Helicobacter pylori와 대장균의 Shuttle Vector 개발

In this study, a vehicle vector using cryptic plasmids was constructed for gene transfer in Helicobacter pylori. pHP51(3.9 kb) and pHP489(1.2 kb) were selected for constructing vectors from cryptic plasmid of H. pylori isolates in Korea. The HindⅢ-digestedDNA fragment(1.2kb) of pHP489 and 1.6kb DNA fragment of pHP51 were ligated with a kanamycin resistance gene(aph3'-Ⅲ) from C. jejuni to produce the recombinant plasmids pHP489K and pHP51K, respectively. Transformation frequency of pHP51K by electroporation was low. But pHP489K could be effectively transformed into various H. pylori strains. In order to design an intermdiate vehicle vector for gene transfer into H. pylori, pBlueHP489K was prepared by recloning pHP489K DNA into pBluescript and pTZ19R vector. This vector permitted the DNA fragment containing pHP489 sequence, aph3'-Ⅲ, and cloned DNA to be cut and self-ligated in the SacⅠ site after cloning. ureA and ureB gene were inserted into pBlueHP489K, resulting in pBlueHP489K/AB. The DNA fragment containing pHP489, kanamycin resistance gene(aph3'-Ⅲ), and urease structural gene was cut away from pBlueHP489K/AB and self-ligated to generate pBlueHP489K/AB. pBlueHP489K/AB made urease-negative H. pylori strains restore their urease activity. By this experiment, pBlueHP489K was confirmed to be the vehicle system for transferring H. pylori genes.

Recently cancer, AIDS, chronic sickness have increased according to the elevation of socioeconomic level and fast change of lifestyle. The number of patients receiving terminal care increased fairly because the span of life is extended by development of medicinal technology. Also necessity of hospice and palliative care was risen according to the request of terminal patients that remove pain and keep calm life by interest about quality of life, However architectural plan and type specialization of facility which can correspond team's composition and supplied nursing program are not consisting. This study researches about care environment of hospice facility plan through investigation into terminal patient's special quality. The purpose of this study is to propose fundamental datas of hospice facility for architectural plan through comparative analysis of cases of domestic and outside facilities.

In this paper, it is proposed to constant power control of solar power system. The solar power system has disadvantage that low power density and variable power output. Proposed strategy is controled by DC/DC converter using phase shift PWM and I-PD type control applying type 1 digital system. The validity of proposed control strategy is verified from simulations results using PSIM.
UASB 반응기에서 제지폐수 처리를 위한 생물학적 속도론 모델에 관한 연구

The granulation of sludge in the UASB reactor is beneficial to stable operation of the reactor at a high volumetric COD loading rate and high hydraulic loading rate, because it improves the setting ability of sludge and the granular sludge acquired has high methanogenic activity. The design of integral biogas?/liquid/solids seperators varies among suppliers of UASB technology. Comples, soluble organic compounds are the first to be hydrolyzed (Stage 1) to simple organics. These simple organics are converted by acid-forming bacteria to C₄H?COOH, C₃H?COOH, CH₃COOH, H₂ and CO₂ in a acidogenic phase(STAGE 2). The higher organic acids are subsequently converted to CH₃COOH and H₂(Stag 3) by acetogenic bacteria. The final step(Stage 4) to produce CH₃ is carried out by me-thano-bacterium. The kinetics of anaerobic digestion of paper wastewaters largely depends on the environmental conditions. The purpose of this study was to elucidate the kinetics of methane production from paper was tewaters in the UASB. A four-step reaction mechanism was proposed for modeling methanogenesis of anaerobic treatment of paper waterwaters.
강섬유보강 콘크리트 슬래브의 내력 및 균열특성에 관한 실험적 연구

This Study is aimed to investigate the effect of load and deflection on steel fiber reinforced concrete slab. Slabs were made with Hooked and Straight types steel fiber and compared a change of steel fiber contents and fiber type. Test were carried out to evaluate the first crack load, maximum load and deflection of slab. AT this result, the first crack load, maximum load and energy absorption capacity were increased remarkably as steel fiber contents were increased. And we found that the deflection of slab at same load were decreased as steel fiber contents were increased, too. As the aspect ratio was increased, the first crack load, maximum load and energy absorption capacity were increased.
토끼에서 근육주사시 입자 크기에 따른 amoxicillin의 비교 약물동태학

To investigate the pharmacokinetic difference between the two amoxicillin (AMX) particles in rabbits after intramuscular injection (i.m.), both of AMX-S (particle size: 10 ㎍) and AMX-L (particle size: 100 ㎍) were injected into New Zealand White rabbits (1.2±0.3 ㎏) at a dose rate of 10 ㎎/㎏ of body weight. In this study, serum AMX concentrations were detected by microbiological assay with E. coli BE 1186 which shows high antibiotic sensitivity. After i.m. administration, AMX-S and AMX-L were best fitted as 1-compartment model with the absorption and elimination phase. The biological half-life (T_1/2, _k10) of AMX-S is 4.06±1.09 h and that of AMX-L 4.76±0.69 h. The serum maximal concentration time (T_max) of AMX-S and AMX-L were 0.88±0.17 h and 0.77±0.11 h, respectively. Maximal AMX concentration (C_max) (AMX-S: 5.71±0.62 ㎍/㎖, AMX-L: 5.04±0.25 ㎍/㎖) in serum showed a significant difference (p<0.05). In terms of bioequivalance, however, there was no difference between the two AMX's after i.m. injection in the basis of C_max and AUC.

Alkaline wastewater can be neutralized by using flue gas, which of method has particular merits, and it is also neutralized without neutral salts and problems by biological treatment. If the pure CO₂ gas is in contact with the alkaline wastewater, the solubility for CO₂ can be presented to the Henry-constant(K_(H)). K_(H) is on the decreased a half by temperature from 10℃ to 50℃. The absorptive moles of CO₂ gas was 1.59×10^(-8)mole/1 at 20℃, and in spite of varing flow rate, the volume of CO₂ gas passed through wastewater was constant to about 1.8L. In these results, it was known that it could be possible to neutralize the alkaline wastewater with flue gas below the pH=9.
Helicobacter pylori 편모 유전자의 클로닝 및 염기서열 분석

A λgt11 expression libary of H. pylori DNA in E. coli Y1090 was screened with flagellin-specific rabbit antiserum for molecular cloning of the flagellin gene of H. pylori. A positive clone, λHPF4, was obtained and the recombinant antigen expressed from λHPF4 was a fusion protein with the molecular weight of 168kd. Sequence analysis of antigen-encoding DNA showed that an open reading frame composed of 1,536 nucleotides encodes a polypepride with a oredicted molecular size of 54kd. This open reading frame did not show the homology with flaA gene encoding 56kd protein of H. pylori and was confirmed as a unique sequence through homoligy searching. Therefore, the cloned antigen is supposed to be the carboxy-terminal region of the other flagellin protein of H. pylori, flaB, with the molecular weight of 58kd.

Fruit are normally kept in a pouch of cold storage room in winter. In this work, a mathematical model that is emulating the fruit respiration in the pouch of the storage has been established. The calculated result is compared to the actual measured gaseous composition change of pear in the packaging pouch LDPE film incorporated with silver coated ceramic powder. The presented model, that is combining polysaccharide digestion kinetics in pear, Fickian diffusion for permeation of the film and sigmoidal adsorption equilibrium between the film and gases, predicts the respiration of the pear in the packing film fairly well.
