      • 신사구방과립세포의 형태학적 연구 : DCA가 부신적출 생쥐에 미치는 영향 Effects of DCA on the Adrenalectomized Mice

        The juxtaglomerular granular cells of the male white mice were investigated by light and electron-microscopies with the aim elucidating the morphological characteristics of the graunles and juxtaglomerular granular index (JGI) and granular cell index (GCI) were calculated under the various experimental conditions. The experimental groups were classified as the normal control, one-, two, and four-week-survived after adrenalectomy, and some of the each group were administered DCA in dose of 1㎎/㎏ body weight. For the light microscopical study, after the fixation in Zenker-formol solution the sections were stained by Wilson's method. And for the ultrastructural study, under the anesthesia of nembutal the kidney was fixed by purfusion method with 2.3% glutaraldehyde in 45mM cacodylate buffer following prerinzing with heparinated 0.1% Ringer-lidocaine solution. After the removal of the tissue, it was postfixed in 0.1% osmium tetroxide in 45mM cacodylate buffer by immersion, and the thin sections were stained with uranyl acetate and lead citrate. The results observed were as follows; 1. Adrenalectomy increased both JGI and GCI markedly, especially in 2-weeks­survived group after adrenalectomy. 2. Increment of JGI was chiefly due to the increase of Type 3 granular cells. 3. There wasn't any significant decrease in both GCI and JGI following administration of DCA to normal animals. 4. By the administration of DCA during the survival period after adrenalectomy, JGI and GCI showed slight decrease but not expected to return to normal value. 5. In the juxtaglomerular granular cells in normal control, well-developed rough surfaced endoplasmic reticulum and round secretory grarnules were found. 6. In the adrenalectomized group, secretory granules showed amorphous and coalesced large granules, and the prototype of secretory granules are found near the Golgi apparatus. 7. In one of the juxtaglomerular cells secretory glanules not surrounded with single membrane are found within the cisternae of rough surfaced endoplasmic reticulum. In the other cell, markedly developed rough surfaced endoplasmic reticulum filled whole cytoplasm. In this case, there were no secretory granules and only small-sized mitochondria were found between them. 8. It is difficult to discuss that the administration of DCA causes ultrastructural changes in juxtaglomerular granular cells.

      • 신 사구방과립세포에 대한 구적법적 연구

        신사구방 과립세포의 미세구조를 Loud(1962)의 구적법적 방법을 사용하여 관찰하여 다음과 같은 결론을 얻었다. 1. 신사구방 과립세포의 핵이 차지하는 영역은 41.27%, 사립체가 차지하고 있는 것은 2.20%, 그리고 분비과립은 23.65%를 차지하고 있었다. 2. Membrane space는 형질내세망, glycogen, ribosome 등이 위치하는 장소로써 이것은 동세포의 31.48%를 점유하고 있었다. 이 결과는 환경조건에 따라 반응하는 신사구방 과립세포의 미세구조를 정량적으로 표시할 수 있는 방법이라고 인정된다. In the study of juxtaglomerular granular cell, the stereomorphometric method was applied and brief discussion was presented comparing with juxtaglomerular granular cell index and juxtaglomerular granular index. 1. The area percentage of the nucleus of juxtaglomerular granular cell was 41.27%, the mitochondria, 2.20% and the secretory granules, 23.65%, respectively. 2. The membrane space consisting of endoplasmic reticulum, glycogen granules, and ribosome occupied the area of 31.4% Above results show that the fractions of the formed elements might be changeable from each other and this stereomorphometric method may be applicable to the quantitative study of the ultrastructures of juxtaglomerular granular cell responding with environmental factors.

      • Ethanol이 신사구방 과립세포에 미치는 영향

        생쥐의 복강을 통하여 ethanol을 투여하여 이에 반응하는 신사구방과립세포의 변화를 현미경하에서 과립세포지수 및 과립지수를 조사하여 관찰하고자 시도하였다. 조직표본은 Smith씨 액에 고정하고 Bowie씨 법의 Smith씨 변법을 사용하여 염색, 제작하였으며 400배의 배율하에서 경검하였다. 그 결과를 요약하면 다음과 같다. 1. 체중 1kg당 3gm을 주여한 군의 과립지수는 시간의 경과에 따라 계속 증가하였으며 체중 1kg당 9gm을 투여한 군의 과립지수는 투여후 30분에 현저히 증가하고 투여후 1시간에도 게속 증가상태를 유지하고 있었다. 2. 체중 1kg당 3gm을 투여한 군의 과립세포지수는 시간의 경과에 /다라 완만한 증가를 보이고 체중 1kg당 9gm을 투여한 군에서는 과립지수 처럼 현저하지는 않아도 증가된 상태를 보여주었다. 3. 각 군에서 출현하는 세포형의 출현빈도는 제Ⅰ형 세포가 제일 높았으며 제Ⅱ형 세포가 그 다음으로 높고 제Ⅲ형, 제Ⅳ형 세포는 극히 소수에 지나지 않는다. 이상의 실험결과로 미루어 보아 과립지수의 증가 현상은 신사구방치내의 다른 세포에서 새로히 상피앙세포화하여 일어난다고 생각할 수 있다고 본다. The relationship between ethyl alcohol, injected intraperitoneally, and juxtaglomerular granular cell was studied by the calculation of granular cell index and juxtaglomerular granular index under light microscope. The histological slides were prepared according to Smith's modification of Bowie's method after fixing in Smith's fluid. The results obtained are as follows: 1. The granular cell indices of the experimental groups which were injected with ethyl alcohol in doses of 3 and 9gm/㎏ body weight were both increased. In case of 3 gm-injected group shows slightly increasing tendency in proportion to time sequence. 2. The juxtaglomerular index of 3 gm-injected group was increased proportionately up to three hours after administration, and that of 9 gm-injected group showed the abrupt increase until one hour later. 3. The most frequently appearing cell type is type Ⅰ cell, and type Ⅱ cell the next. Type Ⅲ and Ⅳ cells were appeared in a very few number. The above results showed that the increase in juxtaglomerular granular index might be caused by the metamorphosis of the other cell types in the juxtaglomerular apparatus into juxtaglomerular granular cells.

        Meprobamate Chlorpromazine 및 電擊療法이 白鼠의 Estrous Cycle에 미치는 影響

        The cyclical activity of the Sprague-Dawley strain rat was observed under the various conditions of administration of meprobamate, chlorpromazine and EST, using the vaginal smear technique. The experimental groups were as followings; GroupⅠ Normal control rats GroupⅡ Rats administered with meprobamate in doses of 60㎎/㎏/day GroupⅢ Rats administered with meprobamate in doses of 400㎎/㎏/day GroupⅣ Rats administered with meprobamate in doses of 400㎎/㎏/day and chlorpromazine, 20㎎/㎏/day GroupⅤ Rats administered with meprobamate in doses of 60㎎/㎏/day and chlorpromazine, 20㎎/㎏/day GroupⅥ Rats administered with meprobamate in doses of 60㎎/㎏/day and chlorpromazine, 5㎎/㎏/day GroupⅦ Rats administered with meprobamate in doses of 60-400㎎/㎏/day combined with EST. In Group Ⅱ and Ⅲ, the average length of the estrous cycle was 9.64 and 10.02 days, which revealed the significant prolongation in comparison with 4.9 days in the normals. Group Ⅳ and Ⅴ showed complete cessation of estrous state of the cycle. Group Ⅵ had irregular cyclical activity, showing 14.4 days interval of the cycle. Group Ⅶ had complete cessation of estrous stage of the cycle immediately after EST. It seems significant that meprobamate has an effect on the cyclical activity of the estrous cycle, which is increased if combined with chlorpromazine or EST.

      • 마우스 고환의 간질세포에 관한 전자현미경적 관찰

        For the study, seven normal adult ICR mice were used as the experimental animals. Two days prior to sacrifice, each animal was treated with 3% trypan blue and the testes were removed under ether anesthesia. Each testis was immersed into 3% glutaraldehyde in 0.1 M collidine buffer, pH 7.4, for 2 hours under cold environment, and then, was sliced into about one cubic mm. The sliced tissue was re-fixed with the same fixative for an hour, and was post-fixed with ferro-cyanide-reduced 1% osmium tetroxide, pH 7.4, for an hour. And after routine treatment, each specimen was embedded into Epon 812 and 2 μm-thick semithin-sections were stained with 'Richardson's stain for identifcation of testicular interstitial cells. Ultrathin sections were stained with lead citrate and uranyl acetate and were examined under the JEOL 200CX transmission electron microscope. As a result, the most outstanding feature of the ultrastructure of the testicular interstitial cell was an abundant agranular endoplasmic reticulum (smooth ER). In some of the examined cells, smooth ER was observed as a concentric lamellar structure and this was interconnected with vesicular smooth ER. Other features were abundant mitochondria, lipid droplets, granular endoplasmic reticulum(rough ER), free ribosomes, and microbodies. The plasma membrane of the testicular interstitial cell was not customary trilaminar in structure but a dense lint, and in some of the examined cells, it has villous projections and interdigitations. Nucleus of the testicular interstitial cell was mostly oval in shape and the chromatin was condensed near the nuclear envelope. Nucleoli were inconspicuous.

      • 부신적출 흰 생쥐의 사구체방세포에 대한 구적법적 연구

        In the adrenalectomized mice, the morphological characteristics of ultrastructure of the juxtaglomerular cells were investigated by the application of the stereomorphometry introduced by Loud(1962) with the aim elucidating its functional changes. The kidney was prefixed by perfusion with 2.3% glutaraldehyde in 45 mM cacodylate buffer, pH7.4, and its specimens were postfixed in cold 1% osmium tetroxide in 45 mM cacodylate buffer, pH 7.4, by immersion. Their thin sections were contrasted with uranyl acetate and lead citrate, and their electron micrographs were taken for the stereomorphometrical study. The results observed were as follows: 1. The fractions of the mitochondria, secretary granles, and membrane space of the normal juxtaglomerular cell show 3.41%, 15.61% and 61.78%, respectively. 2. The fractions of the mitochondria, secretary granules, and membrane space of juxtaglomerular cell in the adrenalectomized mouse are 2.59%, 35.98% and 43.90% respectively. There is no evidence in the change of mitochondrial fraction, but the secretary granules show increase in fraction and that of the membrane space decreases in comparison with normal mouse. 3. In the juxtaglomerular cells of the normal mouse, well developed granular endoplasmic reticulae and matured, round secretary granules with relatively unifrom density are seen in their cytoplasm. On the contrary, the cells of the adrenalectomized mouse contain immatured secretary granules with varying density as well as matured ones with round and irregular contours, and well developed granular endoplasmic reticulae show distended cisternae.

      • Fixation of Heart by Modified Vascular Perfusion

        Langendorff(1895)및 Palay등 (1962)이 고안한 현관관류를 modify 한 방법으로 관류장치를 만들어 2.5%glutaraldehyde로 관류고정한 흰토끼의 심실심근세포와 통상의 침적고정법으로 고정한 같은 세포를 투과전자현미경으로 관찰하고 그 미세구조의 해상도를 서로 비교한 결과, 관류고정으로 처리한 심근세포에서 미세구조가 보다 뚜렷하였으므로 관류장치의 도면과 함께 보고하는 바이다. The rabbit hearts were fixed with 2.5% glutaraldehyde by a modified method of vascular perfusion to compare the ultrastructure of myocardial cells to thase of immersion-fixed hearts. The oerfusion-fixed hearts showed more clearly resolved ultrastructures.
