      • CdSe@ZnS/ZnS quantum dots loaded in polymeric micelles as a pH-triggerable targeting fluorescence imaging probe for detecting cerebral ischemic area

        <P><B>Abstract</B></P> <P>High photoluminescence (PL) quantum yield (QY), photostability CdSe@ZnS/ZnS core/multishell quantum dots (CM-QDs) were first applied for bioimaging. The solubility, stability and biocompatible of the fluorescence imaging probes were constructed by self-assembly of CM-QDs and pH-responsive methoxy poly(ethylene glycol)-poly(β-amino ester/amidoamine)-dodecylamine (mPEG-PAEA-DDA) multiblock copolymers. The resulting CM-QDs-loaded mPEG-PAEA-DDA micelles (CM-QDs-PEG-PAEA-DDA) exhibited lower cell cytotoxicity and higher fluorescence intensity than the core/shell CdSe@ZnS QDs-encapsulated mPEG-PAEA-DDA micelles (CS-QDs-PEG-PAEA-DDA). Moreover, the in vivo fluorescence imaging ability confirmed that the CM-QDs-PEG-PAEA-DDA can be employed as a pH-triggerable targeting imaging probe for detection of a rat bearing cerebral ischemia disease.       • T-SPOT.TB for Detection of Tuberculosis Infection among Hematological Malignancy Patients and Hematopoietic Stem Cell Transplant Recipients

        The diagnosis of latent Mycobacterium tuberculosis infection (LTBI) is recommended in hematological malignancy patients and before hematopoietic stem cell transplantation (Guidelines for the prevention and management of infectious complications of solid organ transplantation, 2004). Compared to traditional methods such as tuberculin skin test (TST), T-SPOT.TB has been shown to be more specific. In the present study we enrolled 536 patients for whom T-SPOT.TB was performed, among which 295 patients also received the TST test. The agreement (79%) between T-SPOT.TB and TST was poor (x=0.274, P<0.001). The patients with positive T-SPOT.TB results numbered 62 (11.6%), in which only 20 (48.8%) of the 41 receiving the TST test had positive results. A majority of the patients with T-SPOT.TB positive results had some other evidence ofTB, such as TB history, clinical symptoms and an abnormal chest CT scan. Active TB was found in 9 patients, in which 2 had negative TST results. We followed up the patients and no one developed active TB. Our study suggested that the T-SPOT.TB may be more useful for screening LTBI and active TB in hematological malignancy patients and hematopoietic stem cell transplant recipients than the TST test.

        JCAD deficiency attenuates activation of hepatic stellate cells and cholestatic fibrosis

        Background/Aims: Cholestatic liver diseases including primary biliary cholangitis (PBC) are associated with active hepatic fibrogenesis, which ultimately progresses to cirrhosis. Activated hepatic stellate cells (HSCs) are the main fibrogenic effectors in response to cholangiocyte damage. JCAD regulates cell proliferation and malignant transformation in nonalcoholic steatoheaptitis-associated hepatocellular carcinoma (NASH-HCC). However, its participation in cholestatic fibrosis has not been explored yet. Methods: Serial sections of liver tissue of PBC patients were stained with immunofluorescence. Hepatic fibrosis was induced by bile duct ligation (BDL) in wild-type (WT), global JCAD knockout mice (JCAD-KO) and HSC-specific JCAD knockout mice (HSC-JCAD-KO), and evaluated by histopathology and biochemical tests. In situ-activated HSCs isolated from BDL mice were used to determine effects of JCAD on HSC activation. Results: In consistence with staining of liver sections from PBC patients, immunofluorescent staining revealed that JCAD expression was identified in smooth muscle α-actin (α-SMA)-positive fibroblast-like cells and was significantly up-regulated in WT mice with BDL. JCAD deficiency remarkably ameliorated BDL-induced hepatic injury and fibrosis, as documented by liver hydroxyproline content, when compared to WT mice with BDL. Histopathologically, collagen deposition was dramatically reduced in both JCAD-KO and HSC-JCAD-KO mice compared to WT mice, as visualized by Trichrome staining and semi-quantitative scores. Moreover, JCAD deprivation significantly attenuated in situ HSC activation and reduced expression of fibrotic genes after BDL. Conclusions: JCAD deficiency effectively suppressed hepatic fibrosis induced by BDL in mice, and the underlying mechanisms are largely through suppressed Hippo-YAP signaling activity in HSCs.

        The Impact of Health Promotion Activities on the Physiological, Psychological, and Social Functions of Inpatients With Chronic Mental Illness

        Objective To explore the impact of the participation of patients with chronic mental illness in health promotion activities on their physical, psychological, and social functions.Methods This study included inpatient with chronic mental illness from a hospital in Eastern Taiwan. According to the experimental research design, the selected subjects were randomly divided into a health promotion group and a control group, with 60 people in each group. The health promotion team conducts health promotion activities twice a week, each for approximately 50 minutes, over the course of eight weeks (16 times in total). The measurement instrument adopts the Adult Mental Health Scale (AMHS) scale. Five experts and scholars in related fields are invited to conduct expert validity.Results Using independent sample t-tests to analyze the changes in the two groups after 8 weeks, the physical, psychological and social problems of the health promotion group were significantly improved compared with the control group.Conclusion Health promotion activities can alleviate the physical illness, anxiety, irritability, depression, and social distress of chronic hospitalized mental patients, and can also increase their positive and optimistic mood. It can provide chronic mental health care institutions to plan patients’ physical activities or exercises.

        Effect of Blending Modifications for Phenylethynyl-terminated Polyimides

        A reactive diluent (ODA-PEPA) and a flexible phenylethynyl-terminated imide oligomer (PEI-PEPA) were designed and synthesized based on the 2,3,3',4'-diphenyl ether tetracarboxylic acid dianhydride (a-ODPA), 3,4'-oxydianiline(3,4'-ODA), and 4-phenylethynylphthalic anhydride (PEPA). Solution blended systems with the addition of 5, 10, and 15 wt% ODA-PEPA to PEI-PEPA oligomer and their cured resin systems were prepared. Results showed that ODA-PEPA is semi-crystalline in nature. The Tg values of cured resins were improved from 273 oC to 280 oC by the addition of ODA-PEPA, due to the higher crosslink densities. In addition, rheological properties of blends showed lower melt viscosity and wider processing window, revealing improved melt processabilities for potential application in making advanced composites. The isothermal viscosity in 280 oC of PEI-PEPA containing 15 wt% ODA-PEPA reactive diluent decreased by two thirds, due to the low molecular weight of ODA-PEPA. The cured blends demonstrated high thermal stability and heat resistance. 5 wt% thermal decomposition temperatures (Td5) of the cured blends were above 549 oC and 547 oC in N2 and air atmosphere, respectively. The char yield reported at 800 oC in N2 atmosphere increased from 58.8% to 63.7 % with the addition of ODAPEPA. Meanwhile, the cured polyimides blends possessed good bond strength (> 9.3 MPa).

        An integrated strategy for production of four anthocyanin compounds from Ribes nigrum L. by deep eutectic solvents and flash chromatography

        This study explored an integrated strategy for extraction and preparative separation of four anthocyaninsfrom Ribes nigrum L. using DES-based microwave assisted extraction andflash chromatography. Theextraction yield of anthocyanins reached 2.03 mg/g after optimization. An efficient preparativeseparation process of four main anthocyanins by macroporous resin andflash chromatography wasestablished afterwards. The yields of D3G, D3R, C3G and C3R reached 0.32 mg/g, 0.47 mg/g, 0.18 mg/g and0.43 mg/g with the purities of more than 95%, respectively. The developed procedure represents anefficient and convenient alternative for the systematic utilization of natural products from plantmaterials.

        Effect of a Mangosteen Dietary Supplement on Human Immune Function: A Randomized, Double-Blind, Placebo-Controlled Trial

        The effect of a mangosteen product containing multivitamins and essential minerals was tested on immune function and well-being in healthy adults. A randomized, double blinded, placebo-controlled study was conducted in 59 healthy human subjects (40–60 years old). Changes from baseline immune function were measured after a 30-day consumption of the mangosteen product and the placebo. The subjects' self-appraisal of their health status was also surveyed. A xanthone-rich mangosteen product intake increased mean values for peripheral T-helper cell frequency (P=.020) and reduced the serum C-reactive protein concentration (P=.014). Increases in peripheral CD4/CD8 double-positive (DP) T-cell frequency and serum complement C3, C4, and interleukin (IL)-1α concentrations were significantly higher in the experimental group than in the placebo group (DP, P=.038; C3, P=.017; C4, P=.031; IL-1α, P=.006). At the end of study, serum IL-1α and IL-1β concentrations in the study group were significantly higher than that in the placebo group (IL-1α, P=.033; IL-1β, P=.04). Furthermore, more participants in the experimental group reported greatly improved overall health status compared with participants receiving placebo (P=.001). The results indicated that the intake of an antioxidant-rich product significantly enhanced immune responses and improved the subject's self-appraisal on his or her overall health status.

        Magnetic imprinted nanomicrosphere attached to the surface of bacillus using miniemulsion polymerization for selective recognition of 2,4,6-trichlorophenol from aqueous solutions

        In the work, we reported an effective method for the preparation of surface molecularly imprintedpolymers based on bacillus as substrate material with superparamagnetic property throughminiemulsion polymerization. Then, the obtained magnetic surface molecularly imprinted polymers(MMIPs) were evaluated as adsorbents for selective recognition 2,4,6-trichlorophenol (2,4,6-TCP) fromaqueous medium. The resulting MMIPs were characterized by several techniques including FT-IR, SEM,TEM, Raman, XRD, VSM and TGA. The results demonstrated rod-shaped MMIPs was covered withimprinted layer, and exhibited chemical stability and good magnetic sensitivity. Batch mode of bindingstudies were carried out to determine the equilibrium isotherm, kinetics, selectivity adsorption andregeneration of MMIPs toward 2,4,6-TCP. The results indicated that the selective adsorption behaviors ofMMIPs were well described by the Langmuir isotherm model and the pseudo-second-order kineticsmodel. The specific adsorption capacity of MMIPs was 45.16 mg g 1 at 298 K, which was 2.15 timeshigher than that of magnetic non-imprinted polymers (MNIPs). The selective recognition studiesdemonstrated the outstanding affinity and selectivity toward 2,4,6-TCP in the presence of competitivephenols. The regeneration study showed excellent adsorption capacity even after five regenerationcycles. In addition, MMIPs were successfully applied to the extraction of 2,4,6-TCP from milk sample.

        miR-98 suppresses melanoma metastasis through a negative feedback loop with its target gene IL-6

        Dysregulated microRNA (miRNA) expression has a critical role in tumor development and metastasis. However, the mechanism by which miRNAs control melanoma metastasis is unknown. Here, we report reduced miR-98 expression in melanoma tissues with increasing tumor stage as well as metastasis; its expression is also negatively associated with melanoma patient survival. Furthermore, we demonstrate that miR-98 inhibits melanoma cell migration in vitro as well as metastatic tumor size in vivo. We also found that IL-6 is a target gene of miR-98, and IL-6 represses miR-98 levels via the Stat3-NF-κB-lin28B pathway. In an in vivo melanoma model, we demonstrate that miR-98 reduces melanoma metastasis and increases survival in part by reducing IL-6 levels; it also decreases Stat3 and p65 phosphorylation as well as lin28B mRNA levels. These results suggest that miR-98 inhibits melanoma metastasis in part through a novel miR-98-IL-6-negative feedback loop.
