Potential Therapeutic Targets for the Primary Gallbladder Carcinoma: Estrogen Receptors
Zhang, Ling-Qiang,Zhang, Xiu-De,Xu, Jia,Wan, Yong,Qu, Kai,Zhang, Jing-Yao,Wang, Zhi-Xin,Wei, Ji-Chao,Meng, Fan-Di,Tai, Ming-Hui,Zhou, Lei,Liu, Chang Asian Pacific Journal of Cancer Prevention 2013 Asian Pacific journal of cancer prevention Vol.14 No.4
Gallbladder carcinoma, the most frequent malignant neoplasm of the biliary tract system, has always been considered to feature late clinical presentation and diagnosis, limited treatment options and an extremely poor prognosis. In recent years, while the incidence of gallbladder cancer has appeared to be on the increase, the available treatment methods have not greatly improved survival of the affected patients. Thus, exploring new therapeutic targets for this devastating disease is an urgent matter at present. Epidemical studies have demonstrated that the incidence of gallbladder carcinoma exhibits a distinct gender bias, affecting females two to three times more than males, pointing to crucial roles of estrogen. It is well known that estrogen acts on target tissues by binding to estrogen receptors (ERs), which are mainly divided into three subtypes, $ER{\alpha}$, $ER{\beta}$ and $ER{\gamma}$. $ER{\alpha}$ and $ER{\beta}$ appear to have overlapping but also unique even opposite biological effects. As important pathogenic mediators, ERs have been considered to relate to several kinds of tumors. In gallbladder carcinoma tissue, ERs have been shown to be positively expressed, and ERs expression levels are associated with differentiation and prognosis of this cancer. Nevertheless, the exact mechanisms of estrogen inducing growth of gallbladder carcinoma remain poorly understood. On the base of the current investigations, we deduce that estrogen participates in promotion of gallbladder carcinoma by influencing the formation of gallstones, stimulating angiogenesis, and promoting abnormal proliferation. Since ERs mediate the carcinogenic actions of estrogen in gallbladder, and therapy targeting ERs may provide new directions for gallbladder carcinoma. Therefore, it should be stressed that ERs are potential therapeutic targets for gallbladder carcinoma.

Surface decarburization behavior and its adverse effects of air-cooled forging steel C70S6 for automobile engine fracture splitting connecting rod were investigated comprehensively by mechanical properties, microstructure and fracture morphology analysis. The results show that the surface decarburization in the outer surface of the fracture splitting at the big end bore and the micro-cracks in the decarburized layer are result in the uneven and spalling fracture surfaces of the waster connecting rod product. Besides, partial decarburization is produced between 900 °C and 1250 °C for heating 2 h, and decarburization sensitivity reach maximum at 1150 °C, but no complete decarburization forms for heating 2 h at 650-1250 °C. The decarburized depth follows a parabolic law with the increase of the heating time from 0.5 h to 12 h, and the decarburization sensitivity coefficient is 2.05×10 -5 m·s -1/2 at 1200 °C. For the connecting rod manufacturing, surface decarburization must be under effective control during the hot forging process but not the control cooling process.

Drying at 100 C is a common practice for processing many biomasses for their further thermochemicalconversion via the routes such as pyrolysis. The drying temperature of 100 C is low, but might not be toolow to cause any impact on pyrolytic behaviors of a biomass feedstock. In this study, varied scenarios ofthermal pretreatment of poplar sawdust (oven-drying, boiling in water and microwave heating) at 100 Cwere screened for probing their potential influence on the pyrolytic behaviors at 500 and 750 C. Comparing with oven-drying, boiling in water (cooking) and microwave heating at 100 C could removesome oxygen-containing species, increasing carbon content of the pretreated sawdust and promotingcarbonization in pyrolysis. This enhanced formation of biochar, heavy p-conjugated organics in bio-oil,but suppressed generation of gas. The cooking or microwave heating pretreatment also promoted aromatizationto form the biochar of higher thermal stability, higher crystallinity, and higher aromatic degree, due to the enhanced deoxygenation reactions in the pyrolysis. The in-situ IR characterization suggestedthat microwave pretreatment promoted generation of unsaturated ketones and also the species with olefinicC = C, favoring their further aromatization. The cooking pretreatment impacted transformationroutes of the oxygen-containing organics, thus modifying pyrolysis behaviors of the pretreated sawdust.

The emergency of surface defect would significantly influence the quality of MPCG (Mobile Phone Cover Glass). Therefore, efficient defect detection is highly required in the manufacturing process. Focusing on the problem, an automatic detection system is developed in this paper. The system adopts backlight imaging technology to improve the signal to noise ration and imaging effect. Then, a modified segmentation method is presented for defect extraction and measurement based on deep neural networks. In the method, a novel data generation process is provided, with which the drawback that huge amount of data is required for training deep structured networks can be overcome. Finally, experiments are well conducted to verify that satisfactory performance is achieved with the proposed method.

Diabetic nephropathy (DN) is an important diabetic complication, and podocyte apoptosis plays a critical role in the development of DN. In the present study, we examined the preventive effect of the total flavone glycosides of Flos Abelmoschus manihot (TFA) on urinary microalbumin and glomerular podocyte apoptosis in experimental DN rats. The preliminary oral administration of TFA (200 mg/kg/day) for 24 weeks significantly decreased the urinary microalbumin to creatinine ratio and 24-h urinary total protein in streptozotocin-induced DN rats. Terminal deoxynucleotidyl transferasemediated dUTP nick end-labeling assay indicated glomerular cell apoptosis in DN rats was significantly improved by pretreatment with TFA. Furthermore, fluorescence-activated cell sorting and Hoechst 33342 staining suggested preincubation with hyperoside (50 and 200 lg/mL), the major active constituent of TFA, could significantly mitigate cultured podocyte apoptosis induced by the advanced glycation end-products (AGEs). Western blot analysis showed that increased caspase-3 and caspase-8 expressions induced by AGEs were also inhibited by pretreatment with hyperoside at both doses. Our results demonstrate that TFA pretreatment can decrease urinary albumin excretion in early-stage DN, which might be accomplished by preventing renal damage and podocyte apoptosis.

Triple-negative breast cancer (TNBC) is the most aggressive subtype of breast cancer and current therapeutic strategies are limited in their effectiveness. The expressions of Rab5 and the M2 tumor-associated macrophage marker CD163 in tissues were detected by Western blot. The migration and invasion of cells were determined using a Transwell assay. The expressions of the exosome markers were evaluated by Western blot. The polarization of human macrophages (THP-1) was determined by incubation of THP-1 cells with conditioned medium or exosomes collected from MDA-MB-231 cells with indicated transfections or by a coculture system of THP-1 and MDA-MB-231 cells. The M1 and M2 macrophage markers were evaluated by qRT-PCR. The expression of Rab5 in TNBC was significantly higher than that in normal breast tissue. Rab5 expressions in triple-negative and luminal A breast cancer were higher than those in other molecular subtypes. Higher CD163 expression was observed in triple-negative breast cancer and in triple-negative and luminal B subtypes. Rab5 knockdown suppressed but Rab5 overexpression promoted the migration and invasion capacity of MDA-MB-231 cells. The levels of CD63 and CD9 in the medium of Rab5 knockdown cells were lower than those in control cells, whereas higher levels of CD63 and CD9 were observed in Rab5 overexpression cells. Rab5 knockdown decreased the excretion but did not alter the diameter of the exosomes. Knockdown of Rab5 facilitated the anti-tumor polarization of macrophages, which was partially reversed by Rab5 overexpression. Therefore, Rab5 is expected to be a potential therapeutic target for triple-negative breast cancer.
