        유체에 의해 유발된 전단력이 치은 섬유아세포 유전자 발현 변화에 미치는 영향에 관한 연구

        The oral cavity is humid environment mainly due to the continuous salivary flow. The reaction of oral mucosa to fluid flow is important for homeostasis and pathogenesis. The objective of this study is the screening the change of gene expression after the application of fluid induced shear stress (FISS) on the gingival fibroblast using cDNA microarray assay. The immortalized human gingival fibroblasts were grown and FISS was applied using a cone viscometer at a rotational velocity of 40 rpm, respectively for periods of 2 and 4 hours. The synthesis of cDNA was done from the extracted total RNA and cDNA microarray assay was done subsequently. The genes that showed over 1.6 in the Cy3/Cy5 or the Cy5/Cy3 value were regarded as genes influenced significantly by the FISS application ion (/M/>0.7). The " RUNX-1" was increased its expression in 2 hours group and " RUN and SH3 domain containing 1" was increased its expression in 4 hours group. The "CC020415", "cyclin L1", "interferon regulatory factor1", "early growth response 1", "immediate early response 2", and "immediate early response 3" genes were increased their expression in 2 and 4 hours after FISS application. In conclusion, we could find many genes that were probably related to the FISS application. Interestingly, most of them were placed in similar molecular pathways and these findings improve the reliability of chip data and usefulness in overall screening. From this experiment, we could find many items for further study and it will make improvement in the understanding of intracellular events in response to FISS.

        녹용약침(鹿茸藥鍼)이 골형성(骨形成)에 미치는 영향(影響)

        Objective : Bone homeostasis is maintained by balance of bone formation and resorption. Therefore, bone related diseases arose by disturbance of this balance between osteoblast and osteoclast activities. To develop a successful screening system of the therapeutic components based on oriental medicine is essential to set out systematic approach for that purpose. Methods : This study was perforated Sprague-Dawley rat femur for bony defects(${\phi}5mm$) by the fissure bur. And experimetal group were treated with Cervi Pantotricuhum Cornu injection at both $Sh{\grave{e}}nsh{\tilde{u}}$(BL23) & $D{\grave{a}}zh{\grave{u}}(BL11)(0.2m{\ell})$. This was evaluated by radiography and histological analysis with in situ hybridization. Results : Cervi Pantotricuhum Cornu Herbal Acupuncture has weak effect on bony defect healing and this was evaluated by X-ray taking and histological analysis with in situ hybridization. Osteocalcin gene expression was not changed by Cervi Pantotricuhum Cornu Herbal Acupuncture in bony defects animal model. Conclusion : Taken together this study show that the Cervi Pantotricuhum Cornu herbal-acupuncture has a weak effect healing of bony defects, this type of approach might give a good chance to explore the favorable effects of Cervi Pantotricuhum Cornu herbal-acupuncture on bone tissue.

        전단력이 연골세포에 미치는 영향에 관한 연구

        The objective of this study is screening the shear stress related proteins in chondrocytes using twodimensional electrophresis and MALDI-TOF. C-28/I2 cell line were grown. The fluid-induced shear stress(FISS) was applied using a cone viscometer at a rotational velocity of 80rpm for periods of 12 hours. Control cultures were tested under identical conditions without mechanical load application. Collected samples were used for the two-dimensional electrophoresis and MALDI-TOF. The identified proteins were calcyclin, RPE-spondin, interleukin-2, extracellular signal regulated kinase (ERK), lamin B2, porA protein, and RET-ELE1 protein. All of them showed a decreased expression. In conclusion, seven proteins were identified as a shear stress related proteins in chondrocytes. As the destruction of articular cartilage is one of main pathogenesis of TMJ internal derangement, this study will give useful information for the understanding of the molecular aspect of TMJ disease.

        Raine 증후군 1례

        Raine 증후군은 안구돌출, 함몰된 코와 후비공 폐쇄 및 구개열과 하악골 발육부전 등의 특징적인 안면기형과 좁은 흉곽구조와 폐 발육 부전으로 인한 호흡부전, 전신 골격의 골경화, 뇌실질의 석회화 등을 특징으로 하는 매우 희귀하고 치명적인 질환이다. 아직까지 유전적인 결함부위는 알려져 있지 않은 상태이며 근친간인 부모 사이에서 발병이 많으며 남녀비의 차이가 별로 없으며 같은 형제 중에서도 이환된다는 사실로 보아 상염색체 열성으로 유전되는 질환이라는 것을 알 수 있고, 뇌 조직 검사상 저산소성 병변이나 괴사, 출혈, 감염이나 염증 등에 의한 2차적인 석회화가 아닌 칼슘 대사 이상에 의한 전신 골격의 골경화 와 두개 내의 석회화가 초래되는 질환으로 추정되고 있다. 본원에서는 상기한 특징적인 기형과 임상경과를 나타내는 매우 드문 증례를 경험하였기에 문헌 고찰과 함께 보고하는 바이다. Raine syndrome was described as an unknown syndrome in 1989. It is characterized by severe craniofacial anomalies with microcephaly, hypoplastic nose, depressed nasal bridge, exophthamos/protosis, gum hypertrophy, cleft palate, low-set ears, small mandible, narrow chest, wide cranial sutures and choanal atresia or stenosis, by generalized osteosclerosis with subperiosteal thickening of ribs, clavicles and diaphysis of long bones, and by intracranial calcifications in the particularly periventricular area. It undergoes an autosomal recessive inheritance. Twelve cases of Raine syndrome have been reported in the literature. However, a case of Raine syndrome in Korea has not been reported yet. Therefore, we describe a female newborn with Raine syndrome with a brief review of the literatures.

        녹용약침액(鹿茸藥鍼液)의 DNA chip을 이용(利用)한 유전자(遺傳子) 발현(發顯) 분석(分析)

Methods : Cervi Pantotricuhum Cornu Herbal-acupuncture extract was prepared by boiling. human osteosarcoma cells(HOS) were treated with Cervi Pantotricuhum Cornu Herbal-acupuncture solution. Then mRNA was extracted and cDNA microarray assay was performed. Results : Human osteosarcoma cells(HOS) treated with Cervi Pantotricuhum Cornu Herbal-acupuncture solution($500{\mu}g/m{\ell}$) showed that thioredoxin, TAFII31 and two novel genes were increased. However many genes decreased their expression by Cervi Pantotricuhum Cornu Herbal-acupuncture. Conclusions : This type of approach will give a good chance to explore the favorable effects of Cervi Pantotricuhum Cornu Herbal-acupuncture. Further study is needed for investigating the effect of Cervi Pantotricuhum Cornu Herbal-acupuncture.

        A squamous odontogenic tumor (SOT) is rare disease and it is believed to originate from epithelial rests of Malassez of the periodontal membrane. Neither sex nor site predilection in either jaw has been established. Some lesion can be shown in juxtaposition in tooth roots. Although most lesions remain smaller than 2 cm, our cases involved a half of left mandibular ramus. The exact pathogenesis is still unknown. We report a case of SOT including the results of immunohistochemical study of pancytokeratin and p53.

        Interferon-γ가 치주인대세포의 Collagen 및 Fibronectin의 합성과 Alkaline Phosphatase 활성에 미치는 영향

        Interferon-γ has been suggested as a cytokine of connective tissue stabilizer. In addition, it has also been demonstrated that this cytokine inhibited bone remodeling activities of the bone derived cells. In order to illuminate the effects of this cytokine in orthodontic force induced bone remodeling, it was administered to primary cultured periodontal ligament cells which have been known to have some osteoblast like characteristics. Interferon-γ slightly decreased [<sup>³</sup>H]thymidine incorporation rate without a significant change in the total cellular DNA content up to 1000 U/ml, which meant these doses were not cytotoxic to the cell. Total protein synthesis was not influenced by various concentration of interferon-γ whether it was determined by the [<sup>³</sup>H]proline incorporation rate or by the Lowry smethod. The effect of interferon-γ on the individual protein was, however, differential, ie, it increased [<sup>³</sup>H]proline incorporation into the noncollagenous protein marginally, while it decreased [<sup>³</sup>H]proline incorporation into the collagen, so that it caused dose-dependent suppression of the relative collagen synthesis. On the contrary, the fibronectin synthesis determined by the ELISA was increased by 1000 U/ml of interferon-γ. The differential effects of the interferon-γ on the collagen and fibronectin synthesis exhibited not only their protein level but also the steady state mRNA level. Interferon-γ decreased steady state level of α1(I) procollagen mRNA significantly, while showing no significant changes in the fibronectin mRNA level. In addition to this, it was also found that indomethacin did not affect on the interferon-γ induced collagen decrease in this cell, which meant prostaglandins were not involved in the process of interferon-γ induced collagen decrease. So it can be concluded that the incubation of periodontal ligament cells with 1000 U/ml of interferon-γ for 24 hr showed differential effects on the type I collagen and fibronectin gene expression. The decrease in relative collagen synthesis in the protein level was related with decrease in the steady state level of mRNA, while the increase in the fibronectin synthesis in the protein level was not correlated with the mRNA level.

        인공고관절 전치환술 후 발생한 무균성 해리에 대한 젤라틴/제4형 콜라젠 분해효소의 역할

        Matrix metalloproteinases(MMPs) are an important group of zinc-dependent enzymes responsible for the degradation of extracellular matrix(ECM) components such as collagen and proteoglycan in normal embryogenesis and in pathologic conditions including rheumatoid arthritis and periodontitis. The purpose of this study is to demonstrate and characterize the gelatinase/type IV collagenase activity in the interface membranes between bone and loose acetabular components by zymographic analysis, and to understand it's role in the cascade of ECM degradation. GelatinasefFype IV collagenase, 72 kDa MMP-2 type and 92 kDa MMP-9 type were analyzed in 5 cases of Miiller type polyethylene acetabular components that had been revised for aseptic loosening in 5 patients. Zymographic analysis revealed elevated production of MMP-2 and induction of MMP-9 in tissue from the interface membrane between bone and implants. These findings suggest that gelatinase/type IV collagenases may have an important role in the degradation of ECM of periprosthetic tissues, subsequent weakening of the connective tissue bed, and loosening of the prosthetic components.

        DICAM에 의한 LPS-매개 대식세포 활성화 억제

        DICAM, a dual Ig domain containing adhesion molecule, is involved in cell-cell adhesion through direct interaction with αvβ3 integrin. In our previous study showing the inhibitory role of DICAM in osteoclast differentiation, we found that DICAM also has a suppressive role in macrophage, the precursor cell of osteoclast, The role of DICAM in macrophage activation at the inflammatory milieu, however, remains obscure, Methods, Expression pattern of DICAM by inflammatory cytokines and lipopolysaccharide (LPS) was studied with RAW264.7, a murine macrophage cell line, To study the role of DICAM on macrophage activation, we stably transduced DICAM, or empty vector, into RAW264.7, and then compared the LPS-mediated activation such as spreading and TNF-a production, Results, DICAM was abundantly expressed in the synovial tissue of collagen-induced arthritis. When we assessed the expression of DICAM in RAW264.7 cells by mediators of inflammation, inflammatory cytokines, such as TNF-α, IL-1β, and IFN-γ, and M-CSF increased the expression of DICAM; however, LPS decreased. Functionally, DICAM that stably transduced-RAW264.7 cells showed attenuation of LPS-mediated macrophage activation including spreading and TNF-α production. DICAM decreased the phosphorylation of JNK MAP kinase by M-CSF and LPS stimulation, which was corroborated by a decrease in the expression of ITAM-associated receptors including Trem2, Pira1, and Oscar. Finally, a recombinant ectodomain of DICAM suppressed LPS-induced activation of RAW264.7 cells, Conclusion. These results indicate that DICAM acts as a negative regulator of LPS-mediated macrophage activation.
