      • 컴퓨터를 이용한 혈액학 교육시스템의 개발

        연구목적 : 전통적인 의학교육 시스템은 최근 개발되고 있는 여러 가지 영상 진단법 등 시각적 자료를 전달하는데 한계가 있으며, 이러한 한계는 모든 분야에 걸쳐서 나타나지만 특히 혈액학이나 심장학 등 시각적인 자료가 진단에 필수적인 분양에서 두드러진다고 할 수 있다.본 연구는 혈액학 교육에 사용되던 기존의 강의실 교육에 시각적 자료가 첨부된 CD-ROM을 개발하여 혈액학 교육 시스템을 개선하기 위한 것이다. 대상 및 방법 : 기존의 혈액학 교육에 있어서 필수적으로 강의 되어오던 자료를 수집하여 체계적으로 정리하였다.강의 자료에 따라서 필요한 영상 자료를 충북대학교 병원에 입원한 혈액질환 환자들의 검사 자료로부터 수집하여 컴퓨터를 기반으로 한 의학영상 처리 및 분석시스템을 통하여 이미지 파일로 변형하였다.또한 입원 환자들 중 대표적인 증례들을 수집하여 실제 임상 진료와 유사한 형태의 증례를 통한 학습을 할 수 있도록 하였다. 결과: 본 CD-ROM은 혈액학 교육에 필요한 대부분의 영역을 포함한다 각 질환의 병태생리, 진단, 임상양상, 치료 및 예후 등 전통적으로 강의되어 오던 강의록에 이미지 파일로 변형된 영상 자료를 필요한 곳마다 링크시켰다.따라서 학습자는 특별한 장치 없이 텍스트 및 영상을 한 화면에서 볼 수 있어 전통적인 교육 시스템에 비하여 시,공간적인 장점을 갖는다 할 수 있다.또한 입원 환자들로부터 수집된 증례를 통하여 실제 임상 진료와 매우 유사한 경험을 하도록하여 교육효과를 증대시켰다. 결론: 혈액학 교육에 있어서 멀티미디어적 효과를 극대화시켜 질환의 병태생리 등 강의실 교육자료뿐 아니라 진단에 필수적인 시각적 자료를 손쉽게 접할 수 있는 CD-ROM의 개발로 전통적인 강의방법이 갖는 한계를 개선시킬 수 있을 것으로 기대된다. Purpose : The traditional medical education system has its limitations in effectiveness of conveying informations in multiple modalities used in diagnosis of patients with hematologic diseases.This study was performed to develop such a multimedia system using computer based CD-ROM, which includes visual assistant systems, several hematological case studies as well as the traditional text-wise education systems. Materials and Methods: The informations about hematologic diseases were compiled and organized.The main visual data was peripheral blood smears, bone marrow data, and several laboratory results accrued from patients admitted to Chungbuk National University Hospital.Each visual data were transformed through computer-based medical image processing and analyzing system and saved as to JPEG files.Case studies were also acquired from admitted patients and the laboratory data of that patients were used unmodified as possible as. Results : The CD-ROM covers almost all kinds of hematologic diseases that must be included in ordinary medical education courses.The text part includes pathophysiology, diagnosis, clinical features, treatment and prognosis of each hematologic diseases.The essential visual data were linked to each text part and accessible with ease, by one click.Case studies were simulated to actual clinical settings and presented step by step from diagnoses to treatments. Conclusion: A multimedia approach for the study of hematologic diseases has been developed and available in CO-ROM format.This system has advantage over traditional education system in conveying all kinds of visual data which is essential in educating hematologic disorders.

      • 도라지가 난소를 절제한 흰쥐의 혈 중 지질 함량 변화에 미치는 영향

        This study was designed to observe the effects of the feeding Platycodon grandiflorum A.DC(PG) extract on the improvement of the lipids on serum in ovariectomized rats. To investigate the effects of ethanol extract, Sprague-Dawley female rats were randomly assigned to groups as follows: Sham, OVX-control and ovariectomized rats supplemented with PG at 50㎎/㎏ bw/day. Body weight gain was not significantly different in groups. The levels of total cholestcrol was higher in ovaiectornized control rats than Sham-operated rats, but supplementation of PG ethanol extracts at 50mg/kg bw/day decreased the level of cholesterol in surum. These results might be expected that ethanol extract of PG is believed to be possible protective effects for the fatty serum increasing serum lipid.

        The Effects of Commercial Alcoholic Drinks Determined Using Scanning Electron Microscopy and Energy-dispersive Electromagnetic Wave (X-Ray) Spectroscopy

        In modern society, drinking is becoming a culture while alcohol consumption continues to increase. Studies show that alcohol consumption in the oral cavity averages 2 hours, and alcohol consumption seems to have a high effect on tooth damage. Therefore, this study was conducted to evaluate the possibility of dental erosion due to intake of some alcoholic beverages in the market. Six types of alcoholic beverages were immersed in 1 ml of each alcohol for 10 min, 60 min, and 120 min to obtain only enamel of the tooth and observe changes over time. The crystal structure of the enamel surface was observed by scanning electron microscopy (SEM), and the changes in calcium and phosphorus, the major constituents of teeth, were analyzed by energy dispersive electromagnetic wave (X-ray) spectroscopy (EDS). Based on the results, the tooth showed a destructive pattern while and loss of calcium (Ca) and phosphorous (P) increased significantly as exposure time to the low pH alcoholic drinks increased. Since this causes the demineralization of inorganic components and greatly affects the risk of tooth erosion, long contact with alcohol should be avoided.

      • Haplotype analysis of major blast resistance (R) genes in rice; Pi9, Pia, and Pib

        Rice blast disease caused by the fungal pathogen Magnaporthe oryzae is one of the most destructive rice diseases worldwide. Resistance to rice blast pathogen mostly shows a quantitative trait controlled by several genes. A total of 13 major blast resistance (R) genes were reported in a number of Korean rice varieties using molecular markers. The Pi-ta gene, which locates near to the centromere of chromosome 12, was haplotyping using 1790 accessions including cultivated and wild varieties in previous research. However, the genetic variations of other R genes in rice still not clear. Three R genes, Pi9, Pia, and Pib on chromosome 6, 11 and 2 respectively, were resequenced among 84 accessions of rice core set. Different types of halotype among the 84 accessions were detected. Some new SNPs and InDels found in exon part of R genes were expected to result into amino acid changes following analysis of the genetic code variations, and the germplam in this rice core set which are resistance to blast were explored. We are expecting to develop the new functional markers and incorporate of resistance genes into existing rice cultivars and finally these apply outcomes in breeding rice resistance to blast diseases.

      • Genome Wide Association Study (GWAS) approach for the blast disease resistance in rice

        One of the biotic stresses in rice production is rice blast disease caused by Magnaporthe oryzae, which is one of the most destructive fungal diseases in rice. We outlined an approach towards genome wide association study for the blast disease resistance in rice. In total, 295 rice accessions including 137 Heuristic Set accessions (HS) and 158 Korean Bred varieties (KB) were screened for the rice blast disease resistance. Firstly, Magnaporthe oryzae were inoculated to the rice seedlings of two weeks after germinations. Then, evaluation of the disease symptoms and checking the crossing point (CP) value were conducted one week after inoculation. To quantify the CP value, real-time polymerase chain reaction (PCR) was employed in combination with the primer pair and Taqman probe specific to Magnaporthe oryzae HYDROPHOBIN class 1 (MHP1) which is an indispensable unigene encoding HYDROPHOBIN for normal virulence expression. Based on these CP values from the PCR reactions containing a series of increasing concentration of cloned amplicon or fungal genomic DNA, correlation among the template’s copy number or its amount and amplification pattern was calculated. Reliability of this equation was further confirmed using the DNA samples from the rice leaves infected with compatible or incompatible strains of M. oryzae. These steps are still being undertaken, and after the complete process of disease resistance phenotyping for the whole population containing 295 accessions, GWAS will be performed to examine the associated genes involving in blast resistance mechanism using the whole genome resequencing data of 295 accessions. This approach would be a useful technique for identifying genetic loci responsible for natural variation in rice blast disease resistance and ultimately, new R genes which can improve the blast resistance in rice.

        자폐 스펙트럼 장애의 산전 및 주산기 합병증

        Objectives:The purpose of this study was to evaluate the prenatal, perinatal, and infancy history of children with autism spectrum disorder (ASD) as compared to unaffected siblings (SIB) and typically developing children (TC). Methods:Subjects with ASD, their SIB, and TC were recruited. All subjects were assessed using both the Korean version of Autism Diagnostic Interview-Revised (K-ADI-R) and the Korean version of Autism Diagnostic Observation Schedule (K-ADOS) and were subsequently identified as affected or unaffected. Prenatal, perinatal, and infancy history was obtained from the primary caregivers and each facet was compared in those with ASD, the SIB, and the TC groups using SPSS ver. 17.0 (p<.05). Results:70 individuals with ASD (63 males, 87.94±37.8months), 53 SIB (27 males, 85.40±48.06 months), and 32 TC (19 males, 104.19±23.409 months) were analyzed. The ASD group showed significantly higher rates of insufficient vaccination as they aged age (χ²=15.54, p=.000). Among the scheduled vaccinations, the DPT vaccination (χ²=10.08, p=.006) was insufficient in ASD groups. The ASD group also showed higher rates of sleep disturbances from infancy. Differences in maternal/paternal age at conception, gestational age, and growth parameters at birth were not significantly difference among the three groups. Conclusion:These results do not support the previous controversies regarding the relationship between prenatal/perinatal complications and ASD. However, these results indicate that perinatal and prenatal factors may contribute to the development of ASD.

      • Thymoquinone-Induced Tristetraprolin Inhibits Tumor Growth and Metastasis through Destabilization of MUC4 mRNA

        <P>Tristetraprolin (TTP), a well-characterized AU-rich element (ARE) binding protein, functions as a tumor suppressor gene. The purpose of this study was to investigate whether a bioactive substance derived from a natural medicinal plant affects the induction of TTP and to elucidate its mechanism. We examined the effects of natural bioactive materials including Resveratrol (RSV), thymoquinone (TQ) and curcumin on the expression of TTP in cancer cell. TQ derived from a natural plant Nigella sativa increased the expression levels of TTP mRNA and proteins in a dose-dependent manner in gastric and breast cancer cells. TQ-induced TTP increased the instability of MUC4 mRNA by direct binding of TTP to ARE in the 3′UTR of MUC4 mRNA. The induction of TTP by TQ also reduced the proliferation, migration and invasion of cancer cells. The expression of the epithelial-mesenchymal (EMT)-related genes, which were target genes of TTP, was also decreased by the TQ treatment. In the in vivo experiments using mouse melanoma cells, TQ-induced TTP inhibited metastasis of tumor cells. We have found that TQ-induced TTP might inhibit metastasis by reducing tumor cell migration and invasion through destabilization of MUC4 mRNA, which suggest the MUC4 as a novel target to TTP.</P>
