        물리적 처리에 의한 강력분 밀가루 Gliadin의 항원성 변화

        본 연구에서는 가압가열 및 microwave 처리가 gliadin의 항원성에 미치는 영향을 살펴보기 위해 강력분에 가압가열과 microwave를 단독 또는 병행으로 처리하여 Ci-ELISA, SDS-PAGE 및 immunoblotting을 실시하였다. 가압가열 처리의 경우 시간이 길어질수록 IgG와의 결합력이 감소하였으며, 특히 50분 처리구에서 약 87%로 가장 낮은 결합력을 보였다. 또한 SDS-PAGE와 immunoblotting 결과에서도 무처리구에서 강하게 보였던 gliadin band가 가압가열처리에 의해 거의 소실되고 항체와 반응하지 않았다. 가압가열 및 microwave를 병행 처리 시도 마찬가지로 gliadin의 결합력이 감소하였으며, 처리구 중 가압가열 50분, microwave 5분 처리구에서 약 93%로 가장 낮은 결합력을 보였다. 반면 microwave를 단독으로 처리한 경우에는 일부 단백질의 변화는 관찰되었으나, 항원성 감소에는 효과가 없음을 확인하여 단백질 변화가 항원성에는 큰 효과를 준 것 같지 않다. 이상의 결과를 통해 가압가열 단독 처리 및 가압가열 및 microwave의 병행처리 시 gliadin의 항원성이 감소함을 확인하였다. This study was conducted to evaluate the effects of physical treatments on the antigenicity of gliadin in strong wheat flour. Strong wheat flour was treated with an autoclave (5, 10, 30, 50 min), a microwave (1, 5, 10 min), or both (10, 30, 50 min/ 5, 10 min), followed by SDS-PAGE, immunoblotting, and Ci-ELISA using anti-gliadin IgG. The results indicated that the binding ability of IgG to gliadin in strong wheat flour slightly decreased after autoclaving or autoclaving/microwaving. In particular, the binding ability was reduced to about 87% after autoclaving for 50 min and to 89% after autoclaving/microwaving (50/5 min). In addition, gliadin bands in the 50 min autoclaved group disappeared in both SDS-PAGE and immunoblotting. On the other hand, the antigenicity of gliadin was unaffected by microwaving alone. In conclusion, the results of this study suggest that autoclaving may reduce the antigenicity of gliadin in strong wheat flour.

      • VIP를 분비하며 신경절신경종의 분화를 가지는 갈색 세포종 1예

        갈색 세포종은 신경내분비종양의 하나로 VIP를 포함한 다양한 신경내분비 펩타이드를 분비할 수 있다. 저자들은 만성 설사와 체중 감소를 주소로 내원한 환자에서 좌측 부신에 VIP를 생성하는 갈색 세포종을 진단하여 성공적으로 제거한 후 증상이 모두 호전된 1예를 경험하였기에 문헌고찰과 함께 보고하는 바이다. Vasoactive intestinal peptide (VIP) is a 28-amino acid peptide, which may cause secretory diarrhea by stimulating the production of adenylate cyclase. Neuroendocrine tumors, secreting vasoactive intestinal peptide (VIP), are almost always of a pancreatic in origin. However, a pheochromocytoma may produce several neuropeptides, containing VIP, as they are considered to be neuroendocrine tumors. A 57-year-old woman, who presented with chronic watery diarrhea, hypokalemia, weight loss and a left adrenal mass, is described. Histologically, the tumor was diagnosed as a pheoch-romocytoma, with ganglioneuronal differentiation, and was histochemically confirmed to produce a vasoactive intestinal polypeptide. A left adrenal VIP-producing pheochromocytoma was successfully resected. After surgery, her diarrhea subsided and the electrolytes, affected neuroendocrine hormone levels, blood pressure and blood sugar level were normalized (J Kor SOC Endocrinol 18:227-231, 2003).

      • Electromagnetic wave absorption properties of multistacked hexaferrite-epoxy composites

        We report on electromagnetic wave (EM) absorption properties of multistacked hexaferrite-epoxy composites. Co-Ti substituted M-type hexaferrite powders, SrFe<sub>12-2x</sub>Co<sub>x</sub>Ti<sub>x</sub>O<sub>19</sub> (x=1.1, 1.2, 1.3) were prepared by conventional solid-state reaction method. Each composition of powder was calcined at 1300°C for 2 hours in air. The calcined powders were ground into fine powders, mixed with 10 wt% of epoxy binder, pressed to form toroidal-shaped (inner and outer diameters of 3.04 and 7.00 mm, respectively) green compacts, and finally cured at 180°C in air for 20 min. The real and imaginary parts of permittivities (ε', ε") and permeabilities (μ', μ") were measured on the toroidal shaped composites using a vector network analyzer (VNA) with an airline kit in the frequency range from 0.1 to 18 GHz. RL spectra for the composites were calculated based on transmission line theory and measured ε', ε" μ', μ" data. Also, the RL spectra were measured from S-parameter method using the VNA with airline kit and one-port Cu threaded hole socket. The minimum RL value (RL<sub>min</sub>) < -20 dB could be obtained for the series of composites samples at the optimal thickness. The frequencies of RL<sub>min</sub> were 14 GHz, 10 GHz, and 6 GHz for x = 1.1, 1.2, and 1.3 samples, respectively. The RL spectra were also measured on the stacked three samples (A, B, C) with varying stacking orders as ABC, ACB, BAC, BCA, CAB, CBA. The RL spectra were slightly varied depending on the stacking order. However, it was very far from the sum of RL spectra in each sample. The measured RL spectra of the stacked samples were well fitted with calculated RL spectra at total thickness of three samples. Here, the RL calculation was based on volume-averaged ε', ε" μ', and μ" values for each sample.

        제 2형 콜라겐에 의해 경구관용 유도된 DBA/1 mice에서의 세포면역반응

        Objective: To investigate the dosage of bovine type II collagen (BnCII) for the induction of oral tolerance in CIA animals, and to verify the changes of immune response and TGF-β production of mesenteric lymph node cells in tolerized CIA animals. Methods: Oral tolerance was induced by feeding of variable doses (5㎍, 10㎍, 20㎍ and 40㎍) of BnCII to DBA/1 mice 4 times per week during 2 weeks, and control mice were given ovalbumin (1000㎍), before immunization. We examed clinical assessment; incidence of arthritis, severity of arthritis, arthritic limb by visual analysis. IgG antibodies to BnCII were measured by ELISA, T cell responses to BnCII and PHA were quantified by antigen (CII)-induced 3H-thymidine incorporation into lymphocytes of mesenteric lymph node, draining lymph node, and spleen. TGF-β in supernatants obtained from lymph node culture medium was measured by ELISA. Results: Arthritis limbs were observed in 100% of control at 5 weeks after subcutaneous BnCII injection. The incidences of CIA in all tolerized group were significantly lower than that in control 5 weeks after immunization (control 100% vs. 5㎍ feeding group: 50%, 10㎍ feeding group: 50%, 20㎍ feeding group: 50%, 40㎍ feeding group: 55.5%, P<0.01). In comparison to control, mean articular indices were lower in all tolerized groups (control 5.13: 5㎍ feeding group 3.50, 10㎍ feeding group 2.75, 20㎍ feeding group 2.87, 40㎍ feeding group 2.63, P<0.05). Arthritic limbs were also significantly lower in tolerized groups (control 58.3: 5㎍ feeding group 20.8, 10㎍ feeding group 16.7, 20㎍ feeding group 20.8, 40㎍ feeding group 20.8, P<0.05). The titers of IgG antibody to CII were lower in tolerized group than that in control [tolerized group; median 10 (min. 0, max. 48), control; median 33 (min. 8.6, max. 101), P<0.05]. The proliferative responses to BnCII were significantly suppressed in tolerized (control 8010±2319cpm, tolerized group 4500±2060cpm, P<0.01). High TGF-β production was noted in tolerized group (control; 28pg/ml, BnCII feeding group; 73pg/ml). Conclusion: Oral tolerance in DBA/1 mice was successfully induced from low doses of BnCII (5㎍) and suppressed T and B cell function in conjunction with increased TGF-β production may play an important role for the induction of CII induced oral tolerance in DBA/1 mice.

      • 리튬 이온 이차전지를 위한 부극용 Si 특성에 관한 연구

        리튬이온이차전지의 부극으로 Si를 사용하였다. 부극용 Si전극을 1회 충방전 시 Li이온이 탈 삽입될 때 각각의 전압에서 XRD 회절분석을 통하여 Li이온이 이동하여 Si와 반응하는 것을 조사하였다. 순환전압전류 및 충방전 시험을 통하여 전기 화학적 특성을 측정해본 결과 부극으로써 Si의 산화환원 반응이 양호하게 이루어졌으며 충방전에 대한 cycle number가 증가할수록 비가역적인 산화환원반응 및 부피변화로 인해 싸이클 특성 감소함을 나타내었다. Si with anode is used Li ion secondary batteries During Li ion intercalating at first cycle, XRD analysis studies at others voltages showed that Li ions are moved and reacted to Si The electrochemical properties of Si anode materials were evaluated by cyclic voltammetry and charge/discharge test. With cycle number increasing, cyclic characteristics are decreased for irreversible reduction/oxidation and volume change.

      • 비타민 C 섭취량과 arterial Pulse Wave Velocity와의 연관성

        Purpose: Several studies suggested that enhanced lipid peroxidation is associated with atherogenesis and cardiovascular disease. Also, many observational studies showed that a high dietary intake of antioxdant vitamins are associated with reduced risk of cardiovascular disease. So we investigated the association vitamin C intake and arterial Pulse wave velocity(aPWV), which assessing aortic arterial stiffness. Subjects & Methods: The 783 adults were recruited into the this cross-sectional study. We exluded subjects who had a history of vascular disease or who were aking medication fot dyslipidemia or vitamin C supplements. Allsubjects examined body mass index (BMI), lipid profile, hs-CRP, uric acid, cystatin C, Homocystein, fasting glucose, and aPWV. aPWV was measured between brachium and ankle by automatic noninvasive apparatus. And daily calorie intake and vitamin C intake examined by food frequency questionaire. Statistical analysis obtained by SPSS. Results: The study consisted of 58% men and 42% women. Vitamin C index was significantly negative correlated with baPWV (P<0.05). Also, homocystein, cystatin C, uric acid, triglyceride and fasting glucose were inverse relationship with vitamin C index (P<0.05). In addition, low vitamin C intake is associated with high baPWV.

      • 가와사끼병에서 면역글로불린 사용 후 ASO 수치의 변동에 관한 고찰

        Objective : Since Kawasaki disease was first described in 1967, the cause and prognostic factors have not been known despite many researches and efforts. The introduction of high-dose intravenous gamma-globulin (IVGG) was an epoch and IVGG is now a standard therapy. But the mode of action of IVGG in reducing inflammatory response is not clearly understood. Understanding of the mechanism should supply the clues that indicate the cause, prognosis, the better treatment of this disease. At present, the correlation of CRP, ESR and change of leukocyte numbers to Kawasaki diseases were known, but the correlation studies of ASO titers are rare. To discover this, we evaluated the interrelationship between the ASO titer and IVGG therapy. Methods and Materials : The subjects included 35 patients with Kawasaki disease who were treated by IVGG at department of pediatrics, Inje university, Busan Paik hospital from June, 2004 to December, 2005. Before and after IVGG administration, ASO titers were evaluated. Results: In the 32 among 35 patients, ASO titers checked before IVGG administration were increased prominently after IVGG therapy. Conclusion : Remarkably increased ASO titers after IVGG administration for the treatment of Kawasaki diseases were found. And changes of ASO titers seem to be due to immunoglobulin instead of streptococcal infection. Further studies will be necessary to clarify the pathogenic mechanisms of increased ASO titers.

      • Rowe Cell 을 利用한 粘土試料 샘플링의 攪亂程道 推定

        Recently many studies which related to effective sampling method have been progressed and valuable data accumulated in foreign country. But, domestic technique is insufficient to evaluate the degree of disturbance so it is difficult to seek soli parameter. Thus the grasp of the degree of disturbance is effential to increase accuracy of reasonable parametern from laboratory tests. Also, the degree of disturbance of it was determined. Oedometer tests and Rowe cell consolidation rests reappeared in -situ state were conducted. And its result was compared and analyzed to deduces the degree of disturbance. From these tests, It can be inferred that the compression index generally is not affected and swelling index(C )and preconsolidation load (P )are increased because of the disturbance of soil. Also, it can be considered that consolidation curves given by two tests is little affected from desturbance. Conclusively, the results of two tests indicated that void ratio(e) and swilling index is independent of the degree of disturbance and it is reasonable to evaluate the degree of disturbance through it.

        한국 성인을 대상으로 한 전산화 Standard Progressive Matrices의 표준화 예비연구

        연구목적 : 전산화 Standard Progressive Matrices(SPM) 검사의 한국인 성인 표준자료를 확립함으로써, 간이로 지능을 평가하는 경우에도 보다 정확하게 지능지수를 추정하기 위해서 본 연구를 수행하였다. 방 법 : 18세 이상의 남녀 정상인 자원자 353명을 대상으로 전산화 SPM 검사를 시행하고, 백분율 및 표준점수(T-점수)를 기준으로 하여 표준화 자료를 작성하였다. 전체 대상군을 성별, 연령별로 구분한 후 평균과·분포가 유사한 집단은 군을 합치는 방법을 사용하였다. 한국인 표준자료에 근거하여 추정한 SPM 지능지수와 외국인 표준자료에 근거하여 추정한 SPM 지능지수를 각각 KWAIS 지능지수와 비교함으로써 타당도를 검증하였다. 결 과 : 18∼30세, 31∼40세, 41∼50세, 51세 이상의 연령군으로 구분된 한국 정상 성인의 SPM 표준화 자료를 산출하였다. 본 연구 결과에 의한 표준화 자료를 적용하여 지능지수를 추정하는 것이 현재 사용하고 있는 외국인의 표준화 자료를 이용하는 것보다 KWAIS 지능지수에 더욱 근접한 결과를 보였다. 결 론 : 18세에서 50세까지의 한국인 남녀에게는 간이지능평가도구로서 전산화 SPM 검사의 한국인 표준화 자료를 유용하고 타당하게 적용할 수 있음을 확인하였다. Objectives : We conducted this study to provide standardized data of the computerized Standard Progressive Matrices(SPM) test in Korean adults. Methods : The computerized SPM test was administered to 353 healthy volunteers aged 18 years over. We provided the standardized data(percentile and standardized T-scores) for the groups with similar mean and distribution of SPM scores, which was originally divided by age and sex. The validity of the new standardized data was tested by comparing IQs estimated by SPM and K-WAIS. Results : The standardized SPM data were provided for four age groups : 18 to 30 years, 31 to 40 years, 41 to 50 years, 51 years and over. IQ estimated by this standardized data tends to be closer to IQ by K-WAIS than the IQ estimated by foreign normative data. Conclusion : The standardized data of the computerized SPM were proven to be a useful and valid tool for measuring IQ briefly, compared with the conventional way of measuring IQ.
