      • Effect of Ionizing Radiation on Rat Tissue: Proteomic and Biochemical Analysis

        <P>Reactive oxygen species (ROS), generated by ionizing radiation, has been implicated in its effect on living tissues. We confirmed the changes in the oxidative stress markers upon irradiation. We characterized the changes in the proteome profile in rat liver after administering irradiation, and the affected proteins were identified by MALDI-TOF-MS and ESI-MS/MS. The identified proteins represent diverse sets of proteins participating in the cellular metabolism. Our results demonstrated that proteomics analysis is a useful method for characterization of a global proteome change caused by ionizing radiation to unravel the molecular mechanisms involved in the cellular responses to ionizing radiation.</P>

      • Newcastle병 면역에 대한 연구

        A study was conducted to determine whether the vaccination programs for the control of Newcastle Disease (ND) would affect the immune status of hens against the disease as a purpose of suggesting the distinguished vaccination programs. Fifteen poultry flocks were surveyed to investigate the level of serum antibody against ND virus and the programs used for the vaccination of hens. Alternatively, vaccination was carried out by the programs designed against ND virus in hens called Manina and investigated the level of HI titer every week, and surveyed the alternation of hematological occurrence following the vaccination programs. The obtained results were summarized as follows: 1. Vaccination history of the investigated 15 poultry flocks were not according by standard vaccination programs and immunization was carried out for random at every poultry farms. Killed and Live vaccin was used together in 7 groups and only Live vaccin was used in 6 groups, and only killed vaccin was used in groups. 2. HI titer of these was 140 in average by killed and live vaccin, and live vaccin was 126 in average. The former was shown higher HI titer. 3. The number of leucocytes was shown much increased hy the designed vaccination program, and 9525/㎣ was shown by 2nd vaccination (9weeks), and 1067O/㎣ was shown highest by 6th vaccination (25weeks). 4. HI titer was 240 by 2nd vaccination and it was continuous to increase. HI titer was 320 by 6th vaccination. The effects of the repeated vaccination were distinguished. 5. Serum globulin value was increased according to the vaccination program. The highest value ware 3.54㎎/㎗ and 3.54㎎/㎗ in 22 weeks and 23 weeks, each γ globulin value of these were 0.95㎎/㎗ and 0.99㎎/㎗.

      • 고액 상평형의 측정과 추산 연구

        정적인 융점측정 방법으로 benzene+n-dodecane, cyclohexane+phenol, phenol+water, water+sulfolane 및 diethylcarbonate+nitrobenzene계에 대한 고액 평형을 측정하였다. 측정된 고액평형 데이터는 문헌값과 modified UNIFAC에 의해 예측된 값과 비교하였으며, 추산값과 실험값은 ±1.61K 범위에서 잘 일치하였다. 본 연구에서 고찰된 diethylcarbonate의 융점은 기존문헌치와 큰 편차를 보여 재검증이 필요하다. Solid-liquid equilibrium(SLE) data were measured for benzene+ n-dodecane, cyclohexane+phenol phenol+water, water+sulfolane, and diethylcarbonate+ nitrobenzene systems by using static melting point measuring method. Measured SLE data were compared with the literature values and also compared with the values predicted by modified UNIFAC(Dortmund) equation. The experimental results are in good agreements with the published and estimated values, and the average deviation between the experimental values and the calculated values is ±1.16K. The measured melting point of diethylcarbonate shows some difference from literature value, and it will be certified through other experimental data for different systems.

      • Pocine Adenovirus-3의 E1B Region의 鹽基序列 分析

        돼지 아데노바이러스(PAV-3). 6618주의 EIB region이 包含되어 있는 map unit 4.0에서 9.7까지의 유전자에 대한 1,984 bp의 염기서열을 決定하였으며, 이 結果를 알려진 여러 아데노바이러스 유전자와 비교하여 다음과 같은 결과를 얻었다. 1. PAV-3의 EIB유전자는 10개의 ORF로 구성되어 있으며, 그 중 아데노바이러스의 단백질과 유사성이 있는 것은 ORF1, ORF2 및 ORF3이었다. ORF1은 Ad41의 19kd 와 BAV-2에서의 EIB ORF2에서의 아미노산의 一致率은 각각 32%와 31%이었다. 2. ORF2는 Ad2 55kd protein과 tupaia adenovirus 44kd protein가 각각 34%로 아미노산 一致率이 가장 높았으며, Ad41의 52kd protein. BAV-3의 EIB ORF3에서도 33%의 一致率을 보였다. 3. ORF1은 61-666 uncleotide (606 bp), ORF 2에서는 429-1,850 uncleotide (1,422 bp)의 부위로 각각 202, 474 a.a로 構成되었으며, 예상되는 분자량은 20 kd와 52 kd이었다. 4. ORF3는 hexon-associated pIX유전자로 추정되며 내부에 1개의 polyadenylation signal(ATAAA)이 1938-1942 uncleotide에 위치하였으며, 이 부위는 TATA box (1937-1942 uncleotide)와 중복되어 존재하였다. Porcine adenovirus type 3 (PAV-3) does not cause severe infection in pigs. Adenovirus has been suggestive of live vaccine vector carrying foreign gene. One of insertion regions is delayed early (EIB) region. However, EIB region of PAV-3 has not been molecularly characterized to date. Nucleotide sequence of EIB of PAV-3 was determined. The EIB region was composed of 1,984 bp and located between 4.0 and 9.7 map units. Three potential open reading frames(ORFs) with low level of homology to other adenoviruses and a polyadenylation signal were identified in the rightward direction of genome. The nucleotide and the predicted amino acid sequences of EIB were compared to those of human and animal adenoviruses. One of the three potential ORFs. ORF1 encoded a polypeptide homologous to bovine adenovirus type 2(BAV-2) ORF2 and human adenovirus type 41(Ad41) 19 kd protein. ORF2 encoded a polypeptide homologous to human adenovirus type 2(Ad2) 55 kd protein, bovine adenovirus type 3(BAV-3) ORF3 and porcine adenovirus type 4(PAV-4) ORF2. The predicted protein of ORF1 had homology to those of Ad41 and BAV-2 with 32 and 31% respectively, whereas the deduced protein of ORF2 had homology to those of Ad2. BAV-3 and PAV-4 with 34, 33 and 29%, respectively.

      • 전남지역 논토양의 미생물상에 관한 연구 : 미생물 분포조사와 불완전 균류의 동정에 대하여 Distribution of the microflora and identification of the imperfect fungi

        전남지역 일원인 광주를 비롯한 8개 지역을 임의로 선정하여 1983년 10월 20일부터 11월 5일 사이에 논토양을 채취하였다. Plate dilution method로 각 지역별 작토층에 서식하고 있는 미생물 취락정도를 조사하고 사상균류만을 선정하여 불완전균류만을 분리동정하였다. monsporic culture로 얻은 순수 균주는 50균주로서 관련된 참고자료들에 의해 39균주를 동정하였으며 동정된 균주는 5속 8종이였다. 본 조사에 의해 밝혀진 미생물 분포정도와 동정된 균주 내력은 다음과 같다. 1. 전남지역 일원의 논토양에서 평균미생물 취락수는 세균이 19.5×10^6이고 방선균이 38.1×10^5이였다. 사상균은 42.5×10^3으로 나타났으며 B/F, B/A 및 A/F율은 각각 458.8, 5.1과 89.6이였다. 2. 전남지역 일원의 논토양에서 분리된 사상균 중 불완전균류 5속 8종을 동정하였으며, 동정된 균주는 다음과 같다. Trichoderma virides, Alternaria alternata, Cladosporium cladosporioides, Penicillium citrinum, Penicillium implication, Penicillium janthinellum, Aspergillus niger, Aspergillus clavatus이다. This investigation was conducted to find out microflora of the paddy soil in Chunnam. Eight paddy soil samples were collected from the different sites. By the dilution plate method, a total number of microorganism was recorded at the upper layer (0∼10cm in depth). The fungi were isolated at the dilution plate from paddy soil samples of eight selected sites. They were obtained after pure cultures with a total of 50 isolated strains. Among these, 39 isolated strains (8 species in 5 genera) were identified and 11 isolated strains were unidentified. The results were summarized as follows; 1. The mean numbers of microorganisms in collected paddy soil were 19.5×10^6 in bacteria (B), 38.1×10^5 in actinomycetes (A), and 42.5×10^3 in fungi (F) per gr paddy soil. The ratios of B/F, B/A and A/F were 458.8, 5.1 and 89.6 respectively. 2. 39 isllated strains (8 species in 5 genera) of imperfect fungi were identified as follows; Trichoderma virides, Alternaria alternata, Cladosporium cladosporioides, Penicillium citrinum, Penicillium implication, Penicillium janthinellum, Aspergillus niger, Aspergillus clavatus.

      • 한국산 돼지의 屠畜檢査例에 의한 臟器 病燮調査 : Ⅰ. 肺의 病燮 Ⅰ. About Lesions of the Lungs

        The lungs of 400 cases which had been collected at the slaughter houses in Pusan, Masan and Chungmu cities were examined histopathologically to clear out the patterns of pathological changes of pigs held in Korea. 1. The most frequent and dominated lesions in the lungs were swine enzootic pneumonia (14%) and lung worm pneumonia(12%). 2. The pneumonic lesions caused by lung worms showing the bronchiolar muscle hypertrophy, eosinophilic infiltration, more limited peribronchiolar lymphocytic cuffing compared with that of swine enzootic pneumonia differentiated each other two diseases histologically. 3. The hyperemia, pulmonary ecema, interstitial pneumonia, and bronchitis occurred also frequently in the lungs, but the occurrences of the broncho pneumonia, abscess, gangrenous or granulomatous lesions were rare.

      • 국내에서 분리된 canine parvovirus의 구조유전자 cloning과 염기서열 분석

        In this study gene encoding structural proteins of a CPV isolate was cloned and saquenced to elucidate the molecular genetical properties of the canine parvoviruses isolated from the field. Six recombinant plasmids of pEP3, p1471, p2070, pEP069, pEP338 and p14711p were constructed from the map positions 22 to 98 of RF DNA to clone the VP1 and VP2 genes of CPV-V20. Sequentialy the gene comprising 3780 nucleotides were sequenced by dideoxy chain termination method. When nucleotide sequence of gene encoding the structural proteins of CPV-V20 was compared with those of other strains, CPV-N, CPV-d and CPV-780929 published previously. DNA homologies to CPV-V20 were 99.87% with CPV-NM, 99.73% with CPV-d, 96.85% with CPV-780929 AND 98.4% with FPLV-Carl, respectively. The DNA sequence data of CPV-V20 showed seven point mutations and also deletion of 135 nucleotides from the nucleotide position 4745 to 4879 located in the 3-noncoding region of CPV-N.

        미래 사회를 대비하기 위한 개인의 개성을 존중과 창의적인 능력을 지닌 인간을 양성을 목적으로 충청남도 지역의 초 ㆍ 중고등학생들을 대상으로 한 창의력 경연대회가 공주대학교 과학교육연구소에서 개최되었다. 충청남도 초 ㆍ중 ㆍ 고등학생들을 대상으로 실시한 창의력 경연대회는 창조적 아이디어를 내어 스스로 문제를 해결해 나갈 수 있는 교육 프로그램을 제공하는 좋은 기회였음이 확인되었으나, 일부의 팀에서만 참신하고 재미있는 아이디어들이 제안되었다. 여학생의 참가자 수가 상당히 증가하였으며, 대체적으로 입상권의 학생들의 상당 부분이 여학생이라는 것이 특이할만하다. 학년이 올라감에 따라서 창의적인 아이디어를 내는 양이나 질에 있어서 뒤떨어지는 것으로 분석되었으며, 토론 학습의 적응력이 미흡하고 발표력 및 청취력에 있어서도 떨어지는 것으로 나타났다. 따라서 중 ㆍ고등학교의 학교교육에서도 학생들에게 프로젝트형 탐구학습 프로그램의 적용은 물론 토론식 수습을 통한 학생들의 사고의 전환의 기회를 제공해야 할 것으로 사료된다. The creativity competition was held to foster the creativity of elementary and secondary school students in the Institute of Science Education, Kongju national University. It was found that the creativity competition effected on the improvement of inventing creative ideas and problem solving activities. But, only some competitors participated in semifinal and final contests, presented original and interesting ideas. More female students participated in competition than last year. More female participants were awarded a prize than males. Totally, participants were defective in discussion and communication, and presentation and listening ideas. Increasing grades correlated inversely with creativity and originality. There is need of inquiry project teaming programs and presenting opportunities of conversion of thinking by discussing instructions in school

        The organs of 400 cases which had been collected at the slaughter houses in Pusan, Masan and Chungmu cities were examined histopathologically to clear out the patterns of pathological changes of pigs kept in Korea. The report about the lesions of the liver succeeding the former report about the lung was summairzed as follows: 1) The most frequent and dominated lesions in the liver were those of hepatic ascariasis(11%) and toxic changes(35%) such as cloudy swelling, hepatic lipidosis, parenchymatous necrosis and liver cirrhosis. 2) The lesions of the liver cirrhosis observed here would be classified as three types; portal cirrhosis subdivided into secondary cirrhosis and Laennec's type. intralobular cirrhosis and biliary cirrhosis.
