      • CD34+ 조혈 모세포 이식 2례

        Background: In most solid tumors, the CD34 antigen has not been detected, so positive selection of CD34+ cells may reduce tumor cell contamination and the CD34+ cells are capable of reconstituting hematopoiesis. We tried CD34+ cell transplantation in two patients. Method: CD34+ cells from chemotherapy + G-CSF mobilized PBPCs or bone marrow were positively selected with an avidin-biotin immunoadsorption column (CEPRATE SC system). Case 1. One course of chemotherapy using cyclophosphamide(200㎎/㎡) and etoposide (4.2g/㎡), combined with G-CSF(5㎍/㎏) S.C. was used in a relapsed lymphoma patient. This patient responded to the induction chemotherapy. CD34+ cells from harvested bone marrow were selected by the CellPro immunoadsorption column. The total number of mononuclear cells loaded onto the CellPro was 2.4×10^(8)/㎏, with 1.1% CD34+ cells. After column separation, the total number of positively selected cells was 5.16×10^(6)/㎏. The number of CFU-GM was 76.8×10⁴/㎏. This patient was treated with melphalan (140㎎/㎡) and TBI (1200cGy) and the positively selected CD34+ cells were infused. The time to neutrophil recovery greater than 0.5×10^(9)/L was 19 days and the time to platelet recovery greater than 50×10^(9)/L was 21 days. Case 2. Two courses of mobilizing chemotherapy were given 4 weeks apart using taxol(210㎎/m2) and adriamycin(60㎎/m2), combined with G-CSF(5㎍/㎏) S.C. in a breast cancer patients with 7 axillary node metastasis. CD34+ cells from each single leukapheresis product were selected by the CellPro immunoadsorption column. In the first collection, the total number of nucleated cells was 4.4×10^(8)/kg, with 0.42% CD34+ cells. In the second collection, the total number of nucleated cell was 2.8× 10^(8)/㎏ with 0.43% CD34+ cells. After colum separation, the total numbers of collected cells were 4.0×106/kg and 4.8×10^(6)/kg, the total number of CD34+ cells were 1.2×10^(6)/㎏ and 0.82×10^(6)/㎏. Colonogenic assays of positively selected CD34+ cells gave rise to myeloid erythroid, and multilineage colonies, with a median of 190 CFU-GM, 190 BFU-E, and 164 CFU-GEMM per 1×10³ adsorbed cells, respectively. High-dose chemotherapy with cumulative doses of 40mg/㎡ mitoxantrone, 750mg/㎡ thioptepa, and 1000mg/㎡ carboplatin was administered. Positively selected CD34+ cells were rapidly infused 24 hours after the end of high-dose chemotherapy. The time to neutrophil recovery greater than 0.5×10^(9)/L was 16 days and the time to platelet recovery greater than 50×10^(9)/L was 20 days.

      • Lignin Peroxidase 에 의한 Azo계 염료 분해

        Lignin peroxidase produced by white rot fungi Phanerochaete chrysosporium, can degrade aromatic compound such a lignin. In this study, the purified lignin peroxidase cultured from P. chrysosporium and commercial HrP as a control was used for determining the degradation ability a of azo dyes. In addition, the effect of immobilization matrix, pH, temperature, dye concentration and type have been studied. As a result, the order of azo dye degradation rate using HrP and LiP was, Congo red, Acid Yellow 9, Orange Ⅱ. LiP showed the highest degradation percentage at 35℃ if among tested reaction temperatures. LiP under the acidic condition was most activated and resulted in the highest degradation rate of azo dyes tested. However, LiP and HrP was unstable for pH 2.0 to 2.5. From the comparison of HrP and LiP for the degradation rate of azo dyes, LiP resulted in more degradation ability than HrP. The comparison of the former martial is more expensive than the latter best the degradation rate and immobilization percentage of both immobilization methods are the same.

      • 韓國 靑少年의 吸煙實態에 關한 硏究

        The purpose of this study lies in examining the smoking experience, smoking reason, difference of smoking status, satisfaction with family, school and friends, smoking extent by survival of parents, smoking extent by total income of family, smoking extent by study time after school between an academic highschool students and technical highschool students and tacking its solution plans. According to this aim, the study divided seoul to the North of the Han River and the South of that, and divided it to the East and the West then conducted questionnaire survey to the total 1,100 students of each 550 male junior students of an academic highschool and technical highschool, and analyzed 1,083 sheets. Research contents were examined using Frequency and Crosstabulation Analysis, ANOVA as to what relation the youth's home, friend and school life has according to school type (humanities line and technical line high schools), and major findings were as follows. (1) The total survey subjects were 1,083 persons, of them, the number of an academic highschool students were 544(50.2%), and that of technical highschool students were 539(49.8%). (2) As to the question of smoking experience, rate of technical school students (77.5%) was higher than that of an academic highschool students(60.7%), and as to the first smoking experience, according to statistics significant an academic highschool students was the highest in their 3rd grade of middle school(25.9%) and technical students was highest in their primary school days(26.5%), and then as to smoking reason, that 58.5% of the former students and 56.2% of the latter students was for contacting friends. according to statistics significant keep smoking status as 58.5% of academic highschool students 67.6% of technical students keep smoking. (3) As to the total family income of smoking academic highschool students, it was indicated that the smoking rate of students with high family income was exceedingly high by showing that 3.01 - 4 million won was 23.5%, 2.02 - 3 million won was 19.4%, 1.01 - 2 million was 17.3%, and less than 1 million won was 5.6% and as to that of smoking technical highschool students, it was indicated that 2,01 - 3 million won was 22.2%. 1.01 - 2 million was 21.4%, 3.10 - 4 million won was 19.4, less than 1 million won was 18.6% and over 4 million won was 18.4%, which showed that the family income of technical highschool students was even compared to academic highschool students. (4) As to the school record of currently smoking academic highschool students, it was indicated that the smoking rate of relatively lower ranking students was higher than that of upper ranking students by showing that 31st - 40th was 23.9%, 21st - 30th was 23.2%, 11th - 20th was 22.9% and below 41st was 18.0% And 1st - 10th was 11.9%, while as to the school record of technical line students, it was indicated that the smoking rate was relatively even regardless of school grades by showing that 31st - 40th was 27.5%, 11th - 20th was 19.8%, 1st - 10th was 19.3%, 21st - 30th was 18.8%, and over 41st was 14.5%, and as to the study hours after school, it was indicated that 31.4% of academic highschool students and 39.1% of technical students studied for about 1 hour.

      • 혈액투석중인 만성신부전 환자에서 골대사 지표로써의 Osteocalcin치

        Background : Serum osteocalcin is synthesized by osteoblast and has been shown to be sensitive indicator of bone turnover inpatients with various metabolic bone disease. In renal osteodystrophy, serum osteocalcin is elevated due to decreased renal clearance and elevated level of PTH. This study was done to evaluate the usefulness of serum osteocalcin as a marker of bone metabolism and the correlation with other biochemical markers of bone metabolism. Methods : We measured serum osteocalcin, calcium, phosphorus, ALP(alkaline phosphatase) and PTH(parathyroid hormone) in 37 patients with end stage renal disease on hemodialysis. Osteocalcin was determined by radioimmunoassay and PTH was determined by radioimmunometric assay. Results : 1) The mean level of serum osteocalcin in ESRD patients was 233.8± 218.2ng/ml which was significantly higher than that of controls(p<0.0001). 2) The mean level of serum PTH in ESRD patients was 40.5± 43.8pg/ml was significantly higher than that of controls(p<0.005). 3) There was a significant positive correlation between the level of serum PTH, ALP and the level of serum osteocalcin in ESRD patients. 4) By using multiple regression, PTH is most reliable factor that affect to elevated level of serum osteocalcin ( beta coefficient = 0.687, Sig T<0.05). Conclusion : Serum osteocalcin as a marker of bone metabolism in ESRD patients is more useful than other biochemical marker such as serum calcium, phosphorus, ALP and PTH is a most reliable factor that affect to elevated level of serum osteocalin.

      • 수포독성 처치용 외용제 제제설계를 위한 약물의 검색

        Abstract-The effects of various drugs on the alleviation of the symptoms of chemical burns were evaluated in mice to formulate topical antiblister preparations. After a chemical burn was induced on the mouse dorsal skin with 2-chloroethylethyl sulfide, the drug was applied on the disease site. The effectiveness of the drug was evaluated by determining blister size, necrosis score of skin and appearance of the chemical burns induced. It showed that steroids and aminoglycoside antibiotics had a tendency to protect skin cell, and antihistamines decreased the size of chemical burns. While oleaginous base resulted in deleterious effect, hydrophilic base didn't show a significant difference on the alleviation of the chemical burn symptoms compared to the control.

        MicroRNA Target Recognition: Insights from Transcriptome-Wide Non-Canonical Interactions

        MicroRNAs (miRNAs) are small non-coding RNAs (~22 nucleotides) regulating gene expression at the post-transcriptional level. By directing the RNA-induced silencing complex (RISC) to bind specific target mRNAs, miRNA can repress target genes and affect various biological phenotypes. Functional miRNA target recognition is known to majorly attribute specificity to consecutive pairing with seed region (position 2-8) of miRNA. Recent advances in a transcriptome-wide method of mapping miRNA binding sites (Ago HITS-CLIP) elucidated that a large portion of miRNA-target interactions in vivo are mediated not only through the canonical “seed sites” but also via non-canonical sites (~15-80%), setting the stage to expand and determine their properties. Here we focus on recent findings from transcriptome-wide non-canonical miRNA-target interactions, specifically regarding “nucleation bulges” and “seed-like motifs”. We also discuss insights from Ago HITS-CLIP data alongside structural and bio-chemical studies, which highlight putative mechanisms of miRNA target recognition, and the biological significance of these non-canonical sites mediating marginal repression.

        기업전략에 따른 경영자 성과-보상 민감도

        기업이 추구하는 전략에 맞는 경영자 유인보상제도의 운영은 경영자에게 전략실행에 대한 동기부여를 제공하여 기업 전략목표 달성에 긍정적인 영향을 미칠 수 있다. 본 연구는 기업이 추구하는 전략에 맞게 경영자 유인보상제도를 설계하여 운영하고 있는지를 살펴보기 위해 기업전략과 경영자 성과-보상 민감도 사이의 관련성을 확인해 보고자 하였다. 2001년부터 2010년까지 유가증권 시장에 상장된 기업을 대상으로 다중회귀분석을 통해 기업전략과 성과-보상 민감도 사이의 관련성을 분석한 결과, 먼저 원가우위 전략을 추구하는 기업의 경우 경영자 보상을 회계성과에 보다 민감하게 대응시키는 것으로 나타났다. 이는 원가우위 전략의 핵심은 제품가격 경쟁력 확보로 기업의 효율성 제고를 통한 원가절감이 무엇보다 중요하므로 경영자의 보상을 회계성과와 보다 밀접하게 대응시키는 것으로 판단된다. 반대로 제품차별화 전략을 추구하는 기업일수록 경영자 보상과 주식성과 사이의 민감도가 높게 나타났는데, 이는 제품차별화 전략을 추구하는 기업의 경우 시장에서의 제품차별화를 위해 연구개발 투자 활동 등 단기 회계성과에는 부정적인 영향을 미치나 미래 기업가치에 긍정적인 영향을 미칠 수 있는 활동이 보다 중요하므로, 경영자 보상을 주식성과와 보다 민감하게 대응시킴으로써 경영자에게 전략실행의 유인을 제공한다고 판단할 수 있겠다. 본 연구는 기업이 추구하는 전략실행에 초점을 맞추어 경영자 보상이 기업이 추구하는 전략에 맞게 설정되어 운영되고 있는지를 실증적으로 확인해 보았다는 데 연구의 의의를 둘 수 있다. This paper investigate the impact of firm’s strategy on the managers’ compensation -performance sensitivity. Specifically, We analyze compensation-accounting performance sensitivity is increased if the corporate pursuit cost leadership strategy and compensation-stock performance sensitivity is increased if the corporate pursuit product differentiation strategy. The purpose mentioned above are empirically tested using 3,569 firm-year data listed on the Korean Exchange(KRX) with multiple regression method. Summarizing the results of analysis as following; We find that managers’ compensation-accounting performance sensitivity is increased In the corporate to pursue a cost leadership strategy. Furthermore, we find that the corporate to pursue a product differentiation strategy increase the sensitivity of managers’ compensation-stock performance. This study contributes to the manager compensation literature by recognizing that corporate strategy influences the sensitivity of managers’ compensation-performance.

        이익유연화와 경영자 보상

        본 연구에서는 경영자 이익유연화 행위와 보상 사이의 직접적인 관련성을 양(+)의 이익유연화 기업과 음(-)의 이익유연화 기업으로 나누어 살펴보았으며, 이익유연화를 통한 경제적 효익이 클 것으로 예상되는 기업에서 경영자에게 보다 강력한 이익유연화를 유도하기 위해 더욱 높은 수준의 보상을 지급하는지를 실증분석 하였다. 2004년부터 2008년까지 280개 상장제조기업, 1400개 기업-연도를 자료를 바탕으로 다중회귀분석을 통해 실증분석한 결과를 요약하면 첫째, 기업의 이익유연화를 양(+)의 유연화와 음(-)의 유연화로 구분하지 않고 전체 표본기업을 대상으로 경영자보상과 이익유연화 사이의 관련성을 분석한 결과 기존의 선행연구와 동일하게 직접적인 관련성은 나타나지 않았으나, 표본을 양(+)의 이익유연화 기업과 음(-)의 이익유연화 기업으로 구분하고, 이익유연화 행위와 경영자 보상 사이의 관련성을 분석한 결과 양(+)의 이익유연화 기업의 경우 이익유연화와 경영자 보상 사이에 아무런 관련성을 확인할 수 없었으나, 음(-)의 이익유연화 기업의 경우 이익유연화 행위와 경영자 보상 사이에 통계적으로 유의한 양(+)의 관련성을 확인할 수 있었다. 둘째, 이익유연화를 통한 경제적 효익이 보다 클 것으로 예상되는 기업(현금흐름 변동성이 큰 기업, 기업의 파산가능성이 높은 기업)의 경우, 경영자 이익유연화 수준과 보상 사이에 매우 유의적인 양(+)의 관련성이 있는 것으로 나타났다. 이상의 연구결과는 이익유연화 방향에 따라 경영자 보상과 이익유연화 수준이 직접적인 관련성을 가질 수 있으며, 이익유연화를 통해 실질적인 효익이 보다 강력할 것으로 예상되는 기업의 경우 경영자에게 적극적인 이익유연화를 유도하기 위해 보다 높은 수준의 보상이 지급되는 것으로 해석할 수 있다. This paper verify whether managers receive incentive compensation for income smoothing. Specifically, after classifying firms according to income smoothing direction(positive income smoothing or negative income smoothing), we analyze how the size of income smoothing affects the level of managers' incentive compensation respectively. Also, we examine that such incentives are more likely to exist in firms where the benefits from smoothing income relatively high. We tests the above mentioned hypotheses using manufacture firm data listed on Korea Stock Exchange(KSE) from 2004 to 2008. Regression models are used to the test the relationship between income smoothing and managers' compensation. The results of this paper provide support for the hypotheses. First, the size of income smoothing significantly influence managers' compensation in negative income smoothing firms. But we can find no relationship between the size of income smoothing and managers' compensation in positive income smoothing firms. Second, if firms have higher benefits from income smoothing(higher cash flow volatility and possibility of bankruptcy), the size of income smoothing positively affects managers' compensation.
