        한우 Band 3 및 CHS 유전병의 분자유전학적 기초연구

        Many bovine genetic diseases are currently unidentified in Korea because of the relatively low monitoring systems in the livestock farms. The molecular detection system using PCR-RFLP of two genetic diseases, namely Band 3 (Erythrocyte Membrane Protein Band Ⅲ) and CHS (Chediak-Higashi Syndrome), have been identified in Japan and used for screening large number of cattle whether each individual has the genetic disease or not. Using the 22 unrelated Korean cattle (Hanwoo) individuals, molecular detection system based on PCR-RFLP have been investigated, which can be distinguishable carriers for the genetic diseases. Even though we could not found the causative mutations for two genetic diseases, the PCR-RFLP techniques used in this study are very valuable for the screening the genetics diseases in Korean cattle, especially for the proven or candidate bulls.

      • 요근 농양이 동반된 Salmonella typhi 척추골수염 1예

        저자들은 아직 국내에 보고된 적이 없는 S. typhi에 의한 척추골수염 환자에서 ciprofloxacin을 5주간 투여하여 임상적 호전을 보였던 증례를 경험하였기에 보고하는 바이다. Vertebral osteomyelitis by Salmonella infection is rare. Although it is known to be associated with sickle cell anemia, various hemoglobinopathies and immunosuppressive states, it may also occur in normal host. To date, vertebral osteomyelitis with Salmonella. typhi has not been reported in Korea. We experienced a case of spinal osteomyelitis due to S. typhi in a 48-year old man who was admitted because of low back pain for two months. MRI scan showed vertebral osteomyelitis in L1 & L2. Needle aspiration was performed by CT guidance. S. typhi was identified from aspirated pus. However, any organism was not identified from blood and urine cultures. He was treated with ciprofloxacin for 5 weeks successfully.

        Resin Jacket Crown을 이용한 법랑질 형성 부전증 환자의 심미적 치험례

        Amelogenesis imperfecta represents a group of hereditary conditions that manifest enamel defects without evidence of generalized or systemic disorders. These enamel disorders are apparently heterogeneous in the basic chemical sturcture, resulting in a diverse presentation of clinical characteristics. The reported prevalence of amelogenesis imperfecta varies from 1 in 14,000-16,000 to 1.4 in 1,000 depending on specific population studied with the autosomal dominant hypocalcification type of amelogenesis imperfecta believed to be the least prevalent. The most widely accepted current classification system for delineating the amelogenesis imperfecta types considers the mode ofinheritance and clinical manifestations. Three major groups are recognized; hypoplastic, hypocalcified, and hypomaturation types. Delineating specific types of amelogenesis imperfecta can be confusing due to the phenotypical similarity of many forms and that the most recent classification lists 14 different types. A 12 year-old female patient came to our pediatric dentistry clinic complaining of the ugly shape and color of her teeth, especially the upper front area. Although the goal of the treatment was mainly focused on the improvement of patient's esthetics, longevity of the restorations was also considered in selecting the appropriate restorative system, resin jacket crown, which can satisfy the both aspects.

      • 한강 인도교지점에서의 이상홍수 해석

        The water level and flow rate data measured at Indogyo measurement point which serves as one of the major measuring points of Han River have been very important in aspect of controlling the flow. Because these data are becoming more and more unreliable due to the recent change in the basin or the narrowing of the river width, these data will be standardized by comparing the recent data of Indogyo measuring point with the past those. To minimize the errors of water level-flow rate curve caused by the yearly changes of basin, the standard water level-flow rate curve with respect to the present basin conditions will be set, and calculated reduced water level will be compared with the past data. And through frequency analysis, frequency-based flood level with respect to return period will be presented. Finally, the total flow rate of the flood measured in September, 1990 at the point of Indogyo measuring point will be compared with the total flow of the past extreme floods.

        낙상으로 인한 좌심실 파열 후 생존한 환자 1례

        Blunt or nonpenetrating chest injuries are common after motor vehicle accidents or falls. Blunt injury to the heart ranges from contusion to rupture. Cardiac rupture, however, is un-common and associated with a very high mortality rate ; left ventricular rupture due to falling is even more rare. We experienced a case of left ventricular rupture of the heart caused by a fall, which the patient survived. A 63-year-old man fell off a ladder and presented with hypotension from left hemothorax and pericardial tamponade. The hemothorax was relieved via an emergency closed thoracostomy but dyspnea and hypotension continued. And so, echocardio-graphy was done and pericardial tamponade was detected. Emergency left thoracotomy was done to repair the rupture site. We believe that patients with cardiac rupture who reach the hospital alive can often be saved by prompt diagnosis and immediate surgical treatment.

      • 대식세포에서 산화질소 생성에 대한 當歸 에탄올 추출물의 억제효과

        Objective : The Angelica gigas Nakai ethanol extract (AGE) was investigated to compare nitric oxide (NO) production and NF-kB activity from RAW 264.7 cells, since NO and nuclear factor-kB(NF-kB) have been shown to be factors implicated In inflammatory disease. Method : AGE was prepared by extracting medicinal herb with 70% (v/v) ethanol solution. We investigated production nitric oxide (NO) and inducible nitric oxide synthase (iNOS) gene expression by ARE in LPS-stimulated RAW 264.7 macrophage cells. We also investigated inhibition of LPS-induced activation of NF-kB on western blot. Result : LPS-induced RAW 264.7 cells increascd NO production and ,iNOS expression. Upon treatment with AGE, nitrite production was significantly inhibited in a concentration-dependent manner compared to the untreated control. AGE inhibited this LPS-induced ,iNOS mRNA and protein in a dose-dependent manner. AGE markedly inhibited the expression of iNOS mRNA and protein at a concentration of 100 μg/ml. LPS-induced RAW 264.7 cells with AGE blocked inhibitory factor-kBα degradation. Conclusion : This study shows that AGE seems to attenuate inflammation through inhibition of NO production and iNOS expression by blockade of NF-kB activation in LPS-stimulated RAW 264.7 cells.

        Anti-Helicobacter pylori Diarylheptanoid Identified in the Rhizome of Alpinia officinarum

        The antibacterial activity of materials derived from the rhizome of Alpinia officinarum (Zingiberaceae) against Helicobacter pylori ATCC 43504, ATCC 700392, and ATCC 700824 was examined using paper-disc diffusion and agar dilution methods. Results were compared with those following treatment with currently used antibiotics: amoxicillin, metronidazole, and tetracycline. The bioactive principle was characterized as the diarylheptanoid 5-hydroxy-7-(4-hydroxy-3-methoxyphenyl)-1-phenyl-3-heptanone by spectroscopic analysis. This compound was isolated from A. officinarum leaves as a new anti-H. pylori principle. Against H. pylon ATCC 43504, ATCC 700392, and ATCC 700824, the antibacterial activity of the diarylheptanoid (48,24, and 24 ㎍/mL) was comparable to that of metronidazole (32, 16, and 16 ㎍/mL) but less effective than that of either amoxicillin (0.06, 0.06, and 0.03 ㎍/mL) or tetracycline (0.5, 1, and 0.5 ㎍/mL), based on minimum inhibitory concentrations. A. officinarum rhizome-derived materials, particularly the diarylheptanoid isolated, merit further study as potential antibacterial functional food products or therapeutic products for prevention or eradication from humans from diseases caused by H. pylori.

      • Monitoring bacterial community structure and variability in time scale in full-scale anaerobic digesters

        <P>Using a high-throughput pyrosequencing technology, this study assessed bacterial community structure and time-scale variability in great detail in seven full-scale anaerobic digesters operated variously in terms of influent substrate, digestion temperature, and reactor configuration. Pyrosequencing generated a total of 83 774 sequence reads from 40 digester sludge samples collected monthly for six months. The highest number of sequence reads were detected within <I>Proteobacteria</I> (20.5%), followed by those within <I>Bacteroidetes</I> (19.7%), <I>Firmicutes</I> (17.8%), and <I>Chloroflexi</I> (4.8%). The relative composition of bacterial populations was varied within the digesters as well as between the digesters, and the bacterial community structures were mainly influenced by digestion temperature. Detailed bacterial community structures were assessed by analyzing the operational taxonomic units (OTUs) based on 97% sequence similarity, which resulted in a total of 9051 OTUs. Among these, a total of 31 core OTUs were analyzed and inferred phylogenetically, which enabled us to classify the sequences within an unclassified phylum. Unclassified sequences were mostly affiliated with the sequences within <I>Spirochaetes</I> and <I>Firmicutes</I>. Interestingly, numerically dominant novel phylotypes (18% of the total sequence reads) presumably involved in anaerobic digestion within <I>Spirochaetes</I> were identified. Temporal variability was further explored using a non-metric multidimensional scaling ordination which demonstrated that the variability of the bacterial community within the digesters was smaller than between digesters. Correlation analysis demonstrated that digester performance and operational conditions affected the pattern of bacterial community in the ordination. Additionally, a multi-response permutation procedure revealed that the bacterial communities within the digesters were more similar than those belonging to other digesters statistically, demonstrating a patchiness of the digesters in the distribution of bacterial populations. Overall, this study revealed the correlation of bacterial community structure and time-scale variability with digester performance and operating conditions.</P> <P>Graphic Abstract</P><P>The present study demonstrated that the bacterial communities within digesters were more similar than those belonging to other digesters, demonstrating a patchiness of the digesters in the distribution of bacterial population. <IMG SRC='http://pubs.rsc.org/services/images/RSCpubs.ePlatform.Service.FreeContent.ImageService.svc/ImageService/image/GA?id=c2em10958a'> </P>
