        Recommendations on management of gynecological malignancies during the COVID-19 pandemic: perspectives from Chinese gynecological oncologists

        The outbreak of coronavirus disease 2019 (COVID-19) caused by the severe acute respiratorysyndrome coronavirus 2 has rapidly spread globally. Cancer patients are at a higher risk ofbeing infected with the coronavirus and are more likely to develop severe complications, ascompared to the general population. The increasing spread of COVID-19 presents challengesfor the clinical care of patients with gynecological malignancies. Concerted efforts should beput into managing gynecological malignancies in an orderly manner by strictly implementingthe measures that are specifically developed for controlling the spread of COVID-19. We havedrafted Recommendations on Management of Gynecological Malignancies during the COVID-19 Pandemicbased on our experience on controlling COVID-19 pandemic in China. We recommendthat patients with gynecological malignancies should be managed in hierarchical andindividualized manners in combination with local conditions related to COVID-19. Medicalcare decision should be balanced between controlling COVID-19 pandemic spread and timelydiagnosis and treatment for gynecologic oncology patients.

      • Prognostic Significance of α5β1-integrin Expression in Cervical Cancer

        The purpose of this study was to evaluate the association of expression of ${\alpha}5{\beta}1$-integrin with clinicopathologic features and prognosis in cervical cancer. Levels of ${\alpha}5{\beta}1$-integrin in normal cervical mucosa and cervical cancer tissue were detected with immunohistochemistry. Survival analysis by the Kaplan-Meier method was performed to assess prognostic significance. ${\alpha}5{\beta}1$-integrin expression was detected in 84.6% (143/169) cervical cancer samples, significantly different from that in normal cervical mucosa (P < 0.05). Positive expression rates of ${\alpha}5{\beta}1$-integrin in patients with poor histologic differentiation, lymph node metastasis, and recurrence were elevated. Using Kaplan-Meier analysis, a comparison of survival curves of low versus high expression of ${\alpha}5{\beta}1$-integrin revealed a highly significant difference in human cervical cancer cases (P < 0.05), suggesting that overexpression of ${\alpha}5{\beta}1$-integrin is associated with a worse prognosis.The ${\alpha}5{\beta}1$-integrin promotes angiogenesis and associates with lymph node metastasis, vascular invasion and poor prognosis of cervical cancer. The current study indicated that ${\alpha}5{\beta}1$-integrin may be an independent prognostic factor for cervical cancer patients.

        We report the annotation and analysis of the draft genome sequence of Brassica rapa accession Chiifu-401-42, a Chinese cabbage. We modeled 41,174 protein coding genes in the B. rapa genome, which has undergone genome triplication. We used Arabidopsis thaliana as an outgroup for investigating the consequences of genome triplication, such as structural and functional evolution. The extent of gene loss (fractionation) among triplicated genome segments varies, with one of the three copies consistently retaining a disproportionately large fraction of the genes expected to have been present in its ancestor. Variation in the number of members of gene families present in the genome may contribute to the remarkable morphological plasticity of Brassica species. The B. rapa genome sequence provides an important resource for studying the evolution of polyploid genomes and underpins the genetic improvement of Brassica oil and vegetable crops.

        Interaction and micellar behavior of ternary mixture of amphoteric amino sulfonate surfactant with traditional anionic and nonionic surfactants: Effect of hydrophilicity

        The micellization and the molecular interaction behaviors for two ternary mixtures constituted by anamphoteric sodium 3-(n-dodecyl ethylenediamino)-2-hydroxypropyl sulfonate (C12AS), an anionicsodium dodecylbenzene sulfonate (SDBS) and a nonionic octylphenol polyoxyethylated ether with thenumber (n) of oxyethylene glycol ethers OP-n (n = 10 or 7) in aqueous solution were investigated usingthe tensiometry and the effect of hydrophilicity on them was also discussed. In the framework of pseudophaseseparation model and based on the regular solution theory, the related micellization parametersincluding the mixed critical micelle concentration (cmc) values in the ideal and real cases, the activitycoefficients and the compositions in mixed micelle, etc. and thermodynamic parameters were estimatedby the Clint’s model and the Rubingh’ model. The mixed cmc value is dependent on the composition inaqueous solution and influenced by the hydrophilicity of nonionic surfactant. With increasing the nonionicin ternary mixture, the mixed cmc value is initially decreased and then slightly rise. An increasingin the hydrophilicity of nonionic will make the minimum value of mixed cmc be increased from1.299 mM to 1.705 mM. The resulting phenomena can be explained reasonably by the electrostatic effect,the steric hindrance, the hydrogen bonding, etc. Thermodynamic data indicate that the contribution ofentropy or enthalpy plays a vital role on the spontaneous process of micellization and the share ofentropy or enthalpy in free energy change is dependent largely on the amount of nonionic surfactantand the hydrophilicity. In ternary mixtures of C12AS/SDBS/OP-10, an increase in an amount of OP-10 willinduce the change from the enthalpy-driven micellization process to the entropically favorable process. Once an abundant amount of OP-7 is added, while, the enthalpy will make a main contribution on themicellization process, which can be described by the drop in the share of entropy at the composition(0.6970/0.0000/0.3030) of ternary mixture from 0.6478 to 0.4901. These findings will help with understandingthe molecular interaction behavior for the ternary surfactant mixture and the effect of the additionof nonionic surfactant and its hydrophilicity.

      • RTN4 3'-UTR Insertion/Deletion Polymorphism and Susceptibility to Non-Small Cell Lung Cancer in Chinese Han Population

        Nogo protein, encoded by gene reticulon-4 (RTN4), includes three major isoforms by different splicing, named Nogo-A Nogo-B and Nogo-C. Nogo proteins play an important role in the apoptosis of cells, especially in tumor cells. RTN4 single nucleotide polymorphisms (SNPs) can influence the efficiency of transcription and translation thus being related with an individual's predisposition to cancer. The CAA insertion/deletion polymorphism (rs34917480) within RTN4 3'-UTR has been reported to be associated with many cancer types. In order to investigate the relationship between this polymorphism and susceptibility to non-small cell lung cancer (NSCLC) in the Chinese population, we conducted the present case-control study including 411 NSCLC patients and 471 unrelated healthy controls. The genotype distributions were significantly different between cases and controls (p=0.014). We found that the del allele could significantly increase NSCLC risk (ins/ins vs ins/del: p=0.007, OR 1.46, 95%CI=1.11-1.93; dominant model: p=0.004, OR 1.47, 95%CI=1.13-1.92 and allele model: p=0.008, OR 1.35, 95%CI=1.08-1.67). This association was stronger in participants over 60 years old, males and smokers. We therefore conclude that the CAA insertion/deletion polymorphism (rs34917480) contributes to non-small cell lung cancer risk in Chinese population. Age, sex and environmental exposure are also related to carcinogenic effects of rs34917480.

        CRA Reputation and Bond Yield: Evidence from the Chinese Bond Market

        The paper examines the reputation effect of credit rating agencies (CRAs) in China. We find a negative association between CRAs’ reputation and public bonds’ offering yield spreads after controlling for endogeneity. Analyses of the correlation between CRA reputation and bond return volatility in the aftermarket and cross-sectional variations of reputation effects alongside a host of information environment proxies for issuers, we identify the information channel through which CRA reputation plays its role. This is further supported when we utilize an exogenous event, the introduction of an independent CRA (the China Bond Rating Co. Ltd), to explore the potential changes of the reputation effect.

        Inhibitory effects of piceatannol on human cytomegalovirus (hCMV) in vitro

        Human cytomegalovirus (hCMV) is a ubiquitous herpesvirus, which results in the establishment of a latent infection that persists throughout the life of the host and can be reactivated when the immunity is low. Currently, there is no vaccine for hCMV infection, and the licensed antiviral drugs mainly target the viral enzymes and have obvious adverse reactions. Thus, it is important to search for compounds with antihCMV properties. The present study aimed to investigate the suppressive effects of piceatannol on hCMV Towne strain infection and the putative underlying mechanisms using human diploid fibroblast WI-38 cells. Piceatannol supplementation prevented the lytic changes induced by hCMV infection in WI-38 cells. Furthermore, piceatannol suppressed the expression of hCMV immediate-early (IE) and early (E) proteins as well as the replication of hCMV DNA in a dose-dependent manner. Moreover, hCMV-induced cellular senescence was suppressed by piceatannol, as shown by a decline in the senescence-associated β-galactosidase (SA-β-Gal) activity and decreased production of intracellular reactive oxygen species (ROS). p16INK4a, a major senescence-associated molecule, was dramatically elevated by current hCMV infection that was attenuated by pre-incubation with piceatannol in a dose-dependent manner. These results demonstrated that piceatannol suppressed the hCMV infection via inhibition of the activation of p16INK4a and cellular senescence induced by hCMV. Together, these findings indicate piceatannol as a novel and potent anti-hCMV agent with the potential to be developed as an effective treatment for chronic hCMV infection.

        IL-33 Promotes ST2-Dependent Fibroblast Maturation via P38 and TGF-β in a Mouse Model of Epidural Fibrosis

        BACKGROUND: Recent evidence suggests that IL-33, a novel member of the IL-1b family, is involved in organ fibrosis. However, the roles of IL-33 and its receptor ST2 in epidural fibrosis post spine operation remain elusive. METHODS: A mouse model of epidural fibrosis was established after laminectomy. IL-33 in the wound tissues post laminectomy was measured with Western blotting, ELISA and immunoflurosence imaging. The fibroblast cell line NIH- 3T3 and primary fibroblasts were treated with IL-33 and the mechanisms of maturation of fibroblasts into myofibroblasts were analyzed. To explore roles of IL-33 and its receptor ST2 in vivo, IL-33 knockout (KO) and ST2 KO mice were employed to construct the model of laminectomy. The epidural fibrosis was evaluated using H&E and Masson staining, western-blotting, ELISA and immunohistochemistry. RESULTS: As demonstrated in western blotting and ELISA, IL-33 was increased in epidural wound tissues post laminectomy. The immunoflurosence imaging revealed that endothelial cells (CD31?) and fibroblasts (a-SAM?) were major producers of IL-33 in the epidural wound tissues. In vitro, IL-33 promoted fibroblast maturation, which was blocked by ST2 neutralization antibody, suggesting that IL-33-promoted-fibroblasts maturation was ST2 dependent. Further, IL-33/ ST2 activated MAPK p38 and TGF-b pathways. Either p38 inhibitor or TGF-b inhibitor decreased fibronectin and a-SAM production from IL-33-treated fibroblasts, suggesting that p38 and TGF-b were involved with IL-33/ST2 signal pathways in the fibroblasts maturation. In vivo, IL-33 KO or ST2 KO decreased fibronectin, a-SMA and collagen deposition in the wound tissues of mice that underwent spine surgery. In addition, TGF-b1 was decreased in IL-33 KO or ST2 KO epidural wound tissues. CONCLUSION: In summary, IL-33/ST2 promoted fibroblast differentiation into myofibroblasts via MAPK p38 and TGFb in a mouse model of epidural fibrosis after laminectomy.

      • MiR-150-5p Suppresses Colorectal Cancer Cell Migration and Invasion through Targeting MUC4

        Growing evidence suggests that miR-150-5p has an important role in regulating genesis of various types of cancer. However, the roles and the underlying mechanisms of miR-150-5p in development of colorectal cancer (CRC) remain largely unknown. Transwell chambers were used to analyze effects on cell migration and invasion by miR-150-5p. Quantitative real-time PCR (qRT-PCR), Western blotting and dual-luciferase 3' UTR reporter assay were carried out to identify the target genes of miR-150-5p. In our research, miR-150-5p suppressed CRC cell migration and invasion, and MUC4 was identified as a direct target gene. Its effects were partly blocked by re-expression of MUC4. In conclusiomn, miR-150-5p may suppress CRC metastasis through directly targeting MUC4, highlighting its potential as a novel agent for the treatment of CRC metastasis.
