        UV-B 조사에 따른 버섯 추출물의 항염증 및 항알레르기 활성

        본 연구에서는 표고버섯(Lentinula edodes)을 자외선 조사를 통한 ergocaciferol (비타민 D₂) 함량을 증진시킨 추출물을 이용하여 염증 및 알레르기 반응에 미치는 효과를 확인하였다. 표고버섯 추출물의 항염증 및 항알레르기 효능은 LPS로 활성화된 대식세포(RAW 264.7)와 PMA와 A23187에 의해 활성화된 비만세포(RBL-2H3)로부터 분비 또는 발현되는 TNF-α, IL-6, IL-1β, IL-4와 같은 cytokine과 histamine 분비량을 측정하여 관찰하였다. LPS에 의해 활성화된 대식세포에서 자외선 조사 표고버섯 추출물 처리에 의해 pro-inflammatory cytokine인 TNF-α와 IL-6의 분비량을 ELISA 방법으로 측정하였을 때 현저히 감소함을 확인하였고 mRNA 발현 또한 감소됨을 확인하였다. 비만세포에 자외선 조사 표고버섯 추출물과 PMA, A23187을 함께 처리한 경우 비만세포의 탈과립에 의해 분비되는 histamine의 양이 유의적으로 감소함을 확인하였고 IL-4의 발현양 또한 감소함을 확인할 수 있었다. 이상의 결과는 자외선 조사에 의해 비타민 D₂ 함량을 증진시킨 표고버섯 추출물이 염증과 알레르기 반응의 cytokine의 발현을 저해하는 것으로 보아 염증 및 알레르기 질환의 예방과 치료에 효과적으로 이용될 수 있을 것으로 사료된다. In this study, the effects of UV irradiation-enhanced ergocalciferol (vitamin D₂) content containing Lentinula edodes extract on inflammation and allergic responses were investigated in vitro. The anti-inflammatory and anti-allergic effects of the mushroom extract were tested by estimating the cytokine secretions, such as TNF-α, IL-6, and IL-1β in LPS-activated macrophages (RAW 264.7), or histamine release in PMA and A23187-activated mast cells (RBL-2H3). Under the condition of macrophage activation with LPS, mushroom extract significantly reduced the secretions of pro-inflammatory cytokines, TNF-α and IL-6, and their mRNA expression also matched the observation. The current mushroom extract also significantly reduced the amount of mast cell degranulation-induced histamine secretion from PMA- and A23187-treated mast cells as well as the reduced secretion of IL-4. These results suggest that mushroom extract, which has increased ergocalciferol content by UV irradiation, inhibits the expression of cytokines in inflammation and allergic reactions; therefore, it can be used effectively for the prevention and treatment of inflammatory and allergic diseases.

        IRAK4 as a Molecular Target in the Amelioration of Innate Immunity–Related Endotoxic Shock and Acute Liver Injury by Chlorogenic Acid

        <P>Mice lacking the IL-1R-associated kinase 4 (IRAK4) are completely resistant to LPS-induced endotoxic disorder or the TLR9 agonist CpG DNA plus D-galactosamine-induced acute liver injury (ALI), whereas wild-type strains succumb. However, translational drugs against sepsis or ALI remain elusive. Lonicerae flos extract is undergoing the clinical trial phase I in LPS-injected healthy human volunteers for sepsis treatment. In the current study, chlorogenic acid (CGA), a major anti-inflammatory constituent of lonicerae flos extract, rescued endotoxic mortality of LPS-intoxicated C57BL/6 mice, as well as ameliorated ALI of LPS/D-galactosamine-challenged C57BL/6 mice. As a mechanism, CGA inhibited various TLR agonist-, IL-1 alpha-, or high-mobility group box-1-stimulated autophosphorylation (activation) of IRAK4 in peritoneal macrophages from C57BL/6 or C3H/HeJ mice via directly affecting the kinase activity of IRAK4, a proximal signal transducer in the MyD88-mediated innate immunity that enhances transcriptional activity of NF-kappa B or AP-1. CGA consequently attenuated protein or mRNA levels of NF-kappa B/AP-1 target genes encoding TNF-alpha, IL-1 alpha, IL-6, and high-mobility group box-1 in vivo under endotoxemia or ALI. Finally, this study suggests IRAK4 as a molecular target of CGA in the treatment of innate immunity-related shock and organ dysfunction following insult of various TLR pathogens from bacteria and viruses.</P>

      • 가감목방기탕(加減木防己湯)이 흰쥐의 Adjuvant 유발 관절염에 미치는 영향

        Objectives : This study was to investigate the effect of Kagammokbanggi-tang on the Freund's Complete Adjuvant(FCA)-induced arthritis in rats. Methods : Arthritis was induced by intradermal injection of FCA into base of tail. Arthritic rats were divided into control(n=10) and sample(n=10) group. Control group was taken normal saline for twenty days and sample group was taken extracts of Kagammokbanggi-tang for same duration. Normal group(n=10), non-arthritic group, was injected with mineral oil and was taken normal saline for twenty days. Body weight, paw edema volume and ankle joint thickness were measured at 0, 10, 15, 20 days after treatment. $TNF-{\alpha}$, $IL-1{\beta}$, IL-6, $PGE_2$ in synovia were analysed by ELISA at 20 days after treatment. Histochemical investigation of NADPH-d in the PAG and histopathological study on the ankle joint were performed at 20 days after treatment. Results : Paw edema volume, ankle joint thickness, $TNF-{\alpha}$, $IL-1{\beta}$, IL-6, $PGE_2$, NADPH-d of sample group were significantly decreased compared with control group. Conclusions : These results indicated that Kagammokbanggi-tang has antiarthritic and analgesic effects, and inhibited expression of NOS on the progression of FCA- induced arthritis in rats.

        임상연구코디네이터의 역할수행과 관련 요인

        Purpose: The purpose of this study was to analyze the role performance and related factors of the Clinical Research Coordinator (CRC). Methods: A quantitative, cross-sectional design was used. Data were collected using a structured questionnaire from 134 Clinical Research Coordinators in 4 hospitals and analyzed using descriptive statistics, t-test, ANOVA, and Pearson correlation coefficient. Results: The scores for role performance of CRC were high (average 336.51 points of a possible 420 points, mean average 5.74±0.67 of a possible 7) and the degree of role performance in the management task sub dimension (mean average 6.12±0.77 of a possible 7) was higher than any other sub dimension. Role performance of the CRC was also related to job satisfaction (r=.198, p=.023). Conclusion: The study results indicate that most CRC are nurses and perceive their role as very important. Further the CRC performance of activities of management task and the Coordination/Advocacy task sub dimension were high and higher levels of job satisfaction were related to higher levels of role performance of CRC was. Therefore, strategies related to improvement in the fringe benefit system and work environment should be developed to increase job satisfaction and thus increase job performance.

        In this thesis, the method for period detection of korean continuous speech recognition, detecting the voice section out of input signal and classification of the voice section by voiced and unvoiced, after period detection in classified voiced. Strong at noise, because of threshold energy stipulate in input signal, possible to detecting the voice section and classification voiced and unvoiced by zero crossing rate(ZCR) and short time energy(STE), period detecting possible as real-time approach by matlab.

      • 군집분석법에 의한 안면비대칭의 분류

        The purpose of this study was to classify the facial asymmetry by cluster analysis using the variables of the posteroanterior cephalogram, and to find out the characteristics of each group. The posteroanterior cephalogram and frontal facial photograph of one hundred orthodontic patients were used. Cluster analysis was executed using the measurements which indicated the degree of facial asymmetry. The resultant groups were compared each other according to the measurements to find out the characteristics of each group. Through the statistical analysis, following results were obtained. 1. Facial asymmetry was classified into five groups, A, B, C, D, and E, on the basis of menton deviation, apical base midline discrepancy and vertical difference of distance between right and left antegonions. 2. The degree of Menton deviation was severe in the group A, moderate in the group C and D, and mild in the group B and E. 3. Apical base midline discrepancy showed high value in the group A and C. 4. Antegonion was superiorly positioned on the same side of menton deviation in the group A and D, whereas it was superiorly positioned on the opposite side of menton deviation in the group B. 5. All measurements except the curvature of mandibular inferior border showed high value in the group A whereas very low in the group E. 6. While the characteristics of the group D were similar with those of the group A, the degree of difference was less in the group D. The group B showed unique characteristics as many measurements showed negative value. The above results suggest that facial asymmetry can be simply classified by cluster analysis on the basis of menton deviation, apical base midiine discrepancy and the vertical positional difference of antegonion between right and left sides.

        In this paper, we has been studied the variable diffusion filter bank. A variable diffusion filter bank which varies the width of pass band is using diffusion network. A variable diffusion filter bank which has advantage of building multi-channel without additional hardware. The simulation result of pass band ripple was very small and the range of slop hand was ignorable.

      • 제2형 당뇨병 환자에서 체내 총항산화능 측정의 의의

        Background: The diabetic patients are at significantly increased risk of developing vascular disease. It's etiology may involve oxidative damage by free radiacals and protection againse such damage can be offered by antioxidants. We investigated that oxidative stress as assessed by measurement of total antioxidant status may play a role in development of diabetes mellitus. Method: We measured total antioxiant status using merchandised kit, glycated hemeglobin(HbA1c) in 46 type 2 diabetes mellitus patients and 50 healthy matched control subjects. Result: The total antioxidant status(TAS) was 2.10(±0.04) mmol/L in uncontrolled type 2 DM patients, 2.60(±0.03) in controlled type 2 DM patients and 2.70(±0.16) in healthy control subjects. TAS was significantly lower(P<0.05) in uncontrolled type 2 DM patients, but no significant association between in controlled type 2 DM patients and healthy control subjects. The TAS was 2.00(±0.17) mmol/L in complicated type 2 DM patients and 2.10(±0.29) uncomplicated type 2 DM patients. There was no significant associations between complicated type 2 DM patients and uncomplicated type 2 DM patients. Conclusion: Poor glycemic control is associated with reduced TAS in type 2 DM patients. TAS was thought indirect index that predict glycemic control of type 2 DM patients.
