Potential roles of D-serine and serine racemase in experimental temporal lobe epilepsy

<P>To confirm the roles of D-serinergic gliotransmission in epilepsy, we investigated the relationship between spatiotemporally specific glial responses and the D-serine/serine racemase system in mesial temporal structures following status epilepticus (SE). In control animals, D-serine and serine racemase immunoreactivities were detected mainly in astrocytes. After SE, D-serine and serine racemase immunoreactivities were increased in astrocytes. Double-immunofluorescence study revealed that up-regulation of serine racemase immunoreactivity was relevant not to D-serine immunoreactivity but to nestin or vimentin immunoreactivity. Neither D-serine nor serine racemase was found in naïve or reactive microglia. In addition, phosphorylated N-methyl-D-aspartate (NMDA) receptor subunit 1 (pNR1-Ser896) immunoreactivity in the hippocampus was increased compared with controls. Increased D-serine immunoreactivity showed direct correlation with the phosphorylation of Ser896 of NR1. Given the findings of our previous study, these findings suggest that D-serine and serine racemase in astrocytes may play roles in neuronal hyperexcitability via a cooperative activation of NMDA receptors. Furthermore, serine racemase may be involved in migration and differentiation of immature astrocytes, which is relevant to reactive astrogliosis. © 2010 Wiley-Liss, Inc.</P>

The regulation of angiogenesis is an interesting area to consider for novel therapeutic approaches to rheumatoid arthritis (RA). Chemically modified heparins have been developed as possible candidates for angiogenesis inhibitor; however, they have a major clinical drawback in exhibiting poor oral bioavailability. Here, orally absorbable O-desulfated low molecular weight heparin (ODS-LMWH) derivatives were newly synthesized by conjugating 2-O- or 6-O-desulfated LMWH with deoxycholic acid (DOCA) or bisDOCA (a dimer of DOCA), and their physicochemical properties, antiangiogenic potency and pharmacokinetic profiles were assessed. After selecting the best candidate among those derivatives, its therapeutic efficacy on arthritis was investigated in a murine collagen antibody-induced arthritis (CAIA) model. ODS-LMWH derivatives significantly inhibited the capillary-like tube formation of human umbilical vein endothelial cells (HUVECs) and basic fibroblast growth factor (bFGF)-induced angiogenesis in the Matrigel plug assay. Among all the compounds, 6ODS-LHbD showed the highest oral bioavailability in rats (19.3%). In the CAIA mouse model, 6ODS-LHbD (10mg/kg, p.o., S.I.D.) significantly inhibited neovascularization in the joint, the increase of hind-paw thickness, and the structural damage in the bone. Therefore, 6ODS-LHbD would be a promising candidate for an orally active drug for the treatment of RA.

[ $0.75delta\%$ ]의 평판광회로(PLC;Planar Lightwave Circuit)소자의 설계 및 제작기술을 가지고 저손실과 높은 누화율을 가진 파장 다중화 및 역다중화 소자를 개발하였다. C-band AWG(Arrayed Waveguide Grating)에서의 삽입손실은 2.503이하, 누화율은 35dB이상, 균일도는 1dB이하이며 L-band에서는 Vernier 디자인을 적용하여 ITU-T의 파장 정확도가 0.04nm이하가 되도록 제작하였다. A 40 channel arrayed-waveguide grating (AWG) filter operating in C-band and L-band wavelength regions has been fabricated using PLC (Planar Lightwave Circuit) processes with 0.75 refractive index difference. Its design was optimized for matching the center wavelength with the ITU-recommended wavelength. The characteristics of the fabricated C-band AWG are as follows; average insertion loss < 2.5 dB, polarization-dependent loss < 0.3 dB, non-adjacent crosstalk >35dB, and the loss uniformity of 0.8 dB. In the L-band AWG, wavelength accuracy is below 0.02nm.
IL-32γ inhibits cancer cell growth through inactivation of NF-κB and STAT3 signals

<P>Several studies have shown physiological functions of interleukin (IL)-32, a novel cytokine. However, the role of IL-32 in cancer development has not been reported. In this study, we showed that IL-32γ inhibited tumor growth in IL-32γ-overexpressing transgenic mice inoculated with melanoma as well as colon tumor growth in xenograft nude mice inoculated with IL-32γ-transfected colon cancer cells (SW620). The inhibitory effect of IL-32γ on tumor growth was associated with the inhibition of constitutive activated nuclear transcription factor-κB (NF-κB) and of signal transducer and activator of transcription 3 (STAT3). The expression of antiapoptotic, cell proliferation and tumor-promoting genes (<I>bcl-2</I>, <I>X-chromosome inhibitor of apoptosis protein</I> (<I>IAP</I>), <I>cellular IAP</I> and <I>cellular FADD-like IL-1β-converting enzyme-inhibitory protein</I>, <I>cyclin D</I>), cyclin-dependent kinase 4, cycolooxygenase-2 and inducible nitric oxide synthase was decreased, whereas the expression of apoptotic target genes (<I>caspase-3</I> and <I>-9</I>, <I>bax</I>) increased. In tumor, spleen and blood, the number of cytotoxic CD8<SUP>+</SUP> T cells and CD57<SUP>+</SUP> natural killer cells and the levels of IL-10 increased, but that of tumor necrosis factor-α (TNF-α), IL-1β and IL-6 decreased. We also found that forced overexpression of IL-32γ inhibited colon cancer cell (SW620 and HCT116) growth accompanied with the inhibition of activated NF-κB and STAT3 <I>in vitro</I>. In addition, when IL-32γ was knocked down by small interfering RNA (siRNA) or neutralized with an anti-IL-32γ antibody, IL-32γ-induced colon cancer cell growth inhibition, the IL-32γ-induced decrease of TNF-α, IL-1 and IL-6 production, and the increase of IL-10 production were abolished. However, siRNA of NF-κB and STAT3 augmented IL-32γ-induced colon cancer cell growth inhibition. These findings indicate significant pathophysiological roles of IL-32γ in cancer development.</P>

This study was carried out to investigate the meat color, fat color and maturity of Hanwoo and imported beef. Meat color and marbling could be observed by the computer images of the digital camera with the naked eye. In histograms, the peaks related with bright cherry red color of beef were concentrated in more than 128 log scale and the peaks of dark red color were concentrated in less than 128 log scale. However, it was difficult to identify the maturity of beef as meat color, for meat color changed during storage. L* and a* values were high along with increasing in myoglobin content and bright cherry red, and b* value was higher at imported beef in Australia than in Hanwoo beef.
성인에서 Bifurcate Plate 를 이용한 상완골 근위부 골절의 치료

Seven patients, range in age from 30 to 65 years(average 49.7), were evaluated minimal 6 months after open reduction and internal fixation of proximal humeral fracture using the Bifurcate blade plate. According to the Neer's classification, there were three 2-part, three 3-part and one 4-part fractures. The methods of threatment and its results were analyzed according to the Neer's classification. l. All fractures were healed. 2. No avascular necrosis was observed at following-up. 3. 6 patients had excellent results, one satisfactory result. 4. The Bifurcate blade plate proved to bc an another method far the treatment of proximal humeral fracture. 5. A prosthetic replacement is recommended for eldery patients with severe osteoporosis, severe commimuted fractures.

Holographic interferometric tomography has been used to reconstruct the three-dimensional temperature field around a single isothermal cube in an infinite medium. The technique, which mainly consists of interferometric fringe enhancement and analysis and computational tomography, becomes user-friendly by utilizing GUI(graphics user interface) environment. It can also reconstruct the reasonably complex, three-dimensional fluid flow in a relative short period of time. More systematic analysis of the three-dimensional field is underway, and the results will be reported shortly.
