay kit. Titers of direct and indirect IgG, IgA and IgM antisperm antibody(% binding) was 0/0.7±0.7, 0/0, 0/0.1±0.1 and 0/1.1±0.5, 0/0.1±0.06, 0/1.1±0.6 in control group and patients with chronic prostatitis. Titers of direct and indirect IgG, IgA and IgM antisperm antibody was 1.8±1.7,0, 0.3±0.3 and 0.3±0.9, 0, 2.8±1.1 in patients with chronic prostatitis whom the antisperm antibody was found in. There was significant difference in indirect IgG and IgM between control group and patients with chronic prostatitis whom the antisperm antibody was found in. The value of seminal IL-6(pg/ml) was 3.2±0.9 and 9.6±2.3 in control group and patients with chronic prostatitis. There was significant difference. The value of seminal IL-6 was 7.0±2.7 and 13.4±5.1 in patients without chronic prostatitis and patients with chronic prostatitis. There was no significant difference. It is suggested that the examination of seminal IL-6 alone can be useful to evaluate the immunologic state of seminal tract but can not take the place of Immunobead test that detect the antisperm antibody formation.
