        Paricalcitol attenuates lipopolysaccharide-induced inflammation and apoptosis in proximal tubular cells through the prostaglandin E<sub>2</sub> receptor EP4

        Background: Vitamin D is considered to exert a protective effect on various renal diseases but its underlying molecular mechanism remains poorly understood. This study aimed to determine whether paricalcitol attenuates inflammation and apoptosis during lipopolysaccharide (LPS)-induced renal proximal tubular cell injury through the prostaglandin E<sub>2</sub> (PGE<sub>2</sub>) receptor EP4. Methods: Human renal tubular epithelial (HK-2) cells were pretreated with paricalcitol (2 ng/mL) for 1 hour and exposed to LPS (1 μg/mL). The effects of paricalcitol pretreatment in relation to an EP4 blockade using AH-23848 or EP4 small interfering RNA (siRNA) were investigated. Results: The expression of cyclooxygenase-2, PGE<sub>2</sub>, and EP4 were significantly increased in LPS-exposed HK-2 cells treated with paricalcitol compared with cells exposed to LPS only. Paricalcitol prevented cell death induced by LPS exposure, and the cotreatment of AH-23848 or EP4 siRNA offset these cell-protective effects. The phosphorylation and nuclear translocation of p65 nuclear factor-kappaB (NF-κB) were decreased and the phosphorylation of Akt was increased in LPS-exposed cells with paricalcitol treatment. AH-23848 or EP4 siRNA inhibited the suppressive effects of paricalcitol on p65 NF-κB nuclear translocation and the activation of Akt. The production of proinflammatory cytokines and the number of terminal deoxynucleotidyl transferase-mediated dUTP nick end labeling-positive cells were attenuated by paricalcitol in LPS exposed HK-2 cells. The cotreatment with an EP4 antagonist abolished these anti-inflammatory and antiapoptotic effects. Conclusion: EP4 plays a pivotal role in anti-inflammatory and antiapoptotic effects through Akt and NF-κB signaling after paricalcitol pretreatment in LPS-induced renal proximal tubule cell injury.

      • 부부의 금전관리 태도와 행동이 재정관리일치 만족도에 미치는 영향

        본 논문은 남편과 부인 금전관리에 대한 태도와 행동, 이에 따른 부부의 일치도를 이해하고, 이 일치정도가 부부의 재정관리일치에 대한 만족도에 미치는 영향을 연구하였다. 연구의 결과, 남편과 부인의 금전관리 태도와 행동은 재정관리 일치수준과 긍정적인 상관관계가 있었다. 그러나 남편과 부인의 태도와 행동은 다른 변수들과 의미있는 상관관계를 보이지않았다. 또 남편과 부인의 재정관리일치에 대한 만족은 그들의 금전관리 태도와 행동 그리고 배우자와의 재정관리의 일치정도를 의미있게 설명하였다. The purpose of this study are to understand wives` and husband` attitudes and behaviors toward money management and to find effects of these on the satisfaction with the agreement of the household financial management. The data collection was processed between sept. 29 and oct. 12, 1992 from student` parents whose children were in middle and high school in Chunju, Iri, and Kunsan. Six hundred questionaires were distributed and 550 of them were analyzed(return rate 91.7%). Five hull hypotheses were specifiedand test and using the statistical software package `SAS` with the confidence levelp<.10. As result of this study, husbands` andwives` attitudes and behaviors toward money management were generally positively correlated to the agreement level of household financial management However those husbands and wives were not significantly correlated to each other. The satisfaction with the agreement of the household financial management of husbands and wives was significantly explained by their money management attitudes and behaviors and the agreement of the financial management between spouses.

        하악에 발생한 과잉치의 치험례 : A CASE REPORT

        과잉치는 정상치판(dental lamina)의 과도한 증식이 결과로 발생되어, 가족적 성향을 보이며, 쇄골두개이형성(Cleidocranial dysplasia), Gardner 증후군 등의 여러 질환과 관련되어 나타나기도 한다. 그 빈도는 유치열에서 0.3~0.8%, 영구치열에서 1.0~3.5% 이고, 남성에서 호발되며(2:1), 상악에서 더 많이 발생한다(9:1). 상악에서는 mesiodens라고 불리는 중절치 사이의 위치가 가장 흔한 부위이고, 그 다음은 fourth molar와 측절치 부위이다. 하악에서는 소구치 부위가 가장 흔한 부위이고 전치부위에서는 약 2%로 매우 드물게 발생한다. 과잉치의 만기 잔존은 치간 이개, 인접한 치아의 맹출 장애, 치근 흡수 등을 야기하고 과잉치와 관련되어 함치성 낭종이 발생될 수 있으므로 조기진단과 적절한 치료가 중요하다. 본 두 증례는 비교적 드물게 나타나는 하악에 발생한 과잉치로, 하악 전치부와 소구치부에 발생한 과잉치를 발거하고 교정치료를 시행하여 양호한 결과를 얻었기에 보고하는 바이다. Supernumerary tooth was resulted from excessive proliferation of dental lamina and associated with familial tendency and a congenital syndrome such as Cleidocranial dysostosis or Gardner's syndrome. Incidence reports identify a range of 0.3~0.8% in primary dentition. 1.0~3.5% in permanent dentistion with males being affected twice as frequently as females. maxilla nine times as frequently as mandible. The most common supernumeraty tooth is the mesiodens, which located between the maxillary central incisors, and the next common site is the fourth molar and lateral incisors. Supernumerary teeth are uncommon in the mendible, but premolars are the most common supernumerary teeth and occurrence is very rare in the incisor region of the mandible of possibility of diastema and cruption failure displacement. rotation of the associated permanent teeth, root resorption and dentigerous cyst with presence of the supernumerary teeth. In this two case, one supernumerary tooth located in the mandibular incisor region, the other supernumerary tooth located in premolar region. We could get normal alignment of mandibular dentition by extraction and orthodontic treatment.

        복잡 치아종으로 인한 매복치아의 교정적 견인

        치아종은 치성 종양 중 가장 흔한 형태로 증상이 없는 경우가 대부분이며 종종 영구치의 매복이나 맹출지연을 유발한다. 이는 치아를 구성하는 법랑질,상아질,백악질,치수로 구성되며,형태에 따라 복합 치아종과 복잡 치아종으로 구분 된다. 복합 치아종은 다수의 작고 치아와 유사한 형태를 지니며 상악 전치부에 호발하고,복잡 치아종은 무정형의 치성 조직으로 구성되어 있으며 전체 치아종의 약 25%를 차지하고 하악 구치부에 호발한다. 치아종의 병인은 확실치는 않으나 국소적인 외상,감염,유전적 요인이 관여할 것이라 추측된다. 치료는 보존적인 외과적 적출술이 추천되며 재발은 거의 없다. 다음의 두 증례는 각각 하악 제 l대구치와 하악 측절치의 맹출 지연을 주소로 본원에 내원한 환아들로,매복치 상방의 복잡 치아종을 외과적 적출술을 시행하고 매복치의 외과적 노출 및 교정적 견인을 시행하여 양호한 결과를 보였기에 보고하는 바이다. Odontomas are the common type of odontogenic tumors and generally are asymptomatic and frequently lead to impaction or delayed eruption of permanent teeth. They are composed of enamel, dentin, cementurn and pulp tissue and are divided into compound and complex according to the morphology of the hard tissues, Compound odontomas consist of varying numbers of small toothlike structure and have a predilection for the maxillary anterior regions. Complex odontomas consist of an unorganized mass of odontogenic tissues and comprise approximately 25 percent of all odontomas and have a predilection for the mandibular posterior regions. The etiology of odontomas is uncertain but hypothesized to involve local trauma, infection and genetic factors. Treatment of odontoma is conservative surgical removal and are little probability of recurrence. These two cases were about the patients with delayed eruption of mandibular first molar and mandibular lateral incisor. We surgically removed odontoma, exposed impacted tooth and guided impacted tooth into normal position by orthodontic traction. At the completion of traction, the mandibular first molar and mandibular lateral incisor was positioned fairly within the arch and complications such as root resorption were not observed.

      • STZ 유발 당뇨쥐에서 시간에 따른 생리적 항산화계의 변화

        The purpose of this study was to investigate change of physiological antioxidative system according to the time in streptozotocin-induced diabetic rats. Sprague-Dawley male rats weighing 150±10gm were randomly assigned to one normal and five STZ-induced diabetic groups. Diabetic groups were classified to experimental period. Diabetes was experimentally induced by intravenous injection of 55 mg/kg of body weight of STZ in citrate buffer(PH 4.3) after 6 weeks feeding of three experimental diets. Animals were sacrificed at 0, 3, 6, 12, 18th days of diabetic states. 1. Activity of superoxide dismutase(SOD) in liver was increased in that of diabetic mellitus(DM) groups at 3th day but that of DM groups was not significant from 6th day according to period. Glutathion peroxidase(GPX) was significantly decreased in DM groups from 6th day 2. Reduced glutathione(GSH) contents in liver significantly decreased in diabetic groups from 3rd day compared with those of DM groups according to the period. Oxidized glutathione(GSSG) was higher from 6th day. GSH/GSSG ratio was significantly lower than that of normal group from 3rd day to all experimental period 3. Contents of vitamin E in liver of DM groups were significantly decreased compared with that of normal group from 6th day. 4. Lipid peroxide(LPO) contents in liver of DM groups were significantly increased compared with that of normal group from 3rd day. The present results indicate that STZ-induced diabetic rats were reduced by antioxidative defense system and taken by peroxidate damage in tissue compared with normal group from 3rd day or 6rd day after injection STZ. It lended to acceleration all diabetic groups but no significance according to the experimental time.

      • Hepatoprotective effects of fermented field water-dropwort (Oenanthe javanica) extract and its major constituents

        <P>Dropwort (Oenanthe javanica) has been used for many years for the treatment of inflammatory conditions, including hepatitis. We investigated the protective effects of fermented field water-dropwort extract (FDE) on tert-butyl hydroperoxide (t-BHP)-induced hepatotoxicity in HepG2 cells and carbon tetrachloride (CCl4)-induced liver damage in rats. Pretreatment with FDE prior to the t-BHP treatment of HepG2 cells inhibited cell death and lactate dehydrogenase (LDH) leakage in a dose-dependent manner. In addition FDE significantly prevented the increase of hepatic enzyme markers (ALT, AST) in vivo. Moreover, FDE administration for 7 days significantly affected CYP2E1, CYP4A2, and PPARγ gene expressions. CYP2E1 and CYP4A2 gene expression in the liver, increased 2 and 22-fold by CCl4 administration, respectively, was attenuated to normal levels by pretreatment with FDE. PPARγ gene expression, completely blocked by CCl4 treatment, was increased by FDE pretreatment compared to normal control group. Histopathological examination of the livers also revealed that FDE reduced the incidence of liver lesions. Caffeic acid and chlorogenic acid were identified as major constituents of FDE. These results demonstrate the protective effects of FDE against hepatocytotoxicity induced by CCl4 and t-BHP in rats and HepG2 cells, thus indicating the potential of FDE as a therapeutic for acute liver diseases.</P>

        Design, Synthesis, Antitumor Activity and Mode of Action of Novel Oxiranyl and Thiiranyl Phenol Derivatives

        Eleven novel oxiranyl and thiiranyl phenolic compounds were synthesized as potential antitumor agents using epichlorohydrin and epithiohydrin in the presence of $K_2CO_3$. Cytotoxicities were found in range of I$C_{50}$ values of 2.5-14.8 $\mu$M, which was partially attributed to topoisomerase II inhibition. Bis-thiiranyl anthraquinone analog, 19 showed more cytotoxicity against MDA-MB-231 (breast cancer cell) and PC3 (prostate cancer cell) after 24 and/or 48 h and more potent topoisomerase II inhibitory activity than etoposide.

        Effects of Transcutaneous Electrical Nerve Stimulation(TENS), Self- Stretching and Functional Massage on the Muscle Fatigue by Maximum Muscular Strength

        The present study purposed to examine the effects of transcutaneous electrical nerve stimulation, self-stretching and functional massage on the recovery of muscle contraction force for muscle fatigue caused by sustained isotonic contraction. The subjects of this study were 45 healthy students. They were divided into transcutaneous electrical nerve stimulation group(n=15), self-stretching group(n=15) and functional massage group(n=15), and using Primus RS. We observed the pattern of changes in maximal voluntary isometric contraction force(MVIC) after causing muscle fatigue in quadriceps femoris muscle through sustained isotonic contraction. Maximal voluntary isometric contraction force(MVIC) were greatly increased after transcutaneous electrical nerve stimulation, self-stretching and functional massage. In the comparison of recovery rate of muscle contraction force for muscle fatigue caused by sustained isotonic contraction among the treatment groups, it did not show any significant differences. However, it showed that each treatment may be effective in recovery of muscle fatigue caused by sustained isotonic contraction.
