위종양에 있어서 H-ras P^(21) 종양유전자단백의 발현에 관한 면역조직화학적 관찰

To observe the increased expression of the human H-ras p21 in tumor cells of gastric neoplasms including 10 cases of adenomatious polyp, 10 cases of cases of early gastic carcinoma and 20 cases of advanced carcinoma, immunohistochemical studies were carried out by using avidin-biotin-complex immnoperoxidase method in formalin fixed tissue sections. The results were as follows: 1. The increased expression of H-ras p21 in gastic carcinoma depended upon the tumor differentiation. 2. Although advanced gastric carcinoma demonstrated stronger positive reaction of H-ras p21 than early carcinoma and adenomatous polyps, no correlation among advanced carcinomas and H-ras p21 expression was observed in relation to depth of tumor size, tumor invasion and lymph node metastasis(p>0.05). 3. Although the adnomatous polyp of stomach showed less positive reaction of H-ras p21 than that of gastric carcinoma, there were positive focally in 3 cases and diffusely in 2 cases. Therefore, the result of this investigation suggested that increased expression of H-ras p21 was involved in the process of multistep carcinogenesis of gastric carcinoma.

The performance of moisture removal system is investigated analytically and experimentally. The program for the moist air calculation and for the condensing tube design are developed to examine the factors which affect moisture removal rate. Experimental apparatus comprises condensing tube, cooling system, vacuum pump, measurement and control devices. Thermoelectric cooling system and vapor compression refrigeration system are used to cool the air, and the performance of two systems are compared each other. Thermoelectric cooling system is compact and free from noise and vibration, but cooling rate of it is relatively slow compared to that of vapor compression refrigeration system. Air velocity, indoor temperature and humidity, temperature of coolant, size and material of condensing tube are found to affect the performance of moisture removal.

Cancer cachexia is a highly debilitating condition characterized by weight loss and muscle wasting that contributes significantly to the morbidity and mortality of pancreatic cancer. The factors that induce cachexia in pancreatic cancer are largely unknown. We previously showed that pancreatic adenocarcinoma upregulated factor (PAUF) secreted by pancreatic cancer cells is responsible for tumor growth and metastasis. Here, we analyzed the relation between pancreatic cancer-derived PAUF and cancer cachexia in mice and its clinical significance. Body weight loss and muscle weight loss were significantly higher in mice with Panc-1/PAUF tumors than in those with Panc-1/Mock tumors. Direct administration of rPAUF to muscle recapitulated tumor-induced atrophy, and a PAUF-neutralizing antibody abrogated tumor-induced muscle wasting in Panc-1/PAUF tumor-bearing mice. C2C12 myotubes treated with rPAUF exhibited rapid inactivation of Akt-Foxo3a signaling, resulting in Atrogin1/MAFbx upregulation, myosin heavy chain loss, and muscle atrophy. The neutrophil-to-lymphocyte ratio and body weight loss were significantly higher in pancreatic cancer patients with high PAUF expression than in those with low PAUF expression. Analysis of different pancreatic cancer datasets showed that PAUF expression was significantly higher in the pancreatic cancer group than in the nontumor group. Analysis of The Cancer Genome Atlas data found associations between high PAUF expression or a high DNA copy number and poor overall survival. Our data identified tumor-secreted circulating PAUF as a key factor of cachexia, causing muscle wasting in mice. Neutralizing PAUF may be a useful therapeutic strategy for the treatment of pancreatic cancer-induced cachexia.

<P>Although epigenetic mechanisms of gene expression regulation have recently been implicated in memory consolidation and persistence, the role of nucleosome-remodeling is largely unexplored. Recent studies show that the functional loss of BAF53b, a postmitotic neuron-specific subunit of the BAF nucleosome-remodeling complex, results in the deficit of consolidation of hippocampus-dependent memory and cocaine-associated memory in the rodent brain. However, it is unclear whether BAF53b expression is regulated during memory formation and how BAF53b regulates fear memory in the amygdala, a key brain site for fear memory encoding and storage. To address these questions, we used viral vector approaches to either decrease or increase BAF53b function specifically in the lateral amygdala of adult mice in auditory fear conditioning paradigm. Knockdown of Baf53b before training disrupted long-term memory formation with no effect on short-term memory, basal synaptic transmission, and spine structures. We observed in our qPCR analysis that BAF53b was induced in the lateral amygdala neurons at the late consolidation phase after fear conditioning. Moreover, transient BAF53b overexpression led to persistently enhanced memory formation, which was accompanied by increase in thin-type spine density. Together, our results provide the evidence that BAF53b is induced after learning, and show that such increase of BAF53b level facilitates memory consolidation likely by regulating learning-related spine structural plasticity.</P>

Immunomodulatory Effect of Allomyrina dichotoma Larva Extract in Macrophage Cells

Macrophages plays an important role in innate immune system. Allomyrina dichotoma is widely used as traditional medicine, however, its immunomodulatory effects is still unknown. In this study, we investigated whether Allomyrina dichotoma larva hot water extracts (ADLE) has immunomodulatory effects in macrophages to explore applicability of ADL as supplements for immune-boosting. Treatment of ADLE did not induce cytotoxicity in RAW 264.7 cells at doses ranging from 1 to 200 μg/mL Treatment of ADLE increased the production of nitric oxide and secretion of pro-inflammatory cytokines (TNF-α and IL-6) in RAW264.7 cells at doses of 100 and 200 μg/mL. Furthermore, ADLE-treated RAW264.7 cells showed phosphorylation of mitogen activated protein kinases (ERK, JNK, and p38) and nuclear factor-κB translocation. Collectively, these results indicate that ADLE exhibits immune-enhancing effects in macrophages, which has a value as an immunomodulatory supplement.
