      • Hepato-protective effect of fucoidan extracted from acid-processed Sargassum fusiformis in ethanol-treated Chang liver cells and in a zebrafish model

        In our previous study, the anticancer effect of the active fucoidan (JHCF4) isolated from acid-processed Sargassum fusiformis was evaluated. In this study, the liver-protective effects of JHCF4 against ethanol-induced Chang liver cell damage and apoptosis-related responses were investigated. Furthermore, the low cytotoxicity and high cell viability of JHCF4 against ethanol-induced cell damage, as well as its protective effect against ethanol-induced cell apoptosis, were observed via nuclear staining with Hoechst 33342 in Chang liver cells. Additionally, the treatment of the 72 h post-fertilization (hpf) zebrafish model with JHCF4 increased the ethanol-stimulated survival rates as well as decreased oxidative stress, lipid peroxidation, and cell death levels. JHCF4 was found to significantly decrease steatosis production in the 128 hpf zebrafish model by Oil Red O staining, as well as attenuate the malondialdehyde and increase the glutathione contents, compared with the untreated group. These results demonstrate that JHCF4 has a potential hepato-protective effect against ethanol-induced damage both in vitro and in vivo.

        意義素性から見たラシイ、ヨウダ ―条件節、理由節における生起状況に注目して―

        In this paper, we investigate two Japanese affixes Rashi and Youda in the modality from and examine the appearances of the hypothetical condition clauses and the cause reason clauses. We find that two affixes can be differentiated by the existence of semantic features. Previous literature indicates that Rashi has at least three semantic features: [+Logical calculation from evidence to inference], [−Assertion] and [+True]. In contrast, there is no semantic feature of [Ture] in the case of Youda. We conclude that Rashi satisfies the feature of the cause reason clauses because it has the semantic feature of [+Ture]. We also notice that Youda does not include the semantic feature of [Ture] and meets the appearance conditions of the hypothetical condition clauses and the cause reason clause.

        Association between Diet and Lifestyle Habits and Irritable Bowel Syndrome: A Case-Control Study

        Background/Aims: Recent papers have highlighted the role of diet and lifestyle habits in irritable bowel syndrome (IBS), but very few population-based studies have evaluated this association in developing countries. The aim of this study was to evaluate the association between diet and lifestyle habits and IBS. Methods: A food frequency and lifestyle habits questionnaire was used to record the diet and lifestyle habits of 78 IBS subjects and 79 healthy subjects. Cross-tabulation analysis and logistic regression were used to reveal any association among lifestyle habits, eating habits, food consumption frequency, and other associated conditions. Results: The results from logistic regression analysis indicated that IBS was associated with irregular eating (odds ratio [OR], 3.257), physical inactivity (OR, 3.588), and good quality sleep (OR, 0.132). IBS subjects ate fruit (OR, 3.082) vegetables (OR, 3.778), and legumes (OR, 2.111) and drank tea (OR, 2.221) significantly more frequently than the control subjects. After adjusting for age and sex, irregular eating (OR, 3.963), physical inactivity (OR, 6.297), eating vegetables (OR, 7.904), legumes (OR, 2.674), drinking tea (OR, 3.421) and good quality sleep (OR, 0.054) were independent predictors of IBS. Conclusions: This study reveals a possible association between diet and lifestyle habits and IBS. (Gut Liver 2015;9:649-656)

        Characterization and screening of anti-tumor activity of fucoidan from acid-processed hijiki (<i>Hizikia fusiforme</i>)

        <P><B>Abstract</B></P> <P>The aim of this study was to investigate the antiproliferative effects of fucoidan from three regional hijiki (<I>Hizikia fusiforme</I>) samples (Zhejiang-China, Jeju-Korea [JH], and Wando-Korea) in East Asia. Hijiki was processed using 1% citric acid to decrease heavy metal content. The JH sample was separated using diethylaminoethyl-cellulose-ion exchange chromatography to obtain four active fractions (JHCF1-JHCF4) and their monosaccharide composition was detected using high-performance liquid chromatography. The structure of the crude polysaccharides and four fucoidan fractions was analyzed using Fourier-transform infrared spectroscopy. JHCF4 showed the highest fucose and sulfate content and decreased Hep3B cell growth in 48 h with a half-maximal inhibitory concentration of 33.53 ± 2.50 μg/ml, which represented the strongest anticancer activity. Further, nuclear staining with Hoechst 33342 and acridine orange-ethidium bromide staining demonstrated that the anticancer activity of JHCF4 was mediated by apoptosis. Moreover, JHCF4 down-regulated B-cell lymphoma extra-large, while up-regulating Bcl-2-associated X protein, caspase-3, and apoptotic bodies to different degrees in Hep3B cells. JHCF4 induced apoptosis via the generation of reactive oxygen species along with the concurrent loss of mitochondrial membrane potential, indicating the potential role of the mitochondria-mediated pathway. Therefore, these results indicate that JHCF4 exhibited antiproliferative effects on the investigated cancer cell lines.</P> <P><B>Highlights</B></P> <P> <UL> <LI> Three regional <I>Hizikia fusiforme</I> samples were processed using 1% citric acid to decrease the heavy metal content. </LI> <LI> Fraction 4 showed the highest fucose and sulfate content and represented the strongest anticancer activity. </LI> <LI> Evidence that fraction 4 induced apoptosis via the mitochondria-mediated pathway was found in Hep3B cells. </LI> </UL> </P>

        Investigation of Indonesian Muslims’ dietary situations in Taiwan to explore the Muslim-friendly standards implementable in the non-Muslim societ

        To probe a Muslim-friendly regulation which may be more implementable than the original Halal standards in Taiwan, this study tried to explore Muslims’ bottom line of acceptance for different dietary cultures when they are exposed to a place where Islam is not the main religion. In view of geographic position and population size, Indonesian Muslims are the most important Muslim customers for Taiwanese market, so this study focused on Indonesian Muslims’ dietary situations in Taiwan. A questionnaire survey was conducted and 233 adequate responses were collected from 3 groups of Indonesian Muslims in Taiwan. The results showed that most of the long/medium-period Indonesian Muslim residents prefer preparing their daily foods to taking the foods supplied by their employers or school canteens. General markets are their major shopping places for foods/food raw materials without special taboos. Multiple choices from 19 categories of foods revealed that they are not used to the Western-style foods. Statistical analyses on the acceptability of the 13 non-Halal scenarios indicated that Indonesian Muslims do not expect the foods/restaurants of 100% Halal qualified in Taiwan. As long as “no pork and no alcohol” can be assured by the credible institutes, many Taiwanese foods and restaurants can be well accepted by the Indonesian Muslims.

        An integrated strategy for production of four anthocyanin compounds from Ribes nigrum L. by deep eutectic solvents and flash chromatography

        This study explored an integrated strategy for extraction and preparative separation of four anthocyaninsfrom Ribes nigrum L. using DES-based microwave assisted extraction andflash chromatography. Theextraction yield of anthocyanins reached 2.03 mg/g after optimization. An efficient preparativeseparation process of four main anthocyanins by macroporous resin andflash chromatography wasestablished afterwards. The yields of D3G, D3R, C3G and C3R reached 0.32 mg/g, 0.47 mg/g, 0.18 mg/g and0.43 mg/g with the purities of more than 95%, respectively. The developed procedure represents anefficient and convenient alternative for the systematic utilization of natural products from plantmaterials.

        Effects of sorbitol on porcine oocyte maturation and embryo development <i>in vitro</i>

        <B>Summary</B><P>In the present study, a porcine system was supplemented with sorbitol during <I>in vitro</I> maturation (IVM) or <I>in vitro</I> culture (IVC), and the effects of sorbitol on oocyte maturation and embryonic development following parthenogenetic activation were assessed. Porcine immature oocytes were treated with different concentrations of sorbitol during IVM, and the resultant metaphase II stage oocytes were activated and cultured in porcine zygote medium-3 (PZM-3) for 7 days. No significant difference was observed in cumulus expansion and the nuclear maturation between the control and sorbitol-treated groups, with the exception of the 100 mM group, which showed significantly decreased nuclear maturation and cumulus expansion. There was no significant difference in the intracellular reactive oxygen species (ROS) levels between oocytes matured with 10 or 20 mM sorbitol and control groups, but 50 and 100 mM groups had significantly higher ROS levels than other groups. The 20 mM group showed significant increases in intracellular glutathione and subsequent blastocyst formation rates following parthenogenetic activation compared with the other groups. During IVC, supplementation with sorbitol significantly reduced blastocyst formation and increased the apoptotic index compared with the control. The apoptotic index of blastocysts from the sorbitol-treated group for entire culture period was significantly higher than those of the partially sorbitol-exposed groups. Based on these findings, it can be concluded that the addition of a low concentration of sorbitol (20 mM) during IVM of porcine oocytes benefits subsequent blastocyst development and improves embryo quality, whereas sorbitol supplement during IVC has a negative effect on blastocyst formation.</P>
