New phenalenone derivatives from the Hawaiian volcanic soil-associated fungus Penicillium herquei FT729 and their inhibitory effects on indoleamine 2,3-dioxygenase 1 (IDO1)

        Jae Sik Yu,Se Yun Jeong,Chunshun Li,Taehoon Oh,Mincheol Kwon,Jong Seog Ahn,Sung-Kyun Ko,Yoon-Joo Ko,Shugeng Cao,Ki Hyun Kim 대한약학회 2022 Archives of Pharmacal Research Vol.45 No.2

Phenalenone derivatives sourced from fungi arepolyketides that have attracted signifi cant interest becauseof their diverse chemical structures and potential bioactivities. As part of our ongoing quest to discover novel naturalproducts with biological properties from diverse naturalresources, three unreported phenalenone derivatives ( 1–3 ),named ent -12-methoxyisoherqueinone ( 1 ), (−)-scleroamide( 2 ), and (+)-scleroamide ( 3 ), together with four known phenalenonederivatives, ent -atrovenetinone ( 4 ), isoherqueinone( 5 ), herqueinone ( 6 ), and ent -peniciherquinone ( 7 ) wereisolated from the Hawaiian soil fungus Penicillium herqueiFT729, collected on the Big Island, Hawaii. Compounds 2 and 3 were enantiomers, which were separated using achiral-phase HPLC column, which provided optically purecompounds 2 and 3 . The structures of the novel compoundswere established by extensive spectroscopic analyses, including 1D and 2D NMR and high-resolution ESIMS. Their absolute confi gurations were determined using quantumchemical electronic circular dichroism (ECD) calculations. The inhibitory activity of the isolated compounds( 1–7 ) against indoleamine 2,3-dioxygenase 1 (IDO1) wasassessed. Compounds 1, 5–7 inhibited IDO1, with IC 50values of 32.59, 36.86, 19.05, and 24.18 μM, respectively. These fi ndings demonstrated that the phenalenone derivatives1 and 5–7, as IDO1 inhibitors, are promising anticancerimmunotherapeutic agents.

      • Src/Syk-Targeted Anti-Inflammatory Actions of Triterpenoidal Saponins from Gac ( <i>Momordica cochinchinensis</i> ) Seeds

Yu, Jae Sik,Kim, Jun Ho,Lee, Seulah,Jung, Kiwon,Kim, Ki Hyun,Cho, Jae Youl World Scientific Publishing Company 2017 The American journal of Chinese medicine Vol.45 No.3

<P><I>Momordica cochinchinensis</I> Spreng (family Cucurbitaceae), also known as gac, or red melon, is an edible Southeast Asian fruit valued for its nutritional and medicinal properties. Specifically, Momordicae Semen, the seeds of the gac fruit, is used in traditional Chinese medicine to treat boils, rheumatic pain, muscle spasm, hemorrhoids, and hemangiomas. In this study, a chemical investigation into a gac seed ethanol (EtOH) extract resulted in the identification of three triterpenoidal saponins (<B>1</B>-<B>3</B>), which were investigated for their anti-inflammatory effects. Among the saponins, momordica saponin I (compound <B>3</B>) reduced the production of nitric oxide (NO) in LPS-activated RAW264.7 cells without inducing cytotoxicity. The mRNA levels of inducible NO synthase (iNOS) and cyclooxygenase (COX)-2 were decreased by momordica saponin I. Additionally, the translocation of p65 and p50 (subunits of the transcription factor NF-<TEX>$ \kappa $</TEX>B) into the nucleus was remarkably inhibited. Furthermore, the phosphorylation levels of inflammatory signaling proteins (I<TEX>$ \kappa $</TEX>B<TEX>$ \alpha $</TEX>, Src, and Syk) known to be upstream regulatory molecules of p65 were decreased under momordica saponin I-treated conditions. The molecular targets of momordica saponin I were confirmed in overexpression experiments and through immunoblot analyses with Src and Syk. This study provides evidence that momordica saponin I could be beneficial in treating inflammatory diseases, and should be considered a bioactive immunomodulatory agent with anti-inflammatory properties.</P>

( Jae Hyuk Choi ),( Woo Jin Chung ),( Yu Jin Lee ),( Yu Jin Kang ),( Jung Min Lee ),( Eun Sung Choi ),( Sang Min Lee ),( Byoung Kuk Jang ),( Jae Seok Hwang ),( Eun Soo Kim ),( Kyung Sik Park ),( Kwang 대한간학회 2013 춘·추계 학술대회 (KASL) Vol.2013 No.1

Background/Aim: The use of oral branched chain amino acid (BCAA) for enriched supplement in liver cirrhotic patients is increasing now. This study aimed to examine the effects of BCAA as well as factors associated with the effectiveness to in crease serum albumin concentration. Methods: We reviewed retrospectively medical records of cirrhotic patients who received oral BCAA from January 2004 to December 2012 at Keimyung University Dongsan Hospital . We analyzed the changes of serum albumin concentration after use of oral BCAA and several factors which affected to the effectiveness of oral BCAA. Results: Total 89 patients were included (64 men; mean age, 63.0 ± 8.7 years). Hepatitis B (57.3%), alcohol (15.7%), hepatitis C (14.6%), and others (12.4%) were the causes of liver cirrhosis. Patients with Child-Pugh class A, B, and C were 28.1, 42.7, and 29.2%. Patients were treated with daily oral BCAA (mean dose ± standard deviation, 8.76 ± 2.25 mg/day; median follow-up period, 95 days). 1. Increased albumin group after use of oral BCAA were 27 and non-increased groups were 62 patients. 2. Antiviral therapy was used in patients with hepatitis B (33.3%) during oral BCAA treatment. Antiviral therapy in patients with hepatitis B (80.0% vs. 55.6%; P=0.123) was not significant difference with albumin level after oral BCAA treatment. 3. According to univariate and multivariate analysis, Patients with CP class A or B (OR, 3.611 ; 95 % CI, 1.168-11.169; P=0.026), initial eunatremia (OR, 6.298 ; 95 % CI, 2.064-19.214 ; P=0.001) were increased albumin concentration after oral BCAA treatment. Conclusion: Oral BCAA in patients with liver cirrhosis is effective both as compensated and uncompensated. Oral BCAA effectively increased serum albumin in cirrhotic patients with CP A or B than C. So, early administration of oral BCAA in cirrhotic patients seemed to be needed for better life quality.

Bioactivity-guided isolation of ginsenosides from Korean Red Ginseng with cytotoxic activity against human lung adenocarcinoma cells

        Jae Sik Yu,노현수,Kwan-Hyuck Baek,Seul Lee,Sil Kim,Hae Min So,Eunjung Moon,Changhyun Pang,Tae-Su Jang,Ki Hyun Kim 고려인삼학회 2018 Journal of Ginseng Research Vol.42 No.4

Background: Lung cancer is the leading cause of cancer-related death worldwide. In this study, we used a bioactivity-guided isolation technique to identify constituents of Korean Red Ginseng (KRG) with antiproliferative activity against human lung adenocarcinoma cells. Methods: Bioactivity-guided fractionation and preparative/semipreparative HPLC purification were used with LC/MS analysis to separate the bioactive constituents. Cell viability and apoptosis in human lung cancer cell lines (A549, H1264, H1299, and Calu-6) after treatment with KRG extract fractions and constituents thereof were assessed using the water-soluble tetrazolium salt (WST-1) assay and terminal deoxyribonucleotidyl transferaseemediated dUTP nick end labeling (TUNEL) staining, respectively. Caspase activation was assessed by detecting its surrogate marker, cleaved poly adenosine diphosphate (ADP-ribose) polymerase, using an immunoblot assay. The expression and subcellular localization of apoptosis-inducing factor were assessed using immunoblotting and immunofluorescence, respectively. Results and conclusion: Bioactivity-guided fractionation of the KRG extract revealed that its ethyl acetatee soluble fraction exerts significant cytotoxic activity against all human lung cancer cell lines tested by inducing apoptosis. Chemical investigation of the ethyl acetatesoluble fraction led to the isolation of six ginsenosides, including ginsenoside Rb1 (1), ginsenoside Rb2 (2), ginsenoside Rc (3), ginsenoside Rd (4), ginsenoside Rg1 (5), and ginsenoside Rg3 (6). Among the isolated ginsenosides, ginsenoside Rg3 exhibited the most cytotoxic activityagainst all human lung cancer cell lines examined,withIC50 values ranging from161.1 mMto 264.6 mM. The cytotoxicity of ginsenoside Rg3 was found to be mediated by induction of apoptosis in a caspaseindependent manner. These findings provide experimental evidence for a novel biological activity of ginsenoside Rg3 against human lung cancer cells. 2018 The Korean Society of Ginseng, Pu
