      • T-SPOT.TB for Detection of Tuberculosis Infection among Hematological Malignancy Patients and Hematopoietic Stem Cell Transplant Recipients

        The diagnosis of latent Mycobacterium tuberculosis infection (LTBI) is recommended in hematological malignancy patients and before hematopoietic stem cell transplantation (Guidelines for the prevention and management of infectious complications of solid organ transplantation, 2004). Compared to traditional methods such as tuberculin skin test (TST), T-SPOT.TB has been shown to be more specific. In the present study we enrolled 536 patients for whom T-SPOT.TB was performed, among which 295 patients also received the TST test. The agreement (79%) between T-SPOT.TB and TST was poor (x=0.274, P<0.001). The patients with positive T-SPOT.TB results numbered 62 (11.6%), in which only 20 (48.8%) of the 41 receiving the TST test had positive results. A majority of the patients with T-SPOT.TB positive results had some other evidence ofTB, such as TB history, clinical symptoms and an abnormal chest CT scan. Active TB was found in 9 patients, in which 2 had negative TST results. We followed up the patients and no one developed active TB. Our study suggested that the T-SPOT.TB may be more useful for screening LTBI and active TB in hematological malignancy patients and hematopoietic stem cell transplant recipients than the TST test.

        Steel Bridge Construction of Hong Kong–Zhuhai–Macao Bridge

        The 55-km-long Hong Kong–Zhuhai–Macao Bridge (HZMB) is the world’s longest sea-crossing bridge, connecting Hong Kong with Zhuhai and Macao at the mouth of the Pearl River Estuary in China, comprising 22.9-km-long steel bridges. HZMB is the leading steel bridge in China, with top-level manufacturing and installation technology. This paper outlines the steel bridge construction experiences of HZMB to provide comparisons for the construction of other long sea-crossing steel bridges at home or abroad. The main considerations of construction constraints, scheme selection, structural and aesthetic design of HZMB are presented, and the following points related to new strategies in the steel bridge construction of HZMB are elaborated: (1) construction quality assurance, (2) automatic manufacturing technology, (3) large segment off shore installation, (4) eco-friendly paint (content limitation of volatile organic compounds) and new multifunctional inspection gantry, and (5) Guss Mastic Asphalt steel deck pavement system. The successful implementation of those strategies shows that the steel bridge construction of HZMB promotes improvement in the overall construction and management level of the Chinese bridge industry. The advanced experience of HZMB has opened up broad prospects for the design and construction of off shore bridge engineering in China.

        Oral administration of Schisandra chinensis extract suppresses Dnmt1 expression in Kunming mice ovaries

        The plant Schisandra chinensis contains a phytoestrogens, a type of naturally occurring estrogens which have multiple functions in a number of biological processes. To investigate the correlation between phytoestrogens and epigenetic modification, especially the effect of phytoestrogens on DNA methylation, sexually healthy female mice were used as an animal model in the present study. Briefly, the total RNA and protein were isolated from the ovary of mice after 7-day oral administration of Schisandra chinensis extract (SCE), while distilled water was given to the animals in the control group. Real-time PCR, Western blotting, and enzyme activity assays were performed to examine the effect of the extract of S. chinensis on Dnmt1 transcription and activity. A promoter assay was further conducted in MCF cells (ER positive) to explore also the influence of SCE on Dnmt1 transcriptional activity. The results revealed that the mRNA and protein expression levels of mouse Dnmt1 were both significantly downregulated in the treated group. The transcription of Dnmt1 was suppressed by SCE and in the E2-added group also. Meanwhile the numbers of oocytes at different stages were increased in the treated group when compared by histological analyses with those in the control group. Taken together, the results indicated that, similarly to the action of estrogen, phytoestrogens affected Dnmt1 transcription in mammals, regulating the related gene expression and cell differentiation. The findings of our examination provided also basic data and understanding for the correlation between phytoestrogens and epigenetic modification.

        Influence of Lighting Schedule and Nutrient Density in Broiler Chickens: Effect on Growth Performance, Carcass Traits and Meat Quality

        The study was conducted to evaluate the effect of lighting schedule and nutrient density on growth performance, carcass traits and meat quality of broiler chickens. A total of 576 day old Arbor Acre male chickens was used with a $4{\times}2$ factorial arrangement. The four lighting schedules were continuous (23 L:1 D, CL), 20 L:4 D (12 L:2 D:8 L:2 D), 16 L:8 D (12 L:3 D:2 L:3 D: 2 L:2 D) and 12 L:12 D (9 L:3 D:1 L:3 D:1 L:3 D:1 L:3 D) and provided by incandescent bulbs. The two nutrient densities were high (H, starter diet: 13.39 MJ/kg apparent metabolisable energy (AME), 23.00% crude protein (CP); finisher diet: 13.39 MJ AME/kg, 19.70% CP) and low energy and protein level (L, starter diet: 12.03 MJ AME/kg, 20.80% CP; finisher diet: 12.14 MJ AME/kg, 18.30% CP). Houses with dark curtains and solid sidewalls were used. Chickens were randomly allocated to the 8 treatments with each treatment comprising 6 replicates of 12 chickens. Feed and water were available ad libitum. Lighting schedules showed no difference (p>0.05) in growth performance at the end of the experiment. 12 L:12 D significantly reduced (p<0.05) the concentration of malondialdehyde (MDA) compared to 23 L:1 D treatment. Intermittent lighting (IL) schedules produced higher protein content (p<0.001) in breast meat. Birds on high density diets had higher body weight (BW), feed intake (FI) (p<0.001), and feed conversion ratio (FCR) (p<0.001) throughout the experiment with the exception of 36 to 42 d. High nutrient density increased (p<0.05) abdominal fat, decreased (p<0.05) the moisture loss of meat, and reduced percentage of wings and legs. There was a significant lighting schedule${\times}$diet interaction (p<0.001) on FCR for days 8 to 14 and 15 to 21. Results indicated that IL can give similar growth performance in comparison with CL, meanwhile with positive effects on meat quality by increasing protein content and decreasing the concentration of MDA. High nutrient density resulted in greater growth performance.

        Chaotic dynamic characteristics of pressure fluctuation signals in hydro-turbine

        The pressure fluctuation characteristics in a Francis hydro-turbine running at partial flow conditions were studied based on the chaotic dynamic methods. Firstly, the experimental data of pressure fluctuations in the draft tube at various flow conditions was de-noised using lifting wavelet transformation, then, for the de-noised signals, their spectrum distribution on the frequency domain, the energy variation and the energy partition accounting for the total energy was calculated. Hereby, for the flow conditions ranging from no cavitation to severe cavitation, the chaos dynamic features of fluctuation signals were analyzed, including the temporal-frequency distribution, phase trajectory, Lyapunov exponent and Poincaré map etc. It is revealed that, the main energy of pressure fluctuations in the draft tube locates at low-frequency region. As the cavitation grows, the amplitude of power spectrum at frequency domain becomes larger. For all the flow conditions, all the maximal Lyapunov exponents are larger than zero, and they increase with the cavitation level. Therefore, it is believed that there indeed exist the chaotic attractors in the pressure fluctuation signals for a hydro-turbine.

      • A Simulated Culture? : Cultural Learning Impact of the One-day Study Tour

        One-day study tour in Taiwan began its establishment in early 2007, inspired by the An-San English Village in Seoul, in order to help students learn English quickly and efficiently. Different situational classrooms, such as a clinic, a hotel, a travel agency etc., are set up and manned with native teachers to teach students appropriate conversations accordingly. The main ideas of setting up such an English immersion program are to provide students with enough exposure to an English speaking environment and the culture related. In language learning, culture is one of the key elements which often determine students' learning interest and therefore cannot be neglected. In this study, based on the previous literature research, a model which suggests the relationships among learners' English ability (EA), learning interest (LI) and the situational classroom teaching (SCT) is discussed and explored. The purpose of the study is to investigate whether the simulated culture in different classroom settings could enhance students' English ability and their learning interest through the examination of the model suggested.

      • Preoperative Neutrophil to Lymphocyte Ratio as a Prognostic Factor in Patients with Non-metastatic Renal Cell Carcinoma

        Background: The neutrophil-to-lymphocyte ratio (NLR) is a strong predictor of mortality in patients with colorectal, lung, gastric cancer, pancreatic and metastatic renal cell carcinoma. We here evaluated whether preoperative NLR is an independent prognostic factor for non-metastatic renal cell carcinoma (RCC). Materials and Methods: Data from 327 patients who underwent curative or palliative nephrectomy were evaluated retrospectively. In preoperative blood routine examination, neutrophils and lymphocytes were obtained. The predictive value of NLR for non-metastatic RCC was analyzed. Results: The NLR of 327 patients was $2.72{\pm}2.25$. NLR <1.7 and NLR ${\geq}1.7$ were classified as low and high NLR groups, respectively. Chi-square test showed that the preoperative NLR was significantly correlated with the tumor size (P=0.025), but not with the histological subtype (P=0.095)and the pT stage (P=0.283). Overall survival (OS) and disease-free survival (DFS) were assessed using the Kaplan-Meier method. Effects of NLR on OS (P=0.007) and DFS (P=0.011) were significant. To evaluate the independent prognostic significance of NLR, multivariate COX regression models were applied and identified increased NLR as an independent prognostic factor for OS (P=0.015), and DFS (P=0.019). Conclusions: Regarding patient survival, an increased NLR represented an independent risk factor, which might reflect a higher risk for severe cardiovascular and other comorbidities. An elevated blood NLR may be a biomarker of poor OS and DFS in patients with non-metastatic RCC.

        Behavior and simplified analysis of steel-concrete composite beams subjected to localized blast loading

        Finite element simulations are increasingly used in structural analysis and design, especially in cases where complex structural and loading conditions are involved. Due to considerable progresses in computer technology as well as nonlinear finite-element analysis techniques in past years, it has become possible to pursue an accurate analysis of the complex blast-induced structural effects by means of numerical simulations. This paper aims to develop a better understanding of the behavior of steel-concrete composite beams (SCCB) under localized blast loading through a numerical parametric study. A finite element model is set up to simulate the blast-resistant features of SCCB using the transient dynamic analysis software LS-DYNA. It is demonstrated that there are three dominant failure modes for SCCB subjected to localized blast loading. The effect of loading position on the behavior of SCCB is also investigated. Finally, a simplified model is proposed for assessing the overall response of SCCB subjected to localized blast loading.

        Numerical study on conjugate heat transfer in a liquid-metal-cooled pipe based on a four-equation turbulent heat transfer model

        Conjugate heat transfer between liquid metal and solid is a common phenomenon in a liquid-metalcooled fast reactor's fuel assembly and heat exchanger, dramatically affecting the reactor’s safety and economy. Therefore, comprehensively studying the sophisticated conjugate heat transfer in a liquidmetal-cooled fast reactor is profound. However, it has been evidenced that the traditional Simple Gradient Diffusion Hypothesis (SGDH), assuming a constant turbulent Prandtl number (Prt, usually 0.85 - 1.0), is inappropriate in the Computational Fluid Dynamics (CFD) simulations of liquid metal. In recent decades, numerous studies have been performed on the four-equation model, which is expected to improve the precision of liquid metal’s CFD simulations but has not been introduced into the conjugate heat transfer calculation between liquid metal and solid. Consequently, a four-equation model, consisting of the Abe k ε turbulence model and the Manservisi kq εq heat transfer model, is applied to study the conjugate heat transfer concerning liquid metal in the present work. To verify the numerical validity of the four-equation model used in the conjugate heat transfer simulations, we reproduce Johnson’s experiments of the liquid lead-bismuth-cooled turbulent pipe flow using the four-equation model and the traditional SGDH model. The simulation results obtained with different models are compared with the available experimental data, revealing that the relative errors of the local Nusselt number and mean heat transfer coefficient obtained with the four-equation model are considerably reduced compared with the SGDH model. Then, the thermal-hydraulic characteristics of liquid metal turbulent pipe flow obtained with the four-equation model are analyzed. Moreover, the impact of the turbulence model used in the four-equation model on overall simulation performance is investigated. At last, the effectiveness of the four-equation model in the CFD simulations of liquid sodium conjugate heat transfer is assessed. This paper mainly proves that it is feasible to use the four-equation model in the study of liquid metal conjugate heat transfer and provides a reference for the research of conjugate heat transfer in a liquidmetal-cooled fast reactor
