        Characterizing Milk Production Related Genes in Holstein Using RNA-seq

        Although the chemical, physical, and nutritional properties of bovine milk have been extensively studied, only a few studies have attempted to characterize milk-synthesizing genes using RNA-seq data. RNA-seq data was collected from 21 Holstein samples, along with group information about milk production ability; milk yield; and protein, fat, and solid contents. Meta-analysis was employed in order to generally characterize genes related to milk production. In addition, we attempted to investigate the relationship between milk related traits, parity, and lactation period. We observed that milk fat is highly correlated with lactation period; this result indicates that this effect should be considered in the model in order to accurately detect milk production related genes. By employing our developed model, 271 genes were significantly (false discovery rate [FDR] adjusted p-value<0.1) detected as milk production related differentially expressed genes. Of these genes, five (albumin, nitric oxide synthase 3, RNA-binding region (RNP1, RRM) containing 3, secreted and transmembrane 1, and serine palmitoyltransferase, small subunit B) were technically validated using quantitative real-time polymerase chain reaction (qRT-PCR) in order to check the accuracy of RNA-seq analysis. Finally, 83 gene ontology biological processes including several blood vessel and mammary gland development related terms, were significantly detected using DAVID gene-set enrichment analysis. From these results, we observed that detected milk production related genes are highly enriched in the circulation system process and mammary gland related biological functions. In addition, we observed that detected genes including caveolin 1, mammary serum amyloid A3.2, lingual antimicrobial peptide, cathelicidin 4 (CATHL4), cathelicidin 6 (CATHL6) have been reported in other species as milk production related gene. For this reason, we concluded that our detected 271 genes would be strong candidates for determining milk production.

      • 착유우의 사료섭취량과 음수량이 산유량과 분뇨 배설량에 미치는 영향

        본 실험은 우리나라에서 사육되고 있는 홀스타인 젖소 착유우의 사료 섭취량과 음수량이 산유량과 분뇨 배설량에 미치는 영향을 구명하기 위하여 수행하였다. 실험은 착유우 10두를 공시하여 수원의 축산연구소 낙농우사에서 봄, 여름, 가을, 겨울 등 4계절에 걸처 실시하였으며 그 결과는 다음과 같다. 1. 시험에 공시된 착유우의 건물섭취량은 평균 19.5kg으로 봄과 가을이 각각 23.9, 22.1kg으로 높았고, 여름과 겨울이 각각 17.0, 15.3 kg으로 낮은 섭취량을 보였다. 2. 착유우의 음수량은 평균 $77.2\ell$로, 여름이 $85.5\ell$로 가장 많았고 겨울이 $62.2\ell$로 가장 낮았으나 계절별 유의차는 보이지 않았다. 3. 착유우의 산유량은 평균 26.0kg으로 봄, 여름, 가을 및 겨울이 각각 30.8, 24.0, 25.4, 23.7kg으로 봄의 산유량이 가장 높았다(p<0.05). 4. 착유우의 분뇨 배설량을 계절별로 조사한 결과, 봄 64.4kg, 여름 63.5kg, 가을 60.4kg, 겨울 51.0kg으로 계절 간에 차이를 보였으며(p<0.05), 전체 평균 분뇨 배설량은 59.8kg이었다. 5. 착유우의 사료섭취량, 음수량, 산유량 및 분뇨 배설량과의 상관관계를 분석한 결과, 산유량과 음수량$(R^2=0.7742)$, 산유량과 사료 섭취량$(R^2=0.7459)$, 음수량과 뇨 배설량 $(R^2=0.7422)$, 사료 섭취량과 분 배설량$(R^2=0.6044)$, 산유량과 분 배설량$(R^2=0.692)$의 상관관계가 비교적 높았으나, 음수량과 분 배설량$(R^2=0.2950)$, 사료 섭취량과 뇨 배설량$(R^2=0.1985)$, 산유량과 뇨 배설량$(R^2=0.2335)$의 상관관계는 낮게 나타났다. 6. 따라서 산유량과 음수량, 산유량과 사료 섭취량의 상관관계식은 $Y=0.1919X_1+11.181(R^2=0.7742),\;Y=0.8568X_2+9.3067(R^2=0.7459)$(Y=milk yield $X_1=water$ consumption, $X_2=feed$ intake)로 추정할 수 있다. This experiment was conducted to determine the effect of feed intake and water consumption on milk yield and manure production in milking cows. The average feed intake(dry matter) of milking cows were 19.5kg/hd/d. Spring(23.9kg) and fall(22.1kg) feed intake rates when higher than in the summer(17.0kg) and winter(15.3kg/hd/d). The average water consumption of milking cows were $77.2\ell/hd/d$. Summer showed the highest value$(85.5\ell/hd/d)$ and winter showed the lowest value$(62.2\ell/hd/d)$. The average milk yield during spring, summer, fall, and winter was 30.8, 24.0, 25.4, and 23.7kg/hd/d, respectively. Milk yield during spring was found to be statiscally greater than for the other seasons. Manure production of milking cows during spring, summer, fall, and winter was 64.4, 63.5, 60.4, and 51.0kg/hd/d, respectively. Consequently, a relatively high correlation between milk yield and water consumption$(R^2=0.7742)$, milk yield and feed intake$(R^2=0.7459)$, water, consumption and urine production$(R^2=0.7422)$, feed intake and feces production$(R^2=0.6044)$, and milk yield and feces production$(R^2=0.6920)$ were observed in milking cows. The other hand, correlation between water consumption and feces production$(R^2=0.2950)$, feed intake and urine production $(R^2=0.1985)$, and milk yield and urine production$(R^2=0.2335)$ were found to be relatively low. Therefore, correlation equation between milk yield and feed intake, milk yield and water consumption can be estimated from : $Y=0.1919X_1+11.181(R^2=0.7742),\;Y=0.8568X_2+9.3067(R^2=0.7459)$(Y=milk yield $X_1=water$ consumption, $X_2=feed$ intake).

        초등학생의 우유 및 유제품 섭취 양상과 소비 성향에 관한 조사

        We conducted a study to investigate milk and dairy products consumption among elementary school students to identify ways to increase their intake of milk and dairy products. We asked 518 students in Seoul to fill out questionnaire. Only 55.6% of the subjects consumed milk daily, and 43.7% of them drank at least 1 cup per day. The frequency and quantity of milk consumption were higher in male students than female students. The majority of the subjects consumed whole milk, and most students(59.5%) preferred flavored milk to whole milk. The reasons given for drinking milk included ``wanting to be tall``(66.1%) and ``for good health``(47.1%), while the main reason given for not drinking milk was stomach pain (68.4%). Records of having an unpleasant experience and upset stomach after drinking milk occurred at a higher rate for those who did not drink milk(84.2%) than for those who did(53.3%). Of the students questioned, 36.9% consumed dairy products other than milk 3~4 times per week, and liquid-type yoghurt(43.8%) and ice cream(35.4%) were the preferred dairy products among students. The reasons given for consuming dairy products were ``good taste``(56.6%), ``good to eat``(46.6%), ``wanting to be tall``(32.4%), and ``for good health``(25.4%). Among those students who did not consumed dairy products, the major reason given was ``family does not eat at home``(33.3%). After consuming the dairy products, 71.4% of participants thought that dairy products did not result in physical improvement. Most students had experienced purchasing milk and dairy products by themselves. When they bought milk or dairy products, the reasons given for making a particular selection were ``shelf-life``(61.6%) for milk, and ``favorite dairy products``(54.1%) for dairy products. According to participants, the factors that need to be considered for milk and dairy product consumption among elementary school students are ``higher nutrition`` (60.8%), ``better sanitation``(57.3%), and ``better quality and taste``(55.2%).

        우유생산비 조사 및 계산상의 문제점과 합리화방안 연구

        Accurate estimation of milk production cost is very important for dairy farmers in establishing strategies for business management (e.g. planning a program for milk production, deciding the size of business and investment, determining the milk price for sale). Since the estimated cost of milk production is used as an important index to determine the basal price of milk in Korea, there has been much interest and debate on the method used to estimate milk production cost among the stakeholder. This study was thus carried out to identify problems in the current methodology for estimating cost of milk production, and to find a better way to improve it. We propose several alternatives and better ways to improve the current method for estimating cost of milk production. Estimation of the income and cost per head should be based on the number of cattle converted to grown cows. Cost estimation per liter of milk should be made for both whole milk and 3.4% milk fat corrected milk. The value of purchased cows and raised replacement heifers should be the same as their market value. The productive life span of cows should be less 4 years, and the terminal or salvage value of cows needs to be 30 to 40% less than her initial value. When calculating depreciation of cows over the productive life span, however, the salvage value should be 0 or 1 Korean won. On calculating labor costs, the farm labor wage corresponding to the average wage of nonfarm industrial workers should be assumed. Beside of these, better estimation procedures for other items are also given. The proposed methods from this study should improve the accuracy of estimation of milk production cost and help to achieve consensus among the stakeholder.

        Production of Milk Clotting Protease by a Local Isolate of Mucor circinelloides under SSF using Agro-industrial Wastes

        Agro-industrial residues, a cheap source of energy have high potential in the area of fermentation for the production of enzymes. Twenty agro-industrial residues were evaluated to check the possibility of potential utilization of substrates in SSF for milk clotting enzyme protease production by Mucor circinelloides. In this study, dhal husk holds the greatest promise for cost effective production of the milk clotting enzyme. The dhal husk supported maximum milk clotting protease production, and yield was improved with the supplementation of sucrose and yeast extract as carbon and nitrogen source, respectively. Among all the physico-chemical parameters tested, the best results were obtained in a medium having moisture content of 20% at pH 7.0, when inoculated with 30% of spore suspension and incubated at 30oC for 5 days. The activity was increased further on addition of Ca²+, Cu²+, and Mg²+ ions. The purified milk-clotting protease obtained from M.circinelloides was successfully applied and compared with commercial rennet in the manufacture of a cheddar cheese.         A Review on Camel Milk Composition, Techno-Functional Properties and Processing Constraints

        Camel milk plays a critical role in the diet of peoples belongs to the semi-arid and arid regions. Since prehistoric times, camel milk marketing was limited due to lacking the processing facilities in the camel-rearing areas, nomads practiced the selfconsumption of raw and fermented camel milk. A better understanding of the technofunctional properties of camel milk is required for product improvement to address market and customer needs. Despite the superior nutraceutical and health promoting potential, limited camel dairy products are available compared to other bovines. It is a challenging impetus for the dairy industry to provide diversified camel dairy products to consumers with superior nutritional and functional qualities. The physicochemical behavior and characteristics of camel milk is different than the bovine milk, which poses processing and technological challenges. Traditionally camel milk is only processed into various fermented and non-fermented products; however, the production of commercially important dairy products (cheese, butter, yogurt, and milk powder) from camel milk still needs to be processed successfully. Therefore, the industrial processing and transformation of camel milk into various products, including fermented dairy products, pasteurized milk, milk powder, cheese, and other products, require the development of new technologies based on applied research. This review highlights camel milk’s processing constraints and techno-functional properties while presenting the challenges associated with processing the milk into various dairy products. Future research directions to improve product quality have also been discussed.

        Duplex PCR을 이용한 유제품 안에 있는 산양유와 우유의 신속한 동정에 대한 연구

        A duplex PCR technique was applied for specific identification of cow and goat milk in milk products by using primers targeting the mitochondrial 12S rRNA gene. Duplex PCR using primers specific for cow and goat generated specific fragments of 223bp and 326bp from cow and goat milk DNA, respectively. Duplex PCR was applied to 15 milk products purchased from the market to verify label statements. The labeling statements of four market milk products, three yoghurt products, and one whole milk powder product were confirmed in the duplex PCR. The labeling statements of five of seven infant milk powder products were also confirmed by duplex PCR but the other two products were shown to be contaminated with either cow or goat milk. The proposed duplex PCR provides a rapid and sensitive approach to detection of as little as 0.1% cow milk in goat milk and one-step detection of cow or goat milk in milk products.

        Effect of milk and milk products consumption on physical growth and bone mineral density in Korean adolescents

        This study was conducted to investigate the relationship among the current status of calcium intake from milk and milk products, physical growth and bone mineral density in 664 male and female middle school and high school students aged 15-17 years. In the study, the current status of calcium intake from milk and milk products was analyzed, and the height, body composition, and bone mineral density of the right heel bone (calcaneus) were measured. The daily calcium intake of milk and milk products was calculated as the ‘dairy equivalent of calcium’, which is the calcium content in 200 mL of white milk. The cutoffs of tertiles of the dairy equivalent of calcium were calculated and then the subjects were categorized into 3 groups according to the tertiles, Q1 group (lower intake group), Q2 group (middle intake group) and Q3 group (upper intake group). The daily calcium intake of milk and milk products in Q1, Q2 and Q3 groups was 16.2 mg, 99.7 mg, and 284.0 mg, respectively, and the ratio of milk and milk product consumption to the daily total calcium intake was 5.4%, 27.4%, and 49.7%, respectively. The ratio of total calcium intake to the daily recommended intake in study subjects was 30.5% in Q1, 42.3% in Q2, and 60.7% in Q3, with significant differences (P < 0.05). Height, body weight, BMI, and % of body fat in three tertile groups (Q1, Q2 and Q3) were not significantly different. However, the T scores for bone mineral density in female students in three tertile groups (Q1, Q2 and Q3) was significantly different (P < 0.05). The study showed that the intake of milk and milk products in adolescents, particularly in girls, can improve the bone mineral density without increasing body weight, and thus confirmed that milk intake is important in adolescence.

        산양유 및 산양유 발효유가 웅성 설치류의 생식기능과 지구력에 미치는 영향에 관한 연구

        Objective: The purpose of this study was to evaluate effects of goat milk and fermented goat milk on reproductive function and stamina of male rodent. Methods: Experiment I: Male ICR mouse was divided into four groups. Group 1 none-treated control; Group 2 received saline; Group 3 received cow milk 10 ml/kg per day for 15 days; Group 4 received goat milk 10 ml/kg per day for 15 days. The cauda epididymal sperm motility and testicular sperm production were investigated. Experiment II: Male SD rat was divided into three groups. Group 1 received saline; Group 2 received goat milk 10 ml/kg per day for 28 days; Group 3 received fermented goat milk 10 ml/kg per day for 28 days. The cauda epididymal sperm motility and testicular sperm production were also investigated. The concentration of testosterone in serum at 1 and 3 weeks after treatment was determined using Immulite 2000 kit. Testes, epididymis, prostate, and seminal vesicle were weighed. Experiment III: Male ICR mouse was divided into four groups. Group 1 none-treated control; Group 2 received saline; Group 3 received goat milk 10 ml/kg per day for 4 weeks; Group 4 received fermented goat milk 10 ml/kg per day for 4 weeks. After treatment, the mouse was forced to swim to test for stamina. Results: In Experiment I, the cauda epididymal sperm motility after in vitro culture for 1 or 3 h was significantly (p<0.05) higher in cow milk and goat milk than in the control and saline. There was no significant difference in the cauda epidymal sperm motility between cow and goat milk. The testicular spermatid number was significantly (p<0.01) higher in goat milk (222.8${\times}10^6$) than in the control (108.6), saline (98.2), and cow milk (118.2). In Experiment II, the cauda epididymal sperm motility after in vitro culture for 1 h was significantly (p<0.05) higher in fermented goat milk than in saline and goat milk. There was no significant difference in the cauda epidymal sperm motility between saline and goat milk but goat milk showed slightly higher sperm motility than saline. After in vitro culture for 3 h, the cauda epididymal sperm motility was significantly (p<0.01) higher in fermented goat milk and goat milk than in saline. The testicular spermatid number was significantly (p<0.05) higher in goat milk than in saline, and significantly (p<0.01) higher in fermented goat milk than in saline. And the serum testosterone levels of rats administered with goat milk or fermented goat milk were increased but were no significant difference among three groups. Also the prostate weight was significantly (p<0.05) increased in the goat and fermented goat milk. In Experiment III, the swimming time in the goat milk and fermented goat milk groups was significantly (p<0.01) longer than in the control and saline. There was no significant difference in the swimming time between goat and fermented goat milk but the fermented goat milk showed slightly longer swimming time than the goat milk. Conclusion: The cauda epididymal sperm motility, the testicular spermatid number and stamina were improved when the mice and rats were drunk with goat milk or fermented goat milk.
