다양한 PCR용 DNA 추출법에 의한 패류 내 Megalocytivirus의 검출

In analysis of DNA viruses from the contaminated shellfish using PCR, preparation method of template DNA is an important factor to get enough copy number of viruses. In this study, we evaluated the efficiency of PCR template of Megalocytivirus (sT50mg-D) DNA obtained from 50 mg digestive gland homogenate of oyster using commercial method, and compared with that obtained from 5 g of the same tissues (T5g-D) after PEG precipitation procedures of virus. Both templates DNA suspended in the same volume of distilled water showed positive results by primary PCR with 35 cycles, and the presence of Megalocytivirus was confirmed in oysters collected from cultured farms in Korea. Moreover, PCR with sT50mg-D allowed us to discriminate the contaminated oyster individually, that can not be done in PCR with T5g-D prepared from the mixture of three different individual oyster to get 5 g digestive gland homogenate. In quantitative analysis with real time PCR, Megalocytivirus concentrations in 50 ㎕ templates prepared using 0.5~50 mg of one positive sample were appeared in the range 6.14E+00~1.2E+02/㎕. We were not able to get positive result using template DNA contained less than 6.14E+00 copies. Consequently, 2-step PCR performed with DNA extracts from oyster homogenate of small amount (sT50mg-D) i) was enough to detect the contaminated Megalocytivirus in shellfish, ii) allowed us to do the analysis for individual shellfish rather than mixture of several shellfish and iii) showed the presence of Megalocytivirus in oyster from Korea.

Objective: Ovarian cancer represents a relatively chemosensitive solid tumor, with responsiveness to a range of agents. Cisplatin is the mainstay of drug treatment and is one of the most active single agent. However, the overall outcome for patients remains unsatisfactory and the emergence of drug resistance is a major factor in treatment failure. Loss of DNA mismatch repair is a common finding in many types of sporadic cancer as well as in patients with hereditary nonpolyposis colon cancer. Cells that lack DNA mismatch repair are resistant to commonly used chemotherapeutic agents. Selection of cells for resistance to cisplatin, a well-recognized mutagen, could result in mutation in genes involved in DNA mismatch repair. Methods: This study evaluated the mutation of hMLH1 and hMSH2, and its relation to the Taxol and Topotecan chemosensitivity in the clones from the ovarian cancer cell line 2008 and cisplatin-resistant cell line 2008/ C13*5.25. Results: 1. Cells from 2008 and 2008/C13*5.25 expressed both hMLH1 and hMSH2 when analysed with immunoblotting. 2. Twenty two out of 100 single-cell clones from 2008 and 27 of clones from 2008/C13*5.25 expressed no hMLH1. hMSH2 was expressed in all clones. 3. There was no difference of Taxol chemosensitivity between 2008 and 2008/C13*5.25 cell lines. In the 2008/C13*5.25 cell line, the hMLH1-deficient clones were more sensitive to Taxol than the hMLH1-proficient clones(P=0.049), but in 2008 cell lines hMLH1-proficient clones were more sesitive to Taxol(P=0.003). 4. There was no difference in Topotecan chemosensitivity between 2008 and 2008/C13*5.25 cell lines. In the 2008/C13*5.25 cell line, the hMLH1- deficient clones were not more sensitive to Topotecan than the hMLH1-proficient clones. In the 2008 cell lines hMLH1-deficient clones were more sesitive to Topotecan(P=0.001). Overall, hMLH1-deficient clones from both 2008 and 2008/C13*5.25 cell lines were significantly more sensitive to Topotecan(P=0.001). 5. Microsatellite instability was not demonstrated in all 4 types of single-cell clones from 2008 and 2008/C13*5.25 cell lines. Conclusions: The present results indicate that there is no relation between mutation of mismatch repair gene and cisplatin resistance. But hMLH1-deficient ovarian cancer cells are more sensitive to Taxol or Topotecan in this study. The latter finding mandates the examination to assess the mutation of hMLH1 in tumor cells before treatment or at the time clinical resistance to cisplatin develops in ovarian cancer.

To investigate the clinical aspects of CHT in atopic dermatitis (AD) treatments, the effect of CHT in anti-oxidative and anti-inflammatory cytokines were tested. 100% or higher cell viability was observed in all tested groups from 25 to 200 ㎍/㎖using Raw 264.7 cells. CHT showed dose-dependent DPPH scavenging activity, with more than 90% scavenging activities at 800 ㎍/㎖concentrations. CHT showed dose-dependent suppression activity of ROS production, especially at 200 ㎍/㎖of 37.5%. CHT decreased NO production activity, with significant decrease of 33.2% at 200 ㎍/㎖. IL-6, MCP-1, TNF-α production rate were decreased by approximately 25% when Raw 264.7 cells were treated with LPS and with CHT of 200 ㎍/㎖. Also, IL-1β production rate was decreased by 25% at 100 ㎍/㎖. The results above indicate that CHT significantly reduces the effect of oxidative and inflammatory cytokines. The use of CHT in dermatitis can be widely suggested.

Recently, as global concerns regarding the environment have surged, changes in material-joining technologies have transpired. Friction stir welding, which is much more environmentally friendly than the existing welding methods, is attracting attention as a new alternative. This paper presents a method for predicting the stiffness using computational analysis when designing a spindle for frictional stir welding by applying air bearings rather than conventional ball bearings. Through fluid analysis, the pressure exerted on the spindle by the air entering through the inlet is calculated. and calculate the 0.005 ㎜. Additionally, the pressure in an eccentric state is calculated. The pressure is converted to stiffness using a formula, the corresponding stiffness is applied to the spindle, and the radial load generated during friction is input at the end of the spindle. Finally, a structural analysis is conducted to determine whether the stiffness is sufficient to determine the feasibility of the design.

블로그 공간은 대표적인 온라인 사회 연결망 중 하나이다. 사회 연결망 분석에서 커뮤니티 구조 파악은 중요한 연구이다. 본 논문에서는 블로그 공간의 분석에 대하여 논한다. 본 논문에서는 실제 블로그 공간을 행렬(matrix)과 텐서(tensor)로 표현하고 SVD(singular value decomposition)와 PARAFAC(parallel factor analysis) 분해를 이용하여 분석한다. 분석 결과에 따르면, SVD는 특정 주제를 나타내는 커뮤니티들을 식별했으며, 각 커뮤니티에서 영향력 있는 블로그들과 권위 있는 포스트들을 식별했다. PARAFAC 분해는 SVD에 비해 더 많은 커뮤니티들을 식별하였다. 또한, PARAFAC 분해는 각 커뮤니티의 영향력 있는 블로그들과 권위 있는 포스트들뿐만 아니라 영향력 있는 키워드들까지 식별할 수 있었다. A blogosphere is a representative example of online social networks. Identifying a community structure is an important issue in social network analysis. In this paper, we address an analysis of a blogosphere. We model a real-world blogosphere as a matrix and a tensor, and then analyze it by using the SVD and PARAFAC decomposition. According to the results, the SVD successfully identified communities, each of which focuses on a specific topic, and found hub blogs and authoritative posts within each community. The PARAFAC decomposition also succeeded in extracting more communities of finer granules than the SVD. In addition, the PARAFAC decomposition could identify the dominant keywords in addition to the hub blogs and authoritative posts honored in each community.
난소암 세포주에서 Mullerian Inhibiting Substance의 증식 억제 효과

Objective: In order to explore Mullerian inhibiting substance (MIS) effects on the ovarian neoplasia, the expression and localization of the MIS type II receptor (MISR II), the growth inhibitory effects of MIS, and the underlying molecular mechanisms were investigated in the ovarian cancer cell lines. Methods: Expression of MISR Ⅱ were studied in SKOV-3, OVCAR-3, and OVCAR-8 cell lines by immunohistochemical staining. The antiproliferative effects of MIS in these cell lines were investigated by methylthiazoletetrazolium (MTT) assay, fluorescence-activated cell sorting (FACS) analysis, annexin-V-FITC binding, and western blot analysis. Results: All cell lines showed strong specific staining for MISR II, although staining in OVCAR-8 cells was more intense than that in SKOV-3 and OVCAR-3. Treatment of OVCAR-8 cells with MIS led to a dose- and time-dependent inhibition of cell growth and survival was determined use by MTT assay. But OVCAR-3 cells exhibited growth inhibition at higher doses after 48 hours of treatment and SKOV-3 cells did not demonstrate response. Using FACS analysis, exposure of OVCAR-8 cells to MIS (71 nM) resulted in G1 arrest after 24 hours of treatment. This pattern was changed by time-dependent increase in the percentage of cells with a sub G0G1 DNA content, suggesting apoptosis, after 48 hours of treatment. These results suggested that cell death be preceded by cell cycle arrest. Time-related induction of apoptosis was also observed in this cell line as measured by annexin-V-FITC binding. In OVCAR-8 cells, the growth inhibitory effects of MIS were mediated through specific induction of CDKI p16 protein expression and via regulation of E2F1 in the absence of detectable levels of pRb. We estimated that OVCAR-3 cells were affected by MIS through p16-independent, alternative mechanistic pathways, since the growth inhibitory effects of MIS were minimal. SKOV-3 cells did not express p16 protein. Conclusion: We have demonstrated that ovarian cancer cells express the MISR II. Epithelial ovarian cancer cells respond to MIS by growth inhibition. Although the precise mechanisms of MIS mediated inhibition of ovarian cancer cell growth have not been fully defined, these data suggest that MIS has activity against ovarian cancers in vitro and may also be an effective targeted therapy for ovarian cancer.

StuntBike is a robot that can stand up on its own using only the torque of the motor. Unlike previous works, StuntBike has the form of a tricycle with unstable characteristics on the pitch axis to achieve standing-up actions with a small torque. As a control scheme, we utilize a 3-stage cascade proportional-integral-derivative (PID) controller which outperforms a conventional 1-stage PID controller in terms of control performance. Based on the proposed design, we demonstrated the feasibility of standing-up action and attitude control from the simulation. Next, we present the control result from experiments, which suggests that stand-up action and attitude control is possible. Furthermore, we discuss some factors that impact the performances of the system and an overview of supplementary work.
