Objective : Immunotoxin therapy is a novel approach for the treatment of tumor, and it has been successfully used in the central nervous system. The purpose of this study is to evaluate the cytotoxicity of OKT9 ScFv-Diphtheria toxin fusion immunotoxin on various human brain tumor cell lines. Methods : Immunotoxin which was composed of OKT9 ScFv and Diphtheria toxin was made. Its cytotoxicity on glioblastoma cell lines(U87MG, U118MG) and medulloblastoma cell line(TE671) was tested and compared with anti-cancer chemotherapeutic agents. And we also examined the relationship between its cytotoxicity and transferrin receptor expression. Results : It showed most cytotoxicity on U87MG cell line and nearly no effect on U118MG cell line, moderate cytotoxicity on TE671 cell line in sixteen hours exposure experiment. In continuous exposure experiment, it showed moderate cytotoxicity on U118MG cell line, but showed strong cytotoxicity on other cell lines comparable or higher than anti-cancer chemotherapeutic agents. The relationship between its cytotoxicity and transferrin receptor expression was tested using flow cytometry, but no direct relationship could be found. Conclusion : Collectively, the result shows the cytotoxic effects of OKT9 ScFv-Diphtheria toxin fusion immunotoxin against various human brain tumor cell lines in continuous exposure experiment. Therefore, we suggest that this immunotoxin could be developed as a potential immunotherapeutic agent in the treatment of various human brain tumor clinically.

The mechanism of the protective action of acetate against heat-inactivation of the L-amino acid oxidase (L-amino acid:oxygen oxidoreductase (deaminating, EC 1.4.3.2) of Crotalus adamanteus venom has been investigated. The entropy of activation for the heat-inactivation of the enzyme in the presence of acetate was 224 e.u. compared to 114 e.u. in its absence. The Km value of the enzyme for L-arginine was decreased from 3. 86 mM to 0. 77 mM in the presence of 330mM acetate. The Vmax was reduced from 227 to 161 ㎕O₂ consumed per hour per ㎎ of enzyme protein. These results are taken to mean that the protective action of acetate is by preventing the enzyme protein from assuming a less ordered state, at high pH, by combining at a site different from the substrate-binding site.

Testicular teratoma is characterized that has more than one germ cell layer in various stages of maturation and differentiation. The incidence of mature teratoma varies from 2 to 9% of all germinal testicular tumor. Only 3 cases of bilateral teratoma have been reported in the literature to date. The teratoma can occurs at all ages but is most common between the age of 15-35 year. It is relatively infrequently seen in childhood and infancy. In these age group, teratoma tends to be a benign. Recently, we experienced one case of bilateral testicular teratoma in 4 month-old infancy. We report a case of bilateral testicular teratoma found infancy with review of related literatures.

수산화나트륨 용액에서 니켈 표면에 형성되는 양극 산화막의 성질과 두께를 알기 위하여 反射率 측정과 楕圓偏光反射測定을 同時에 하는 實驗方法을 썼다. 높은 순도의 다결성 니켈을 연마한 후 부동화를 일으키는 피텐셜 범위 내의 여러 피텐셜에서 전기화학적으로 표면막이 생기도록 한 후, 자동화된 타원편광반사법측정기를 써서 반사율(R)과 타원편광반사법 파라미터 (△, Ø)들의 변화를 기록하였다. 이 기록된 실험치 들로 부터 표면 막의 두께와 광학상수(n, k)들을 계산한 결과 n=2.0∼2.5, k=0∼0.5, 두께=10∼20Å의 결과를 얻었다. 이러한 결과와 電氣化學的 측정치를 함께 고려하여 표면막은 Ni(OH)₂와 비슷한 조성을 가지되 일부는 탈수되어 NiO로 된 것으로 추정되었다.
