        양격산화탕(凉膈散火湯)의 항염증(抗炎症) 효과에 대한 연구

        Objective: Yang Geouk San Hwa - Tang (YGSHT) has been widely used in Sasang Constitutional Medicine of Korea for treatment of acute inflammatory symptom, such as palatine tonsillitis, polydipsia, headache, papule, pimple however, the mechanism of its anti-inflammatory activity has not been clarified. In this study, therefore, we investigated the mechanism of the inhibitory effect of YGSHT on LPS-induced inflammation. Materials and methods: The effect of YGSHT was analyzed by ELISA, RT-PCR and Western blotting in LPS-stimulated RAW264.7 cells. Results: We found that YGSHT suppressed not only the production of pre-inflammatory cytokines (IL-$1{\beta}$ and TNF-$\alpha$), the generation of nitric oxide (NO) and prostaglandin E (PGE)2, but also the mRNA expression of pre-inflammatory cytokines, inducible nitric oxide synthase (iNOS), and cyclooxygenase (COX)-2. Furthermore, YGSHT was shown to inhibit the phosphorylation of ERK1/2 and JNK1/2 and the activation and translocation of NF-kB from cytosol to nuclear in LPS-stimulated RAW264.7 cells. Conclusions: These results suggest that YGSHT exerts an anti-inflammatory effect through the regulation of the ERK1/2 and JNK1/2 pathway and NF-kB pathway, thereby decreasing production of pre-inflammatory cytokines, NO, and PGE2.

      • 중학생의 태권도에 대한 인식과 참여욕구에 관한 연구

        This study is attempted to survey the recognition of and desire for participation in Taegwondo for 920 students of 3 middle schools located in Myeonmok-dong of Seoul city as of 2003, to examine how they, our juveniles, recognize our proud Taegwondo and what they have desire for participation in Taegwondo, and to ultimately provide fundamental data for efficient Taegwondo education. Subjects of this study were selected randomly in 3 middle schools located in Myeonmokdong of Seoul City currently and totally 920 students from 1st year, 2nd year, and 3rd year students. Questionnaires were prepared on the basis of self-administration and, as a result of questionnaire, 10 questionnaires were excluded under the decision that they were not reliable and 910 valid samples were gained. For data analysis, SPSS 10.0 statistic program was applied and reliability analysis, frequency analysis, ANOVA, and pearson correlation were used as statistic analysis methods. Findings of this study from data analysis are as follows. First, it was shown that the recognition of middle school students for Taegwondo was high as a whole and, especially, the recognition of position and general recognition were significantly high. But, it was shown that they did not know well the cognitive area. Second, it was found that their desire for participation in Taegwondo was negative wholly while they showed high desire for participation with respect to psychomotor area and definitive area. Third, it was revealed that the higher their recognition of Taegwondo was, the higher their desire for participation in Taegwondo was.

      • 정맥내 통증자가조절법을 이용한 Nalbuphine-Ketorolac과 Butorphanol-Ketorolac의 술후 진통효과 비교

        This study aimed to compare analgegic efficacy, satisfaction score and side effect of nalbuphine-ketorolac and butorphanol-ketorolac by using intravenous patient controlled analgesia(IV-PCA) for postoperative anlgesia following a gastrectomy for stomach cancer. Ninety patients who had undergone gastrectomy for stomach cancer under general anesthesia were randomly divided into two groups. Each group recieived nalbuphine 50mg with ketorolac 150mg(Group 1, n=45) and butorphanol 5mg with ketorolac 150mg(Group 2, n=45) by using IV-PCA during postoperative 48hrs. Assessments for pain with numerical rating scale(NRS), and side effects were evaluated at 2hr, 6hr, 12hr, 24hr, 36hr and 48hr after the operation. There were no significant difference in NRS for pain during rest, but Group 2 requested significantly greater amount of supplementary diclofenac during first 24 hours. Side effects were higher Group 1 in pruritus, nausea and vomiting and Group 2 in sedation, nausea and vomiting. This study suggests that adding ketorolac with intravenous nalbuphine or butrophanol in using an intravenous PCA can decrease analgesics requirement and improve analgesic property without the major morbidity like respiratory depression, but needs for the careful observation and treatment on the side effects like nausea. vomiting, pruritis and sedation...

      • 미세절제술과 비교 유전자 보합법에 의한 각종 종양에서의 유전자 변화에 관한 연구

        For the evaluation of oncogenesis, progression and prognosis of cancer, CGH is an important technique, because this technique is economic due to utilization of only one probe and lack of culture, screening mathod of whole genome and possibility of retrospective and prospective study. By the CGH, genornic variation of 20 breast cancer tissues, 23 stomach cancer tissues and 16 bladder cancer tissues were analyzed. The results were as followes ; 1. breast cancers The CGH results showed gains on chromosomes 8q(40%), lq(30%), 17q(15%), 20q(15%), 18q (15%), 5p(15%), and 13q(15%). The Deletions were on chromosomes 17p(45%) and 22q(20%). High-level amplifications(green/red ratio >1.5) were noted on chromosomes 1p31, iq, 3q25-qter, 5p, 7q31-qter, 8q, 9q22-qter, 10p, l1p, 11q22-qter, 12p, 12q24, 14g21-qter, 15q23-qter, 17q, 18p, 18q12-qter, 20p, and 20q. By comparison with infiltrating ductal carcinoma, the two medullary carcinomas showed high-level amplification on chromosomes iq3l, lq, 8q, 10p, 11p and 12p. 2. stomach cancers 1) Usual amplification sites of genome were lq, 13q, 17q, 20p,q. 2) 17p was the most common deletion site. The other sites of the deletion were lq, 4q. 3) In intestinal type of stomach cancer, genomic variation is more common than diffuse type. 4) In the cases of no evidence of lymph node metastasis, deletion of 17p is absent but amplification of 8q is obvious in the case of lymph node metastasis. 3. bladder cancers Common amplification of copy numbers of DNA sequences by CGH were seen at 1q, 3q, 4q, 5p, 6pq, 7p, 8q, 11q, 12q, 13q, 17q, 18q and 20pq(more than 20% of cases). High level amplification was noted at 1p32, 3p2l, 3q24, 4q26, 8q21-ter, 11q14-22, 12q15-21, 12q21-24, 13q 21-31, 17q22, and 18q22. Deletions were noted at 2q21-qter, 4q13-23, 5q, 8p12-22, 9pq, 11p13-15 (more than 20% of cases).

        1q21.1 microdeletion identified by chromosomal microarray in a newborn with upper airway obstruction

        A 1q21.1 microdeletion is an extremely rare chromosomal abnormality that results in phenotypic diversity and incomplete penetrance. Patients with a 1q21.1 microdeletion exhibit neurological-psychiatric problems, microcephaly, epilepsy, facial dysmorphism, cataract, and thrombocytopenia absent radius syndrome. We reported a neonate with confirmed intrauterine growth restriction (IUGR), micrognathia, glossoptosis, upper airway obstruction, facial dysmorphism, and eye abnormality at birth as well as developmental delay at the age of 1 year. These clinical manifestations, except for the IUGR and upper airway obstruction, in the neonate indicated a 1q21.1 microdeletion. Here, we report a rare case of a 1q21.1 microdeletion obtained via paternal inheritance in a newborn with upper airway obstruction caused by glossoptosis and tracheal stenosis.

        Facilitation of AMPA receptor-mediated steady-state current by extrasynaptic NMDA receptors in supraoptic magnocellular neurosecretory cells

        In addition to classical synaptic transmission, information is transmitted between cells via the activation of extrasynaptic receptors that generate persistent tonic current in the brain. While growing evidence supports the presence of tonic NMDA current (I<sub>NMDA</sub>) generated by extrasynaptic NMDA receptors (eNMDARs), the functional significance of tonic I<sub>NMDA</sub> in various brain regions remains poorly understood. Here, we demonstrate that activation of eNMDARs that generate INMDA facilitates the α-amino-3-hydroxy-5-methylisoxazole-4-proprionate receptor (AMPAR)-mediated steady-state current in supraoptic nucleus (SON) magnocellular neurosecretory cells (MNCs). In low-Mg<sup>2+</sup> artificial cerebrospinal fluid (aCSF), glutamate induced an inward shift in I<sub>holding</sub> (I<sub>GLU</sub>) at a holding potential (V<sub>holding</sub>) of –70 mV which was partly blocked by an AMPAR antagonist, NBQX. NBQX-sensitive IGLU was observed even in normal aCSF at V<sub>holding</sub> of –40 mV or –20 mV. IGLU was completely abolished by pretreatment with an NMDAR blocker, AP5, under all tested conditions. AMPA induced a reproducible inward shift in Iholding (I<sub>AMPA</sub>) in SON MNCs. Pretreatment with AP5 attenuated I<sub>AMPA</sub> amplitudes to ~60% of the control levels in low-Mg <sup>2+</sup> aCSF, but not in normal aCSF at V<sub>holding</sub> of –70 mV. I<sub>AMPA</sub> attenuation by AP5 was also prominent in normal aCSF at depolarized holding potentials. Memantine, an eNMDAR blocker, mimicked the AP5-induced I<sub>AMPA</sub> attenuation in SON MNCs. Finally, chronic dehydration did not affect I<sub>AMPA</sub> attenuation by AP5 in the neurons. These results suggest that tonic INMDA, mediated by eNMDAR, facilitates AMPAR function, changing the postsynaptic response to its agonists in normal and osmotically challenged SON MNCs.

        최근에 (주) 녹십자가 개발한 일본뇌염 및 신증후 출혈열을 동시에 예방할 수 있는 혼합 백신(KGCC-95VI)의 아급성 독성을 조사하였다. ICR 마우스의 피하에 10, 50 및 250㎖/Kg(사람의 임상투여량의 20, 100 및 500배에 해당)의 KGCC-95VI를 매일 1회씩 30일간 투여하면서 임상증상을 관찰하였다. 시험기간동안 폐사한 동물을 없었고 KGCC-95VI에 의한 것으로 생각되는 임상증상도 없었다. 30일간의 투여후에 요검사, 혈액학적 검사, 혈액 화학적 검사 및 병리학적 검사를 실시한 결과 대조군과 비교할 때 아무런 이상이 확인되지 않았다. 따라서 KGCC-95VI는 마우스에서 아급성 독성을 갖고 있지 않는 것으로 생각된다.

      • Association between colony-stimulating factor 1 receptor gene polymorphisms and asthma risk

        <P>Colony-stimulating factor 1 receptor (<I>CSF1R</I>) is expressed in monocytes/macrophages and dendritic cells. These cells play important roles in the innate immune response, which is regarded as an important aspect of asthma development. Genetic alterations in the <I>CSF1R</I> gene may contribute to the development of asthma. We investigated whether <I>CSF1R</I> gene polymorphisms were associated with the risk of asthma. Through direct DNA sequencing of the <I>CSF1R</I> gene, we identified 28 single nucleotide polymorphisms (SNPs) and genotyped them in 303 normal controls and 498 asthmatic patients. Expression of <I>CSF1R</I> protein and mRNA were measured on CD14-positive monocytes and neutrophils in peripheral blood of asthmatic patients using flow cytometry and real-time PCR. Among the 28 polymorphisms, two intronic polymorphism (+<I>20511C</I>><I>T</I> and +<I>22693T</I>><I>C</I>) were associated with the risk of asthma by logistic regression analysis. The frequencies of the minor allele at <I>CSF1R</I> +<I>20511C</I>><I>T</I> and +<I>22693T</I>><I>C</I> were higher in asthmatic subjects than in normal controls (4.6 vs. 7.7%, <I>p</I> = 0.001 in co-dominant and dominant models; 16.4 vs. 25.8%, <I>p</I> = 0.0006 in a recessive model). <I>CSF1R</I> mRNA levels in neutrophils of the asthmatic patients having the +<I>22693CC</I> allele were higher than in those having the +<I>22693TT</I> allele (<I>p</I> = 0.026). Asthmatic patients with the +<I>22693CC</I> allele also showed significantly higher <I>CSF1R</I> expression on CD14-positive monocytes and neutrophils than did those with the +<I>22693TT</I> allele (<I>p</I> = 0.045 and <I>p</I> = 0.044). The +<I>20511C</I>><I>T</I> SNP had no association with <I>CSF1R</I> mRNA or protein expression. In conclusion, the minor allele at <I>CSF1R</I> +<I>22693T</I>>C may have a susceptibility effect in the development of asthma, via increased <I>CSF1R</I> protein and mRNA expression in inflammatory cells.</P><P><B>Electronic supplementary material</B></P><P>The online version of this article (doi:10.1007/s00439-010-0850-3) contains supplementary material, which is available to authorized users.</P>

      • Mammalian Status of Mt. Cheomchalsan in Jindo, Korea

        In order to analyze a status of animal in an area where has been became a land cause of Jindo Grand Bridge established on Jindo island area, mammal status in the biggest mountain in Jindo province, Mt. Cheomchalsan has been investigated for four times from May to October of 2016. According to the investigation, 5 orders, 9 families and 15 species of inhabitation was confirmed, and water deer (Hydropotes inermis), raccoon (Nyctereutes procyonoides), cat (Felis catus), and etcetera were dominant. Biodiversity and evenness index were likely to be high as 2.24 and 0.83, respectively and it is considered that favorable condition of natural ecosystem for inhabitation of mammalian is established. Meanwhile, since the cat designated as control species is confirmed as a dominant species, it is supposed that continuous management is necessary.
