        감마선 조사에 의한 총목피(Aralia elata Cortex)의 추출수율 증대 및 항산화 효과

        This study was designed to investigate the effects on anti-oxidative activities and increasing extraction yield of Aralia elata Cortex by gamma irradiation. Electron spin resonance (ESR) analysis as physical techniques for irradiation identification of Aralia elata showed that a pair of peak appeared on a space of 6.0 mT at the left and right of symmetric unspecific central ESR spectrums, confirming that the plant was gamma-irradiated. The optimum extracting conditions for preparing gamma irradiated samples from Aralia elata Cortex were to extract with 50% ethanol for 15 hrs after 10 kGy irradiation. DPPH scavenging activity and ABTS radical cation inhibitory activity of the water and 50% ethanol extracts from non irradiated and irradiated Aralia elata Cortex was very high as over 80% and 98%, respectively, at tested low concentration of 50 μg/mL. Antioxidant protection factor (PF) as anti-oxidation indicator of lipophilic compounds showed a very high level of activity as 2.18~2.78 PF. As for TBARs, water and ethanol extracts showed high level. Increase of TBARs inhibitory activity of water extracts was not shown by gamma-ray irradiation but ethanol extracts showed slight increasement of TBARs inhibitory activity with 10 kGy gamma-ray irradiation. These results shown confirmed increasement of extraction yield for phenolic compounds and anti-oxidative activity from Aralia elata. Thus, the treatment of gamma- irradiation can be used a way to amplify a solubility for biological active compounds and anti-oxidative activity in plants. 본 연구에서는 감마선 조사가 총목피 추출물의 생리활성에미치는 영향을 검토하였다. 감마선 조사한 총목피의 감마선 조사여부를 확인하기 위하여 전자스핀공명(ESR) 분석을 시행한결과 대칭적인 unspecific central ESR 신호 좌우로 cellulose를 함유한 조사 식품에서 나타나는 한 쌍의 peak가 6.0 mT의공간을 두고 나타나 감마선을 조사한 시료로 판별되었다. 감마선 조사된 총목피의 최적 추출조건은 10 kGy를 조사한 시료를50% ethanol을 사용하여 15시간 추출이었다. 감마선을 조사한총목피의 항산화 효과를 확인한 결과 DPPH radical 소거효과와 ABTS radical 저해는 총목피의 물 추출물과 50% ethanol 추출물의 경우, 비조사구와 조사구를 저 농도인 50 ㎍/㎖ 처리하였을 때 각각 80%와 98% 이상의 매우 높은 저해 효과를 나타내었다. 지용성 물질의 항산화 활성의 지표인 antioxidant ProtentionFactor (PF)는 2.18~2.78 PF로 매우 높은 PF값을 나타내었다. TBARs의 경우 물과 ethanol 추출물 모두 높은 TBARs값을 나타내었다. 물 추출물은 감마선의 조사에 의한 TBARs 저해능의 상승은 관찰되지 않았으나, ethanol 추출물은 10 kGy의 감마선조사에 의해 TBARs 저해능의 상승이 약하게 관찰되었다. 이상의 결과 총목피는 감마선의 조사에 의해서 총목피의 phenolic성분들의 용출율과 항산화 효과가 증대되는 현상을 확인하였다. 또한 감마선 조사가 식물체로부터 생리활성물질의 용출 증대와 항산화 활성을 증폭시킬 수 있는 수단으로 활용이 가능할것이라 판단되었다.

        UPLC-DAD-ESI(+)-QToF/MS를 이용한국내산 두릅나무 및 음나무 순 내 페놀산 특성 비교

        BACKGROUND: In this study, shoots of Aralia elata and Kalopanax pictus which belong to the Araliaceae family were analyzed using UPLC-DAD-ESI(+)-QToF/MS to characterize of individual phenolic acids. METHODS AND RESULTS: Total thirteen phenolic acids were identified, and nine hydroxycinnamic acid derivatives have been identified for the first time in shoots of Aralia elata and Kalopanax pictus. For total phenolic acid content (mg/100g dry weight), shoots of Aralia elata and Kalopanax pictus showed 754.8 and 845.3 mg/100g, respectively. 5-O-Caffeoylquinic acid (49%) and 3,5-di- O-caffeoylquinic acid (44%) were found as major phenolic acids in Aralia elata, while 5-O-caffeoylquinic acid (91%) was a major component in Kalopanax pictus. CONCLUSION: On comparing the two plants, it was considered that the biosynthesis of 3,5-di-O-caffeoylquinic acid can be affected by 5-O-caffeoylquinic acid in Aralia elata.

        랫드에서 두릅 추출물의 4주 반복투여 독성시험

        Aralia elata, a common medicinal and culinary herb, has been consumed for centuries without any reported adverse effects. However, due to its limited safety information, we decided to investigate the repeated-dose toxicity of ethanolic extract of Aralia elata. The test was administered once daily by the gavage to male and female rats at doses of 0, 250, 500 and 1,000 ㎎/㎏/day for four weeks. Throughout the study, no treatment-related deaths or clinical signs were observed. Also, no apparent changes were detected in ophthalmoscopy, urinalysis, serum biochemistry, hematology and gross necropsy. The test result showed a significant decrease in body and heart weight of males treated with 250 ㎎/㎏ of extract of Aralia elata compared to normal control, a significant increase in relative brain weight and adrenal weight in females treated with 250 ㎎/㎏ of extract compared to normal control. However, all these changes were not considered toxicologically important due to irrelevant dose-response relationship to gross and microscopic findings. Histopathologically, abnormal changes were not observed in any target organs. On the basis of these results, the NOAEL of extract of Aralia elata was estimated to be more than 1,000 ㎎/㎏/day under the tested conditions.

        두릅 순에서 분리된 화합물의 항산화 활성

        Objectives: This study was performed to investigate the antioxidant activities of isolated compounds from the shoot of Aralia elata. Methods: The methanol extract from the shoot of Aralia elata was fractionated into ethyl acetate, n-BuOH and H2O layers through solvent fractionation. Repeated silica gel, ODS column chromatography of n-BuOH layer afforded four flavonol glycosides. Their antioxidant activity was determined by measuring free radical scavenging activity by DPPH, ABTS and superoxide dismutase (SOD) like activity assay. Results: They were identified as quercetin 3,7-di-O-α-rhamnopyranoside (1), quercetin 3-O-β-D-galactoside-7-O-α-L-rhamnoside (2), kaempferol 3-O-β-glucosyl(1→2)-α-rhamnoside-7-O-α-rhamnoside (3) and quercetin 3-O-β-glucosyl(1→2)-α-rhamnoside-7-O-α-rhamnoside (4) on the basis of spectroscopic data. The result showed that 1 is the most active compound in the DPPH and ABTS radical scavenging test. Conclusions: Isolated Compounds from the shoot of Aralia elata showed anti-oxidative effect.

        참두릅과 개두릅을 첨가한 국수의 제조특성

        참두릅과 개두릅가루의 단백질과 지방함량은 개두릅가루가 참두릅가루보다 많았으며, 회분과 식이섬유 함량은 참두릅가루보다 적었다. 참두릅과 개두릅가루는 높은 식이섬유 함량을 나타냈다. 두릅가루를 혼합하여 제조한 색도는 L값은 낮았으며, a값은 증가하였고 b값은 감소하였다. 조리면의 cutting force는 두릅가루를 첨가한 조리면이 대조군보다 낮게 나타났고 두릅가루의 첨가량이 많을수록 감소하는 것으로 나타났으며, 참두릅가루를 첨가한 조리면이 개두릅가루를 첨가한 조리면보다는 높은 것으로 나타났다. 국수의 기능성을 높이고 기호도를 향상시키기 위하여 참두릅가루와 개두릅가루를 3, 5, 7% 첨가한 조리면의 관능검사를 실험한 결과 5% 첨가한 국수가 가장 높은 관능적 평가를 받았다. The objective of this study was to evaluate the quality characteristics of noodles made with a dried powder of the shoots and leaves from Aralia elata and Kalopanax pictus. Ash, protein, and dietary fiber contents of A. elata and K. pictus were higher than those of wheat flour. The protein and fat contents of K. pictus powder were higher than those of the A. elata powder, but ash and dietary fiber contents were less than those of A. elata. When the amount of A. elata powder increased, brightness (L) values of dough decreased, and redness (a) and yellowness (b) values of dough increased. The weight and volume of the cooked noodles also increased, as did the turbidity of the soup. Addition of A. elata and K. pictus powders in noodles reduced the cutting forces needed to cut the cooked noodles. The sensory evaluation indicates that noodles with 5% A. elata and K. pictus powder added were the most preferable.

        두릅 에탄올 추출물의 Chlorogenic acid 함량 분석 및 생리활성

        This study was conducted to quantify chlorogenic acid content and evaluate biological activity, such as antioxidant, antibacterial, anti-inflammatory, and digestive enzyme activity of Aralia elata ethanol extract (AEE). The SC<sub>50</sub> of DPPH and ABTS radical scavenging activities of AEE were 4.79±0.05 mg/mL, 5.79±0.05 mg/mL; total polyphenol and total flavonoid contents were 170.0±1.8 mgGAE/g, 105.5±4.1 mgQE/g, respectively. Nitric oxide (NO) was increased in RAW 264.7 cells and Caco-2 cells with treatment of LPS, and production of NO was inhibited by AEE in a concentration-dependent manner. Production of NO was reduced by 60.0±1.1% in RAW 264.7 cells and 50.7±2.8% in Caco-2 cells at of AEE. Similarly, the production of inflammatory cytokines (TNF-α, IL-1β and IL-6) was inhibited in a concentration dependent manner. Antibacterial activity increased as the dose concentration of AEE increased, and the MIC was 75 mg/mL for L. monocytogenes, and 100 mg/mL for S. typhimurium and H. pylori. In addition, amylase and protease enzyme activity was observed in AEE and increased enzyme activity was observed according to the concentration of the extract. AEE contained 7.06±0.01 mg/g of chlorogenic acid. As a result of the experiment, it is judged that it can be used as basic data for the development of health food using Aralia elata.

        Saponins and Other Constituents from the Leaves of <i>Aralia elata</i>

        <P>A new triterpenoid saponin, together with five known saponins, were isolated from the nonpolar <I>n</I>-hexane fraction of the leaves of <I>Aralia elata</I>. The structure of the new saponin, durupcoside C, was elucidated as hederagenin 3-<I>O</I>-β-<SMALL>D</SMALL>-glucopyranosyl(1→3)-β-<SMALL>D</SMALL>-glucopyranosyl(1→3)-α-<SMALL>L</SMALL>-arabinopyranoside on the basis of spectroscopic analysis. The known saponins were characterized as 3-<I>O</I>-α-<SMALL>L</SMALL>-rhamnopyranosyl(1→2)-α-<SMALL>L</SMALL>-arabinopyranosyl hederagenin 28-<I>O</I>-β-<SMALL>D</SMALL>-xylopyranosyl(1→6)-β-<SMALL>D</SMALL>-glucopyranosyl ester, hederagenin 3-<I>O</I>-β-<SMALL>D</SMALL>-glucopyranosyl(1→3)-α-<SMALL>L</SMALL>-rhamnopyranosyl(1→2)-α-<SMALL>L</SMALL>-arabinopyranoside, oleanolic acid 3-<I>O</I>-β-<SMALL>D</SMALL>-glucopyranosyl(1→3)-α-<SMALL>L</SMALL>-rhamnopyranosyl(1→2)-α-<SMALL>L</SMALL>-arabinopyranoside, hederagenin 3-<I>O</I>-α-<SMALL>L</SMALL>-rhamnopyranosyl(1→2)-α-<SMALL>L</SMALL>-arabinopyranoside (α-hederin), and hederagenin 3-<I>O</I>-β-<SMALL>D</SMALL>-glucopyranosyl(1→3)-α-<SMALL>L</SMALL>-arabinopyranoside (collinsonidin).         An antihyperglycemic compound in a model of alloxan induced diabetic rats was isolated from the root bark of Aralia elata. The compound identified to be oleanolic acid $28-O-{\beta}-_D-glucopyranoside$ was active at a dose of 100 mg/kg p.o. in the rats. It also has increased the contents of liver glycogen which were lowered by injection of alloxan in the rats.

        반응표면분석법(RSM)을 이용한 두릅의 유용성분 환류추출공정의 최적화

        Response surface methodology (RSM) was used to monitor the characteristics of a return current extracts from Aralia elata. A central composite design was used to investigate the effects of the independent variables of extraction time (X1), ethanol concentration (X2) and extraction temperature (X3) on the dependent variables yield (Y1), total phenolics (Y2), electron donating ability (Y3), browning color (Y4) and reducing sugar contents (Y5) of extracts. The maximum yield was at 316.58 min (X1), 15.68% (X2) and 93.49oC (X3) in saddle point. Total phenolics were highly affected by ethanol concentration. The maximum total phenolics were at 94.78oC (X1), 22.9% (X2), 323.05 min (X3) in saddle point. The maximum electron donating ability was at 94.78oC (X1), 29.82% (X2), 323.05 min (X3) and was highly affected by ethanol concentration. The maximum browning color was at 97.00oC (X1), 38.35% (X2), 321.82 min (X3). The browning color was highly affected by ethanol concentration. The ruducing sugar content was at 97.00oC, 38.35%, When the extraction temperature, ethanol concentration, and extraction time were 87.62oC (X1), 24.62% (X2), 339.39 min (X3). The optimal conditions of extraction conditions for response variables by superimposing of contour maps were 263.5 min (X1), 14% (X2) and 89.5oC (X3).
