Objectives : This study evaluated the effect of Dioscoreae Rhizoma on antioxidant and antimicrobial activity in vitro. Methods : In vitro experiments were made on antioxidant by DPPH radical scavenging activity, enzyme activity of SOD, protective effect about $H_2O_2$ and DNA fragmentation in RAW264.7 cells, as well as antimicrobial by Cox-2 expression in LPS-induced RAW264.7 cells. Results : 1. Dioscoreae Rhizoma's antioxidant ability was improved by DPPH radical scavenging activity. 2. Antioxidant ability of Dioscoreae Rhizoma produced good results at more than 0.025 mg/$m{\ell}$. 3. On protective effect of Dioscoreae Rhizoma about $H_2O_2$ on RAW264.7 cells, with addition of Dioscoreae Rhizoma extract the cells' survival increased compared to adding $H_2O_2$ only. 4. About revelation-control of Cox-2 at RAW264.7 cells which induce by LPS, when Dioscoreae Rhizoma's concentration was higher from 0.0125 mg/$m{\ell}$ to 1 mg/$m{\ell}$, its control activity was stronger. Thus we found Dioscoreae Rhizoma has antimicrobial activity. 5. The power of DNA fragmentation control of Dioscoreae Rhizoma was time-independent. Conclusions : From this study we conclude that Dioscoreae Rhizoma has effects on antioxidant and antimicrobial activity. However, considerable work still needs to be done studying antioxidants by concentration, as well as a variety of experiments about antimicrobial activity.
산약의 멜라노마 세포(B16F10)에서 MITF, TRP-1, TRP-2, Tyrosinase, PKA, ERK 발현 억제 효과

Objectives: The purpose of this study was to research the whitening effects and developing by cosmetics of the extract from Dioscoreae Rhizoma, which is one of the most popular health-promoting herb in herbal medications. Methods: We performed tyrosinase inhibition assay, reverse transcription-polymerase chain reaction (RT-PCR) and western blot for whitening effects. Also we measured MTT assay for cell viability. Results: The results were obtained as follows : For whitening effect, tyrosinase inhibition rate of extract from Dioscoreae Rhizoma showed more than 42.28% at 1,000 ㎍/㎖concentration. Cell toxicity effect on melanoma cells (B16F10) of extract from Dioscoreae Rhizoma showed 81.97% with toxicity at 50 ㎍/㎖concentration. So we were measured at a concentrations of 5, 10 and 50 ㎍/㎖in all experiments involving cell. In addition, whitening related mRNAs including microphthalmia associated transcription factor (MITF), tyrosinase related protein-1 (TRP-1), tyrosinase related protein-2 (TRP-2), tyrosinase were reduced by Dioscoreae Rhizoma. We also found Dioscoreae Rhizoma transiently decreased protein kinase A (PKA) which is known to be upstream to the down regulation of MITF and tyrosinase. But phosphorylation of extracellular signal related kinase (pERK) were increased by Dioscoreae Rhizoma. These results imply that Dioscoreae Rhizoma decrease melanogenesis via ERK activation and subsequent down regulation of MITF and tyrosinase. Conclusions: Therefore, all these findings suggested the potent usage of Dioscoreae Rhizoma as materials of functional cosmetics by confirming whitening activity related with melanin content.

This study was designed to investigate the optimum recommendation level on Gastrodia rhizoma and the effects on the improvement of the lipids in the dietary hypercholesterolemic rats. Experimental diets mixed with 5% Dioscorea batatas(Group 2), 10% Dioscorea batatas(Group 3), 15% Dioscorea batatas(Group 4), 5% Gastrodia rhizoma(Group 5), 10% Gastrodia rhizoma (Group 6) and 15% Gastrodia rhizoma(Group 7) were administered to the male rats of the Sprague Dawley for 3 weeks. Concentration of total cholesterol in serum was lower in the Gastrodia rhizoma groups than in the other groups, especially total cholesterol concentration of 10% Gastrodia rhizoma(Group 6) was the lowest in the Gastrodia rhizoma groups. Concentration of HDL-cholesterol in serum was higher in the 10% Gastrodia rhizoma and 15% Gastrodia rhizoma than in the other groups. Concentrations of cholesterylester, LDL, LDL-cholesterol in serum were the lowest in the 10% Gastrodia rhizoma. Concentration of glucose and activity of GPT in serum were the lowest in the 10% Gastrodia rhizoma group. The activity of GOT in serum was lower in the 10% Gastrodia rhizoma group and 15% Gastrodia rhizoma group than in the other groups. Therefore, we consider that there are effects on the improvement of the lipids in the Gastrodia rhizoma and the optimum recommendation quantity of Gastrodia rhizoma is 10% to quantity of food composition.

It is well known that Dioscoreae Rhizoma tonifies and benefits the Spleen and Stomach, benefits the Lung and nourishes the Kidney. A major component isolated from this herb consist of dioscin, diosgenin, allantoin, starch, alanin, mucilage, choline, amylase, and glycoprotein. In this study, we aimed to measure the content of allantoin, one of major component, to determine the quality of Dioscoreae Rhizoma.

An easy and reliable HPLC method was developed to determine allantoin in Dioscorea Rhizoma using cyano columns. Qualitative and quantitative analyses of allantoin were performed successfully by cyano columns (YMC-Pack CN column, Zorbax SB-CN column and Discovery® Cyano column). The intraday precision were 0.58 - 3.33% for YMC-Pack CN, 0.41 - 2.20 for Zorbax SB-CN and 0.45 - 1.93% Discovery® Cyano columns, while interday variations were 0.09 - 1.84%, 0.04 - 2.59% and 0.87 - 5.18% for YMC-Pack CN, Zorbax SB-CN and Discovery® Cyano columns. The recoveries of allantoin were in the range at 98.8 - 102.6% (RSD 1.1 - 1.6%) for YMC-Pack CN column, 99.7 - 110.5% (RSD 1.3 - 4.9%) for Zorbax SB-CN column, and 97.2 - 110.1% (RSD 1.8 - 5.7%) for Discovery® Cyano column. The contents of allantoin in four Dioscorea Rhizoma samples were determined by cyano columms and ranged at 4.1-7.1 mg/g dry weight. The present study indicated that HPLC method using cyano column for determining allantoin is a reliable method and this method can be applied to verify allantoin in Dioscorea Rhizoma.

Objectives: The aim of this study is to evaluate the efficacy and the safety of Sanyak pharmacopuncture therapy for the treatment of peripheral facial paralysis patients. Methods: This study was a retrospective investigation of a total of 70 patients who were inpatients of the Oriental Hospital of Daejeon University between January 1,2011, and May 31, 2012, and who were diagnosed as having peripheral facial paralysis by physical examination,the patients received three different interventions. Eleven (11) patients were treated with acupuncture and alcohol Dioscorea rhizoma pharmacopuncture (ADG), 25 patients with acupuncture and distillation Dioscorea rhizoma pharmacopuncture (DDG), and 34patients with acupuncture and non-Dioscorea rhizoma pharmacopuncture (NDG). The resulting data were analyzed. Results: The changed H-B grades indicated significant improvements in all three groups, and the ADG and the DDG groups showed significant results after two weeks of treatment when compared to the NDG group. The changed y-Scores indicated significant improvements in all three groups, and the ADG group showed significant results after 10 and 15 days of treatment when compared to the NDG group. Dioscorea rhizoma pharmacopuncture may be safe for the human body because in most cases, the only abnormal finding was the pain could by the application of pharmacopuncture. Objectives : This report describes 45 studies related to the use of Rhizoma dioscoreae main blended prescription from Donguibogam. Methods : The following conclusion were reached through investigations on the prescriptions that use Rhizoma dioscoreae as a key ingredient. Prescriptions that Rhizoma dioscoreae was taken as a monarch drug are utilized for 18 therapeutic purposes, for example, internal injury, stool disease, pediatric disease. In paticular, 24% of prescriptions appear in the chapter of internal injury, and 18% of those appear in the chapter of stool. Results : Prescriptions that utilize Rhizoma dioscoreae as the main ingredient are used in the treatment of indigestion, diarrhea and paralysis. Rhizoma dioscoreae is used in pathogenic factors such as water, water with damp, and used in pathology related to stomach system. Conclusions : The dosage of Rhizoma dioscoreae is 8li(about 0.29 gram) to 3jeon(about 11.25 gram), however 1jeon(about 3.75 gram) has been taken the most for clinical application. Sagunjatang and Siljangsan is the most useful base prescription which use the Rhizoma dioscoreae as the main ingredient.

The purpose of this study was to examine the antiobesity effects of lactic acid bacteria-fermented Dioscoreae Rhizoma powder (LDR) in Sprague-Dawley rats with high-fat diet (HFD)-induced obesity. Rats were divided into 5 groups: normal diet group, control (high fat diet-vehicle) group, 0.4 g/kg LDR (high fat diet + 0.4 g/kg lactic acid bacteria-fermented Dioscoreae Rhizoma powder) group, 1 g/kg LDR (high fat diet + lactic acid bacteria-fermented Dioscoreae Rhizoma powder) group, 2.5g/kg LDR (high fat diet + lactic acid bacteria-fermented Dioscoreae Rhizoma powder) group. Our results indicate that LDR administration has effects on decreasing of body weight, fat weight, blood parameters and adipocyte size in the obesity animals. In addition, verified that fat degeneration and ballooning degeneration were alleviated in the experimental group fed on a high fat diet combined with LDR groups. These results indicate that lactic acid bacteria-fermented Dioscoreae Rhizoma powder may reduce elevating body weight and lipid accumulation in rat fed a high fat diet, suggesting its usefulness as a functional food for reducing body fat and obesity.

Objectives: To investigate the quantitative determination of marker constituents of Dioscoreae Rhizoma by the packaging methods. Methods: HPLC for the determinations of allatoin in the Dioscoreae Rhizoma, the separation method was performed on an Luna NH2 column (250 mm×4.6 mm, 5 μm, Phenomenes) using solvent water-acetonitrile (2:8, v/v%) with photo diode array detector (210nm). The flow rate was 2.0 mL/min. Results: Retention time of allatoin in HPLC chromatogram was about 4.8 min and calibration curve showed good linearity (R2=0.9994) at concentrations from 50.0 to 1000.0 μg/mL of allantoin. Average content of allantoin by packaging methods was 0.16~0.40%. In addition, weight loss rate (%) of Dioscoreae Rhizoma according to the packaging methods was -0.60~1.80%. Conclusions: The variation on content of the marker constituent of Dioscoreae Rhizoma by the packaging methods didn`t show difference.
