      • Protective effects of <i>Centella asiatica</i> leaf extract on dimethylnitrosamine-induced liver injury in rats

        <P>Oxidative stress in liver injury is a major pathogenetic factor in the progression of liver damage. <I>Centella asiatica</I> (L.) Urban, known in the United States as Gotu kola, is widely used as a traditional herbal medicine in Chinese or Indian Pennywort. The efficacy of <I>Centella asiatica</I> is comprehensive and is used as an anti-inflammatory agent, for memory improvement, for its antitumor activity and for treatment of gastric ulcers. The present study investigated the protective effects of <I>Centella asiatica</I> on dimethylnitrosamine (DMN)-induced liver injury in rats. The rats in the treatment groups were treated with <I>Centella asiatica</I> at either 100 or 200 mg/kg in distilled water (D.W) or with silymarin (200 mg/kg in D.W) by oral administration for 5 days daily following intraperitoneal injections of 30 mg/kg DMN. <I>Centella asiatica</I> significantly decreased the relative liver weights in the DMN-induced liver injury group, compared with the control. The assessment of liver histology showed that <I>Centella asiatica</I> significantly alleviated mass periportal ± bridging necrosis, intralobular degeneration and focal necrosis, with fibrosis of liver tissues. Additionally, <I>Centella asiatica</I> significantly decreased the level of malondialdehyde, significantly increased the levels of antioxidant enzymes, including superoxide dismutase, glutathione peroxidase and catalase, and may have provided protection against the deleterious effects of reactive oxygen species. In addition, <I>Centella asiatica</I> significantly decreased inflammatory mediators, including interleukin (IL)-1β, IL-2, IL-6, IL-10, IL-12, tumor necrosis factor-α, interferon-γ and granulocyte/macrophage colony-stimulating factor. These results suggested that <I>Centella asiatica</I> had hepatoprotective effects through increasing the levels of antioxidant enzymes and reducing the levels of inflammatory mediators in rats with DMN-induced liver injury. Therefore, <I>Centella asiatica</I> may be useful in preventing liver damage.</P>

        Improving effect on Oral Conditions with Dentifrice Combined by Extracts of Dipotassium Glycyrrhizinate, Bamboo Salt and Quantitative Extracts of Centella Asiatica

        Objective: This experiment was contrived to find out the changed in oral condition index when standardized and regular use of the dentifrice is done according to the usage of dosage of the dentifrice. Methods: After inspecting the result of acceleration of collagen forming and suppressing effect of inflammatory reaction by using the titrated extracts of Centella asiatica and Dipotassium glycyrrhizinate with In-vitro experiment (done inside the lab), the remedying effect on oral cavity condition was to be obtained with dentifrice made with combined bamboo salt and those above through in-vivo experiment (clinical) worked with 90 healthy bodied examinees for over 6 months. Results: With the result of the experiment done at the lab, when the changes of improved index of oral cavity condition, MMP-1 formative inhibition analysis, and collagen analysis were obtained during the experiment periods, the titrated extract of Centella asiatica suppresses the damage of the tissue due to inflammatory response at the gingiva cell and after the inflammatory response, accelerated regeneration of the gingiva were clearly shown. Also, as with the result in clinical experiment, the effect was acceptable in reducing effect of dental plaque index and gingivitis index with dentifrice that only contains the titrated extract of Centella asiatica and also showed outstanding result in antibacterial activity. Conclusion: The most effective improvement especially shown on the oral conditions were dentifrice that contains Dipotassium glycyrrhizinate, bamboo salt and titrated extracts of Centella asiatica.

        제주용암해수로 숙성된 병풀 추출물의 항염 및 보습 효과

        병풀의 잎과 줄기는 오랜 기간 궤양, 건선과 같은 피부질환 치료를 위한 약재로 사용되어왔으며, 병풀의 유효성분인 asiaticoside, madecasosside, asiatic acid, madecassic acid와 같은 triterpenoids는 피부 항염 및 피부 광 노화 개선 등의 효능이 밝혀져 있다. 본 연구의 목적은 인체에 유익한 미네랄을 많이 함유하고 있는 제주용암해수로 병풀을 숙성한 뒤 추출하여 피부 항염 및 보습 효능을 확인하는 것이다. 제주용암해수로 병풀을 숙성한 뒤 추출물을 HPLC에 분석하였을 때 triterpenoids의 농도가 더 증가하는 것을 관찰하였다. 숙성한 병풀 추출물의 항염증 효능을 확인하기 위해 LPS로 활성화된 RAW 264.7 세포에서 NO의 생성과 PM10 혹은 UV에 의해 유도된 HaCaT 세포에서 염증성 사이토카인들의 발현을 확인하였다. 실험 결과, 숙성한 병풀 추출물이 NO와 IL-6, IL-8, TNF-a의 발현을 저해하였고 숙성하지 않은 군에 비해 숙성한 군에서 억제 효능이 더 높은 것을 확인하였다. 또한, 피부 보습 효능을 확인하기 위해 HaCaT 세포에서 HA의 합성을 확인한 결과 숙성하지 않은 군에서는 효능이 없는 것에 반해 숙성한 군에서 농도 의존적으로 HA 생성이 증가하는 것을 확인하였다. 따라서 본 연구 결과를 바탕으로 제주용암해수로 숙성한 병풀 추출물은 항염과 보습에 유효한 소재임을 확인하였고 숙성하지 않은 병풀 추출물보다 효능이 우수할 것으로 판단된다. The leaves and stems of Centella asiatica have a long history of their usage as a medicine for the treatment of skin diseases such as ulcers and psoriasis, especially in Asia. Triterpenoids, the active components of Centella asiatica including asiaticoside, madecasosside, asiatic acid and madecassic acid, have shown to inhibit skin inflammation as well as improve skin photoaging. The main objective of this study is to investigate whether the Centella asiatica ripened with lava seawater which is rich in minerals known to be beneficial to human body can provide anti-inflammatory and moisturizing effects to skin. HPLC analysis showed that the concentration of triterpenoids increased further after ripening Centella asiatica with lava seawater. In order to confirm the inflammatory efficacy of the extract of the extract of the ripened Centella asiatica, the production of NO in LPS-activated RAW 264.7 cells and the expression of inflammatory cytokines in PM10 or UVB-induced HaCaT cells were observed. We found that the extract of the ripened Centella asiatica inhibited the expression of NO, IL-6, IL-8, and TNF-a and had higher inhibitory effect compared to the extract of the non-ripened Centella asiatica. In order to confirm the skin moisturizing effect, we investigated the synthesis of HA in HaCaT cells. The result showed HA production was enhanced in a concentration-dependent manner from the ripened group, while there was no efficacy from the non-ripened group. Taken together, it is concluded that the extract of the Centella asiatica ripened with lava seawater was effective in anti-inflammation and moisturization.

        This study examined the effects of Centella asiatica (C. asiatica) extracts on anti-oxidant and collagen synthesis in both cell-free and normal human dermal fibroblast (NHDF) cell culture systems and compared differences in the effects of extracts obtained under varying extraction conditions. Extracts were prepared from either fresh or dried C. asiatica and using either H2O, absolute ethanol, or 60% ethanol as a solvent. The contents of C. asiatica extracts were analyzed by high-performance liquid chromatography, and extracts prepared from dried C. asiatica in 60% ethanol showed the highest content levels of madecassoside, asiaticoside, madecassic acid and asiatic acid. All extracts except those prepared in H2O demonstrated effects on increasing NHDF cell viability. Extracts prepared from dried C. asiatica in 60% ethanol showed the most prominent effects on NHDF cell viability and radical scavenging activity. Among the extraction conditions tested, extracts prepared fresh C. asiatica in H2O had the greatest effect on type I collagen synthesis. These results suggest that the efficacy of C. asiatica extracts can be improved by altering extraction conditions.

        Centella asiatica (C. asiatica) plays a key role in traditional Ayurvedic medicine, treating a myriad of conditions. C. asiatica`s ancient reputation earned its place in modern alternative medicine and it has even been examined in a number of clinical studies. C. asiatica has many other practical uses because of its ability to improve circulation and strengthens vessel walls, varicose and spider veins. Also it is often used as an active ingredient in the cosmetic industry. The purpose of this study is to analyse the effect of obesity treatment on abdominal obese with C. asiatica extracts. This research divided 20-30`s women who have more than 30% higher percent body fat. Here we present that the group treated with C. asiatica extracts showed significant differences in changes of body compositions and concentrations of triglyceride in blood in comparison with the control group. The C. asiatica treatment has positive influence upon physical slimming, body circumference reduction and so on. These imply that if this ingredient is largely used for abdominal obesity management, it could be more helpful to preventing obesity related disease.

        Enhanced biosynthesis of triterpenoids in Centella asiatica hairy root culture by precursor feeding and elicitation

        Centella asiatica contains various triterpenoids, including madecassoside, asiaticoside, madecassic acid, and asiatic acid. Among them, they are mainly contained in leaf, and among them asiaticoside is known as tissue-specific compound that are mainly accumulated in the leaves. Therefore, in the present study, C. asiatica hairy root cultures were established from petiole and leaf explants using Agrobacterium rhizogenes-mediated transformation. To promote the biosynthesis of these triterpenoids, hairy root cultures were treated with precursors (squalene and pyruvic acid) and elicitors [methyl jasmonate (MeJA) and salicylic acid (SA)] at various concentrations. No significant difference was detected in the growth index of hairy root cultures between precursor feeding and elicitation, except pyruvic acid feeding, suggesting that elicitors do not affect the growth of hairy roots. Treatment with 2.5 mM squalene and 5 mM pyruvic acid enhanced the biosynthesis of target triterpenoids (57.53 and 29.13 mg g−1 DW, respectively) compared with other treatments. However, compared to squalene, MeJA showed a greater effect on the triterpene saponin content in C. asiatica. The highest quantity of triterpenoids (60.25 mg g−1 DW) was produced in hairy root cultures treated with 400 μM MeJA. These results indicate that precursorand elicitor-treated C. asiatica hairy root cultures rapidly accumulate biomass and could be used to enhance the production of triterpenoids for industrial purposes.

        재배조건 및 관수방법에 따른 병풀 (Centella asiatica) 계통의 생육, Asiaticoside 함량과 항산화활성

        Background: Centella asiatica is a well-known medicinal plant having a wound healing effect. In this study, the growth, asiaticoside content, and antioxidant components and activity were investigated in C. asiatica resources under different cultivation and irrigation conditions. Methods and Results: The cultivation and irrigation methods were divided as greenhouse and open field, and sprinkler and sub irrigation. respectively. Growth characteristics were measured in aerial parts of C. asiatica. Asiaticoside content was analyzed using high performance liquid chromatography. Total polyphenol, total flavonoid and 2,2-diphenyl-1picryl hydrazyl (DPPH) radical scavenging activity were analyzed for antioxidant activities. Growth was higher under greenhouse and sub irrigation than open field and sprinkler conditions. In the leaves, asiaticoside content was 53.45 ㎎/g (open field) and 34.38 ㎎/g (sub irrigation), total polyphenol was 41.14 ㎎/g (open field) and 25.73 ㎎/g (sub-irrigation), and total flavonoid was 27.26 ㎎/g (open field) and 23.72 ㎎/g (sub-irrigation). DPPH radical scavenging activity in the leaves was 85.97 ㎎·ascorbic acid equivalent (AAE)/g (open field) and 54.83 ㎎·AAE/g (sub irrigation). Conclusions: Asiaticoside and antioxidant components and activity were not accompanied with high yield, although high growth was observed under greenhouse conditions. Therefore, cultural requirements of C. asiatica should be sufficiently considered to suit each purpose.

        Cloning and Molecular Analysis of cDNA Encoding Cycloartenol Synthase from Centella asiatica (L.) Urban

        cDNA for oxidosqualene cyclase was cloned by a homology-based PCR method and sequenced from Centella asiatica. In a sequences analysis, the putative polypeptide of C. asiatica cycloartenol synthase (CaCYS) deduced from the 2,274 bp nucleotide sequence, consisted of 758 amino acids and had a molecular mass of 86.3 kD. The predicted amino acid sequence exhibited high homology to that of PNX (cycloartenol synthase) from Panax ginseng ($89\%$). Southern blot analysis suggests that CaCYS may be present in one copy of the C. asiatica genome. If methyl jasmonate (MJ) is applied exogenously to plants, not only triterpene saponins are accumulated in tissues, but also it produces effects such as growth inhibition and the promotion of ethylene production. In order to investigate the effect of MJ and thidiazuron (TDZ), a cytokinin that plays a role as an antisenescence agent in several plants, on the level of CaCYS mRNA, we performed northern blot analysis. When MJ is alone treated by adding to culture medium, CaCYS transcripts were inhibited. However, sustained levels of the expression of CaCYS, by adding TDZ to the medium despite MJ treatments, were demonstrated in C. asiatica leaves.
