        광합성세균 Rhodopseudomonas gelatinosa 의 시토크롬 c 산화효소의 정제 및 특성

        화학 영양성으로 배양한 Rps. gelatinosa 에서 2회의 시토크롬 c 친화성 크로마토그래피와 DEAE-Sephacel 이온 교환 크로마토그래피 등 3 단계의 크로마토그래피를 수행하여 시토로콤 c 산화효소를 정제하였다. 정제된 시토크롬 c 산화효소는 Sephacryl S-300 에 의한 분자걍이 약 110,000 Da 이고 SDS-gel 전기영동에 의한 분자량이 약 52.000 Da 으로써 이량체일 것으로 보인다. 전제된 시토크롬 c 산화효소는 온도데 매우 불안정하고 말 심장 시토크롬 c 를 기질로 사용했을때 Km 값은 $20\mu$M, Vmax 값은 44unit/mg prot. 이며 pH 6.4 의 효소방응 최적 pH 와 25.deg.C 의 최적 온도를 보였다. 환원된 시토크롬 c 산화효소는 554, 523, 421 nm 에서 .alpha., .betha. soret 흡수대를 보였고 chromatophore 에서와 마찬가지로 KCN 과 $NaN_{3}$ 에 의해서는 효소 활성도가 저해를 받았지만 CO 와 antimycin A, myxothiazol 에 의해서는 효소 활성도가 저해를 받지 않았다. 빛을 에너지원으로 배양하거나 또는 화학영양성으로 배양하든지 모두 시토크롬 c-551 이 생성되었고 환원된 시토크롬 c-551 은 시토크롬 c 산화효소에 의해 산화되었다. 시토크롬 c-551 을 기질고 이용하였을 때 시토크롬 c 산화효소의 Km 값은 $26\mu$M 이었고 Vmax 값은 31.unit./mg prot. 로써 말심장의 시토크롬 c 를 기질로 이용할때 보다 오히려 낮았다. 이와 같은 결과로 보아 화학 영양성은 배양한 Rhodopseudomonas gelatinosa 에서 호흡에 의한 전자전달은 시토크롬 c-551 이 시토크롬 $bc_{1}$ 복합체로 부터 전자를 받아 b-형 시토크롬 c 산화효소에 전자를 전달해 주고 최정적으로 산소를 환원시킬 것으로 생각된다. Cytochrome c oxida5e from chemotrophically grown R p , geliitinosu was purified by cytochrome c affinity chromatography and DEAE-Sephacel ion exchange chromatography. The molecular weight of the cytochrome c oxidase was approximately 110.000 Da by sephacryl s-300 gel chromatography and approximately 52, 000 Da by SDS-gel electrophoresis, respectively. Therefore. cytochrolne c oxidase of Rps. gehtinosu seems to be dimer. The cytochrome c oxidasc was very sensitive to temperature. It's Km and Vmax were 20 pM and 44 unitlmg protein for horsc heart cytochrome c as a substrate. respectively, and its optimum pH and temperature were 6.4 and 25$^{\circ}$C. respectively. The absorption peaks of the reduced cytochrome c oxidase showed at 554 nm, 523 nm. and 422 nm. The activiiy of cytochrome c oxidase was inhibited by KCN, and NaN3, but not by CO, antimycir~ A. and myxothiazol. The cytochrome c-551 was produced either in phototrophically or chemotrophically grown Rps. gelaiinosci. The rcduced cytochrome c-551 was oxidized by b-type cytochrome c oxidase from Rp.v. gc.lrtino.sc~. Km and Vmax of cytochrome c oxidase was 26 pM and 31 unitlnlg protein For cytochrome c-551 as a substrate. respectively. Thercfore. thc electron transfer chain of chemotrophically grown Rps. glatinosa seems lo be ubiquinol cytochrome bc, complex -'cytochrome c-55lMb-type cytochrome c oxidase+02.

        Rhodopseudomonas gelatinosa의 chromatophore에서 시토크롬 $bc_1$ 복합체와 시토크롬 c 산화효소의 확인 및 특성연구

        The chromatophore from the chemotrophically grown facultative anaerobic photosynthetic bacterium, Rhodopseudomonas gelatinosa ATCC 17013 was isolated through stepwise sucrose gradient centrifugation. The isolated chromatophore showed high activities of the cytochrome $bc_{1}$ complex and cytochrome c oxidase. The activity of cytochrome $bc_{1}$ complex was completely inhibited by .5$\mu$M antimycin A,10$\mu$M myxothiazol, and that of cytochrome c oxidase was completely inhibited by .$50\mu$M KCM and $100\mu$M $NaN_{3}$but not inhibited by carbon monoxie. The activity of cytochrome c oxidase of th chromatophore was increased by addition of ionophores or protonophores. The reduced-oxidised difference sspectrum of cytochrome $bc_{1}$ complex isolated by affivity chromatography showed the absorption maxima at 553 nm(shoulder at 547 nm), 520 nm, and 418.5 nm, on the other hand, that of cytochrome c oxidase showed .alpha., .betha. and soret peaks at 554 nm, 523 nm, and 421 nm, respectively. The cytochrome c oxidase from chemotrophically grown Rhodopseudomonas gelatinosa seems to be a b-type cytochrome c oxidase.

        Anti-atherosclerotic Effect of the Methanol Extract of Sorbus commixta Cortex in the High Cholesterol-Diet Rats

        Hypercholesterolemia is a pivotal pathogenic factor for the development and maintenance of atherosclerosis. The present study was designed to evaluate whether the methanol extract of Sorbus commixta cortex (MSC) restores vascular dysfunction in association with the aortic expressions of proinflammatory and adhesion molecules in high cholesterol (HC) diet-rats. Chronic treatment with low (100 mg/kg/day) or high doses (200 mg/kg/day) of MSC lowered the increase in plasma levels of triglyceride (TG) and low-density lipoprotein (LDL) cholesterol induced by a cholesterol-enriched diet without affecting on the plasma level of high density lipoprotein (HDL)-cholesterol. Vascular tone attenuated in the HC-diet rats was restored by administration with MSC. Treatment with MSC also suppressed the HC-induced increase in the monocyte chemoattractant protein-1 (MCP-1) and nuclear factor-κB (NF-κB) p65 expressions as well as expressions levels of adhesion molecules including intercellular adhesion molecule-1 (ICAM-1), vascular cell adhesion molecule-1 (VCAM-1), and E-selectin in aorta. The present study also showed that MSC inhibited the HC-mediated induction of ET-1 and ACE expression. In histopathological examination, aortic segments in the HC-diet rat revealed thickening intima and media, which were blocked by administration with MSC. Taken together, MSC could suppress the development of atherosclerosis in the HC-diet rat model through the inhibition of the aortic expression levels of pro-inflammatory and adhesion molecules.

        심폐소생술후 발생한 전종격동 출혈 1례

        Anterior mediastinal hematoma is often reported as a complication of cardiopulmonary resuscitation (CPR). CPR can be performed as a result of myocardial infarction, and early percutaneous coronary intervention (PCI) and anticoagulant,antiplatelet agent can improve outcome. As use of antiplatelet agents, like glycoprotein IIb/IIIa inhibitors,becomes more widespread, occurrence of complications such as bleeding may be increased. The mediastinal hematoma usually resolves itself without complications;however, a large amount of hematoma can cause cardiac tamponade. Therefore, rapid diagnosis is very important. We describe a case of anterior mediastinal hematoma detected by echocardiography after CPR and PCI.

      • 한약을 이용한 당뇨성 혈관계 병변 억제제 개발 연구

        The final purpose of this study is the development of herbal medicine for diabetes mellitus-induced vascular diseases. Thus, present purpose is the selection of candidate extracts for treatment of diabetic anti -vascular diseases using medicinal plants traditionally used in the Korea. Throughout in vitro screening test, candidate extracts (simple or combination) against anti-vascular inflammatory activity were selected in primary cultured human umbilical vein endothelial cells (HUVECs). Now we confirm the effect of candidate extracts on high glucose-induced vascular inflammatory process and its related signal pathways in HUVECs. Summaries of results are as follows : 1. Experimental conditions by high glucose were established prior to the measurement of effect of candidate extracts. 2. Aqueous extract of Buddleia officinalis could suppress high glucose or TNF-a-induced vascular inflammatory process, which may be closely related with the inhibition of oxidative stress and NF -кB activation and in primary cultured HUVECs. 3. Aqueous Extract of Benincasa hispida Cogniaux inhibited high glucose-induced increase of rCAM-I, VCAM-I, and E-se1ectin as well as l\1MP-2/-9 expression level in HUVECs. 4. Aqueous Extract of Zanthoxylum Schinifolium, Lonicera japonica, Gastrodia elata, and Lycopus coreanus also inhibited vascular inflammation process. Thus, the potent materiels of anti -vascular process were confirmed using cell-based experiments in this study. The further study is necessary to confirm these effects of candidates using various cell or animal models and, in addition, develop drug for treatment of vascular disease associated with diabetes mellitus from these candidates.

        청소년 태권도지도자의 자율성지지와 선수들의 번아웃 및 스포츠 도덕행동의 관계

        The purpose of this study was to investigate the The relationship between support for autonomy of youth Taekwondo instructors, burnout and sports moral behavior of athletes. To this end, Taekwondo athletes registered by the Korea Taekwondo Association selected as a population, a survey was conducted on 280 youth Taekwondo athletes living in Gyeonggi, Daejeon, Jeonnam, and Gwangju for about six months from September 2020 to March 2021. Among them, 277 data were used for the final analysis after excluding the data of 3 athletes whose response information was omitted. As a result of this study, first, autonomy support had a negative effect (-) on burnout. Second, autonomy support had a positive effect (+) on team prosocial behavior, opponent prosocial behavior, and opponent antisocial behavior. Third, burnout had a negative effect (-) on opponent prosocial behavior, while had a positive effect (+) on team antisocial behavior and opponent antisocial behavior. Based on the above results, this study identified the relationship of perceived autonomy support, burnout, and Sport moral behavior.

        광합성 세균인 Rhodopseudomonas gelatinosa ATCC 17013에서 Cytochrome c-551의 정체

        The soluble cytochrome c-551 of photosynthetic bacterium, Rhodopseudomonas gelatinosa ATCC 17013 was purified through a sequene of four step chromatography including CM-cellulose ion-exchange chromatography, DEAE-Sephacel chromatography, Sephacryl s-200 gel permeation chromatography, and HPLC (SP-5PW). The molecular weight of the purified cytochrome c-551 was 14, 600 Da, and this protein shows the absorption peak at 551 nm, 522 nm, and 417 nm as the reduced form, and at 412 nm as the oxidized form. The cytochrome c-551 seems to be a substrate for the terminal oxidase in the electron transport chain.

        쪽파 에탄올 추출물이 果糖 誘導 고혈압 백서의 腎臟 기능에 미치는 영향

        The present study was aimed to investigate whether ethanol-extract of Allium wageki has an ameliorative effect on the renal function in high fructose-diet induced hypertensive rats. The urine osmolality (Uosmol) was decreased in rats with high fructose-diet (60%) during the whole experiment period without change of the urine volume (UV). The urinary excretion of sodium (UNaV) and chloride (UCIV) were decrease significantly in rats with fructose-induced hypertensive rats, whereas urinary excretion of potassium (UKV) was increased. The creatinine clearance (Ccr) and solute-free water reabsorption were also decreased by treatment of fructose-rich diet. Among these renal functional parameters, Ccr was partially restored by the administration of ethanol-extract of Allium wageki. The Uosmol was also partially restored by the administration ethanol-extract of Allium wageki at the end of the experimental period. Taken together, ethanol-extract of Allium wageki has the ameliorative effect on glomerular filtration rate in rats with high fructose-diet induced hypertension.
