        A cytochrome C - specific protein methylase Ⅲ was purified approximately 63 - fold from Saccharomyces cerevisiae. The unmethylated cytochrome C served as substrate for the enzyme. Horse heart apocytochrome C was found to be a better substrate than the native horse heart cytochrome C. This observation suggests that the in vivo methylation of cytochrome C in the yeast probably occurs before cytochrome C become attached to the mitochondria. In partly purified protein methylase Ⅲ the optimun pH is about 9.0, the enzyme activity is linearly proportional to the amount of enzyme protein up to about 30㎍ and also to time up to 15min. The Km values for horse heart cytochrome C and S - adenosyl - L- methionine were 1.33mM and 40㎛, respectively. S - adenosyl - L - homocysteine was found to be a competitive inhibitor for S - adenosyl - L - methionine, and the Ki value was 2.7 μM. The molecular weight of the enzyme was approximately 97,000, determined on sephadex S- 200 gel filtration. The regulation of cellular cytochrome C and the protein methylase Ⅲ are not coordinately related in a parallel manner.

      • 산성강하물의 침착량과 동태 해명에 관한 연구 -춘천과 서울 강우의 화학조성 비교 : Comparison of Chemical Composition of Rain Water between Chunchon and Seoul

        The rain water samples were collected at Chunchon and Seoul by using wet only automatic sampler from January 1996 through December 1997. The daily base rain water samples collected over than 95% rainy events during the whole period. The samples were measured both pH and conductivity, and were analysed major ionic components, SO₄^(2-), NO₃^(-), Cl^(-), NH₄^(+), Ca^(2+), Na^(+) and K ^(+), by ion chromatography. In 1996, about 77% sampled rain water showed below pH 5.6 and the 60% of rain water was lower than pH 5.0. The volume weighted average pH was 4.7 at all sites. In 1997, the volume weighted average pH was 4.6 and 4.9 at Seoul and Chunchon, respectively. Among the rain water samples, 87% and 55% of samples showed below than pH 5.6 and 5.0, respectively. The pH value of Chunchon was significantly (p<0.05) lower than Seoul at the rain samples for less than 20 ㎜ rainfall. However conductivity of the rain samples were 20.9 μS/㎝ for 1996 and 27.7 μS/㎝ for 1997 at Seoul, and 19.1 μS/㎝ for 1996 and 14.1 μS/㎝ for 1997 at Chunchon. H₂SO₄AND HNO₃CONTRIBUTED 65.9% and 29.6% of free acidity at Seoul, respectively. The ratio of [NO₃^(-)]/[nss-SO₄^(2-)] were 0.43 at Seoul and 0.51 at Chunchon for rain samples for less than 20 ㎜ rainfall. The annual wet deposition of Cl^(-), NO₃^(-), SO₄^(2-), H^(+), Na^(+), NH₄^(+), K ^(+), Mg^(2+) and Ca^(2+), respectively, 568.8 ㎏/㎢, 1489.3 ㎏/㎢, 3184.8 ㎏/㎢, 20.9 ㎏/㎢, 249.4 ㎏/㎢, 1091.2 ㎏/㎢, 189.8 ㎏/㎢, 90.2 ㎏/㎢, and 702.4 ㎏/㎢ at Seoul for 1996; 656.4 ㎏/㎢, 2029.7 ㎏/㎢, 3280.7 ㎏/㎢,27.2 ㎏/㎢, 229.4 ㎏/㎢, 1063.9 ㎏/㎢, 106.9 ㎏/㎢, 7802 ㎏/㎢, and 645.3 ㎏/㎢, at Seoul for 1997; 116.9 ㎏/㎢, 983.3 ㎏/㎢, 1797.0 ㎏/㎢, 21.4 ㎏/㎢, 83.2 ㎏/㎢, 648.1 ㎏/㎢, 78.0 ㎏/㎢, 22.2 ㎏/㎢ and 368.8 ㎏/㎢ at Chunchon for 1996; 100.2 ㎏/㎢, 1077.6 ㎏/㎢, 1754.0 ㎏/㎢, 13.4 ㎏/㎢, 146.0 ㎏/㎢, 602.3 ㎏/㎢, 88.8 ㎏/㎢, 16.2 ㎏/㎢ and 206.8 ㎏/㎢ at Chunchon for 1997.

        In the rectifying single sampling inspection, 9-sampling inspection methods are briefly described, and then the optimal inspection plan to minimize the expected total cost is derived. The results of this experiment through numerical example are: (1) The optimal inspection plan of sampling to minimize the cost with several variables is derived. The result is TC(134, 4)=2321. (2) The optimal inspection plan is obtained when the LTPD is increased, and the sample size, acceptance number and total cost are decreased. (3) The optimal inspection plan is obtained when the lot size is increased, and sample size and acceptance number are also increased. (4) When e₁ is fixed and e₂ is increased, p_(e) is decreased with very small erefore, the type -Ⅰ error must influences on sample size more than the type-Ⅱ error does.

      • 南部 畓地帶에서 發生하는 올챙이고랭이(Scirpus juncoides Roxb.)의 雜草生態的 硏究

        This study on the occurrence, regeneration and reproduction of the problematic weed, Scirpus juncoides Roxb., in the paddy field in the southern area was carried on for 12 months in order to investigate the various adaptability. The results obtained are as follows: 1. In O₂condition, a slightly insufficient condition treated by 0.02mm polyethylene film was the most favorable for germination. 2. In temperature, the more favorable condition for the germination of S.juncoides Roxb. seed was 35℃, 30℃, 25℃ in order, and the emergence rate of their stumps was high from 20℃ to 30℃. 3. In light intensity, germinating rate of S.juncoides Roxb. seed was higher in shade conditions than in perfect exposure, and so was the emergence rate of stump. 4. In light quality, germinating rate of S.juncoides Roxb. seed was high in order of black, yellow, green, and colorless film, while the emergence rate of stump was high in green film treatment. 5. In hydrogen-ion concentration of soil, the most optimun pH value for germination was 5.7, while in the extemely acid and alkaline soil, the growth was checked after the sixth week. 6. The emergence rate of stump was high in order of 3g, 4g, 5g, 2g, 1g in stump weight. 7. Initial growth of S.juncoides Roxb. shoot at two weeks after its cutting depended on underground nutrition. 8. Regeneration vigour of S.juncoides Roxb. shoot became dull since a growth period, and only one cutting after 25, May was enough to control S.juncoides Roxb. 9. Cutting of S.juncoides Roxd. shoot in the lagvegetative period around the end of May did not affected its regrowth. 10. In soil moisture content, the number of flower-bud was more in 100% saturated condition than in 80% saturated. In the submergence condition, 2cm water flooded depth was the most favorable for flower-bud formation of S.juncoides Roxb. 11. There was no constant tendency in soil condition and poor aeration condition was well adapted to S.juncoides Roxb. 12. As the shade rate increased, the flower-bud formation of S.juncoides Roxb. was suppressed. 13. The flower-bud formation of S.juncoides Roxb. was increased in yellow polyethylene film treatment. 14. with the MPK fertilizer level increased, the number of flower-bud was rapidly increased.

      • T 세포활성항원 CTLA-4의 기능에 관한 연구 : T 세포에서 표적세포 살해능과 CTLA-4 발현과의 연관성 Target Cytotoxicity of T Cell Correlates with CTLA-4 Production

        CTLA-4, a T cell activation antigen and a homologue of CD28, was originally identified as a gene by a series of reverse genetic approaches. While CD28 molecule has been characterized well as a stimulator of T-cell function via enhanced lymphokines production and stablilization of those mRNA, most of the functions of CTLA-4 remain unknown. It has been widely accepted that CTLA-4 functions as an immune suppressor which is down-regualting the function of CD28. We previously showed that 34 KD form of this antigen mainly expressed CD8+ subset, a cytotoxic or suppressor, of activated peripheral blood lymphocyte. Based on our previous finding this study was conducted to further characterize immunological function of CTLA-4 especially in terms of involvement of this molecule in T-cell effector function mediating target cell cytotoxicity. 4 human T cell clones with different target cytotoxicities were employed in this study. NBL46 (CD4+) and NBL77 (CD8+) were cytotoxic and NBL32 (CD4+) and NBL58 (CD 8+) were non-cytotoxic to target LBF cell in target cell chromium release assay. And in Western blot assay 34 kD antigen was detected in NBL46 (CD4+) and NBL77 (CD8+) clones but not in NBL32 (CD4+) and NBL58 (CD8+) clones. It was summarized that expression of the CTLA-4 antigen was associated with cytotoxicity but not with subset phenotypes of T cells. In conclusion CD8+ T subset of PHA-stimulated PBL was major and only CTLA-4 producer and this molecule was induced during mid to late stage of T cell activation. The cytotoxicity of T cell clones to its target cell was directly correlated with its CTLA-4 production and vice versa. And it was highly suggested that primary function of CTLA-4 may involved in T cell effector function which mediates target cell killing.

      • 간질환에서의 lipoprotein분획에 관한 연구

        There were studied on the chronic liver diseases, 38 patients for determination of lipoprotein fractions and serum lipids visiting to the Chunan Soonchunhyang Hospital. The results are obtained as followings: 1. The lipoprotein fractions in liver diseases were shown as:α-Lp 28.21±7.22%, pre-β-Lp 28.57±10.00%. β-Lp42.94±7.26%. 2. Pearson correlation between serum cholesterol and lipoprotein fractions was shown that chronic liver diseases are significant except β-Lp, and blood triglyceride with all lipoprotein fractions. 3. To compare between chronic liver diseases and lipoprotein fractions pre-β-Lp was only shown significantly positive. 4. From the above results chronic liver diseases were not determined from blood lipids and lipoprotein fractions.

        Tuberculosis of duodenum is so rare and is usually secondary to pulmonary or intestinal tuberculosis . It is difficult to diagnose duodenal tuberculosis in clinical and radiologic findings due to its non-specific symptoms and inconsistent lesion. The diagnosis was made on the basis of the histological study of the endoscopic biopsy and resected surgical specimen. so we report a case of tuberculosis of duodenal bulb and of ascending colon with, the brief review of the literatures.

        The Presence of Urinary Ketones according to Metabolic Status and Obesity

        Background: Recently, new concepts about obesity and normal weight subtypes with metabolic conditions are rising and ketone bodies are emerging as a significant indicator of metabolic health. This study aimed to find a relationship between ketonuria and those subtypes. Methods: The data of 19,036 subjects were analyzed in this cross-sectional study (2013–2017 Korea National Health and Nutrition Examination Survey, KNHANES). Based on body mass index and adult treatment panel III with modification of waist circumference, individuals were categorized into 4 groups: metabolically healthy normal weight (MHNW), metabolically healthy obese (MHO), metabolically unhealthy normal weight (MUNW), and metabolically unhealthy obese (MUO). Individuals were divided into 2 groups, positive and negative ketonuria groups, and the metabolic parameters were compared. Results: The metabolic indicators of the positive ketonuria group showed better results than those of the negative ketonuria group and the MHNW group showed the highest proportion of positive ketonuria. The MHNW group showed higher urinary ketones than the MUO group (odds ratio [OR], 0.391; 95% confidence interval [CI], 0.254–0.601) in men. In women, OR of having ketonuria was 0.698 (95% CI, 0.486–1.002) in the MHO group and 0.467 (95% CI, 0.226–0.966) in the MUNW group compared to the MHNW group, respectively. Conclusion: Compared to the MHNW group, the MUO group showed lower presence of ketonuria in men, and tendency to have less ketonuria in women.

      • 박의 RAPD 분석을 위한 PCR 최적 조건 연구

        This study was conducted to identify the best combinations of factors(template DNA concentration, MgCl2 concentration, and amount of taq polymerase) for the optimization of PCR in gourd. 3×3×3 factorial experiment for template DNA concentration, MgCl2 concentration, and amount of taq polymerase was conducted. Another factorial experiment for denature, annealing, extension temperature on the optimal PCR condition was also conducted. The most clear and reproducible bands were appeared in 40ng(1unit taq polymerase and 2.5mM MgCl2, and lunit taq polymerase and 4.5mM MgCl2, 1unit taq polymerase and 7.0mM MgCl2), 60ng(0.5unit taq polymerase and 2.5mM MgCl2, 0.5unit taq polymerase and 4.5mM MgCl2, 1unit taq polymerase and 2.5mM MgCl2, 1unit taq polymerase and 4.5mM MgCl2, and 1unit taq polymerase and 7.0mM MgCl2). The best MgCl2 concentration was 4.5mM(40ng template DNA and 1unit taq polymerase, 60ng template DNA and 0.5unit taq polymerase, and 60ng template DNA and lunit taq polymerase), and 7.0mM(40ng template DNA and 0.5unit taq polymerase DNA). The combinations which showed the best bands from the amount of taq polymerase were 0.5unit(60ng template DNA and 4.5mM MgCl2) and lunit(40ng template DNA and 2.5mM MgCl2, 40ng template DNA and 4.5mM MgCl2, 40ng template DNA and 7.0mM MgCl2, 60ng template DNA and 2.5mM MgCl2, 60ng template DNA and 4.5mM MgCl2, 60ng template DNA and 7.0mM MgCl2). When we consider results from template DNA concentration, MgCl2 concentration, and amount of taq polymerase, 1 condition(40ng of template DNA, 4.5mM MgCl2 and 1unit taq polymerase) was the best combination for the optimal PCR condition. Reaction temperatures for the optimal PCR condition were 84℃, 32℃, 62℃ and 86℃, 35℃, 64℃.
