Thus we performed the analysis of dynamic characteristics through the experiment about developed system in order to reduce such problems and discuss the alternative methods.

      • Src/Syk-Targeted Anti-Inflammatory Actions of Triterpenoidal Saponins from Gac ( <i>Momordica cochinchinensis</i> ) Seeds

Yu, Jae Sik,Kim, Jun Ho,Lee, Seulah,Jung, Kiwon,Kim, Ki Hyun,Cho, Jae Youl World Scientific Publishing Company 2017 The American journal of Chinese medicine Vol.45 No.3

<P><I>Momordica cochinchinensis</I> Spreng (family Cucurbitaceae), also known as gac, or red melon, is an edible Southeast Asian fruit valued for its nutritional and medicinal properties. Specifically, Momordicae Semen, the seeds of the gac fruit, is used in traditional Chinese medicine to treat boils, rheumatic pain, muscle spasm, hemorrhoids, and hemangiomas. In this study, a chemical investigation into a gac seed ethanol (EtOH) extract resulted in the identification of three triterpenoidal saponins (<B>1</B>-<B>3</B>), which were investigated for their anti-inflammatory effects. Among the saponins, momordica saponin I (compound <B>3</B>) reduced the production of nitric oxide (NO) in LPS-activated RAW264.7 cells without inducing cytotoxicity. The mRNA levels of inducible NO synthase (iNOS) and cyclooxygenase (COX)-2 were decreased by momordica saponin I. Additionally, the translocation of p65 and p50 (subunits of the transcription factor NF-<TEX>$ \kappa $</TEX>B) into the nucleus was remarkably inhibited. Furthermore, the phosphorylation levels of inflammatory signaling proteins (I<TEX>$ \kappa $</TEX>B<TEX>$ \alpha $</TEX>, Src, and Syk) known to be upstream regulatory molecules of p65 were decreased under momordica saponin I-treated conditions. The molecular targets of momordica saponin I were confirmed in overexpression experiments and through immunoblot analyses with Src and Syk. This study provides evidence that momordica saponin I could be beneficial in treating inflammatory diseases, and should be considered a bioactive immunomodulatory agent with anti-inflammatory properties.</P>

Bioactivity-guided isolation of ginsenosides from Korean Red Ginseng with cytotoxic activity against human lung adenocarcinoma cells

        Jae Sik Yu,노현수,Kwan-Hyuck Baek,Seul Lee,Sil Kim,Hae Min So,Eunjung Moon,Changhyun Pang,Tae-Su Jang,Ki Hyun Kim 고려인삼학회 2018 Journal of Ginseng Research Vol.42 No.4

Background: Lung cancer is the leading cause of cancer-related death worldwide. In this study, we used a bioactivity-guided isolation technique to identify constituents of Korean Red Ginseng (KRG) with antiproliferative activity against human lung adenocarcinoma cells. Methods: Bioactivity-guided fractionation and preparative/semipreparative HPLC purification were used with LC/MS analysis to separate the bioactive constituents. Cell viability and apoptosis in human lung cancer cell lines (A549, H1264, H1299, and Calu-6) after treatment with KRG extract fractions and constituents thereof were assessed using the water-soluble tetrazolium salt (WST-1) assay and terminal deoxyribonucleotidyl transferaseemediated dUTP nick end labeling (TUNEL) staining, respectively. Caspase activation was assessed by detecting its surrogate marker, cleaved poly adenosine diphosphate (ADP-ribose) polymerase, using an immunoblot assay. The expression and subcellular localization of apoptosis-inducing factor were assessed using immunoblotting and immunofluorescence, respectively. Results and conclusion: Bioactivity-guided fractionation of the KRG extract revealed that its ethyl acetatee soluble fraction exerts significant cytotoxic activity against all human lung cancer cell lines tested by inducing apoptosis. Chemical investigation of the ethyl acetatesoluble fraction led to the isolation of six ginsenosides, including ginsenoside Rb1 (1), ginsenoside Rb2 (2), ginsenoside Rc (3), ginsenoside Rd (4), ginsenoside Rg1 (5), and ginsenoside Rg3 (6). Among the isolated ginsenosides, ginsenoside Rg3 exhibited the most cytotoxic activityagainst all human lung cancer cell lines examined,withIC50 values ranging from161.1 mMto 264.6 mM. The cytotoxicity of ginsenoside Rg3 was found to be mediated by induction of apoptosis in a caspaseindependent manner. These findings provide experimental evidence for a novel biological activity of ginsenoside Rg3 against human lung cancer cells. 2018 The Korean Society of Ginseng, Pu

        Jae Sik Yu,Jiwon Baek,Hyun Bong Park,Eunjung Moon,Sun Yeou Kim,Sang Un Choi,Ki Hyun Kim 대한약학회 2016 Archives of Pharmacal Research Vol.39 No.12

A new rearranged eudesmane sesquiterpene, named eudeglaucone (1), and five known sesquiterpenesincluding (?)-faurinone (2) and four eudesmane-typesesquiterpenes (3–6), were isolated from the twigs of Linderaglauca (Sieb. et Zucc.) Blume. The structure of 1 waselucidated by a combination of extensive spectroscopicanalyses, including extensive 2D NMR (1H-1H COSY,HMQC, HMBC, and NOESY) and HR-MS. Compound 1was a relatively rare rearranged eudesmane sesquiterpenein terpenoids. All isolates were evaluated for theirantiproliferative activities against four human tumor celllines (A549, SK-OV-3, SK-MEL-2, and HCT-15). Compounds3 and 6 showed significant cytotoxicity against SKMEL-2 and HCT-15 cell lines with IC50 values rangingfrom 9.98 to 12.20 lM. We also investigated the antineuroinflammatoryactivities of the isolates (1–6) in thelipopolysaccharide (LPS)-stimulated murine microglia BV-2 cell line by measuring nitric oxide (NO) levels. All isolatessignificantly inhibited NO production with IC50 valuesof 3.67–26.48 lM without inducing cell toxicity.

      • HCV, Acute, LT : Significance of Homeostasis Model Assessment-Insulin Resistance Level to Assess Insulin Resistance in Chronic Hepatitis C Patients

( Jae Hyuk Choi ),( Woo Jin Chung ),( Yu Jin Lee ),( Yu Jin Kang ),( Jung Min Lee ),( Eun Sung Choi ),( Sang Min Lee ),( Byoung Kuk Jang ),( Jae Seok Hwang ),( Eun Soo Kim ),( Kyung Sik Park ),( Kwang 대한간학회 2013 춘·추계 학술대회 (KASL) Vol.2013 No.1

Background/Aim: Homeostasis model assessment-insulin resistance (HOMA-IR) is commonly used for assessment of insulin resistance (IR) and IR is associated with chronic hepatitis C (CHC) virus infection. Recently, several study presented data about irrelevance between HOMA-IR and IR. This study aimed to examine the significance of HOMA-IR as representative of IR. Methods: We retrospectively reviewed data from CHC patients who receive standard of care (SOC) from September 2009 to March 2013 at Keimyung University Dongsan Hospital. We analyzed the changes of HOMA-IR score before treatment, at the time of end of treatment and at 6 months after treatment. Results: 53 patients were included (26 men; mean age, 51 ± 10.10 years) for this study. The prevalence of genotype 1a, 1b, 2a, 2b, and 3a were 1.9%, 35.8%, 54.7%, 3.8%, and 3.8%. Before treatment, initially high HOMA-IR group (>2) was 25 patients (47.2%). 1. There were no significant difference of ETR (83% vs. 92.9%, P=0.404) and SVR rate (87.5% vs. 96.0%, P=0.505) in initially high (>2) vs low (≤2) HOMA-IR groups. 2. There were no significant differences of ETR and SVR according to changes of HOMA-IR level between before and after treatment (high to high vs. high to low vs. low to high vs. low to low HOMA-IR; 23.4%, 21.3%, 6.4%, 48.9%, P=0.750 at ETR, and 13.2%, 23.7%, 7.9%, 55.3%, P=0.424 at SVR). 3. Initial HOMAIR levels had no significant differences between ETR group and non-ETR group (3.30 ± 6.29 and 2.99 ± 1.90; P=0.795), and between SVR group and non-SVR group (2.33 ± 2.90 and 3.34 ± 2.77; P=0.795). Conclusion: HOMA-IR levels were not inversely correlated or predicted of SVR after SOC for CHC patients. So, another assessing modalities such as hyperinsulinaemic euglycaemic clamp may be needed to assess IR in CHC patients.
