천연항균성 물질을 이용한 Vibrio vulnificus의 살균 및 독소생성 억제효과

생선회용 해산물에 오염되어, 생식한 경우 발생되는 패혈병의 원인균주인 Vibrio vulnificus의 생육 및 독성생성을 억제할 목적으로 천연 항균제인 Grapefruit종자추출물(GPSE)을 이용하여 그 항균효과를 검토하였다. In vitro 시험결과, Vibrio vulnificus에 대한 GFSE의 생육최소저해농도는 50~100 ppm 정도이었으며, Vibrio vulnificus 균체세포틀 100 ppm 농도의 GFSE 용액으로 처리하고 전자현미경 촬영시료로 조제하여 촬영한 후, 미생물형태을 무처리대조구와 비교, 검토한 결과, GFSE 처리로 세포막 기능이 파괴되어 세포내용물이 균체외부로 유출되어 균체의 생육이 억제되고 균체내부가 빈 ghost 형태의 균체수가 증대되어 GFSE의 항균효과를 확인할 수 있었다. 한편, 생선회용 해산물에 GFSE를 근육주사하고 Vibrio vulnificus를 접종한 수조내에서 사육하여 채취한 어육부를 마쇄하여 일정량씩을 표준사료에 첨가해서 통닭용 병아리를 실험동물로 하여 사양실험을 실시한 결과. 체중증가율, 단백질이용효율, 혈청내 효소활성, 혈청의 주요성분함량 등에 있어서, GFSE 처리수조내에서 사육한 해산물을 첨가한 시료시험구의 경우, 무첨가사료처리구와 비교할 때 뚜렷한 차이를 목격할 수 없었으며, GFSE 첨가에 따른 실험동물의 성장저해 또는 독성현상을 유발하지 않음을 알 수 있었다. 아울러, 각 처리구에서 사양한 해산물의 어육부에 오염된 Vibrio vulnificus의 균수를 측정한 결과, 250ppm 이상의 GFSE 처리구에서는 Vibrio vulnificus를 전혀 검출할 수 없었다. To prevent food-poisoning outbreaks by Vibrio vulnificus the antimicrobial efficacy of grapefruit seed extract (GFSE) was examined. Minimal inhibitory concentration (in vitro) for the microorganism was found to be 50~100 ppm. Transmission electron micrographs of Vibrio vulnificus showed the biocidal action of this natural antimicrobial agent would be related to specific respiratory effect coupled with the destruction of permeable function of microbial cell membrane. After Anguilla japonia GFSE-injected to the body was incubated in the seawater contaminated by Vibrio vulnficus the fish meats were taken up, mixed with control diet and used as a diet in the feeding experiment. In this experiment the effect of GFSE treated with fish muscle on body weight protein efficiency ratio, serum enzymes and serum blood component of broiler chicks was investigated. It is proved from this study that there is neither Vibriosis nor toxicity associated with GFSE itself and fish meats treated with it when it is injected to the fish body at a level of 250 ppm or less.

아크리놀은 살균,소독에 쓰이는 약물로서 연로제 및 지사제용도의 경구투여제제등게 이용되고 있다. 공정서에 수재된 아크리놀의 분석법은 적정법이고 제제에서늬 분석법은 확립되지 않았고 원료의 적정법을 제제에 적용시 다른 주성분이나 부형제 또는 안정제 드으이 영향으로 분석오차가 생길 우려가 있다. 따라서 아크리놀을 보다 신속·정확하게 확인 및 정량분씬을 할 수 있고 강도와 분리능이 우수한 HPLC를 이용하여 아크리놀을 분석하고자 하였다. MPLC 분석조건은 이동상 :아세료니트릴 · 1% 인산일수소나트륨혼합액(40:60)(p템 5.2), 칼랏 :옥타데실실릴화한 실리카칼럼,검출기 :자외부흡광광도계(측정파장 : 254nm)을 사용하여 양호한 결과를 얻었다. 틴PLC법의 분석 결과, 검량선의 상관계수는 0.9999, 재현성(aSD(%))은 0.5234. 정균회수율은 100.28%(RSD=0.89%)로 정량성 및 재현성이 높온 분석 결과를·얻었다. 또한 시판품에 대하여 려PLC분석법과 공정서에 수재된 적정법의 시칩결과를 비교하였을 때 핀Plf법인 더욱 정확한 결과를 얻을 수 있었으므로, 조작이 간편하고 재현성이 높은 딘PLC분석법을 약전차정의 기초자료로 활용하고자 한다. We describe a method for the determination of acrino1 and its preparations by HPLC. The optimum analrtical coriditions were as follows;column : u-Bondapak C_(18) mobile phase : acetonitrile 1% Na_(2)HPO_(4), buffer (40:60)(pH 5.2), detector : 254nm, flow rate 1.OmL/min, injection volumn : 5uL, The calibration curve at 254nm was linear within the range of 10 ~ 140㎍/mL. This HPLC method has been validated to be precise, accurate and linear. This method has also been found to be applicable to other formulations containing acrinol.
옻나무(Rhus verniciflua STOKES) Flavonoid 분획 투여가 정자생성 및 생식관련 장기에 미치는 영향

Rhus verniciflua Stokes (RVS) has been used as a food supplement and a traditional herbal medicine. In this study, we prepared various flavonoid fractions (RS, RW1, RW2 and RWE) from a hot water extract of RVS and their influence on male reproductive organs and spermatogenesis were studied in rats which were orally administered 200 ㎎/㎏ of them for 8 weeks. All experimental groups did not show any significant changes in body weight and blood clinical chemistry for liver function. Plasma testosterone level was elevated about 3.7, 5.2 and 6.3 folds in RW1, RW2 and RWE groups, respectively. The weights of testes and epididymides tended to increase slightly without the statistical significance in RW2 and RWE. The spermatozoon motility and epididymal sperm concentration were significantly increased (P < 0.05) in RWE and RW1, respectively, when compared to the control group. There was no significant difference in histology and apparent shape of testes and epididymides among the control and the experimental groups. Collectively, RWE showed effectively the elevation of plasma testosterone level, spermatozoon motility and the epididymal sperm concentration without the significant increase of testis and epididymides weights. When the component HPLC profile among the flavonoids fractions of RVS was compared, the ratio of components were only different. These findings suggest that the Rhus flavonoid fraction, particularly RWE, can stimulate the androgen-dependent male sexual function and it can be applied to the material of functional food for enhancing the sexual function.

It has been reported that there are two distinct phenotypes in Frarinus mandshurica Rupr. growing in Korea. Recently developed polymerase chain reactioniPCR) was used to detect DNA sequence polymorphism in the species. Using a thermostable DNA polymerase and synthetic DNA primers, unknown DNA sequences from the species were randomly amplified. The two types of the species produced different DNA amplification pattern with three different primers tested, Although DNA polymorphism was detected among individuals within types, each type has its own distinct pattern. The two types could be easily differentiated by trier characteristic predominant bands.

Rhus verniciflua Stokes (RVS) has been used as a food supplement and a traditional herbal medicine for a men's sexual enhancement. In this study, we prepared a flavonoid fraction (RWE) from a hot water extract of RVS and its influence on sexual behavior was studied in male rats which were orally administered varying doses of RWE for 2 weeks. All doeses of RWE stimulated sexual behavior in male rats such as reducing mounting latency and enhancing mount frequencies. However, intermission numbers were not changed and none of group can observe the ejaculation during behavioral testing time 5 min. Testosterone levels were increased about 0~53% and 92~164% by the treatment of RWE for 1 and 2 weeks, respectively. However, estrogen levels in male rats teded to decrease in a dose dependent manner of RWE. At the dose of RWE 200㎎/㎏, estrogen level was reduced to 77.5% and 70.3% of control after 1 and 2 weeks treatment. These findings suggest that the Rhus flavonoid fraction can stimulate the androgen-dependent male sexual behavior and it can applied to the material of functional food for enhancing the sexual function.

Cholestatic liver cirrhosis (CLC) eventually proceeds to end-stage liver failure by mediating overwhelming deposition of collagen, which is produced by activated interstitial myofibroblasts. Although the beneficial effects of Rhus verniciflua Stokes (RVS) on various diseases are well-known, its therapeutic effect and possible underlying mechanism on interstitial fibrosis associated with CLC are not elucidated. This study was designed to assess the protective effects of RVS and its possible underlying mechanisms in rat models of CLC established by bile duct ligation (BDL). We demonstrated that BDL markedly elevated the serological parameters such as aspartate aminotransferase, alanine transaminase, total bilirubin, and direct bilirubin, all of which were significantly attenuated by the daily uptake of RVS (2 mg/kg/day) for 28 days (14 days before and after operation) via intragastric route. We observed that BDL drastically induced the deterioration of liver histoarchitecture and excessive deposition of extracellular matrix (ECM), both of which were significantly attenuated by RVS. In addition, we revealed that RVS inhibited BDL-induced proliferation and activation of interstitial myofibroblasts, a highly suggestive cell type for ECM production, as shown by immunohistochemical and semi-quantitative detection of a-smooth muscle actin and vimentin. Finally, we demonstrated that the anti-fibrotic effect of RVS was associated with the inactivation of Smad3, the key downstream target of a major fibrogenic cytokine, i.e., transforming growth factor b (TGF-b). Simultaneously, we also found that RVS reciprocally increased the expression of Smad7, a negative regulatory protein of the TGF-b/Smad3 pathway. Taken together, these results suggested that RVS has a therapeutic effect on CLC, and these effects are, at least partly, due to the inhibition of liver fibrosis by the downregulation of Smad3 and upregulation of Smad7.
폐주변부 병변 진단 시 기관지솔질에 의한 출혈이 병소확인에 주는 유용성 연구

Background : Diagnosis of peripheral pulmonary lesion by conventional bronchoscopy is still challenging. The sheathed bronchial brush has been used to collect samples from the visible endobronchial lesion. Recently developed endobronchial ultrsonography (EBUS). can obtain image of peripheral pulmonary lesion at bronchus suspected by bleeding sign of sheathed brush. This study evaluated availibility of detected lesions by bleeding after bronchial brushing in pheripehal pulmonary lesions before transbronchial lung biopsy Methods : Between August 2006 and November 2008, 50 patients(38 men and 12 women, median age, 65 yrs) whose peripheral pulmonary lesions could not be detected with flexible bronchoscopy were included in this study. Among 50 patients, 40 cases were malignant lesion(non-small cell carcinoma 35, small cell carcinoma 5), and 10 cases were benign lesions(inflammatory disease 7, others 3) Results : Of the 50 patients, lesions detected bleeding after bronchial brushing was 34 cases(68%) and lesions detected by EBUS was 33 cases(66%). Sensitivity was 90.0-95.4%, Specificity was 83.3-80%, Positive predictive value(PPV) was 83.3-95.4% and Negative predictive value(NPV) was 80-90.9%. Diagnostic rates in visualized lesion by EBUS and otal peripheral lesions were 75%(25/33), 58%(29/50) respectively. There was no significant complication related with bronchial brushing. Conclusions : Bleeding sign by bronchial brushing was safe and effective method to predict location of peripheral pulmonary lesion with sufficient diagnostic yield
