        FK506이 T 림프구 사멸에서 활성산소 생성에 미치는 영향

        Purpose: Tacrolimus (FK506) has been widely used as an immunosuppressant in organ transplanted recipients to suppress organ rejection phenomenon. We investigated the role of oxidative stress and heme oxygense-1 by FK506 on human Jurkat T cells. Methods: The cells viability was examined by DAPI stain, enzyme activity of caspase family proteins, and western blotting for Baks, PUMA, iNOS, HO-1. Cells were cultured in the absence or presence of CoPPIX or ZnPPIX and the fluorescence intensity was analyzed using a flow cytometry. Results: Treatment with FK506 increased the generation of reactive oxygen species (ROS), including hydrogen peroxide and superoxide anion, and NO in Jurkat cells in a dose-dependent manner. Immunohistochemistry and Western blot analysis data revealed the hemoxygenase-1 (HO-1) was induced by the addition of FK506 in Jurkat cells. Induction of CoPP, HO-1 inducer, resulted in decreased intracellular H₂O₂ and NO concentrations. Instead ZnPP, an HO-1 competitive inhibitor did it reversely. In addition, ZnPP regulates iNOS protein synthesis by inhibition of HO-1. Conclusion: Increase of HO-1 expression would induce to decrease the intracellular H₂O₂ and NO concentrations. Also, HO-1 would regulate iNOS protein synthesis. Consequently, we can expect the regulation of HO-1 expression with concomitants use of FK506 to suppress organ rejection phenomenon by enhancing apoptosis.

        Mycophenolic Acid에 의해 유도된 Jurkat 세포주 세포자멸사에서 Heme Oxygenase-1의 발현이 미치는 영향

        Purpose: This study demonstrates that pharmacologic induction of heme oygenase-1 (HO-1) along with catalytic activation significantly modulated apoptosis of Jurkat cells induced by mycophenolic acid (MPA). Methods: Cells were cultured with the presence or absence of MPA. Flow cytometric analysis was performed after propidium iodide staining. Western blotting of HO-1, Bcl, and Bax was also performed. Cells were stained 4’-6-Diamidino-2-phenylindole (DAPI) and measured by flow cytometry in the absence or presence of CoPPIX. Results: Treatment of MPA decreased cell viability in a dose- and time-dependent manner. MPA-induced cell death was confirmed as apoptosis characterized by sub G0/G1 phase arrest. Expression of HO-1 assumes a pattern of decline after rising at the initial phase. CoPPIX, HO-1 inducer, significantly inhibited the cisplatin-induced apoptosis. Treatment of MPA resulted in reactive oxygen species (ROS) generation in Jurkat cells. CoPPIX attenuated ROS production in MPA-treated cells. Conclusion: This result suggests that the protective mechanism of HO-1 on MPA-induced cytotoxicity is associated with direct inhibition of ROS generation and mitochondrial permeability transition.

      • HEMA와 CA 및 PVA공중합체의 막제조와 물성에 관한 연구

        The grafting of 2-hydroxyethyl methacrylate(HEMA) and cellulose acetate(CA) and synthesis polymer poly(vinylalcohol) as a biocompatible material for medical application was investigated in the various condition. The structure of graft copolymer was indentified by using infrared spectroscopy.(IR) The membrane of sytnesis copolymer CA-g-p(HEMA) and PVA-g-p(HEMA) were prepared for a skin substitute and water content, tensile strength, elongation, water vapor transimmision rate(WVTR) and biodegradation the membranes was determind. And the surface structure of graft copolymer membrane characterized by SEM. The water content was about 100% and then tensile strength was 166∼661kg/㎠, 210∼88kg/㎠ at dry condition, 60∼328kg/㎠, 54∼23kg/㎠ at wet condition in the PVA-g-p(HEMA) and CA-g-p(HEMA) membrane respectively. Water vapor transimmision rate(WVTR) was 123.59∼128.54kg/㎠/day like that of the normal skin. From the result of the biodegradation by protein hydrolysis pepsin the biodegradation rate was that at the 4∼5day and accessed to the equilibrium state. From the results obtained in this study, syntheis membrane was found to satisfy most of the basic properties of a skin substitute.

      • 主要作物의 微量要素에 關한 硏究 : 第1報 硼素 및 有機物이 배추 生育에 미치는 影響 1. Effects of boron and organic matter on the chinese cabbage

        硼素缺乏地帶의 배추圃場에서 硼素의 施用方法 및 施用量을 달리하여 얻은 收量으로 肥效을 追求하고, 또 完熟堆肥의 效果를 調査한 結果를 要約하면 다음과 같다. 1. 硼砂 土壤施用에 依한 배추의 收量은 分散分析結果 1% 水準의 高度의 有意性을 보였으며, 硼砂 10a當 1,500㎏施用의 것이 가장 많은 收量이었고, 그 以上의 硼砂施用에서는 오히려 減少했다. 2. 배추의 硼酸 葉面施用의 差異에 依한 收量差는 土壤施用 때처럼 뚜렷한 差를 볼 수 없었다. 大體로 硼酸 0.25%∼0.5%의 濃度로 葉面撒布한 것이 좋은 結果를 거두었다. 3. 完熟堆肥의 增施에 따라 배추의 收量은 增加되었다. 10a當 1,800㎏ 施用에서 最大値의 收量에 이르고 그 以上의 增施에서는 별다른 收量의 增加를 볼 수 없었다. This experiment was designed to make clear the effects of the application of Boron and organic matter on chinese cabbage growing in Boron dificient soil. The results obtained were as follows. 1. The application of Borax in increased remarkably the yield of the chinese cabbage. The yield was increased by the application of Borax in the soil. It reached maximum amounts at 1,500g per 10a, but decreased above 1,500g per 10a. 2. The various concentration of Boron on the leaf spray was no more effective than the application of Boron in soil. The adequate concentration of Boric Acid on the leaf spray was from 0.25 percent solution to 0.5 percent solution. 3. The yield of the chinese cabbage was increased by the application of the compeletely decayed compost. It reached maximum amounts at the application if 1,800㎏ per 10a, but there was no effects above 1,800㎏ per 10a.

      • Ciprofloxacin 제제의 생물학적 동등성에 관한 연구

        국내 시판중인 ciprofloxacin 제제인 Citopcin®(250㎎ tablet)의 동등성을 검토하기 위해 제조원인 Bayer사의 Citopcin®(250㎎ tablet)를 기준제제로 하여 18명의 건강한 남성 피험자를 대상으로 500㎎ 1회 교차 경구 투여후 약동학적 성상을 분석비교한 결과는 다음과 같다. 시험제제의 생체 이용율의 지표인 AUC, C_(mao), T_(max) 및 MRT들의 평균치는 백분율차이에 있어 모든 기준제제 지표의 ± 20% 이내였으며, 이들 생체 이용율 지표들은 분산분석 검정에서 차이를 인지할 수 없었다.생체 이용율 지표들의 기준제제에 대한 백분율 90% 대칭 신뢰구간 검토시 모든 경수들에서 ± 20% 이내의 조건을 만족하였으며 AUC의 경우에는 대칭형 신뢰구간으로 변환시 95% 신뢰구간에서도 동등성의 조건을 만족하였다, 이상의 시험결과로 시험제제인 Citopcin®은 기준제제인 Ciprobay®와 생물학적 동등한 제제로 판단되었다. The pharmacokinetics of ciprofloxacin was studied for the evaluation of the bioequivalence of the generic ciproflocacin products. Two single doses of 500㎎ each of ciprofloxacin(Ciprobay® 250㎎ tablet as a reference compound and Citopcin® 250㎎ tablet s a test compound) were administered orally to ighteen male volunteers in a balanced, randomized crossover design. Pek plasma levels of ciprofloxacin were observed about 1 hour after the doses and the peak concentrations of both products were similar(Ciprobay®, 3.18㎍/㎖; Citopcin®, 2.89㎍/㎖). The values of other pharmacokinetic parameter of ciprofloxacin computed for Ciprobay® are presented in that order: AUC=13.53:12.47㎍·h/㎖, T_(max)=1.28:1.14 hours; MRT=5.30:5.08 hours; t_(1/2)β=3.65:3.47 hours. No satistically significant, differences were detected when AUC and the other parameters were compared with the method of ANOVA. Using the criteria of 90% confidence interval for the assessment of bioequivalence al the parameters were acceptable. The products were found to be equivalent on the premise that no significant difference was detected when the relevant pharmacokinetic parameters were compared, and the confidence limit analysis showed acceptable results.

        As the machine tool industry progresses, the performance of cutting tools also needs to be of good quality and specialized. If the existing metal cutting tool tip is coated properly, its life would be longer and the machining of a difficult-to-cut material also could be possible. For the development of the high quality cutting tool, the applicabilities of TiCN coating deposited by PVD on the cutting insert were experimentally investigated. The various analyses such as SEM micrograph, XRD pattern, AFM surface morphology, and hardness were performed to cross-check the possibility and availability of TiCN coated tool in the field.

      • 한국인의 온열쾌적감 평가 및 쾌적지표의 적용성에 관한 연구 : Part Ⅱ: 고령자 및 청년층의 겨울철 체감실험

        Recently aged people are highly increased. Engineers are needed to supply architectural environment suitable to aged because they are easily affected by the environment. Generally aged people hardly adjust themselves to a new environment and it gives them heavy loads mentally and physically. So it is necessary to consider the comfortable temperature of environment fitting body function of aged people. Many studies show that there are no differences in thermal comfort temperature between young and aged people in normal state. However, several researchers say that aged people are late to fit themselves to a changing thermal environment. They usually dislike low temperature after exposed in a cold environment. Experiments on thermal and comfort sensation have been done in winter. Most subjects were over 60 years old and healthy. The subjects were exposed to various environments. and they were indicated to vote Thermal Sensation Vote, TSV, and Comfort Sensation Vote, CSV, for the given environments. PMV and SET* has been used as the estimation standard of the thermal environment, and Mean Skin Temperature, MST, has been used for the estimation of the physical index. Through the experiments the comfort zone was SET* 25.7 to 28.2 in the heating season, as the range of TSV was over -0.5 and under +0.5. The value of SET* through the experiments were higher than that of SET*. 20.0 to 26.1, which the ASHRAE Comfort range has recommended. These data could be helpful to set the optimal environmental temperature and humidity for elderly people and to review the comfort index of indoor thermal environment for comfortable lives of them.

      • 重金屬이 Chlorella 세포의 生長및 呼吸能에 미치는 影響

        Chlarella ellipsoidea를 Hg, Cu, Pb, Zn과 같은 중금속 처리배지에서 배양하고 배양중 일정량의 세포를 收穫하여 생장률 및 엽록소 함량을 측정하고 여러농도에서 呼吸能을 측정 비교하였다. 1) 중금속 처리구에서의 세포의 生長, 엽록소함량 호흡能은 정상세포에 비해 정도의 차이는 있으나 모두 감소하였다. 2) 생장장해는 엽록소형성能과 비슷한 저하현상을 보였으며 ??서는 Hg가 가장 심한 장해 를 나타내었고 다음으로 Cu Zn Pb의 順이었다. 그러나 ?? Zn처리구에서는 ?? Hg나 Cu및 ?? Pb처리구보다 심하였다. 3) ??에서의 Hg및 Cu처리구에서는 각각 접종후 3시간 및 41시간후에 황백화현상과 더불어 사멸하였다. 4) 呼吸能은 대체로 ??이상의 농도에서 억제되는 경향을 나타내었다. Chlorella ellipsoidea cells were cultured in a Mercury, Lead, Copper or Zinctreated medium. Some portions of the cells were taken out at intervals during the culture, and the growth rate, the chlorophyll content and the respiratory activity of the cells were meaured. 1. Growth rate, chlorophyll content and respiratory activity of the heavy metaltreated cells decreased more or less, compared with these of the normal cells. 2. The growth retardation showed the similar tendency to the lowering of biosynthesis of chlorophyll, which in ?? medium came out to gradually severe in follow order, Lead, Znic, Copper, and Mercury. But the growth of the cells in ?? Zinc-treated medium was retarded more severely than in ?? Mercury or Copper and ?? Lead-treated medium. 3. The cells in the medium containing 1?? Mercury died out with chlorosis 3 hours later after inoculation, and those in ?? Copper-treated medium, 41hours later. 4. Generally the respiratory activity showed a tendency to de controlled in ?? and over.

        當歸類가 혈관신생에 미치는 영향에 대한 비교연구

        A rat sponge implant model was used to examine the angiogenic effect of water extract of Angelica Gigantis Radix (EAG). Angelica Sinensis Radix (EAS) and Angelica Radix (EAA). It was based on subcutaneous implantation of sterile polyester sponges in the Sprague Dawley (SD) rat and we have characterized a dye method that incorporates 5% carmine red in 10% gelatin. Vascularity index (V.I.) expressed as ㎍ dye/㎎ dry tissue. The result of study is as follows; 1. Subcutaneous implantation of sterile polyester sponges in SD rats enhanced the angiogenesis over 14 days, as determined by measurements of relative angiogenic effects by a vascularity index. 2. Daily oral administration (100 ㎎/㎏ for 14 days) of EAS and EAA produced weak effect on sponge-induced angiogenesis but EAG caused intense neovasculalization. 3. PBS (50 μ1) and EAG (100 ㎎/㎏) - induced angiogenesis represents a special form of angiogenesis. 4. The vasoconstrictor peptidem, angiotensin Ⅱ(AⅡ), in daily doses of 100 nmol, caused an intense neovascularization and coadministration of the AⅡ (100 nmol) and EAG (100 ㎎/㎏), AⅡ and EAS (100 ㎎/㎏) and AⅡ and EAA (100 ㎎/㎏) produced angiogenic effect. Especially, AⅡ (100 nmol) and EAG (100 ㎎/㎏) caused very potent angiogenesis. We have shown that oral administration of EAG (100 ㎎/㎏) induced angiogenesis. Thus the sponge model offers a new means for continuous and reproducible studies on the controlling mechanisms of angiogenesis.
