The object of this study was to optimize PCR condition for RAPD analysis in adzuki bean. The best template DNA concentration was 20ng(0.25unit taq polymerase and 2.5mM MgCl2), 40ng(1unit taq polymerase and 2.5mM MgCl2, 1unit taq polymerase and 4.5mM MgCl2 and lunit taq polymerase and 7.0mM MgCl2), and 60ng(0.5unit taq polymerase and 2.5mM MgCl2, 0.5unit taq polymerase and 4.5mM MgCl2, 1unit taq polymerase and 2.5mM MgCl2, and 1unit taq polymerase and 4.5mM MgCl2) The best MgCl2 concentration was 2.5mM(40ng template DNA and 1unit taq polymerase, 60ng template DNA and 0.5unit taq polymerase, and 60ng template DNA and 1unit taq polymerase), 4.5mM(20ng template DNA and 0.25unit taq polymerase, 60ng template DNA and 0.5unit taq polymerase, and 60ng template DNA and 1unit taq polymerase), and 7.0mM(40ng template DNA and 1unit taq polymerase). Amount of taq polymerase was 0.25unit(20ng template DNA and 2.5mM MgCl2), 0.5unit(60ng template DNA and 2.5mM MgCl2, 60ng template DNA and 4.5mM MgCl2) and 1unit(40ng template DNA and 2.5mM MgCl2, 40ng template DNA and 4.5mM MgCl2, 40ng template DNA and 7.0mM MgCl2, 60ng template DNA and 2.5mM MgCl2, 60ng template DNA and 4.5mM MgCl2, and 60ng template DNA and 7.0mM MgCl2). When we consider results from template DNA concentration, MgCl2 concentration, and amount of taq polymerase, the best condition for PCR optimization was 60ng template DNA, 4.5mM MgCl2, and 1unit taq polymerase. Reaction temperatures for the optimal PCR condition were 84℃, 32℃, 62℃; 90℃, 40℃, 72℃; and 92℃, 36℃, 72℃.

The purpose of this study was to evaluate the effect of immediate or delayed composite resin filling on dentinal microtensile bond strength (μTBS) after applied the adhesive. The coronal dentin of human third molars was exposed. Single-Bond or One-Step was applied on the dentin surfaces, and composite resin were constructed immediately (group 1) or 5 min., 10 min., 15 min., 20 min. and 30 min. (groups 2-6) after an adhesive was applied. The specimens were sectioned and made bar-shaped. Each surface area of them was about l ㎟. The RBS test was performed by EZ test. The results were analysed by One-way ANOVA and Tukey's test at 95 % significance level. The results suggested that the μTBS of Single-Bond to dentin was decreased when the composite resin was constructed 20 min. and 30 min. after Single-Bond was applied. But the μTBS of One-Step was not affected by delayed composite resin filling.

<P>Unreinforced masonry (URM) structures represent a significant portion of existing historical structures around the world. Recent earthquakes have shown the need for seismic retrofitting for URM structures. Various types of strengthening methods have been used for URM structures. In particular, a strengthening technique using externally bonded (EB) fiber reinforced polymer (FRP) composites has attracted engineers since EB FRP materials effectively enhance the shear strength of URM walls with negligible change to cross-sectional area and weight of the walls. Research has been extensively conducted to determine characteristics of URM walls strengthened with EB FRP materials. However, it is still difficult to determine an appropriate retrofitting level due to the complexity of mechanical behavior of strengthened URM walls. In this study, in-plane behavior under lateral loading was, therefore, investigated on a full-scale nonstrengthened URM wall and URM walls retrofitted with two different FRP materials: carbon (CFRP) and hybrid (HFRP) sheets. The test results indicated that both FRP composites were effective in increasing shear strength in comparison with the control specimen. However, better performance was obtained with HFRP compared to CFRP. In addition, an equation for estimating effective strain was proposed, and the theoretical results were in good agreement with the experimental ones.</P>
Retrofit of Artificially Perforated Shear Wall in Existing Dominant Wall Structure

<P>A series of three shear wall specimens were tested under constant axial stress and reversed cyclical lateral loading in order to evaluate the seismic retrofit that had been proposed for the shear wall with the opening induced by remodeling. One of these specimens was tested in the as-built condition and the other two were retrofitted prior to testing. The retrofit involved the use of carbon fiber sheets and steel plates (a thickness of 3mm) over the entire face of the wall. The test results showed that the failed specimens had shear fractures and that two different types of retrofitting strategies had different effects on the strengths of each specimen.</P>

Minimal hepatic dysfunction can be common in acute Q fever, but severe acute cholestatic hepatitis is rarely reported. We report on a 55-year-old male with acute Q fever and severe acute cholestatic hepatitis. He complained of fever, jaundice, ascites, and restlessness on admission. A liver biopsy revealed the presence of compact fibrin-ring granulomas. Serologic titers for C. burnetii IgM and IgG were 2048:1 and 1024:1, respectively. C. burnetii DNA was detected by a nested polymerase chain reaction on the liver tissue. (Gut and Liver 2009;3:141-144)

Many engineers find the way of improving the old building’s structural behavior in the remodeling project which is performed using artificial openings for merging two houses. This test was performed to verify the characteristics of coupling beams according to shape of the openings. One of the test specimens had rectangular shape openings, another one had circular shape openings, and the other one had coupling members only as slabs. Three specimens with openings had 23% ratio of opening area to total wall area. A solid type specimen which had no opening area showed shear failure. In the case of CW-RBS which had rectangular shape openings, cracks were developed in coupling beam significantly. CW-CS which had circular opening failed in shear showing development of diagonal cracks at the wall toes and wall mid-height. It was believed that degradation of wall strength was under the control regardless of the openings and coupling beam-wall connection area.

Flat plate slab has been widely used in high rise buildings for its remarkable advantages. However, punching shear failure has become increasingly important issue with the spread of flat plate slab. The results of an experiment program on punching shear performance of RC slab-column connection are presented in this paper. Specimens were full-scale representations of a one-story slab in a Korean apartment. The test results showed that failure mechanism of specimens governed by shear fracture and the strength of specimens were different according to the retrofitting strategies.
