      • Aberrant Expression of Markers of Cancer Stem Cells in Gastric Adenocarcinoma and their Relationship to Vasculogenic Mimicry

        Background: Gastric cancer is the second leading cause of cancer-related death in Asia, and the majority type is gastric adenocarcinoma (GAC). Most GAC patients die of recurrence and metastasis. Cancer stem cells (CSCs) have been thought to be responsible for the initiation, development, metastasis, and ultimately recurrence of cancer. In this study, we aimed to investigate expression and clinical significance of CSCs markers, CD133 and Lgr5, and vasculogenic mimicry (VM) in primary GAC. Materials and Methods: Specimens from 261 Chinese patients with follow-up were analyzed for CD133, Lgr5 protein expression and VM by immunohistochemical and histochemical staining. The Pearson Chi's square test was used to assess the associations among the positive staining of these markers and clinicopathological characteristics. Postoperative overall survival time was were studied by univariate and multivariate analyses. Results: In GAC tissues, positive rates of 49.0%, 38.7%, and 26.8% were obtained for CD133, Lgr5, and VM, respectively. The mean score of microvessel density (MVD) was $21.7{\pm}11.1$ in GAC tissues. There was a significantly difference between the positive and negative groups. There was a positive relationship between the VM, the expression of CD133 and Lgr5, and the score of MVD and the grades of tumor, lymph node metastasis, TNM stages (all p<0.05). The overall mean survival time of the patients with CD133, Lgr5, VM, and MVD (${\geq}22$) positive expression was lower than that of patients with negative expression. The score of MVD, positive expression of CD133 and VM were independent prognostic factors of GAC (p<0.05). Conclusions: VM, and expression of CD133, Lgr5, and the score of MVD are related to grades of tumor, lymph node metastasis, TNM stages, and overall mean survival time. It is suggested that CSCs and VM could play an important role in the evolution of GAC.

        Detection and Characterization of PCR-SSCP Markers of the Bovine Lactoferrin Gene for Clinical Mastitis

        A total of 80 cows, including 40 top mastitis resistant and 40 top mastitis susceptible animals as Group I and Group II, were selected from a population of 520 cows based on clinical mastitis occurrence. PCR-SSCP analysis on four fragments within the 5'region and two fragments of Exons 4,15 of bovine lactoferrin (bLF) revealed that four fragments-P1,P4,E4,E15-had polymorphisms which totally included six base mutations, and only two of them had significant differences in allele frequencies between resistant and susceptible groups, P1 (53.7% vs. 70.0%, p<0.05) and P4 (55.0% vs. 68.8%, p<0.05). Further study on these two promising markers combined with the milk performance traits of cows demonstrated that their selection would result in higher fat percentage (p<0.05), lower Somatic Cell Score (SCS) (p<0.05) and Clinical Mastitis Residuals (CMR) (p<0.01) indicating higher mastitis resistance and lower milk yield (p<0.05). The putative transcription factor binding sites in the 5'region were also studied by using MatInspector 7.2.2 software, and two signal pathways regulating the expression of bLF including the NF-${\kappa}B$ pathway and nuclear hormone receptor pathway were predicted.

      • Study on shear fracture behavior of soft filling in concrete specimens: Experimental tests and numerical simulation

        In this paper, the shear behavior of soft filling in rectangular-hollow concrete specimens was simulated using the 2D particle flow code (PFC2D). The laboratory-measured properties were used to calibrate some PFC2D micro-properties for modeling the behavior of geo-materials. The dimensions of prepared and modeled samples were 100 mm×100 mm. Some disc type narrow bands were removed from the central part of the model and different lengths of bridge areas (i.e., the distance between internal tips of two joints) with lengths of 30 mm, 50 mm, and 70 mm were produced. Then, the middle of the rectangular hollow was filled with cement material. Three filling sizes with dimensions of 5 mm×5 mm, 10 mm×5 mm, and 15 mm×5 mm were provided for different modeled samples. The parallel bond model was used to calibrate and re-produce these modeled specimens. Therefore, totally, 9 different types of samples were designed for the shear tests in PFC2D. The shear load was gradually applied to the model under a constant loading condition of 3 MPa (σc/3). The loading was continued till shear failure occur in the modeled concrete specimens. It has been shown that both tensile and shear cracks may occur in the fillings. The shear cracks mainly initiated from the crack (joint) tips and coalesced with another one. The shear displacements and shear strengths were both increased as the filling dimensions increased (for the case of a bridge area with a particular fixed length).

      • Risk Effects of GST Gene Polymorphisms in Patients with Acute Myeloid Leukemia: A Prospective Study

        Glutathione S-transferase (GST) enzyme levels are associated with risk of many cancers, including hematologic tumours. We here aimed to investigate the relationships between GSTM1, GSTT1 and GSTP1 polymorphisms and the risk of AML. Genotyping of GSTs was based upon duplex polymerase-chain-reactions with the confronting-two-pair primer (PCR-CTPP) method in 163 cases and 204 controls. Individuals carrying null GSTT1 genotype had a 1.64 fold risk of acute leukemia relative to a non-null genotype (P<0.05). A heavy risk was observed in those carrying combination of null genotypes of GSTM1 and GSTT1 and GSTP1 Val allele genotypes when compared with those carrying wild genotypes, with an OR (95% CI) of 3.39 (1.26-9.26) (P<0.05). These findings indicate that genetic variants of GST and especially the GSTT1 gene have a critical function in the development of AML. Our study offers important insights into the molecular etiology of AML.

      • PLAXIS 3D simulation, FLAC3D analysis and in situ monitoring of Excavation stability

        Near-surface excavations may cause the tilting and destruction of the adjacent superstructures in big cities. The stability of a huge excavation and its nearby superstructures was studied in this paper. Some test instruments monitored the deformation and loads at the designed location. Then the numerical models of the excavation were made in FLAC3D (a threedimensional finite difference code) and Plaxis-3D (a three-dimensional finite element code). The effects of different supporting and reinforcement tools such as nails, piles, and shotcretes on the stability and bearing capacity of the foundation were analyzed through different numerical models. The numerically approximated results were compared with the corresponding in-field monitored results and reasonable compatibility was obtained. It was concluded that the displacement in excavation and the settlement of the nearby superstructure increases gradually as the depth of excavation rises. The effects of support and reinforcements were also observed and modeled in this study. The settlement of the structure gradually decreased as the supports were installed. These analyses showed that the pile significantly increased the bearing capacity and decreased the settlement of the superstructure. As a whole, the monitoring and numerical simulation results were in good consistency with one another in this practically important project.

      • "Farming" in osteosarcoma

        Osteosarcoma may be the result of malignant transformation of mesenchymal stromal cell and characterised by direct formation of osteoid or bone. They are morphologically heterogenous with histological diversity. They can therefore be classified be classified into fibroblastic (spindle cell predominant), osteoblastic (bone predominant), chondroblastic (cartilage or chondroid differentiation) or telangiectatic (vascular differentiation). This heterogenicity may reflect the various developmental stages of single cell strain or the mutation of single cell. Being cancer cell, they have escaped from normal cell cycle regulation. They invade and colonise territories with unregulated growth. They have predilection site in metaphyseal of long bone(e.g. distal femur and proximal humerus) arising de no-va as primary osteosarcoma of secondary to pre-existing conditions (Paget's disease, multiple enchondroma, bone infarct, bone irradiation, etc). They can also occur in extraskeletal tissue. The tissue that have high growth and reparative process appear to form an excellent environment or "soil" for the invasive osteosarcoma to develop with its uncontrolled growth and ultimately metastases to other organs.

      • A Low Power Consumption Frequency Adaptation Mechanism Based on the Traffic and Implementation on NetFPGA

        Both in academic and industrial research, building a green energy-saving network has become a hot topic. Combined with hardware division and adaptive algorithm, this paper proposes an adaptive frequency control mechanism based on traffic to reduce power consumption. By calculating real-time traffic, the network device can adjust its operating frequency. On the premise of maintaining network performance, this mechanism could reduce the power consumption significantly. Besides, the adaptive frequency mechanism has been verified and realized in the NetFPGA Reference Router. Experimental data show that under different ranges of traffic loads and without affect its performance, the Router embedded this mechanism can reduce power consumption by more than 20%.

        The effect of radial cracks on tunnel stability

        The surrounding rock mass contains cracks and joints which are distributed randomly around tunnels, and in the process of tunnel blasting excavation, radial cracks could also be induced in the surrounding rock mass. In order to clearly understand the impact of radial cracks on tunnel stability, tunnel model tests and finite element numerical analysis were implemented in this paper. Two kinds of materials: cement mortar and sandstone, were used to make tunnel models, which were loaded vertically and confined horizontally. The tunnel failure pattern was simulated by using RFPA2D code, and the Tresca stresses and the stress intensity factors were calculated by using ABAQUS code, which were applied to the analysis of tunnel model test results. The numerical results generally agree with the model test results, and the mode II stress intensity factors calculated by ABAQUS code can well explain the model test results. It can be seen that for tunnels with a radial crack emanating from three points on tunnel edge, i.e., the middle point between tunnel spandrel and its top with a dip angle $45^{\circ}$, the tunnel foot with a dip angle $127^{\circ}$, and the tunnel spandrel with $135^{\circ}$ with tunnel wall, the tunnel model strength is about a half of the regular tunnel model strength, and the corresponding tunnel stability decreases largely.
