폐암종에서의 H-ras p^21과 p^53양유전자 발현에 관한 면역조직화학적 연구
박언섭,박용욱,유재형 중앙대학교 의과대학 의과학연구소 1993 中央醫大誌 Vol.18 No.4
To observe the expression of H-ras P^21 and p^53 in cancer cells with clinical data and different pathologic findings of lung cancer, immunohistochemical studies were carried out by using avidin-biotin complex immunoperoxidase method in paraffin-embedded tissue sections from 27 cases. The studies consisted of 18 cases of bronchogenic squamous cell carcinoma, including 4 cases of well differentiated type and 14 cases of moderately differentiated type, 7 cases of primary adenocarcinoma, and 2 cases of metastatic carcinoma, including 1 case of metastatic adenocarcinoma from stomach and 1 case of metastatic transitional cell carcinoma from urinary bladder. The results were as follows: 1. Positive reactions of H-ras p^21 in lung carcinoma were observed in 72% of the squamous cell carcinomas and 87% of the adenocarcinomas. 2. Positive reactions of p^53 in the carcinoma were observed in 34% of the squamous cell carcinomas and 62.5% of the adenocarcinomas. 3. Correlation between patient's age and oncogene expression was statistically not significant 4. Correlation between tumor size and oncogene expression was statistically not significant 5. Correlation between stage and oncogene expression was statistically not significant 6. Correlation between tumor size and stage was statistically significant (Kendall's tau-b Coefficients=0.04405, P<0.05O. 7. Correlation between H-ras P^21 and p^53 expression was statistically significant (Kendall's tau-b Coefficients=0.6123, P<0.01). 8. Therefore, authors suggested that H-ras P^21 and p^53 expression was involved in the progression of lung cancer.
위선암종의 예후인자로서 p53, CD44v6과 VEGF 단백 발현

Purpose: The p53 protein is a tumor supressor gene, and its mutation is associated with biologic aggressiveness. CD44v6, one of the CD44 family, is a cell surface glycoprotein that plays a role in cancer invasion and metastasis. Vascular endothelial growth factor (VEGF) is another recently identified growth factor with significant angiogenic properties. The purpose of this study was to investigate p53, CD44v6, and VEGF expressions to determine whether degree of expression was related to pathological parameters such as Lauren's classification, depth of invasion, and lymph node metastasis. Materials and Methods: Immunohistochemical stains of p53, CD44v6, and VEGF in formalin-fixed paraffin-embedded tissue sections of 125 gastric adenocarcinomas were done. Results: The overall expression rates of p53, CD44v6, and VEGF were $54.4\%$ (68/125), $36.8\%$ (46/125), and $48.0\%$ (60/125), respectively. The p53, not CD44v6 and VEGF was higher in intestinal-type gastric carcinomas by Lauren's classification. The expressions of p53, CD44v6, and VEGF were statistically correlated with depth of tumor invasion. The expression of CD44v6 was higher in the lymph node metastatic group than in the negative group. The p53 expression was significantly associated with VEGF expression. Conclusions: These data suggest that the expressions of p53, CD44v6, and VEGF are biologically related to malignancy. The p53 and CD44v6 expressions are independent; however, p53 gene mutation is one of the contributing factors to VEGF expression in gastric adenocarcinoma.
Vol.20 No.3
To investigate the transforming potential in various hyperplastic nodules developed during hepatocarcinogenesis induced by N, N-diethylnitrosamine(DEN), we analyzed the sequential histopathology, foci of localization of Glutathione S-Transferase(GST-P), numbers of AgNORs and DNA ploidy in various nodules as well as in carcinoma. Male rats, Wister, with body weight of 150-170gm were used and fed daily oral intak by 150ppm DEN. Five animals were killed at an interval of two weeks during experimental 16 weeks period and at the same date two control rats were killed for comparison. Areas of Gluathione S-transferase foci were analyzed by image analysis, numbers of AgNORs were analyzed by direct counting under the light microscope and DNA ploidy by flow cytometry. Immunohistochemical staining was done using avidin biotin complex with nonspecific polyclonal GST-P antibody. The clear cell nodule appeared first at the 3rd weeks after DEN administration followed by eosinophilic and basophilic nodules whit mixed cell nodules consisting respective cells in between, and carcinoma appears at the 14 weeks. GST immunostaining were revealed a tendency of progressive increase in positive foci, strong reaction were noted especially in clear cells and eosinophilic cell nodules as well as in addenoma and carcinoma. Mean numbers of AgNORs per nucleus in control, clear cell, eosinophilic cell, basophilic cell nodule, addenoma and carcinoma were 1.59, 2.86, 2.66, 2.72, 2.92 and 3.43, respectively. And the perentage of cells containing over five AgNORs were 0.07%, 7.30%, 6.20%, 6.50%, 8.80% and 12.40%(p<0.01), respectively. The percentage of aneuploidy were 14.3%, 36.4%, 30.0%, 40.0% and 45.5%, in respective nodules. As above results it could be suggested that carcinoma night be developed from the eosinophilic cell nodule through the aberration of DNA ploidy as in basophilic cell nodule and adenoma in the hepatocarcinogensis.
Intravascular Leiomyosarcoma of the Femoral Vein - A Case Report -
박언섭,홍순억,홍민의,권귀영,이태진,유재형 대한병리학회 2008 Journal of Pathology and Translational Medicine Vol.42 No.4
Intravascular leiomyosarcomas of the femoral vein are extremely rare. Our patient was initially diagnosed with a deep vein thrombosis based on ultrasonography and venography. The thrombectomy specimen consisted of typical spindle cells with variable anaplasia arranged in a fasciculating and interlacing pattern. The final diagnosis was proved to be an intravascular leiomyosarcoma confirmed by immunohistochemical studies for smooth muscle actin, desmin, vimentin, CD34 and CD68.
