        Serratia liquefaciens AL-11이 생산하는 Alkaline Lipase의 특성 및 작용양상

        본 효소의 최적 반응온도는 약 45℃이고, 최적 pH는 10.0 정도였고, pH 7.0~10.0 범위와 30~50℃의 범위에서 안정하였다. 금속이온중 Mn^2+, Ca^2+ 등에 의하여 활성이 증대되었으나 Fe^2+, Pb^2+와 Zn^2+ 등에 의해서는 효소 활성이 저해되었고, 효소활성 저해제 중 ethylenediaminetetraacetic acid(EDTA)에 의해 강한 저해작용을 나타내어 본 효소는 효소분자 중 금속이온이 관여하는 것으로 추정되었다. 효소반응 처리한 olive oil 가수분해물을 박충크로마토그래피 분석한 결과 Serratia liquefaciens AL-11이 생산하는 지방분해효소는 기질특이성이 비특이적이었으며, sodium cholate, sodium edoxychol-ate, sodium taurocholate 등의 담즙산염에 의해 효소활성이 증대되었다. The optimum temperature and pH for the enzyme activity were 45℃ and 10.0, respectively. The enzyme was stable in a pH range of 5 to 10, and 62% of its activity was lost on heat treatment at 60℃ for 20 min. The activity of the purified enzyme was inhibited by Fe^2+, Zm^2+ and Pb^2+, and slightly activated by Mn^2+ and Ca^2+. γ-Chloromercuribenzoic acid, 2,4-dinitrophenol and H_2O_2 did not show inhibitory effect on the lipolytic activity of the alkaline lipase but ethylenediaminetetraacetic acid inhibited the enzyme activity. This suggested that the enzyme have metal group in its active site. Sodium salts of bile acids stimulated the enzyme activity. Analysis of hydrolyzates of olive oil after the lipase reaction revealed that Serratia liquefaciens AL-11 produced non-specific lipolytic enzyme.

      • Hurdle Technology방법에 의한 식품저장

        Hurdle technology에 의한 방법으로 식품을 저장하는 방법이 논의되었다. 이 방법은 다른 처리방법과 함께 사영하거나하여 미생물이 성장하는데 장애가되는 저해 요인들을 유도하여 한 가지 방법이 아니라 여러방법에 의해 가공 처리하는 방법이다. 이 방법은 냉장이나 냉동 없이도 안정하게 유지되며 가벼운 처리 가공방법이 적용되므로 관능면이나 영양가면에서도 매우 우수한 것으로 나타났다. 이에 기초를 둔 대표적인 식품으로는 F-SSP, A-SSP, pH-SSP, Combl-SSP, IMF 그리고 방사선조사 식품에 다른 처리를 한 것등이 있다. 이 기술방법은 주로 육류제품, 유가공제품, 해산물등에 응용되고 있으나 이를 다른 분야에 적용하는 방안을 앞으로 모색하여 나아가야할 것이다. The concept of hurdle technology is reviewed. This technique is the intelligent use of combinations of different preservation factors or technique in order to achieve multitarget, mild but reliable preservation effect. Hurdle-preserved foods are F-SSP, A_wSSP. pH-SSP, combi-SSP,IMF and irradiated foods with other treatments. This technology is mainly applied meat products, fish, and dairy products. Recently qualities of irradiated foods have been improved by combination of irradiation with other treatments such as cryogenic temperature or heating. The future work for application to various foods is discussed.

      • 4-nitro-N-(triphenyl phosphoranylidene)의 전기화학적 환원에 관한 연구

        The electrochemical reduction of 4-nitro-N-(triphenyl-phosphoranylidene) in nonaqueous media has been examined by polarography, cyclic voltammetry and controlled-potential coulometry. The reduction of 4-nitro-N-(triphenylphosphoranylidene) proceeds by a one electron-transfer to form anion radical which undergoes both protonation and a second one electron reduction followed by cleavage of a phosphorous-phenyl bond was also observed after reduction of triphenylphosphine oxide which is one of the major products of the chemical reaction which follow the primary electrode process.

      • 도시내 타워건축에 따른 경관영향 분석방안에 관한 연구

        Tower architectures affect their city-scape by their size, shape and height among other buildings and natural elements. Therefore the tower become the landmark of the town and symbolic and cognition element of the city scape. In this thesis we suggest a way to analyze the influence of tower construction to the city scape and we explain the concept and processes of visibility analysis, the analysis of view point and analysis of aerial view as the way of study of the city scape. Finally we propose the evaluation methods for the city tower to establish evaluation indices and elements following characteristics of the tower.

        We numerically investigate stable propagation regimes of 10 ㎓ train of optical solitons with 17.6 ps widths to determine the acceptable variations of periodic amplifier gain(△G), dispersion(△D), and initial chirp(C), as a function of the amplification period LP. We fine that, when LP = 25 km, the acceptable variations are △G = ±4%, △D= ±12%, and C= -0.36 ∼ 0.39. The variations decrease as the amplification period LP increases.

      • 년령 증가에 따른 췌장도 내분비세포 분포에 관한 조직학적 연구

        It is well known that the islet changes of pancreas could be induced by various factors, for example, obesity, hormones, pregnancy, age, etc. For the observation of the effect of age and sex, the size of the pancreatic islets and endocrine cells composing them were calculated. As an experimental animal the albino rats of Wistar strain were used. During breeding for fifty weeks, the rats were sacrificed with the interval of ten weeks and the pancreatic tissues were fixed in Bouin's solution and stained with hematoxylin and eosin, and aldehyde fuchsin and trichrome. The results were as follows : 1. At the age of fifty weeks of the male, the islets showed marked hypertrophy with peri-and intra-insular fibrosis. 2. The changes of the islets are found more earlier and severe in the male than in the female. 3. The frequency of appearance of pancreatic endocrine cells seemed not to change greatly during the increase of age.

      • 담즙정체성 간염의 임상적 양상

        Background: Cholestatic hepatitis is failure of bile to reach the duodenum with hepatocellular damage and no demonstrable obstruction of the major bile ducts. The prognosis is usually good with recovery in less than 4 weeks after withdrawal of the offending drug. However, a prolonged course lasting over 3 months is possible and, in rare cases, progression to ductopenia with development of a vanishing bile duct syndrome occurs. A differential diagnosis with other causes of Chronic liver disease is needed. Materials and Methods: From January 1991 through Jaunary 2000, 14 patients diagnosed as cholestatic hepatitis by liver biopsy were inclouded. The possible causative drug, clinical features, laboatory findings, and progression of cholestatic hepatitis were evaluated. The semiquantitative study of liver lesions was performed by two independent observers. Results: Causes of cholestatic hepatitis are 5 cases of oriental medicine, 3 cases of anti-tuberculosis medication, 1 case of ticlopidine and antibiotics and 4 cases of unknown causes. The clinical features of cholestatic hepatitis were jaundice, itching, urine color change, and general weakness. During 6 to 30 months, LFT of 5 patients showed prolonged elevation. Elevated total cholesterol ≥250 mg/dL in 6 patients, pheripheral blood eosinophilia in 5 patients, auto-antibody positive in 6 patients were observed respectively. The biopsies showed intralobular bilirubinostasis with a mixed portal inflammatory infiltration. Conclusion: In cholestatic hepatitis. durations of abnormal LFT are variable regardless of causative drugs. If cholestatic hepatitis progresses toward chronic course, viral hepatitis, primary biliary cirrhosis, and autoimmune hepatitis should be differentially diagnosed and sequential liver biopsies are needed.

      • 이락촌 폐흡충이 흰쥐 폐장의 비만세포에 미치는 영향

        In order to observe the changes in number of mast cell in lungs of albino rats against Paragonimus iloktsuenensis infection, rats of three experimental groups were infected with 20 metacercariae of Paragonimus iloktsuenensis, and they were kept for 3 days, 3 weeks and 4 weeks. Right after the each end of the infection period the lung tissues were fixed in 10% neutral formalin and embedded in paraffin. Their sections were stained with Hematoxylin and Eosin for general observation, and toluidine blue for mast cells. All the sections were examined under light microscope and the results obtained were as follows: 1. In the early stages of infection with P. iloktsuenensis there were noticeable changes in lung parenchyma and mast cell numbers in comparing with normal control. 2. After four weeks of infection the invasion of lungflukes into lung parenchyma was noticed. The lesions were represented by thick and fibrosclerotic cavern, granuloma due to eggs, and the increase of peribroncheal lymphatic tissue. A number of mast cell in this infection period were found in these lesions.

      • Isoproterenol이 심근에 미치는 영향에 대한 광학 및 전자현미경적 연구 : Light and Electron Microscopic Study

        For the study, twenty young rats of Wistar strain, weighing approximately 250gm, were sacrificed. For the experimental group, 8 ㎎ of isoproterenol HCI(1­/3',4'­dihydroxyphenyl/­2­isopropylaminoethanol hydrochloride) per 100gm of body weight was administered for the frequency of two times subcutaneously at intervals of 24 hours, and extraction of the hearts of the experimental animals were performed under ether anesthesia 24 hours after the second injection of isoproterenol. After extraction, the volume of each heart was measured by the Archimedes principle, and the wet weight of that was also measured. Paraffin sections of the heart tissue fixed with 10% neutral fromalin were stained with hematoxylin and eosin for routine study, and those fixed with absolute ethanol with von Kossa staining for histochemical study of calcium ion. Stained section were examined under light microscope. For electron microscopic study, the remainder part of the heart tissue was cut into one mm^3 in size, and was fixed with 2% glutaraldehyde in 0.2M phosphate buffer, pH7.4 and post-fixed with 1% osmium tetroxide in 0.2M phosphate buffer, pH7.4. After dehydration with ascending alcohols from 70% and resin infiltration, each tissue specimen was embedded into epon 812. Ultrathin sections were stianed with 4% uranyl acetate and lead citrate and were examined under JEOL200-CX transmission electron microscope. The results obtained were as follows: 1. There was a significant increase in the weight of heart of the experimental group. 2. The major site of injury was the left ventricle, especially the apex of the experimental group. 3. In the experimental group, there was widespread inflammatory cell infiltration, swelling of myocardial fibers and focal wavy appearance of them in the myocardium of the left ventricle including apex. 4. In the experimental group, there was widespread calcium ion deposit in the myocardium. 5. By electron microscopic study, there was loss glycogen granules, mitochondrial swelling, destruction of mitochondrial cristae, and infiltration of macrophages at the margin of inflummed area of myocardium of affected area.
