        핵치환 생쥐의 생산

        Pronuclear transplantation between ICR and Fl hybrid(C57BL×CBA) mouse eggs was achieved by using a method that combines microsurgical removal of the zygote pronuclei with the introduction of donor pronuclei by a virus-mediated cell fusion technique. The technical efficiency of this procedure composed of zona cutting, enucleation, karyoplast injection and fusion step was more than 90 % at each step. Developmental rate of these nuclear transplanted embryos to blastocyst was about 60 % and when these embryos were transferred to foster mother mice. 4 nuclear transplanted mice were born.

        백렴 추출물의 Matrix Metalloproteinase-1 발현 저해 효과

        Matrix metalloproteinases (MMPs) are known to play an important role in photoaging by mediating the degradation of extracellular matrix proteins. In this study, to develop a n ew anti-aging agent, we investigated the antioxidant activity and the inhibitory effect of Melothria heterophylla extract on expression of MMP-1 in UVA-irradiated human dermal fibroblasts and MMP-1 activity. The M.heterophylla extract was found to scavenge radicals and reactive oxygen species (ROS) with the $SC_{50}$ values of $13{\mu}g/ml$ against 1,1-diphenyl-2-picrylhydrazyl (DPPH) radicals and $20{\mu}g/ml$ against superoxide radicals in the xanthine/xanthine oxidase system, respectively. UVA-induced MMP-1 expression was reduced about 80% by $100{\mu}g/ml$ of the M.heterophylla extract but MMP-1 Mrna expression was not inhibited. Therefore, we conclude that the M.heterophylla extract significantly inhibits MMP-1 expression at the protein level. Also, the M.heterophylla extract inhibited MMP-1 activity in a dose dependent manner. From these results, we suggest that the M.heterophylla extract can be used as a new anti-aging agent by antioxidant activity, regulation of UVA-induced MMP-1 production, and inhibition of MMP-1 activity.

        Streptozotocin - 유도 당뇨모델을 이용한 쥐눈이콩 분말의 혈당강하 및 인슐린 감수성의 효과

        본 연구에서는 임실특산물인 쥐눈이콩및 쥐눈이식초콩의 혈당강하 및 인슐린 감수성에 대한 효과를 검정콩, 메주콩과 비교하여 조사하였다. 식이물질의 성분검사 결과 쥐눈이콩과 쥐눈이식초콩이 검정콩과 메주콩에 비하여 당뇨개선의 효과가 있는 여러 아미노산 및 식이섬유 함량이 유의적으로 높게 측정되었다. 실험을 위해 평균체중 195.5±0.98 g의 숫컷쥐를 정상군(NC), 당뇨대조군(DI, DC), 검정콩식이군(DB), 메주콩식이군(DY), 쥐눈이콩식이군(DS), 쥐눈이식초콩식이군(DSS)으로 총 7개군으로 나누어 해당식이첨가 시료로 4주간 사육하였다. 당뇨유발을 위해 STZ를 1회(70 mg/kg/rat) 투여하였으며 그 결과 체중변화는 당뇨대조군(DC)에서 21.12 g 감소함에 반하여 각각의 시료를 첨가식이한 당뇨실험군에서 특히 쥐눈이콩(DS)과 쥐눈이식초콩(DSS) 식이군은 높은 식이이용 효율(FER)을 보여 각각 14.73±3.65 g 및 16.71±5.54 g의 몸무게 증가를 보였다. 음용수 및 뇨량의 측정결과 역시 당뇨실험군에서 유의적인 감소(p<0.05)를 보였다. 혈당량은 당뇨대조군(DC)에 비하여 검정콩(DB), 메주콩(DY), 쥐눈이콩(DS), 쥐눈이식초콩(DSS) 식이군이 각각 17.9%, 16.9%, 10.35%, 19.54%씩 유의하게 감소하였으나 정상대조군(NC)보다는 높은 수치를 나타내었다. 또한 당뇨쥐의 인슐린 감수성에 대한 관찰결과 당뇨대조군 중 인슐린 무처지군(DC)에 비하여 모든 당뇨실험군에서 높은 값을 보였다. 결론적으로 임실산 쥐눈이콩과 쥐눈이식초콩은 당뇨쥐의 혈당강하 및 인슐린감수성에 대한 개선작용의 가능성이 있으므로 당뇨합병증 예방의 기능성 식품으로 기대된다. Beans are acknowledged to be food resources, which have more abundant proteins and fats. The constituent parts of beans (i.e. aspartic aid, glycine, arginine) are effective against diabetes, and dietary fiber contained in the beans has an important property to maintain insulin sensitivity. Based on these, using streptozotocin (STZ)-induced diabetic rats, this study examined how the rat-eye soybean, which is principal products of the Imsil province, is effective to attenuate and/or prevent the development of diabetes mellitus. We divided rats into the non-diabetic and diabetic group, and diabetic group was further subdivided into six experimental groups [DC, diabetic control; DI, diabetes with insulin treatment (4~6 IU/rat); DB, diabetes with black bean;DY, diabetes with yellow soybean; DS, diabetes with rat-eye soybean; DSS, diabetes with vinegar-fermented rat-eye soybean. All bean treatment (1.5 ㎎/1 g body weight).]. Food efficiency ratio (FER), body weight and insulin sensitivity in diabetic rats were significantly reduced compared to those in normal control animals. These reductions were obviously attenuated by administration of a variety of beans used in this study (20~30%), and the recovery effects were comparable to the results obtained by insulin treatment. Taken together, this study suggests that all beans used may have an essential property to improve and/or attenuate the development of diabetes mellitus in rats.

      • 브로콜리 줄기의 callus배양 및 항노화 효과

        Broccoli (Brassica oleracea var. italica) is well known of excellent antioxidant food so that we researched on its anti-aging and callus differentiation from the tissue of broccoli stem by MS medium with exo-phytohormone as IAA, kinetin. The functional materials of callus tissue were extracted with water and 70% EtOH after 16 days progressed callus formation. As a result, callus differentiation and formation were showed higher efficiency at B5 condition medium (0.1 mg/L IAA, 0.4 mg/L). The callus cultured on B3, B6 and B7 medium were observed the reaction of enzymatic browning to callus tissue. The anti-aging effect on the extract was experiment with DPPH radical scavenging and catalase(CAT). The activity of DPPH radical scavenging was showed high value in water extract of B1, B6, B8, B9 and was increased depending on sample concentration. However, in case of the extract of 70% EtOH, DPPH activity was showed different effect depending on the sample kind. CAT activity was found the highest enzyme unit in the extract of natural broccoli and B1(no add exo-phytohormone). It is to express that a broccoli itself is great food for anti-aging. Therefore, callus formation of broccoli considers having no economic efficiency to do development of anti-aging product.

      • 고추(Capsicum annuum) callis와 검은 무늬병 유발균의 elicitor를 이용한 항균물질 생산

        We described the effects of antimicrobial activity on a produced material by the co-culture of elicitor inducing black rot's disease and callus of Capsicum annuum that callus was induced in MS medium containing IAA 0.5 mg/ml and kinetin 2.0 mg/ml. the bacterial elicitor used 0.5% Yeast extract and the fungal elicitor made by Alternaria alternata KACC 40019 with PDB medium. In order to produce antimicrobial agent protective balck rot's disease, we co-cultured the fungal elicitor or bacterial elicitor with the red pepper callus tissue for 7days repectively. The result, an antifungal agent was produced extracellular materials into medium during co-culture red pepper callus with the fungal elicitor. The antifungal activity of the extracellular material was showed 23% inhibition rate on Alternaria alternata KACC 40019 on the other hand the intracellular material was showed 2% inhibition rate. A chitinase activity of extracellular material was also showed higher value than a chitinase of the intracellular material. The extracellular material produced by elicitor was had more 15% of andtibacterial activity than the intracellular material. As a result, a biotic stress as a fungal elicitor and a bacterial elicitor brings a defence system in the red pepper callus and induce to synthesis in extracellular material an antimicrobial agent as a chitinase and phytoalexin.

      • Asparagus와 shallot, 방울 양배추의 항노화 효과

        We experimented to investigate the anti-aging effect on the extracts of Asparagus officinalis L. (asparaguse), Eleutherine Americana Merr. (shallot) and Brassica oleracea var. gemmifera (Brussels sprouts). It was respectively extracted for 7 days at 4 ℃ with 70% EtOH and water as solvent and those extracts were determined the concentration of total polyphenolic compound, flavonoid, free radical scavenging as DPPH and catalase activity. In a result, the water and 70% EtOH extract of asparagus were showed over 50% activity of DPPH radical scavenging. The extracts of shallot were had high concentration of polyphenol and flavonoid than the extracts of other sample, but DPPH radical scavenging was found more low activity than the other sample. The extracts of Brussels sprouts were showed lowest concentration of polyphenol and flavonoid among samples, but DPPH radical scavenging was measured highest activity over 60%. And the extract of asparagus and Brussels sprouts were reversely increased the activity of DPPH radical scavenging in a case of the treatment of low concentration. Catalase was confirmed high activity the extract of asparagus tissue and the extract of Brussels sprouts tissue in order. Therefore, Asparagus officinalis L. and Brassica oleracea var. gemmifera have the effect of anti-aging and it is good garnish vegetable eating with meat.

      • Fungal Elicitor와 Exo-phytohormone이 청양고추(Capsicum annuum L.) 종자발아와 2차 대사산물에 미치는 효과

        We studied growth rate and synthesis of secondary metabolite as capsaicin and polyphenolic compound during seed germination of Capsicum annuum L. on condition media with fungal elicitor and exo-phytohormone. The condition media was consisted MS (Murashige & Skoog) medium and sucrose for carbon source with different concentration of elicitor and exo-phytohormone. The fungal elicitor was made by Aspergillus niger KCCM 11240 and exo-phytohormone used kinetin, IAA (indole-3-acetic acid). The secondary metabolites were extracted from the seeding tissue by acetone as solvent after 20 days. As a result, the germination of seed was found on the third day and growth of seeding was observed on the seventh day. The capsaicin concentration was had the highest concentration in R0’’ (no exo-phytohormone, 180 mg/ml of fungal elicitor) extract among all extract solutions. The polyphenolic compound was had high concentration in R1’’ (0.1 mg/L of IAA and kinetin, 180 mg/ml of fungal elicitor) extract. The inhibition rate of lipid peroxidation was showed in proportion to the concentration of capsaicin. The effect of NO radical scavenging was measured more activity to natural pepper fruit than seeding cultured in vitro and that activity considered to be phytonutrient included capsaicin and polyphenolic compound. The fungal elicitor inhibited growth and differentiation of seeding, but it induced the synthesis of secondary metabolites. The secondary metabolites were in inverse proportion to total length of seeding. Therefore, capsaicin as phytonutrient can mass-produce by plant tissue culture with biotic stress.

      • 식물 추출물을 이용한 초파리 기피제 개발

        To study repellent of Drosophila melanogaster, we extracted plant materials from wild sesame leaf, cinnamon, fig leaf, ginko leaf, quince with 100% ethanol or water as an extract solvent and described an insecticidal effect on the plant extract respectively. In the result, the 100% ethanol extract had higher insecticidal effect than the water extract. The ethanol extract of wild sesame leaf showed 58% extinction effect on Drosophila and the extracts of cinnamon and fig leaves had the effect of repellent. Each ethanil extract and the extract of cinnamon water were mixed with the ethanol extract of wild sesame leaf and a secondary test on the repellent effect has been conducted. In case of cinnamon extract mixed with wild sesame leaf, it showed no larva development and low Drosophila influx and also confirmed the same effect in the ethanol extract of fig leaf. The ethanol extract of wild sesame leaf has aromatic compound and high level of flavonoid concentration that affect like lure materials and accelerate a repellent effect of Drosophila. The cinnamon extract has high concentration of phenol compound that reasons material on repellent effect. However, the effect of fig leaf can not describe because it is low concentration of phenol compound and flavonoid compared with wild sesame leaf and cinnamon. Therefore, in case of mixed plant extracts with the ethanol extract of wild sesame leaves it can make repellent product to induce larva inhibition and insecticidal effect.

      • 여주(Momoridica charantia)열매 추출물의 향균효과 검정

        We conducted experiments on the different development stage of Momoridica charantia fruit and different types of solvent, water and 70% EtOH, affect the content of total polyphenol and flavonoid and its antimicrobial activity. The total polyphenol and flavonoid of each extracted solution from Momordica charantia fruit was determinated by colorimetric method. The antibacterial and antifungal activity was studied by paper disc diffusion method and inhibition rate with potato dextrose agar (PDA) respectively. The result, total polyphenol was determinated higher content to 70% EtOH extracted unripe fruit, and 70% EtOH extracted seed hull. Flavonoid concentration was desterminated the highest content to water extracted ripe fruit. The results of the antibacterial activities was showed that water extracted unripe fruit had 43% antibacterial activity against Neisseria gonorrheoae KCTC 2746. While all extracted solution was showed the highest antifungal activity against balck rot's disease. 10 mg/ml of extracted solutions were particularly exhibited an average 40% and over anti-fungal activity. Therefore, the fruit extracts of Momordica charantia possess the effect of biological prevention on the black rot's disease of plant.
