        Control synthesis of magnetic Fe3O4–chitosan nanoparticles under UV irradiation in aqueous system

        Novel magnetic Fe3O4–chitosan nanoparticles were synthesized via photochemical method in an emulsifier-free aqueous system at room temperature for the first time. The scanning electron microscopy (SEM)and transmission electron microscopy (TEM) results showed that the Fe3O4–chitosan nanoparticles were in regular shape with a mean diameter of 41 nm, whereas the average size in aqueous solution measured by photocorrelation spectroscopy (PCS) was 64 nm, which indicated that the nanoparticles had waterswelling properties. X-ray diffraction (XRD) patterns indicated that the Fe3O4 nanoparticles were pure Fe3O4 with a spinel structure, and the irradiation under UV light did not result in a phase change. The Fe3O4–chitosan nanoparticles were also characterized by Fourier transform infrared (FTIR) spectra, thermogravimetric analysis (TGA) and vibrating sample magnetometer (VSM). Magnetic measurement revealed that the saturated magnetization (Ms) of the Fe3O4–chitosan nanoparticles reached 48.6 emu/g and the nanoparticles showed the characteristics of superparamagnetism. The stability test showed these novel nanoparticles had high magnetic stability. The PCS and TGA results indicated that the size and chitosan content of Fe3O4–chitosan nanoparticles formed was pH- and chitosan/Fe3O4 ratio-dependent,which could be used to synthesize magnetic Fe3O4–chitosan nanoparticles with different size to meet the requirements of different applications.

        Effectiveness and Safety of Dabrafenib in the Treatment of 20 Chinese Children with BRAFV600E-Mutated Langerhans Cell Histiocytosis

        Purpose We sought to investigate the effectiveness and safety of dabrafenib in children with BRAFV600E-mutated Langerhans cell histiocytosis (LCH). Materials and Methods A retrospective analysis was performed on 20 children with BRAFV600E-mutated LCH who were treated with dabrafenib. Results The median age at which the patients started taking dabrafenib was 2.3 years old (range, 0.6 to 6.5 years). The ratio of boys to girls was 2.3:1. The median follow-up time was 30.8 months (range, 18.9 to 43.6 months). There were 14 patients (70%) in the risk organ (RO)+ group and six patients (30%) in the RO– group. All patients were initially treated with traditional chemotherapy and then shifted to targeted therapy due to poor control of LCH or intolerance to chemotherapy. The overall objective response rate and the overall disease control rate were 65% and 75%, respectively. During treatment, circulating levels of cell-free BRAFV600E (cfBRAFV600E) became negative in 60% of the patients within a median period of 3.0 months (range, 1.0 to 9.0 months). Grade 2 or 3 adverse effects occurred in five patients. Conclusion Some children with BRAFV600E-mutated LCH may benefit from monotherapy with dabrafenib, especially high-risk patients with concomitant hemophagocytic lymphohistiocytosis and intolerance to chemotherapy. The safety of dabrafenib is notable. A prospective study with a larger sample size is required to determine the optimal dosage and treatment duration.

        The Impact of Risk Perception on Developing Incentive Systems for Relational Contracting

        Financial incentives, specifically refer to risk/reward sharing arrangements, have been seen as an important way of facilitating relational contracting. Although there has been a great interest in developing appropriate incentive systems, still very little literature focuses on the impact of risk perception on risk/reward sharing arrangements. Taking into account risk characteristics in relational contracting, this study proposes a theoretical model in order to extend the understanding of relational and performance risk perception to the formulation of financial incentives from social and individual perspectives, and then validates the model for assessing the relationships among social and individual factors, risk perception and financial incentives with structural equation modeling in the context of integrated project teams. Data collected from 238 members in China can provide support for the proposed model. The results demonstrate that both relational and performance risk perception are significantly related to financial incentive, and social interaction ties, trust, shared vision, control ability and self-efficacy can reduce participants’ perceived level of risk. This study suggests that participants’ social capital and individual cognition should not be ignored when developing incentive systems for relational contracting.

        Comparison of Two Cases of Strong Increase in the Bottom Temperature in the Yellow Sea in Winter

        Mooring observations were carried out on the western slope of the southern Yellow Sea (YS) to observe the Yellow Sea Warm Current (YSWC). Strong increases in the bottom temperature (about 3°C within 1 day) were observed at mooring M5 (shelf break) on 8 January 2007, and at mooring A2 (mid-slope) on 5 December 2008. The strong temperature increase of bottom water at M5 was closely related to the burst of the YSWC. The bottom temperature at mooring A2 increased and decreased alternately from 7 November to 14 December 2008, and the strong increase (about 3°C within 1 day) occurring on 5 December was one of the four rises during that period. The significant semi-diurnal-oscillation during that period indicated that the thermal fronts outlining the Yellow Sea Cold Water Mass (YSCWM) boundary was very close to location A2. The notable rises in the bottom temperature at A2 were associated with bottom eastward currents, while the distinct falls in temperature coincide with bottom westward currents. The distinctive distribution in the bottom temperature associated with the YSCWM and the bottom eastward currents were mainly responsible for the strong increase in the bottom temperature at A2. The variations in the bottom temperature described here are valuable for understanding the time evolution of the YSWC in winter and the YSCWM from late autumn to early winter.

      • A Novel Area and Time Efficient Digital Pixel Sensor

        This paper presents a Digital Pixel Sensor(DPS) system with accurate Photo-gate (PG) model, Multi-Channel Bit Serial (MCBS) Analog-to-Digital convertor (ADC) and 1-bit 2-T memrory shared among a block of 2×2 pixels and novel conversion and a new readout timing scheme designed using 0.18-㎛ process with a supply voltage of 3.3V. With 160㎒ readout frequency, a 512×640 pixel array can have a frame rate higher than 10,000 frames/s. The layout of the block has been drawn with a size of 24×24㎛ with a fill factor of 56%. A Bandgap biasing circuit for the ADC designed has been implemented with layout size of 66×105.5㎛ and stable performances under variable temperature and supply voltage.

        LncRNA A2M-AS1 lessens the injury of cardiomyocytes caused by hypoxia and reoxygenation via regulating IL1R2

        Background: Myocardial ischemia and reperfusion injury (MI/RI) is a complex pathophysiological process, which can lead to severe myocardial injury. The long noncoding RNA alpha-2-macroglobulin antisense RNA 1 (A2M-AS1) has been revealed to be abnormally expressed in MI, However, its function in MI and the potential mechanism are still unclear. Objective: To evaluate the functional role of A2M-AS1 in hypoxia/reoxygenation (H/R)-induced neonatal cardiomyocytes and its potential molecular mechanism. Methods: Dataset GSE66360 was obtained from GEO database for analyzing the RNA expression of A2M-AS1 and interleukin 1 receptor type 2 (IL1R2). KEGG pathway enrichment analysis of the genes that co-expressed with A2M-AS1 was performed. Human neonatal cardiomyocytes were subjected to H/R to construct in vitro models. QRT-PCR and Western blot were adopted to test the levels of mRNA and protein. The viability and apoptosis of cardiomyocytes were tested by CCK-8 and flow cytometry assays, respectively. Results: The expression of A2M-AS1 was notably downregulated in H/R-treated cardiomyocytes. Overexpression of A2M-AS1 can notably enhance the cell viability of H/R-damaged cardiomyocytes, whereas knockdown of A2M-AS1 showed the opposite outcomes. Besides, a negative correlation was showed between A2M-AS1 and IL1R2 expression. In H/R-treated cardiomyocytes, overexpression of IL1R2 weakened the promoting proliferation and anti-apoptosis effects caused by overexpressing A2M-AS1, however, IL1R2-knockdown abolished the anti-proliferation and pro-apoptosis effects caused by silencing A2M-AS1. Conclusion: This study demonstrates the potential regulatory role of A2M-AS1/ IL1R2 axis in cardiomyocytes suffered from H/R, and provides insight into the protection of MI/RI.

        CADMIUM TELLURIDE NANOCRYSTALS: SYNTHESIS, GROWTH MODE AND EFFECT OF REACTION TEMPERATURE ON CRYSTAL STRUCTURES

        A series of cadmium telluride (CdTe) nanocrystals were synthesized by a modified organometallic synthesis method at various reaction temperatures ranging from 130 to 250°C. In this method, octadecylamine (ODA) was introduced as an additional coordinating component to the mixture of trioctylphosphine oxide (TOPO) and trioctylphosphine (TOP). CdO was used as a precursor. The prepared CdTe nanocrystals were studied by the absorption and emission spectra as well as the powder X-ray diffraction (XRD) patterns. The result shows that besides the traditional continuous-growth mode observed frequently at relatively high reaction temperature, a discontinuous-growth mode was confirmed at the initial growth stage of CdTe nanocrystals, arising from the change of the absorption spectra of CdTe nanocrystals with the reaction time at relatively low reaction temperature. The structures of CdTe nanocrystals, e.g., the cubic zinc blende structure at 160°C and the hexagonalwurtzite structure at 250°C, were characterized by XRD.

      • Anti-proliferation Effects of Interferon-gamma on Gastric Cancer Cells

        IFN-${\gamma}$ plays an indirect anti-cancer role through the immune system but may have direct negative effects on cancer cells. It regulates the viability of gastric cancer cells, so we examined whether it affects their proliferation and how that might be brought about. We exposed AGS, HGC-27 and GES-1 gastric cancer cell lines to IFN-${\gamma}$ and found significantly reduced colony formation ability. Flow cytometry revealed no effect of IFN-${\gamma}$ on apoptosis of cell lines and no effect on cell aging as assessed by ${\beta}$-gal staining. Microarray assay revealed that IFN-${\gamma}$ changed the mRNA expression of genes related to the cell cycle and cell proliferation and migration, as well as chemokines and chemokine receptors, and immunity-related genes. Finally, flow cytometry revealed that IFN-${\gamma}$ arrested the cells in the G1/S phase. IFN-${\gamma}$ may slow proliferation of some gastric cancer cells by affecting the cell cycle to play a negative role in the development of gastric cancer.

        High­risk human papillomavirus genotype distribution and attribution to cervical cancer and precancerous lesions in a rural Chinese population

        Objective: To explore the genotype distribution of high-risk human papillomavirus (HR-HPV) and its attribution to different grades of cervical lesions in rural China, which will contribute to type-specific HPV screening tests and the development of new polyvalent HPV vaccines among the Chinese population. Methods: One thousand two hundred ninety-two subjects were followed based on the Shanxi Province Cervical Cancer Screening Study I (SPOCCS-I), and screened by HPV DNA testing (hybrid capture® 2 [HC2]), liquid-based cytology (LBC), and if necessary, directed or random colposcopy-guided quadrant biopsies. HPV genotyping with linear inverse probe hybridization (SPF10-PCR-LiPA) was performed in HC2 positive specimens. Attribution of specific HR-HPV type to different grades of cervical lesions was estimated using a fractional contribution approach. Results: After excluding incomplete data, 1,274 women were included in the final statistical analysis. Fifteen point two percent (194/1,274) of women were HR-HPV positive for any of 13 HR-HPV types (HPV16, 18, 31, 33, 35, 39, 45, 51, 52, 56, 58, 59, and 68) and the most common HR-HPV types were HPV16 (19.1%) and HPV52 (16.5%). The genotypes most frequently detected in HR-HPV-positive cervical intraepithelial neoplasia grade 1 (CIN1) were HPV52 (24.1%), HPV31 (20.7%), HPV16 (13.8%), HPV33 (13.8%), HPV39 (10.3%), and HPV56 (10.3%); in HR-HPV-positive cervical intraepithelial neoplasia grade 2 or worse (CIN2+): HPV16 (53.1%), HPV58 (15.6%), HPV33 (12.5%), HPV51 (9.4%), and HPV52 (6.3%). HPV52, 31, 16, 33, 39, and 56 together contributed to 89.7% of HR-HPV-positive CIN1, and HPV16, 33, 58, 51, and 52 together contributed to 87.5% of CIN2+. Conclusion: In summary, we found substantial differences in prevalence and attribution of CINs between different oncogenic HPV types in a rural Chinese population, especially for HPV16, 31, 33, 52, and 58. These differences may be relevant for both clinical management and the design of preventive strategies.
