        Glutathione S-transferases (GSTM1, GSTT1 and GSTP1) and N-acetyltransferase 2 Polymorphisms and the Risk of Gastric Cancer

        Objectives : Polymorphisms of genes from glutathione Stransferases (GSTs) and N-acetyltransferase 2 (NAT2) have been associated with increased susceptibility to various cancers. Previous results showed that East Asians such as Koreans, Japanese and Chinese have a much higher frequency of the GSTM1 and GSTT1 null genotypes and NAT2 rapid acetylator type. Therefore, we investigated the association between the polymorphic types of GSTs (GSTM1, GSTT1, GSTP1) and NAT2 and the incidence of gastric cancer which is one of the most prevalent cancers among the East Asians. Methods : It was performed in a case-control study consisting of 238 healthy subjects and 108 cancer patients (54 distal and 54 proximal carcinomas). We also evaluated the association between GSTs and NAT2 and the risk factors for gastric cancer such as alcohol consumption, smoking, H. pylori infection, family history of gastric cancer, and tumor location. Results : In our study, the percentage of cases whose hometown was rural was higher than those of controls (odds ratio (OR) =2.88; 95% CI=1.72-4.76), and the frequency of the lower socio-economic status increased significantly in patients (OR=2.53; 95% CI=1.59-4.02). There was no significant difference in the GST polymorphic types between the cases and controls. However, NAT2 rapid or intermediate acetylator types were frequently detected in the cases with family history of gastric cancer (OR=1.92; 95% CI=1.79-26.0). Conclusions : These results suggest that the hometown and socio-economic status are important environmental factors for gastric carcinogenesis, and NAT2 polymorphic types could be associated with familial gastric carcinoma.

        항생물질을 생산하는 Clostridium sp. KH-431의 동정 및 항생물질의 정제

        순수분리된 2,000여 주의 혐기성 세균으로 부터 선발된 stain KH-431은 bacteria 및 fungi에 대하여 재현성있는 항균활성을 보이는 물질을 생산하였다. Strain KH-431은 그 형태 및 생리, 생화학적 특성으로 미루어 보아 Clostridium fallax의 근연종으로 동정되었다. Strain KH-431의 항생물질 생산을 위한 최적 배양조건에 대해 고찰한 결과 PY배지를 기초배지로 사용하고 sorbitol, yeast extract, d-biotin 그리고 CaCl_2를 영양원으로 첨가하였을 때가 양호한 것으로 나타났다. 배지 초기 pH 및 배양온도, 배양시간에 대한 최적조건은 각각 pH7과 30℃, 108시간으로 나타났다. 20ℓ의 배양액으로 부터 solvent extraction, silica gel column chromatography를 거쳐 얻은 sample을 재결정한 결과 KG-431B는 순수한 결정상태로 얻어졌으나 KG-431A의 완전정제에는 많은 문제점이 뒤따른 바 현재 다른 정제방법의 도입을 고려중에 있다. A strain showing antibiotic activities against various bacteria and fungi was selected from approximately 2,000 microorganisms obtained from soil samples. This strain, designated as KH-431, was identified as a Clostridium sp. by its morphological, physiological and biochemical characteristics. The highest production of the antibiotics was achieved in a fermentation medium containing sorbitol, yeast extract, d-biotin and CaCl_2. The antibiotics were isolated from the culture broth by solvent extraction using ethyl acetate, silica gel column chromatography and recrystallization. Two kinds of antibiotics, KG-431A and KG-431B were obtained after the purification procedure, and only KG-431B was successful to recrystallize.

        Clostridium sp.가 생산하는 항생물질 KG-1167A 와 KG-1167B의 정제 및 특성

        순수분리된 2,200여주의 혐기성 세균으로부터 선별된 Clostridium sp. KH-1167은 그람양성 및 음성균에 대하여 재현성있는 항균활성을 나타내는 물질을 생산하였다. Solvent extraction, silica gel column chromatography에 의하여 배양액으로부터 항생물질 KG-1167A와 KG-1167B를 부분정제 하였다. 이들은 pH4∼11의 범위에서는 비교적 안정하였으며, 온도에 대해서는 50℃ 정도에서부터 점차 불안정해지는 것을 알 수 있었다. UV λ_max 분석결과로부터 이들은 aromatic ring을 포함하고 있는 화합물로 추정되는데, 현재 진행 중인 MS, ^1H-NMR 등의 분석결과와도 상당히 일치되었다. 19종의 그람양성 및 음성균에 대한 부분정제된 KG-1167A와 KG-1167B의 MIC값은 32∼128㎍/㎖로 나타났으며 mouse에 대한 LD_50값은 150∼200㎎/㎏이었다. The antibiotics KG-1167A and KG-1167B were isolated from the fermentation broth of the bacterial strain KH-1167, identified as Clostridium sp. The individual antibiotics, KG-1167A & KG-1167B were separated and purified by solvent extraction followed by silica gel column chromatography. These antibiotics showed antibacterial activities against broad spectrum of gram positive and gram negative bacteria. The physico-chemical properties and UV spectra suggest that they possess aromatic moiety in their structures.

        사물인터넷의 발달에 따라 언제 어디서나 건강관리 및 의료서비스가 가능한 스마트 헬스케어 서비스가 증가하고 있다. 이에 본 연구에서는 사물인터넷(IOT)기반 스마트 헬스케어 시스템 및 헬스케어 모니터링 장치 구현 사례를 살펴보고, 이와 관련된 개인건강기기 국제표준기구인 IEEE 11073 PHD 정의 및 구성, Device에 대해 알아보았다. 또한, 의료 서비스 및 의료 산업에 적용된 사례를 분석하여 당면 문제점을 살펴보고, 스마트 헬스케어 산업의 발전방안을 모색하였다.

        DNA Hypermethylation of Tumor-Related Genes in Gastric Carcinoma

        The hypermethylation of the CpG islands is a common mechanism for the inacti-vation of tumor-related genes. In the present study, we analyzed the methylation status of genes for cell repair such as hMLH1, MGMT, and GSTP1, and a gastric cancer-specifically methylated DNA fragment, MINT 25 in gastric cancer cases and control groups. The study population consisted of 100 gastric cancer patients (50 distal and 50 proximal carcinomas), and 238 healthy controls. All genes showed more frequent hypermethylation in the cases than in the control group (p<0.0001). We investigated the association between promoter hypermethylation and relevant parameters including age, gender, alcohol consumption, smoking, and family his-tory. There was a common hypermethylation of hMLH1 (p=0.008), MGMT (p= 0.0001), and GSTP1 (p=0.0003) in females. This study also demonstrates that hypermethylation was strongly associated with non-drinkers (MGMT, p=0.046 and MINT 25, p=0.049) and non-smokers (hMLH1, p=0.044; MGMT, p=0.0003; MINT 25, p=0.029). Moreover, the frequency of MINT 25 hypermethylation increased with age (p=0.037), and MGMT methylation was frequently detected in distal gas-tric cancer than in proximal type (p=0.038). Our study suggested that promoter hypermethylation of the genes involved in cell repair system and MINT 25 is asso-ciated strongly with some subgroups of primary gastric carcinoma.

        Clostridium sp. KH-431이 생산하는 항생물질의 특성 및 구조

        순수분리된 2,000여주의 혐기성 세균으로부터 선발된 Clostridium sp. KH-431은 bacteria 및 fungi에 대하여 재현성있는 항균활성을 나타내는 항생물질 KG-431A와 KG-431B를 생산하였다. 이들 중 KG-431B는 항암활성의 1차 검색에서도 양성반응을 보인 바 이에 대한 후속연구를 계획중에 있다. 이들의 MIC 값을 비교해 보면 완전정제가 가능하였던 KG-431B 보다는 차라리 정제과정에서 많은 어려움이 따라 불완전 정제된 상태로 실험을 행한 KG-431A의 활성이 더 뛰어난 것을 알 수 있었다. 정제된 KG-431B의 여러가지 이화학적 특성과 spectra data가 기지물질인 3-IPA와 일치하나 본 실험을 통해 밝혀진 항암활성의 검색은 정제된 3-IPA 존재에 대한 결과는 아직 정제가 완료되지 않은 KG-431B에 관한 연구와 함께 후속적인 확인 연구가 진행 중에 있다. Antibiotics KG-431A and KG-431B were isolated from the fermentation broth of the Clostridium sp. KH-431. As we have shown previously, only KG-431B was successful to recrystallize. These antibiotics showed antimicrobial activities against broad spectrum of bacteria and fungi. KG-431B also showed anticancer activity against some animal tumor cells according to the SRB method. Physico-chemical properties of KG-431B were determined using UV, IR, NMR and Mass spectra. It was identified to be 3-Indole propionic acid and the structure of the KG-431A is currently under investigation.

        Pseudomonas aeruginosa KGM-100이 생산하는 항생물질의 특성 및 구조

        미생물 대사산물로부터 항 곰팡이성 항생물질 탐색연구를 수행하던 중 광범위한 식물 병원성 곰팡이에 강한 억제활성을 보이는 Pseudomonas aeruginosa KGM-100을 선발하였으며, 이 균주가 생산하는 항생 물질의 분리, 정제 및 생물학적, 이화학적 특성 및 구조분석을 행하였다. 균 배양액으로부터 HP-20 column chromatography, ethyl acetate extraction, silica gel column chromatography, preparative TLC, recrystallization 등을 통해 KGM-100A와 KGM-100B의 두 종류의 항생물질을 순수분리하였으며, 항균 spectrum을 조사한 결과 methanol로 재결정된 KGM-100A가 다양한 종류의 bacteria와 곰팡이에 강한 활성을 보였다. 이중 KGM-100A를 UV, IR, ^1H NMR, ^(13)C NMR, Mass spectrum, 원소분석 등의 기기분석을 한 결과 phenazine-1-carboxylic acid로 동정되었다. During the screening of antifungal antibiotics from microbial metabolites, we selected Pseudomonas aeruginosa KGM-100 showing powerful antagonistic activity against various phytopathogenic fungi. Antibiotics KGM-100A and KGM-100B were purified from the culture broth of Pseudomonas aeruginosa KGM-100 by diaion HP-20 column chromatography, ethyl acetate extraction, silica gel column chromatography, preparative TLC and recrystallization. KGM-100A which was recrystallized in MeOH showed antimicrobial activities against a broad spectrum of fungi and bacteria. Physico-chemical properties of KGM-100A were determined and identified to be phenazine-1-carboxylic acid by UV, IR ^1H-NMR, ^(13)C-NMR, mass spectrum, and elemental analyses.

        Ribosomal RNA와 M13 probe에 의한 clostridium thermocellum 균주들의 RFLP(Restriction Fragment Length Polymorphism)비교

        The degree of the genetic variations among Clostridium thermocellum ATCC 27405 and the wild type strains was investigated by the mehtod of GC ratio, DNA-DNA hybridization and RFLP (Restriction Fragment Length Polymorphism) patterns by ribosomal RNA and M13 probe. GC ratio and KNA homology values of th three isolates were approximately equal to those of ATCC type strain. The RFLP patterns by the rRNA and M13 probe showed some differences among C. thermocellum ATCC 27405, wild type strains and Clostridium thermohydrosulfuricum ATCC 33223, indicating that the two probes can be useful in subspecies- and apecies-identification.

        표준 요 시료 중 Oxalate의 측정을 위한 FT-NIR 분광기의 유용성 검정

        Objectives : The determination of oxalate in urine is required for the diagnosis and treatment of primary hyperoxaluria, idiopathic stone disease and various intestinal diseases. We examined the possibility of using Fourier transform near-infrared (FT-NIR) spectroscopy analysis to quantitate urinary oxalate. The practical advantages of this method include ease of the sample preparation and operation technique, the absence of sample pre-treatments, rapid determination and noninvasiveness. Methods : The range of oxalate concentration in standard urine solutions was $0-221mg/{\ell}$. These 80 different samples were scanned in the region of 780-1,300 nm with a 0.5 nm data interval by a Spectrum One NTS FT-NIR spectrometer. PCR, PLSR and MLR regression models were used to calculate and evaluate the calibration equation. Results : The PCR and PLSR calibration models were obtained from the spectral data and they are exactly same. The standard error of estimation (SEE) and the % variance were $10.34mg/{\ell}$ and 97.86%, respectively. After full cross validation of this model, the standard error of estimation was $5,287mg/{\ell}$, which was much smaller than that of the pre-validation. Furthermore, the MCC (multiple correlation coefficient) was 0.998, which was compatible with the 0.923 or 0.999 obtained from the previous enzymatic methods. Conclusions : These results showed that FT-NIR spectroscopy can be used for rapid determination of the concentration of oxalate in human urine samples.
